Abstract:A new glucose sensing strategy has been developed by using diffraction-gratings of a stimuli-responsive hydrogel bearing phenylboronic acid groups. The sensing gratings were fabricated through soft-lithographic duplication of photoinduced surface-relief-gratings (SRGs) on azo polymer films to the hydrogel surfaces. For the preparation, elastomeric stamps were obtained by casting PDMS prepolymer against the azo polymer masters and curing properly. The hydrogel gratings containing phenylboronic acid groups were shaped by contact printing with the PDMS stamps and a specially prepared “ink”, which was a precursor solution curable by UV light irradiation. The gratings showed a fast and highly sensitive response to glucose in buffer solutions as read out by the increase of the diffraction efficiency (DE/DE0). The saturated DE/DE0 value was linearly correlated to the glucose concentration of the solutions. The gratings showed ability for the continuous detection and were durable for repeated applications. The sensing ability of the gratings was stable when kept in the neutral buffer at room temperature for more than one month. Owing to the versatile nature, this approach can be further explored to prepare various grating sensors for the saccharide recognition and detection.

Abstract:The display of glucose oxidase (GOx) on yeast cell surface using a-agglutinin as an anchor motif was successfully developed. Both the immunochemical analysis and enzymatic assay showed that active GOx was efficiently expressed and translocated on the cell surface. Compared with conventional GOx, the yeast cell surface that displayed GOx (GOx-yeast) demonstrated excellent enzyme properties, such as good stability within a wide pH range (pH 3.5-11.5), good therrnostability (retaining over 94.8% enzyme activity at 52 degrees C and 84.2% enzyme activity at 56 degrees C), and high D-glucose specificity. In addition, direct electrochemistry was achieved at a GOx-yeast/multiwalled-carbon-nanotube modified electrode, suggesting that the host cell of yeast did not have any adverse effect on the electrocatalytic property of the recombinant GOx. Thus, a novel electrochemical glucose biosensor based on this GOx-yeast was developed. The as-prepared biosensor was linear with the concentration of D-glucose within the range of 0.1-14 mM and a low detection limit of 0.05 mM (signal-to-noise ratio of S/N = 3). Moreover, the as-prepared biosensor is stable, specific, reproducible, simple, and cost-effective, which can be applicable for real sample detection. The proposed strategy to construct robust GOx-yeast may be applied to explore other oxidase-displaying-system-based whole-cell biocatalysts, which can find broad potential application in biosensors,, bioenergy, and industrial catalysis.

Abstract:We report a glucose oxidase-containing polyacrylamide hydrogel-coated microcantilever sensor for the measurement of glucose. This enzymatic reaction of glucose results in swelling of the hydrogel due to formation of charged ions (gluconate molecules and protons). The microcantilever undergoes reversible and reproducible bending deflection upon exposure to solutions containing various glucose concentrations due to swelling or shrinking of the hydrogels. The microcantilever deflections increase when the glucose concentrations increase. A theoretical model has been built to correlate volume changes of the gel with microcantilever bending. The calculated data matched with the experimental results very well. Such hydrogel-coated microcantilevers could potentially be used to prepare microcantilever-based chemical and biological sensors when other enzymes are immobilized in the hydrogel.

Abstract:In this study, a glucose biosensor based on a hydrogel optical fiber immobilized with a fluorescence indicator and glucose oxidase (GO) is presented. The Hydrogel optical fiber was fabricated by mixing 2-hydroxy-2-methyl-propiophenone as a photo initiator, polyethylene glycol diacrylate (PEGDA) as a base, and deionized water with a certain ratio, irradiated under UV light. The oxygen-sensitive fluorescent indicator, o-phenanthroline ruthenium, and GO were introduced into the core of the hydrogel optical fiber. The sensitivity and efficiency of the system were optimized by varying the concentration of GO and the fluorescent indicator, and the sensing characteristics at different pH and temperatures were investigated. The experimental results showed that the performance of the sensor head was best at 25°C and a pH within the range of 4.5–6. The relationship between the glucose concentration and fluorescence ratio I/I0 within the range of 20–100 μg/mL is linear with a correlation coefficient of R2=0.9934. Moreover, the repeatability of the sensing system is good. Due to its good molecular permeability and biocompatibility, a hydrogel optical fiber glucose sensing system can be widely used in biomedical fields.

