Abstract:Gestational diabetes is the condition with an elevated blood glucose level during the pregnancy period. Approximately 10% of women have been affected by gestational diabetes in USA every year. Babies of pregnant mothers are with diabetes have increased risk of developing type-2 diabetes and obesity in their later life. In addition, uncontrolled diabetes causes the death to infant either before or immediate after the birth. It is mandatory to monitor the condition of diabetes during the pregnancy period for a healthy delivery. Researches are developing various simple biosensing methods to monitor the glucose level using glucose oxidase. Recently nanomaterials are focused by researchers to develop high-performance glucose sensors due to its high surface area, biocompatibility, higher sensitivity and selectivity. Nanomaterials are proved for improving the detection with glucose oxidase by various sensors to identify gestational diabetes. This review is discussed on sensing methods development for gestational diabetes and the incorporation of different nanomaterial in monitoring the glucose level during the pregnancy period.

Abstract:Diabetes is a dominating health issue with 425 million people suffering from the disease worldwide and 4 million deaths each year. To avoid further complications, the diabetic patient blood glucose level should be strictly monitored despite there being no cure for diabetes. Colorimetric biosensing has attracted significant attention because of its low cost, simplicity, and practicality. Recently, some nanomaterials have been found that possess unexpected peroxidase-like activity, and great advances have been made in fabricating colorimetric glucose biosensors based on the peroxidase-like activity of these nanomaterials using glucose oxidase. Compared with natural horseradish peroxidase, the nanomaterials exhibit flexibility in structure design and composition, and have easy separation and storage, high stability, simple preparation, and tunable catalytic activity. To highlight the significant progress in the field of nanomaterial-based peroxidase-like activity, this work discusses the various smart nanomaterials that mimic horseradish peroxidase and its mechanism and development history, and the applications in colorimetric glucose biosensors. Different approaches for tunable peroxidase-like activity of nanomaterials are summarized, such as size, morphology, and shape; surface modification and coating; and metal doping and alloy. Finally, the conclusion and challenges facing peroxidase-like activity of nanomaterials and future directions are discussed.

Abstract:Photoelectrochemical detection represents a unique signal transducing modality, where the photogenerated charge carriers are modulated by the redox reactions of molecular targets over the electrode surface. In this work, we report a novel glucose sensor based on hydrothermally-grown, single-crystalline TiO2 nanowires with surface-functionalized glucose oxidase (GOx), which can oxidize glucose to gluconic acid. The key feature of this glucose sensor design is that the photogenerated holes over the TiO2 anode are utilized to form O-2, which directly serves as an efficient electron shuttling mediator between the enzymatic redox center of GOx and the sensor surface, thus leading to an increase of the photocurrent. As a proof-of-concept, the GOx-functionalized TiO2 (TiO2-GOx) nanowires exhibit a high sensitivity of similar to 0.9 nM in the detection of glucose in buffer. The capability of detecting glucose in mouse serum is also demonstrated. Our work suggests the potential of developing these nanowire-based photoelectrochemical biosensors for convenient, efficient and low-cost biomarker detection and disease diagnosis.

Abstract:Glucose oxidase (GOx) is an endogenous oxido-reductase that is widely distributed in living organisms. Over recent years, GOx has attracted increasing interest in the biomedical field due to its inherent biocompatibility, non-toxicity, and unique catalysis against beta-D-glucose. GOx efficiently catalyzes the oxidization of glucose into gluconic acid and hydrogen peroxide (H2O2), which can be employed by various biosensors for the detection of cancer biomarkers. Various cancer therapeutic strategies have also been developed based on the catalytic chemistry of GOx: (1) the consumption of glucose provides an alternative strategy for cancer-starvation therapy; (2) the consumption of oxygen increases tumor hypoxia, which can be harnessed for hypoxia-activated therapy; (3) the generation of gluconic acid enhances the acidity of the tumor microenvironment, which can trigger pH-responsive drug release; (4) the generation of H2O2 increases the levels of tumor oxidative stress, and the H2O2 can be converted into toxic hydroxyl radicals that can kill cancer cells upon exposure to light irradiation or via the Fenton reaction. More importantly, GOx can be combined with other enzymes, hypoxia-activated prodrugs, photosensitizers or Fenton's reagents, to generate multi-modal synergistic cancer therapies based on cancer starvation therapy, hypoxia-activated therapy, oxidation therapy, photodynamic therapy, and/or photothermal therapy. Such multi-modal approaches are anticipated to exert a stronger therapeutic effect than one therapeutic mode alone. Thus, maximizing the potential of GOx in a biomedical context will offer novel clinical solutions to diagnose and treat cancer. In this tutorial review, we introduce the recent advances of GOx in cancer diagnosis and treatment. We then emphasize the design principles and biomedical applications of GOx-based biosensors and cancer therapeutic approaches. Finally, we discuss the challenges and future prospects of GOx-based catalytic systems in biomedicine.

