Abstract:We report a glucose oxidase-containing polyacrylamide hydrogel-coated microcantilever sensor for the measurement of glucose. This enzymatic reaction of glucose results in swelling of the hydrogel due to formation of charged ions (gluconate molecules and protons). The microcantilever undergoes reversible and reproducible bending deflection upon exposure to solutions containing various glucose concentrations due to swelling or shrinking of the hydrogels. The microcantilever deflections increase when the glucose concentrations increase. A theoretical model has been built to correlate volume changes of the gel with microcantilever bending. The calculated data matched with the experimental results very well. Such hydrogel-coated microcantilevers could potentially be used to prepare microcantilever-based chemical and biological sensors when other enzymes are immobilized in the hydrogel.

Abstract:A conceptually new "light-up" biosensor with a high specificity for d-glucose (Glu) in aqueous media has been developed. The emission from a tetraphenylethene (TPE)-cored diboronic acid (1) was greatly boosted when the fluorogen was oligomerized with Glu because of restriction of the intramolecular rotations of the aryl rotors of TPE by formation of the oligomer. Little change in the light emission was observed when 1 was mixed with D-fructose, D-galactose, or D-mannose, as these saccharides are unable to oligomerize with the fluorogen.

Abstract:Diabetes mellitus is a chronic metabolic disorder, requiring vigilant monitoring of blood glucose levels. In this study, an injectable fluorescent enzymatic hydrogel was designed for rapid glucose detection. The leakage-free glucose-responsive hydrogel was constructed by the covalent linkage of a multi-arm poly-(ethylene glycol) (PEG), bovine serum albumin (BSA), glucose oxidase (GOx), and 4-(aminomethyl)-6,7-dimethoxycoumarin (Coumarin-NH2). The GOx serves as glucose-recognition element and the pH-sensitive Coumarin-NH2 as a fluorescence turn-on reporter. The material properties of the fluorescent hydrogel were systematically characterized which show high elasticity with good mechanical strength. Upon the addition of glucose, the as-developed fluorescent hydrogel shows a fast response time, good sensitivity, and good reproducibility at physiological pH and ambient temperature. The glucose-sensing mechanism is based on the oxidation of the glucose by GOx that generates protons to change the local pH. Consequently, protonation of the covalently immobilized and pH-sensitive Coumarin-NH2 turns on the fluorescence of the coumarin. The fluorescence hydrogel developed holds great promise as an injectable, implantable glucose-sensing biomaterials for in vivo continuous glucose monitoring.

Abstract:Hydrogels, water-filled networks that can adapt to external stimuli by altering their volume, are known for their high flexibility and biocompatibility. DNA, a critical biomolecule renowned for its exceptional characteristics including information transmission, molecular recognition, and editability, has found widespread applications in the biosensing field as well. The integration of these two biomaterials offers promising opportunities for the development of novel biosensors with enhanced sensitivity, specificity, and adaptability. Therefore, by virtue of the collective features, researchers have recently focused on the construction of responsive DNA hydrogel systems. This feature article describes recent developments in fabricating DNA hydrogels and their applications in the biosensing area. Initially, it focuses on the design strategies employed in preparing DNA hydrogels, encompassing both pure DNA hydrogels and hybridized DNA hydrogels. Subsequently, it summarizes the use of DNA hydrogels in biosensing applications, highlighting their applications in visual detection, electrochemical sensing, and optical biosensing analyses. Furthermore, the underlying responsive mechanisms within these biosensing systems are also described. Lastly, this article presents a comprehensive discussion on the existing challenges and prospects of responsive DNA hydrogels, offering insights into their potential to revolutionize the field of biosensing.

Abstract:Analyte-sensitive DNA-based hydrogels find multiple applications in the field of biosensors due to their adaptable nature. Here, the design of DNA-based hydrogel and its application as sensing platform for the detection of a specific target sequence are presented. DNA-functionalized hydrogel structures were formed via a free radical co-polymerization process. A simple one-step probe immobilization procedure is reported: DNA probe molecules are added to the photoactive polymer mixture, dispensed onto a solid support, or a mold, and covalently attached while the hydrogel is formed through UV light exposure. Such hydrogels can be synthesized with desired recognition ability through the selection of a certain nucleotide sequence. Here we show the application of DNA-based hydrogel to detect the target with high performance in fluorescence microarray format and, additionally, to fabricate holographic surface relief gratings for label-free sensing assays.

Abstract:Food safety has always been a major global challenge to human health and the effective detection of harmful substances in food can reduce the risk to human health. However, the food industry has been plagued by a lack of effective and sensitive safety monitoring methods due to the tension between the cost and effectiveness of monitoring. DNA-based hydrogels combine the advantages of biocompatibility, programmability, the molecular recognition of DNA molecules, and the hydrophilicity of hydrogels, making them a hotspot in the research field of new nanomaterials. The stimulus response property greatly broadens the function and application range of DNA hydrogel. In recent years, DNA hydrogels based on stimulus-responsive mechanisms have been widely applied in the field of biosensing for the detection of a variety of target substances, including various food contaminants. In this review, we describe the recent advances in the preparation of stimuli-responsive DNA hydrogels, highlighting the progress of its application in food safety detection. Finally, we also discuss the challenges and future application of stimulus-responsive DNA hydrogels.