Abstract:Hydrogel biosensors usually suffer from a slow response, which severely hinders their practical applications. Here a new optical glucose biosensor was designed, using glucose-sensitive hydrogel films as both glucose-sensing material and Fabry-Perot cavity. The film was fabricated by layer-by-layer assembly from partially oxidized dextran (PO-Dex), chitosan, and glucose oxidase (GOD). The film responds to glucose because the incorporated GOD converts glucose to gluconic acid, and thus lowers the local pH in the film, and, in turn, triggers the pH-sensitive film to swell. The glucose-induced swelling causes a shift of Fabry-Perot fringes on the reflection spectra of the film, from which the glucose concentration can be reported. The new sensor works well under physiological conditions. Potential interferents, such as diols for phenylboronic acid-based sensors and electroactive compounds for electrochemical sensors, do not influence the new sensor. The sensor can respond reversibly over a wide range of glucose concentration. Particularly, it responds linearly within the clinically relevant glucose range (0-20 mM). More importantly, because the film is very thin, the new sensor can respond quickly, making it potential for real-time, continuous glucose monitoring. (C) 2016 Elsevier Ltd. All rights reserved.
Label-free Glucose Biosensor Based on Enzymatic Graphene Oxide-Functionalized Tilted Fiber Grating

Abstract:A label-free biosensor based on graphene oxide (GO) and glucose oxidase (GOD) functionalized tilted fiber grating (TFG) with large tilted angle is proposed for low concentration glucose detection. Taking advantages of sufficient binding sites of the GO with oxygen-containing groups, the enzymes (GOD) are covalently immobilized onto GO-deposited TFG via 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide and N-hydroxyl succinimide cross-liner. Surface characterizations with optical microscopy, scanning electron microscopy, Raman and infrared spectroscopy provide detailed assessments and evidences about the homogeneity of GO deposition and the effectiveness of enzyme modification. Through the specific catalysis reaction of GOD on the glucose, a considerable refractive index change in local microenvironment around the TFG results in the resonant wavelength shifts of cladding modes. The detection results of the low-concentration glucose demonstrate that the resonant wavelength has a linear response to the glucose concentration in the range of 0-8 mM with a response coefficient of 0.24 nm/mM, showing an enhanced sensitivity and bio-selectivity compared with the pristine TFG. The miniaturized size and remote label-free sensing capacity of the proposed device permit a multitude of opportunities for single-point measurement in harsh conditions and hard-to-reach spaces, presenting a promising candidate for label-free glucose detection for disease diagnosis, pharmaceutical research and bioengineering applications. A Hydrogel-Based Biosensor for Stable Detection of Glucose

Abstract:Glucose detection is vital in the food industry for safety and quality management. As a healthy ingredient, the flavor of honey is frequently impacted by the crystallization of glucose. Therefore, determining the glucose level can offer precise reference data for the manufacture of honey. Various approaches have been tried, and the enzyme-based electrochemical analytical method is one of the most important and widely used strategies. However, there are still challenges for most electrochemical methods to achieve stable detection resistant to temperature variation due to the easy inactivation of the enzyme, the poor anti-interference capacity of the detection techniques and other influences from the external environment. Herein, a hydrogel-based electrochemical biosensor is proposed to stably detect glucose even at wide ranges of temperatures via electrochemical impedance spectroscopic (EIS) measurement. The key factor for stable detection relies on the metal-organic framework nanoparticles' protective layer to guarantee the robustness of glucose oxidase (GOx), thereby achieving stable and specific detection for glucose. Moreover, a cascade reaction-induced hydrogel formation in a 3D structure can be used as an impedance readout, which not only amplifies but also further stabilizes the GOx-induced response. The prepared hydrogel-based electrochemical biosensor showed a linear response to the glucose concentration in the range of 0.75-4 mg/mL. Furthermore, the biosensor has excellent anti-interference and temperature stability. High performance liquid chromatography analysis also validated the accuracy of this biosensor in detecting glucose in the honey sample.

Abstract:A diffraction grating of a stimulus-responsive hydrogel was fabricated for the detection of CO2 with a low detection limit. The hydrogel consisted of dimethylaminopropyl methacrylamide (DMAPMA), methyl methacrylate, and 2-hydroxyethyl methacrylate. The sensing gratings were fabricated through soft-lithographic duplication of photoinduced surface-relief-gratings on azo molecular glass films on the hydrogel surfaces. The detection of an analyte was achieved by the reaction of tertiary amino groups of DMAPMA in the hydrogel with CO2 in water and swelling of hydrogel gratings to cause the variation of the optical-diffraction efficiency (DE). The relationship between DE and CO2 concentration, controlled by the gas-injection rate and volume, was systematically investigated. The performance of continuous detection of CO2 was also evaluated. The sensing device showed high sensitivity, reliable readability, and excellent repeatability for CO2 detection. The CO2-sensing performance of the hydrogel grating was proved to be almost unaffected by relative humidity variations of the environment in a wide range.
Novel Glucose Sensor Based on Enzyme-Immobilized 81° Tilted Fiber Grating.