Abstract:As an endogenous oxido-reductase, glucose oxidase (GOx) has been widely used in biomedical fields due to its inherent biocompatibility, non-toxicity, and unique catalysis against β -D-glucose. GOx can effectively catalyze the oxidation of glucose into gluconic acid and hydrogen peroxide (H2O2) in the presence of O2. This process depletes glucose nutrients and oxygen levels, resulting in elevated acidity, hypoxia and oxidative stress in the tumor microenvironment. All of these changes can be readily harnessed to develop a multimodal synergistic cancer therapy by combining GOx with other therapeutic approaches. Herein, we introduce the research progress of GOx-based multimodal synergistic cancer therapy, and highlight the current challenges and future prospects of GOx-based catalytic systems in nanomedicine.

Abstract:Diabetes mellitus, known as diabetes, is a type of common metabolic disease in which glucose is build up in the blood, caused either by pancreas's failure to produce insulin or by insulin resistance from the body tissue. This chapter focuses on glucose oxidase (GOx) anchored nanomaterials that are used for glucose detection and advanced insulin delivery and discusses nanomaterials that can be potentially an alternative to GOx nanosystems. Insulin is a peptide hormone produced by beta cells in pancreas for regulation of glucose metabolism. Nonenzyme nanosensors constructed from metal oxides are cost-effective and highly sensitive in glucose detection affording minimum fabrication costs and longer storage lifetimes. The closed-loop system, functioned by automatically feedback to blood glucose fluctuation, offers obvious advantages over the open loop system. Glucose-binding proteins, mainly lectins, are a group of natural carbohydrate-binding proteins known to interact with glycosylated receptors or lipids for cell signal transduction.

Abstract:Nanozymes with high catalytic efficiency have been widely investigated and studied in various fields. Compared with natural enzymes, nanozymes with enzyme-mimicking properties possess the advantages of low-cost, scalability, high stability, and tunable catalytic performance. In this paper, platinum nanoparticles loaded on oxygenated nanodiamonds (Pt-ONDs) were synthesised through a one-step method as a highly efficient nanozyme to mimic the function of peroxidase. Based on this, a novel sensing strategy was developed by combining Pt-ONDs and gold bipyramids (AuBPs) for sensitive detection of glucose. The detection limit of this method was established at 0.2 μM (a few thousand times before the lowest detection limit of commercial glucometers), with a wide detection range from 0.2 to 400 μM, highlighting its potential for ultra-high sensitive glucose detection. This study may provide a novel strategy for the application of nanozymes and gold nanostructures for the sensitive detection of glucose.

Abstract:Glucose monitoring is essential to evaluate the degree of glucose metabolism disorders. The enzymatic determination has been the most widely used method in glucose detection because of its high efficiency, accuracy, and sensitivity. Noble metal nanomaterials (NMs, i.e., Au, Ag, Pt, and Pd), inheriting their excellent electronic, optical, and enzyme-like properties, are classified as noble metal nanozymes (NMNZs). As the NMNZs are often involved in two series of reactions, the oxidation of glucose and the chromogenic reaction of peroxide, here the chemical mechanism by employing NMNZs with glucose oxidase (GOx) and peroxidase (POD) mimicking activities is briefly summarized first. Subsequently, the regulation strategies of the GOx-like, POD-like and tandem enzyme-like activities of NMNZs are presented in detail, including the materials, size, morphology, composition, and the reaction condition of the representative NMs. In addition, in order to further mimic the enantioselectivity of enzyme, the design of NMNZs with enantioselective recognition of d-glucose and l-glucose by using different chiral compounds (DNA, amino acids, and cyclodextrins) and molecular imprinting is further described in this review. Finally, the feasible solutions to the existing challenges and a vision for future development possibilities are discussed.

