Abstract:We have developed an enzymatic glucose biosensor that is based on a flat platinum electrode which was covered with electrophoretically deposited rhodium (Rh) nanoparticles and then sintered to form a large surface area. The biosensor was obtained by depositing glucose oxidase (GOx), Nafion, and gold nanoparticles (AuNPs) on the Rh electrode. The electrical potential and the fractions of Nafion and GOx were optimized. The resulting biosensor has a very high sensitivity (68.1 μA mM−1 cm−2) and good linearity in the range from 0.05 to 15 mM (r = 0.989). The limit of detection is as low as 0.03 mM (at an SNR of 3). The glucose biosensor also is quite selective and is not interfered by electroactive substances including ascorbic acid, uric acid and acetaminophen. The lifespan is up to 90 days. It was applied to the determination of glucose in blood serum, and the results compare very well with those obtained with a clinical analyzer.
One-pot Electrodeposition of 3-Aminopropyltriethoxysilane–chitosan Hybrid Gel Film to Immobilize Glucose Oxidase for Biosensing

Abstract:We report on the electrodeposition of a 3-aminopropyltriethoxysilane-chitosan (APTES–CS) hybrid gel film for in situ immobilization of glucose oxidase (GOx) on an Au or platinized Au (Ptnano/Au) electrode for biosensing of glucose. Controllable electroreduction of p-benzoquinone is used to lift the electrode-surface pH for the GOx–APTES–CS codeposition, which was monitored by an electrochemical quartz crystal microbalance. The fabrication procedures of the biosensor and the parameters influencing the biosensing performance were optimized. The prepared porous GOx–APTES–CS/Ptnano/Au and GOx–APTES–CS/Au electrodes can be used to detect the enzymatically generated H2O2 at 0.5 and 0.7V vs SCE, respectively. The enzyme electrodes exhibited linear responses to glucose concentration from 0.2μM to 8.2mM (R=0.998, at Ptnano/Au substrate) and from 0.2μM to 5.5mM (R=0.998, at Au substrate), with current sensitivities of 69.5 (Ptnano/Au) and 65 (Au) μAmM−1cm−2, respectively, and a detection limit of 0.2μM (S/N=3) was achieved for each electrode. The response time was less than 5 (Ptnano/Au) or 8 (Au) s. It is striking that the enzyme electrodes remained their initial response sensitivity after storage for 5 (Au) and >6 (Ptnano/Au) months in 0.10M PBS (pH 7.0) at 4°C.
Study of Electrochemical Biosensor for Determination of Glucose Based on Prussian Blue/Gold Nanoparticles-Chitosan Nanocomposite Film Sensing Interface

Abstract:A highly electroactive bilayer composite film sensing interface of Prussian Blue (PB)/gold nanoparticles-chitosan (AuNPs-CS) was modified on Au electrode through electrochemical deposition and coating method followed by integrating glucose oxidase (GOx) into the interfacial matrix to fabricate a high-performance glucose biosensor. The excellent electrocatalytic ability of the PB/AuNPs-CS composite film sensing interface for H2O2 was evaluated, which has a broad linear response of 0.01-7.95 mM, with a low detection limit (LOD) 0.269 μM and a high sensitivity of 511.82 μA·mM-1·cm-2. The enhanced electrocatalytic activity of this sensing interface for H2O2 is attributed to the protection from the network CS film to PB and the synergistic effect of PB and AuNPs. Consequently, an electrochemical biosensing interface was constructed with GOx immobilized as a model enzyme. The PB/GOx-AuNPs-CS biosensing nanocomposite film was capable of a fast steady-state response time (within 2s) and high sensitivity to glucose with a wide linear range of: 0.025~2.00 mM (R2=0.99), with a sensitivity of 40.41 μA·mM-1·cm-2 and a LOD of 1.62 μM; and 2.00~6.50 mM (R2=0.98), with a sensitivity of 8.90 μA·mM-1·cm-2 and a LOD of 7.16 μM. The biosensor has good anti-interference and selectivity, which provides a promising wide linear range platform for clinical blood glucose detection in the future.