Abstract:We demonstrate a novel glucose sensor based on an optical fiber grating with an excessively tilted index fringe structure and its surface modified by glucose oxidase (GOD). The aminopropyltriethoxysilane (APTES) was utilized as binding site for the subsequent GOD immobilization. Confocal microscopy and fluorescence microscope were used to provide the assessment of the effectiveness in modifying the fiber surface. The resonance wavelength of the sensor exhibited red-shift after the binding of the APTES and GOD to the fiber surface and also in the glucose detection process. The red-shift of the resonance wavelength showed a good linear response to the glucose concentration with a sensitivity of 0.298 nm·(mg/ml)-1 in the very low concentration range of 0.0~3.0mg/ml. Compared to the previously reported glucose sensor based on the GOD-immobilized long period grating (LPG), the 81° tilted fiber grating (81°-TFG) based sensor has shown a lower thermal cross-talk effect, better linearity and higher Q-factor in sensing response. In addition, its sensitivity for glucose concentration can be further improved by increasing the grating length and/or choosing a higher-order cladding mode for detection. Potentially, the proposed techniques based on 81°-TFG can be developed as sensitive, label free and micro-structural sensors for applications in food safety, disease diagnosis, clinical analysis and environmental monitoring.

Abstract:Phenylboronic acids have emerged as synthetic receptors that can reversibly bind to cis-diols of glucose molecules. The incorporation of phenylboronic acids in hydrogels offers exclusive attributes; for example, the binding process with glucose induces Donnan osmotic pressure resulting in volumetric changes in the matrix. However, their practical applications are hindered because of complex readout approaches and their time-consuming fabrication processes. Here, we demonstrate a microimprinting method to fabricate densely packed concavities in phenylboronic acid functionalized hydrogel films. A microengineered optical diffuser structure was imprinted on a phenylboronic acid based cis-diol recognizing motif prepositioned in a hydrogel film. The diffuser structure engineered on the hydrogel was based on laser-inscribed arrays of imperfect microlenses that focused the incoming light at different focal lengths and direction resulting in a diffused profile of light in transmission and reflection readout modes. The signature of the dimensional modulation was detected in terms of changing focal lengths of the microlenses due to the volumetric expansion of the hydrogel that altered the diffusion spectra and transmitted beam profile. The transmitted optical light spread and intensity through the sensor was measured to determine variation in glucose concentrations at physiological conditions. The sensor was integrated in a contact lens and placed over an artificial eye. Artificial stimulation of variation in glucose concentration allowed quantitative measurements using a smartphone's photodiode. A smartphone app was utilized to convert the received light intensity to quantitative glucose concentration values. The developed sensing platform offers low cost, rapid fabrication, and easy detection scheme as compared to other optical sensing counterparts. The presented detection scheme may have applications in wearable real-time biomarker monitoring devices at point-of-care settings.

A 2-D Photonic Crystal Hydrogel For Selective Sensing Of Glucose

Abstract:A novel 2-D photonic crystal (PC) sensing material for the visual detection of glucose with high selectivity at a physiological ionic strength (150 mM) is developed. A monolayer polystyrene crystalline colloidal array (CCA) is embedded in a phenylboronic acid (PBA) functionalized hydrogel film to diffract light to sensitively report on the hydrogel surface area. This 2-D PC sensor is superior to the previously reported 3-D PC sensors due to its fast preparation and simple detection. The binding of glucose would increase the cross-linking of the hydrogel that could shrink the hydrogel to increase the Debye diffraction ring diameter. At a physiological ionic strength of 150 mM, the 2-D PC sensor exhibits significant sensitivity for glucose across the entire human physiological glucose range. Additionally, the 2-D PC sensor shows high selectivity for glucose rather than other sugars (fructose and galactose). The diffraction color change of the 2-D PC sensor can be observed. For validation, the prepared 2-D PC hydrogel sensor is applied for the sensing of glucose in the artificial tear fluid.

Abstract:Diabetes mellitus is a chronic metabolic disorder, requiring vigilant monitoring of blood glucose levels. In this study, an injectable fluorescent enzymatic hydrogel was designed for rapid glucose detection. The leakage-free glucose-responsive hydrogel was constructed by the covalent linkage of a multi-arm poly-(ethylene glycol) (PEG), bovine serum albumin (BSA), glucose oxidase (GOx), and 4-(aminomethyl)-6,7-dimethoxycoumarin (Coumarin-NH2). The GOx serves as glucose-recognition element and the pH-sensitive Coumarin-NH2 as a fluorescence turn-on reporter. The material properties of the fluorescent hydrogel were systematically characterized which show high elasticity with good mechanical strength. Upon the addition of glucose, the as-developed fluorescent hydrogel shows a fast response time, good sensitivity, and good reproducibility at physiological pH and ambient temperature. The glucose-sensing mechanism is based on the oxidation of the glucose by GOx that generates protons to change the local pH. Consequently, protonation of the covalently immobilized and pH-sensitive Coumarin-NH2 turns on the fluorescence of the coumarin. The fluorescence hydrogel developed holds great promise as an injectable, implantable glucose-sensing biomaterials for in vivo continuous glucose monitoring.