Abstract:This review will unveil the development of a new generation of electrochemical sensors utilizing a transition-metal-oxide-based nanocomposite with varying morphology. There has been considerable discussion on the role of transition metal oxide-based nanocomposite, including iron, nickel, copper, cobalt, zinc, platinum, manganese, conducting polymers, and their composites, in electrochemical and biosensing applications. Utilizing these materials to detect glucose and hydrogen peroxide selectively and sensitively with the correct chemical functionalization is possible. These transition metals and their oxide nanoparticles offer a potential method for electrode modification in sensors. Nanotechnology has made it feasible to develop nanostructured materials for glucose and H2O2 biosensor applications. Highly sensitive and selective biosensors with a low detection limit can detect biomolecules at nanomolar to picomolar (10-9 to 10-12 molar) concentrations to assess physiological and metabolic parameters. By mixing carbon-based materials (graphene oxide) with inorganic nanoparticles, nanocomposite biosensor devices with increased sensitivity can be made using semiconducting nanoparticles, quantum dots, organic polymers, and biomolecules.

Abstract:The development of a glucose sensor that can monitor blood glucose levels is particularly important with the increasing number of diabetes patients. Here, a glucose biosensor based on Nafion (NF), glucose oxidase (GOx), multi-walled carbon nanotubes (MWCNTs), hydroxy fullerene (HFs) composite modified glassy carbon electrode (GCE) is presented. The transmission electron microscopy, field emission scanning electron microscope, UV-Visible spectroscopy, cyclic voltammetry, square wave voltammetry, and electrochemical impedance spectroscopy methods are used to study the characteristics of the modification materials and to evaluate electrochemical properties of the modified electrodes. The heterogeneous electron transfer constant (ks) is calculated to be 4.49 s-1. The response to glucose is linear in the range of 0.02–0.25 mM and 0.25–4.0 mM with a sensitivity of 80.59 μA·mM-1·cm-2 and detection limit (LOD) of 9.93 μM. The NF/MWCNTs-HFs-GOx/GCE biosensor demonstrate high sensitivity to glucose with excellent storage stability and anti-interference capability.

Abstract:Promoting the electrocatalytic oxidation of glucose is crucial in glucose biosensor design. In this study, nanoporous gold (NPG) was selected for glucose oxidase (GOx) immobilization and glucose biosensor fabrication because of its open, highly conductive, biocompatible, and interconnected porous structure, which also facilitates the electrocatalytic oxidation of glucose. The electrochemical reaction on the surface of the resulting GOx/NPG/GCE bioelectrode was attributed to the co-catalysis effect of GOx and NPG. A surface-confined reaction in a phosphate buffer solution was observed at the bioelectrode during cyclic voltammetry experiments. Linear responses were observed for large glucose concentrations ranging from 50μM to 10mM, with a high sensitivity of 12.1μAmM(-1)cm(-2) and a low detection limit of 1.02μM. Furthermore, the GOx/NPG/GCE bioelectrode presented strong anti-interference capability against cholesterol, urea, tributyrin, ascorbic acid, and uric acid, along with a long shelf-life. For the detection of glucose in human serum, the data generated by the GOx/NPG/GCE bioelectrode were in good agreement with those produced by an automatic biochemical analyzer. These unique properties make the GOx/NPG/GCE bioelectrode an excellent choice for the construction of a glucose biosensor.

Abstract:Novel glucose oxidase (GOX) sensor arrays with nanoelectrode population densities of up to 1×1010/cm2 nanoelectrodes are reported. These nanoelectrode sensors, polypyrrole (PPy) nanofibers containing entrapped GOX, were fabricated via a two-step process, which consisted of an electroadsorption of GOX followed by an electrochemical codeposition of PPy with GOX within the nanopores of an anodic aluminum oxide (AAO) membrane. It was found by SEM observation that GOX could intercalate in PPy nanofibers, especially in the region close to their side surface. The GOX/PPy nanofiber array electrode demonstrates a good biocatalytic activity to glucose with an apparent Michaelis–Menten constant (KMapp) of 5.7 mM and a response time of about 15 s. The PPy/GOX nanofiber array electrode may have potential applications in monitoring and diagnostic tests in practical use in the future.