Abstract:We examine here the electropolymerization of electrochemically or chemically preoxidized catecholamines in glucose oxidase (GOx)-containing neutral solutions to efficiently immobilize the enzyme at Prussian blue-modified Au electrodes for sensitive amperometric biosensing of glucose. The electrochemical quartz crystal microbalance (EQCM) was used to track various electrode-modification processes. The optimized poly(dopamine)-based glucose biosensor displayed a sensitivity of 35μAmM−1cm−2 and a limit of detection of 0.3μM at 0.7V vs. SCE, and similar results were obtained at −0.05V vs. SCE, which are obviously better than those from preoxidation-free conventional electropolymerization. The immobilized GOx retained high enzymatic specific activity, as quantified by UV–vis spectrophotometry and EQCM. l-Noradrenalin could similarly electropolymerize and the resultant enzyme film gave equivalent biosensing characteristics, but the electropolymerization of EP was less efficient and the resultant enzyme film showed notably poorer performance.
Efficient Immobilization of Glucose Oxidase by in Situ Photo-Cross-linking for Glucose Biosensing.

Abstract:A glucose biosensor was fabricated based on electrostatic self-assembly in combination with in situ photo-cross-linking of glucose oxidase (GOx) and diazoresin-chitosan (DAR-CS) on Prussian blue deposited multi-walled carbon nanotubes (PB-MWNTs) backbone. It was demonstrated that GOx was initially ionically deposited and subsequently covalently photo-cross-linked onto the PB-MWNTs backbone using photosensitive DAR-CS as the assembly interlayer. The modified electrode exhibited good electrical conductivity and effective electron transfer mediation toward H(2)O(2) reduction due to the employment of PB-MWNTs as the fabrication backbone. The biosensor showed high sensitivity of 77.9 μA mM(-1) cm(-2) to glucose in the linear concentration range from 1.0×10(-5) to 1.1×10(-3) M with fast response time of 10s, detection limit of 3.1×10(-6) M, and good anti-interference ability. More importantly, the biosensor exhibited greatly improved biosensing stability in comparison with the non-photo-cross-linked biosensor attributed to the conversion of weak ionic bonds to strong covalent ones for enzyme immobilization by the proposed strategy. The results for glucose determination in real serum samples with the biosensor were found to be in good agreement with those obtained by the conventional clinical procedure.

Abstract:An improved amperometric glucose biosensor was constructed by in situ incorporating glucose oxidase (GOD) within the electrodeposited chitosan–gold nanoparticle (chitosan–AuNP) hybrid film on a Prussian Blue (PB) modified electrode. The method is simple and controllable. It combined the merits of in situ immobilizing biomolecules in the chitosan–AuNP hybrid film by electrochemical method and the synergic catalysis effects of PB and GOD molecule. Scanning electron microscopy showed that the chitosan–AuNP hybrid film was homogeneous with small pores, which improved the stability and sensitivity of the biosensor. The biosensor prepared under optimal conditions showed fast response time (3s), high sensitivity (69.26μAmM−1cm2), long-term operational stability, good suppression of interference and low detection limit (6.9×10−7M). A linear calibration plot was obtained in the wide concentration range from 1.0×10−6 to 1.6×10−3M. The biosensor was successfully applied to determine the glucose concentration in human serum samples.

Abstract:A novel complex material was fabricated by three steps. In the first step, gold nanoparticle (Au-nano) was prepared with the method of chemistry and dialysis. In the second step, 4-aminothiophenol (AT) was encapsulated in the cavity of P-cyclodextrin and formed inclusion complex, cyclodextrin/4-aminothiophenol (CD/AT). And then inclusion complex was adsorbed to the surface of Au-nano based on the bond of Au-S interaction. In the last step, a complex material, cyclodextrin/poly(4-aminothiophenol)-Au nanoparticles (CD/PAT-Au-nano) was obtained by the polymerizing in the acid solution initiated by chlorauric acid. The CD/PAT-Au-nano has spherical nanostructure with the average diameter of 55 nm. Glucose oxidase (GOx) was anchored with this complex material and direct electrochemistry of GOx was achieved. A couple of stable and well-defined redox peaks were observed with the formal potential (E-0') of -0.488 V (vs. SCE) in a pH 6.98 buffer solution. The GOx modified electrode also exhibited an excellent electrocatalytic activity to the reduction of glucose, a linearity range for determination of glucose is from 0.25 mM to 16.0 mM with a detection limit of 0.09 mM (S/N = 3). This protocol had potential application to fabricate the third-generation biosensor.