Abstract:We present an easy and fast way to fabricate glucose oxidase-diphenylalanine-reduced graphene oxide (GOx-FF-rGO) film by using pulsed potentiostatic method. Glassy carbon electrode (GCE) modified by the obtained GOx-FF-rGO nanocomposite film is systematically characterized for its application in glucose detection. Under optimized condition, the biosensor exhibits a wide linear response in the range from 0.1 to 30.0 mmol L-1, with a low detection limit down to 0.01 mmol L-1. Our results indicate that the pulsed potentiostatic deposition approach may lead to production of highly sensitive and stable electrochemical biosensors. (C) 2017 Elsevier Ltd. All rights reserved.

Abstract:A sensitive nano-device for D-glucose detection is prepared by modifying a single conical polymer nanochannel with GOx enzymes. The current-voltage (I-V) characterization suggests that the nano-device responds to D-glucose rather than its enantiomer at concentrations down to 1 nM (10(-9) mol L-1). Moreover, the nano-device exhibits good reproducibility and specificity for D-glucose and is an ideal candidate for commercial non-invasive blood glucose meters in the future.

Abstract:Promoting the electrocatalytic oxidation of glucose is crucial in glucose biosensor design. In this study, nanoporous gold (NPG) was selected for glucose oxidase (GOx) immobilization and glucose biosensor fabrication because of its open, highly conductive, biocompatible, and interconnected porous structure, which also facilitates the electrocatalytic oxidation of glucose. The electrochemical reaction on the surface of the resulting GOx/NPG/GCE bioelectrode was attributed to the co-catalysis effect of GOx and NPG. A surface-confined reaction in a phosphate buffer solution was observed at the bioelectrode during cyclic voltammetry experiments. Linear responses were observed for large glucose concentrations ranging from 50μM to 10mM, with a high sensitivity of 12.1μAmM(-1)cm(-2) and a low detection limit of 1.02μM. Furthermore, the GOx/NPG/GCE bioelectrode presented strong anti-interference capability against cholesterol, urea, tributyrin, ascorbic acid, and uric acid, along with a long shelf-life. For the detection of glucose in human serum, the data generated by the GOx/NPG/GCE bioelectrode were in good agreement with those produced by an automatic biochemical analyzer. These unique properties make the GOx/NPG/GCE bioelectrode an excellent choice for the construction of a glucose biosensor.
Immobilizing glucose oxidase on AuCu hydrogels for enhanced electrochromic biosensing

Abstract:Development of highly sensitive and accurate biosensors still faces a great challenge. Herein, glucose oxidase (GOx) is efficiently immobilized on the AuCu hydrogels owing to their porous structure and interfacial interaction, demonstrating enhanced catalytic activity, satisfactory stability and recyclability. Besides, by integration of AuCu@GOx and electrochromic material of Prussian blue, a sensitive and stable biosensing platform based on the excellent electrochromic property of Prussian blue and the enhanced enzyme activity of AuCu@GOx is developed, which enables the electrochemical and visual dual-mode detection of glucose. The as-constructed biosensing platform possesses a wide linear range, and good selectivity for glucose detection with a limit of detection of 0.82 μM in visual mode and 0.84 μM in electrochemical mode. This easy-to-operate biosensing platform opens a door for the practical application of the multi-mode strategy for glucose detection.

Abstract:Optical fiber microprobe functionalized with Glucose oxidase (GOD) has been proposed for bio-selective and high-sensitive glucose detection. Taking advantages of the free amine groups of 3-aminopropyl-triethoxysilane (APTES), the enzymes (GOD) are immobilized on the multimode microfiber through covalent interaction. Surface characterization offered by optical microscopy, scanning electron microscope and multiphoton laser scanning microscopy provide detailed evidences about the effect of the bio-functionalization. The fabricated microfiber sensor is immersed into glucose solutions at different concentrations to record and analyze the transmission spectrums from an optical sensing interrogator. The experimental results demonstrate that the resonant wavelength shift is linearly correlated with the glucose concentration in the range of 0-3.0 mg/ml with a response coefficient of 1.74 nm/mg(-1) ml(-1). Meanwhile, the reported sensor is also proved to be of practical utility by accurately detecting glucose content in animal serum samples. Due to the small size, label-free sensing capacity and excellent practicality, the proposed device has great potential to be applied in the fields like disease diagnosis, clinical analysis and food safety.