Abstract:Accurate and rapid determination of blood glucose levels is essential for real-time monitoring and management of diabetes mellitus, and nanomaterials-based nonenzymatic glucose sensing plays an increasingly significant role in this regard. Here, a simple two-step electrodeposition technique that was efficient, environmentally friendly, easily manipulated, and exceedingly controllable was adopted for the synthesis of metal-organic framework (MOF)/carbon material composites on a titanium mesh (TM), namely α-cyclodextrin functionalized reduced graphene oxide/nickel-based MOF (α-CD-rGO/Ni-MOF/TM), in which Ni-MOF served as the electrocatalyst for glucose oxidation, and rGO greatly enhanced the electrochemical performance of Ni-MOF, benefiting from α-CD effectively preventing aggregation of rGO nanosheets while also improving the stability of the composites, so that these endow the obtained nanomaterials exhibited remarkable electrocatalytic ability for glucose. Consequently, the as-prepared glucose sensor revealed two linear dynamic ranges of 0.65 μM− 4.828 mM with a sensitivity of 1395 μA mM−1 cm−2 and 4.828 − 9.178 mM with a sensitivity of 760 μA mM−1 cm−2 together with a rapid response time of only 1.9 s and a low detection limit of 0.3 μM as well as distinguished reproducibility, selectivity, and stability, further demonstrating the synergistic effects of this composite. The newly manufactured glucose sensing platform was also successfully used to detect glucose in real serum. Considering the convenience and controllability of two-step electrodeposition, the α-CD-rGO/Ni-MOF/TM composite holds great promise for commercial potential, and also provides direction and technical reference for the synthesis of MOF/carbon material composites.

Abstract:Due to unhealthy lifestyles, the population of diabetics is rapidly increasing, therefore, the demand of glucose meter is also increasing. However, not all the people could overcome the fear of jabbing a needle during blood glucose tests. In this regard, we report a non-invasive glucose sensing electrode composed of conductive and bio-compatible Nafion-carbon nanotubes (Nf-CNT) nanocomposite and glucose oxidase (GOx), which are sequentially coated on screen-printed electrode (SPE). The Nf-CNT nanocomposite is prepared through ultrasonication followed by drop casting it onto the SPE. Subsequently, Nf-CNT/SPEs are subjected to plasma treatment followed by drop-casting of GOx to fabricate GOx/Nf-CNT/SPE electrodes. The plasma treatment alters the hydrophobic nature of Nf-CNT/SPE, which not only exposes the surface of Nf-CNT layer, but also enables it for uniform coating of GOx. The results confirm that the mixing ratio of Nafion and CNT as well as plasma treatment time significantly affects sensing performance. The GOx/Nf-CNT/SPE subjected to 1 min of plasma treatment prior to the coating of GOx shows the best performance with a sensitivity of 99.13 μA/mMcm 2 and a linear range of 20–700 μM. Moreover, the GOx/Nf-CNT/SPE electrodes are also tested under artificial saliva, which shows their high potential in the diagnosis of diabetes.

Abstract:This review overviews the recent development of nanomaterials for the application of electrochemical non-enzymatic glucose biosensors. The electrocatalytic mechanism and glucose sensing performance of a variety of nanostructured materials including metallic nanoparticles, metal oxides, metal complexes, alloys and carbon nanomaterials are discussed. The merits and shortfalls of each nanomaterial as electrocatalyst for non-enzymatic biosensing are evaluated and the prospects of non-enzymatic glucose biosensors are presented.

The first generation of glucose sensors had limitations in terms of sensitivity and selectivity. In order to overcome these problems, scientists have used a range of new materials to produce new glucose electrochemical sensors with higher sensitivity, selectivity and lower cost. A variety of different electrochemical sensors including enzymatic electrochemical sensors and enzyme-free electrochemical sensors have been extensively investigated. We discussed the development process of electrochemical glucose sensors in this review. We focused on describing the benefits of carbon materials in nanomaterials, specially graphene for sensors. In addition, we discussed the limitations of the sensors and challenges in future research.

A glucose biosensor based on direct electrochemistry of glucose oxidase immobilized on nitrogen-doped carbon nanotubes.

Abstract:A novel biosensor for glucose was prepared by immobilizing glucose oxidase (GOx) on nitrogen-doped carbon nanotubes (CNx-MWNTs) modified electrode. The CNx-MWNTs membrane showed an excellent electrocatalytic activity toward the reduction of O2 due to its diatomic side-on adsorption on CNx-MWNTs. The nitrogen doping accelerated the electron transfer from electrode surface to the immobilized GOx, leading to the direct electrochemistry of GOx. The biofunctional surface showed good biocompatibility, excellent electron-conductive network and large surface-to-volume ratio, which were characterized by scanning electron microscopy, contact angle and electrochemical impedance technique. The direct electron transfer of immobilized GOx led to stable amperometric biosensing for glucose with a linear range from 0.02 to 1.02mM and a detection limit of 0.01mM (S/N=3). These results indicated that CNx-MWNTs are good candidate material for construction of the third-generation enzyme biosensors based on the direct electrochemistry of immobilized enzymes.