Abstract:Mass detection of glucose, which is required in many applications, remains challenging. The commercial enzyme-based glucose test strips cannot be reused, and current non-enzymatic glucose sensors exhibit a narrow range of detection and slow glucose oxidation kinetics. Herein, controlled etching of Prussian blue analogue (PBA) nanocubes at the vertices is conducted and Au nanoparticles (Au NPs) are subsequently inlaid in the etched cavities by in-situ reduction of HAuCl4. The unique AuNP-PBA nanocomplexes exhibit low electrochemical potential for glucose oxidation, high electrocatalytic activity, and rapid redox electron transfer rate. Covalent immobilization of the Au-inlaid nanomaterials on a fine Au wire leads to a non-enzymatic glucose sensor with a particularly wide linear detection range (10 µM to 16 mM), excellent anti-interference, and fast response. More importantly, the sensor is reusable, and its sensitivity is well maintained even after 150 times of detection. This new-concept material promises to enable high-throughput glucose detection at a low cost, which is essential in diabetic management and other healthcare applications.

Abstract:Nanozyme/natural enzyme hybrid plays a vital role in biosensing, therapy, and catalysis owing to the integrated advantages in the selectivity of natural enzymes and controllable catalytic activity of nanozymes. Herein, Fe-MIL-88B-NH2 [(Fe-metal-organic framework (MOF)] with remarkable peroxidase-like activity, ultrahigh stability, and high biocompatibility was utilized for immobilization of glucose oxidase (GOx) via an amidation coupling reaction. On the basis of the excellent selectivity and catalytic activity of Fe-MOF-GOx, a cascade catalysis was performed for the colorimetric detection of glucose. The integrated Fe-MOF-GOx not only exhibited higher stability and reusability than their mixtures including Fe-MOF and free GOx system but also possessed a wide linear range (1-500 μM), with a low detection limit of 0.487 μM for glucose detection.

Abstract:A novel electrochemical glucose biosensor was developed based on in situ covalent immobilization of glucose oxidase (GOx) by one-pot chitosan (CS)-incorporated sol–gel process in the presence of Prussian blue deposited multi-walled carbon nanotubes hybrids (PB/MWNTs) using 3-isocyanatopropyltriethoxysilane (ICPTES) as both a sol–gel precursor and a covalent coupling agent for GOx and CS. The electrode modified with the PB/MWNTs-GOx-CS-ICPTES sol–gel composite film showed good electrical conductivity and effective low-potential electron transfer mediation toward H2O2 reduction attributed to the incorporation of PB/MWNTs. The biosensor exhibited a linear response to glucose in the concentration range from 2.5×10−5 to 1.3×10−3M with a correlation coefficient of 0.9998, a detection limit of 7.5×10−6M, a low response time (10s), good sensitivity and high anti-interference ability. Compared with the control biosensor based on the traditional tetraethoxysilane derived sol–gel composite film, the biosensor showed a similarly small apparent Michaelis–Menten constant of 3.67mM but much higher electrochemical and biosensing stability.

Abstract:Rapid and accurate detection of glucose is of great significance for diabetic management. Highly sensitive glucose sensors promise to achieve noninvasive detection technology, enabling more convenient and efficient means for large-scale screening and long-term dynamic monitoring of diabetes patients. In this work, we demonstrate a sensitive glucose electrochemical biosensor through the synergetic labelling strategy utilizing PbS colloidal quantum dots (CQDs) and Au nanospheres (AuNSs). The PbS CQDs/AuNSs/glucose oxidase (GOx) mixture could be stably immobilized on the carbon electrode surface via the one-step dip-coating method. The electrochemical biosensor employing PbS CQDs/AuNSs/GOx-modified electrode integrates the functions of specific molecule recognition, signal transduction as well as signal amplification. The sensor is capable of transducing the glucose enzyme-catalyzed reaction into significant current signals, exhibiting a good linear response in the glucose concentration range of 0.1 μM—10 mM with the limit of detection being 1.432 nM.

Abstract:A simple and controllable electrodeposition method is described to fabricate a homogeneous chitosan-ferrocene/Au nanoparticles/glucose oxidase (CS-Fc/Au NPs/GOx) nanocomposite film. The morphologies and electrochemistry of the nanocomposite film were investigated by using scanning electron microscopy (SEM) and electrochemical techniques including electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV), respectively. The ferrocene group (Fc) covalently bounded chitosan hybrid (CS-Fc) can effectively avoid the leakage of Fc and retain its electrochemical activity. Further immobilization of Au NPs into the CS-Fc matrix enhances both the charge-transport properties of the composite and bioaffinity to enzyme. Biosensor based on this CS-Fc/Au NPs/GOx film has advantages of fast response, excellent reproducibility and high stability. This biosensor shows a linear response to glucose in the concentration range from 0.02 to 8.66mM with a detection limit of 5.6μM at a signal-to-noise ratio of 3. The present method offers a facile way to fabricate biosensors and bioelectronic devices.

Abstract:The MOFs (ZIF-8) coated Fe3O4/PPy magnetic nanocomposite (Fe3O4/PPy@ZIF-8) with large surface area and high conductivity was successfully prepared by polymerizing the pyrrole on Fe3O4 nanoparticle and self-assembling the ZIF-8 on the composite. Due to the outstanding specific area and pore volume of MOFs on the resulted nanocomposite, the glucose oxidase (GOx) could be effectively immobilized on the Fe3O4/PPy@ZIF-8 to fabricate a novel amperometric glucose biosensor. The as-prepared GOx/Fe3O4/PPy@ZIF-8/GCE biosensor exhibited extraordinary electro-detection performance for glucose with wide linear range from 1 μM to 2 mM and the limit of detection was 0.333 μM (S/N = 3). Furthermore, the prepared biosensor also showed good selectivity for glucose detection and satisfactory result in real samples detection.

Abstract:A method for immobilizing proteins in a carbon mesoporous material (CMM) containing platinum nanoparticles (Pt-NPs) is demonstrated. Compared to pure CMM or carbon nanotubes, CMM containing Pt-NPs enhances the electron transfer and redox properties of redox enzymes, such as glucose oxidase (GOx), due to a cooperative effect of Pt-NPs and CMM. The quasi-reversible electron transfer of GOx in this system is probed, and the apparent heterogeneous electron transfer rate constants are found to be 66% larger than in pure CMM. The GOx/Pt-CMM based glucose biosensor enables the determination of glucose at a potential of 600 mV (vs. SCE). Its detection limit is 10 times lower, and the sensitivity is 16 times higher than that of the respective biosensor without Pt-NPs.

Abstract:<p>Sulfide minerals (galena, pyrite, chalcopyrite and sphalerite) were employed as a matrix for glucose oxidase (GOx) immobilization in glucose sensing. Multilayered GOx biosensors were fabricated using glutaraldehyde (GA) modified glassy carbon electrode (GCE). The sensors made using this method were capable of detecting glucose with a linear range of 0.5-90 mM, 0.8-80 mM, 1.0-25 mM, and 1.5-70 mM for galena, pyrite, chalcopyrite and sphalerite, respectively. And the detection limits are 0.06 mM, 0.1 mM, 0.2 mM and 0.35 mM for galena, pyrite, chalcopyrite and sphalerite, respectively. Unlike traditional biosensors, the novel biosensors could detect the glucose under air-saturated buffer solutions. In addition, they showed acceptable reproducibility, good repeatability and applicability. These results indicated that the proposed sulfide minerals are good platforms for enzyme immobilization, thus creating a new and high value application in biosensor for the sulfide minerals.</p>

Abstract:Glucose oxidase (GOx) is one of the typical enzymes widely used in glucose sensors. In recent years, novel nanomaterials have been developed to immobilize the enzyme, which is of great importance for the development of biosensors. Here we constructed 3D porous Ti3C2Tx MXene/Graphene/AuNPs (MGA) composites by a simple mixing-freeze-drying and self-reduction process. The abundant hydrophilic groups on the surface of Ti3C2Tx MXene nanosheets endow the MGA composites with highly hydrophilic microenvironment. The 3D porous composites provide a more open structure and promote the entry of GOx into the internal pores, which may enhance the stable immobilization and retention of GOx in the film. The unique electrocatalytic properties and synergistic effects between AuNPs and Ti3C2Tx MXene/Graphene are used to further enhance the electron transport in the 3D porous composites and reduce the redox potential. The MGA/GOx glucose sensor shows linear current in the range of glucose concentration from 2 μM to 0.4 mM and 0.4 mM to 6 mM, with sensitivities of 169.49 μA/(mM·cm2) and 34.13 μA/(mM·cm2) respectively, showing high sensitivity at low concentrations. The detection limit of glucose sensor reaches 2 μM and the response time is less than 3 s. This sensor can be applied to the detection of glucose in human sweat.

Electrochemically fabricated nanoelectrode ensembles for glucose biosensors

Abstract:Novel glucose oxidase (GOX) sensor arrays with nanoelectrode population densities of up to 1×1010/cm2 nanoelectrodes are reported. These nanoelectrode sensors, polypyrrole (PPy) nanofibers containing entrapped GOX, were fabricated via a two-step process, which consisted of an electroadsorption of GOX followed by an electrochemical codeposition of PPy with GOX within the nanopores of an anodic aluminum oxide (AAO) membrane. It was found by SEM observation that GOX could intercalate in PPy nanofibers, especially in the region close to their side surface. The GOX/PPy nanofiber array electrode demonstrates a good biocatalytic activity to glucose with an apparent Michaelis–Menten constant (KMapp) of 5.7 mM and a response time of about 15 s. Porous Nitrogen-Doped Reduced Graphene Oxide-Supported CuO@Cu2O Hybrid Electrodes for Highly Sensitive Enzyme-Free Glucose Biosensor

Abstract:Constructing high-performance enzyme-free biosensors for detecting glucose is essential to preliminary diabetes diagnosis. Here, copper oxide nanoparticles (CuO@Cu2O NPs) were anchored in porous nitrogen-doped reduced graphene oxide (PNrGO) to construct CuO@Cu2O/PNrGO/GCE hybrid electrode for sensitive detection of glucose. Benefiting from the remarkable synergistic effects between the multiple high activation sites of CuO@Cu2O NPs and the dramatic properties of PNrGO with excellent conductivity and large surface area with many accessible pores, the hybrid electrode possesses outstanding glucose sensing performance that is far superior to those of pristine CuO@Cu2O electrode. The as-fabricated enzyme-free glucose biosensor displays prominent glucose sensitivity of 2,906.07 μA mM−1 cm−2, extremely low limit of detection of 0.13 μM, and wide linear detection of 3 μM–6.772 mM. In addition, excellent reproducibility, favorable long-term stability, and distinguished selectivity are obtained in the glucose detection. Importantly, this study provides promising results for continuous improvement of non-enzyme sensing applications.
Cu₂O-Au nanocomposites for enzyme-free glucose sensing with enhanced performances

Abstract:A facile method for the synthesis of Cu(2)O-Au nanocomposites has been reported by injecting Cu(2)O nanocubes into Au precursor directly with the assistance of ultrasound radiation at room temperature. The ultrasound radiation is not a necessary requirement but can make the distribution of Au nanoparticles more homogenous. The formation of Cu(2)O-Au nanocomposites is attributed to following two reasons. The first one is the difference in the reduction potential between Cu(2+)/Cu(2)O and AuCl(4)(-)/Au, which can also be considered as the driving force for the redox reaction. The other one is the low lattice mismatch between (200) planes of Cu(2)O and (200) facets of Au, which is favorable for the formation of heterostructure. The electrochemical investigation demonstrates that the performances of Cu(2)O nanocubes in enzyme-free glucose sensing have been improved significantly after the decoration of Au nanoparticles which may be derived from the polarization effect provided by Au nanoparticles. As-prepared Cu(2)O-Au nanocomposites have great potential in enzyme-free glucose sensing.