Abstract:In this study, a glucose biosensor based on a hydrogel optical fiber immobilized with a fluorescence indicator and glucose oxidase (GO) is presented. The Hydrogel optical fiber was fabricated by mixing 2-hydroxy-2-methyl-propiophenone as a photo initiator, polyethylene glycol diacrylate (PEGDA) as a base, and deionized water with a certain ratio, irradiated under UV light. The oxygen-sensitive fluorescent indicator, o-phenanthroline ruthenium, and GO were introduced into the core of the hydrogel optical fiber. The sensitivity and efficiency of the system were optimized by varying the concentration of GO and the fluorescent indicator, and the sensing characteristics at different pH and temperatures were investigated. The experimental results showed that the performance of the sensor head was best at 25°C and a pH within the range of 4.5–6. The relationship between the glucose concentration and fluorescence ratio I/I0 within the range of 20–100 μg/mL is linear with a correlation coefficient of R2=0.9934. Moreover, the repeatability of the sensing system is good. Due to its good molecular permeability and biocompatibility, a hydrogel optical fiber glucose sensing system can be widely used in biomedical fields.

Abstract:Fluorescent biosensors have been widely applied in DNA detection because of their reliability and reproducibility. However, low kinetics in DNA hybridization often brings out long test terms, thus restricting their practical use. Here, we demonstrate unexpected fast DNA fluorescence detection on the confined surface of hydrogel particles. When the pore size and surface charge of hydrogel particles are tailored, DNA molecules can be confined in the outer water layer of hydrogel particles. We fabricated a fluorescence-on DNA sensor based on the hydrogel particle array by utilizing the fluorescence quenching property of graphene oxide and its different adsorption behaviors toward single-strand DNA or double-strand DNA. Benefiting from the confinement effect of hydrogel particle surface and the enrichment effect of water evaporation, the DNA-recognition time was descreased significantly from 3000 s to less than 10 s under the target concentration of 400 nM. Moreover, rapid detection can be achieved at concentrations between 50 and 400 nM. Abstract:DNA hydrogels play an increasingly important role in biomedicine and bioanalysis applications. Due to their high programmability, multifunctionality and biocompatibility, they are often used as effective carriers for packing drugs, cells, or other bioactive cargoes in vitro and in vivo. However, the stability of the DNA hydrogels prevents their in-demand rapid release of cargoes to achieve a full therapeutic effect in time. For bioanalysis, the generation of signals sometimes needs the DNA hydrogel to be rapidly degraded when sensing target molecules. To meet these requirements, stimulus-responsive DNA hydrogels are designed. By responding to different stimuli, self-degradable DNA hydrogels can switch from gel to solution for quantitative bioanalysis and precision cargo delivery. This review summarizes the recently developed innovative methods for designing stimuli-responsive self-degradable DNA hydrogels and showed their applications in the bioanalysis and biomedicines fields. Challenges, as well as prospects, are also discussed.

Abstract:A new glucose sensing strategy has been developed by using diffraction-gratings of a stimuli-responsive hydrogel bearing phenylboronic acid groups. The sensing gratings were fabricated through soft-lithographic duplication of photoinduced surface-relief-gratings (SRGs) on azo polymer films to the hydrogel surfaces. For the preparation, elastomeric stamps were obtained by casting PDMS prepolymer against the azo polymer masters and curing properly. The hydrogel gratings containing phenylboronic acid groups were shaped by contact printing with the PDMS stamps and a specially prepared “ink”, which was a precursor solution curable by UV light irradiation. The gratings showed a fast and highly sensitive response to glucose in buffer solutions as read out by the increase of the diffraction efficiency (DE/DE0). The saturated DE/DE0 value was linearly correlated to the glucose concentration of the solutions. The gratings showed ability for the continuous detection and were durable for repeated applications. The sensing ability of the gratings was stable when kept in the neutral buffer at room temperature for more than one month. Owing to the versatile nature, this approach can be further explored to prepare various grating sensors for the saccharide recognition and detection.

Abstract:A novel glucose-responsive hydrogel system based on dynamic covalent chemistry and inclusion complexation was described. Hydrogels are formed by simply mixing the solutions of three components: poly(ethylene oxide)-b-poly vinyl alcohol (PEO-b-PVA) diblock polymer, alpha-cyclodextrin (alpha-CD) and phenylboronic acid (PBA)-terminated PEO crosslinker. Dynamic covalent bonds between PVA and PBA provide sugar-responsive crosslinking, and the inclusion complexation between PEO and alpha-CD can promote hydrogel formation and enhance hydrogel stability. The ratios of the three components have a remarkable effect on the gelation time and the mechanical properties of the final gels. In rheological measurements, the hydrogels are demonstrated to possess solid-like behaviour and good structural recovery ability after yielding. The sugar-responsiveness of the hydrogels was examined by protein loading and release experiments, and the results indicate that this property is also dependent on the compositions of the gels; at a proper component ratio, a new glucose-responsive hydrogel system operating at physiological pH can be obtained. The combination of good biocompatibility of the three components and the easy preparation of hydrogels with tunable glucose-responsiveness may enable an alternative design of hydrogel systems that finds potential applications in biomedical and pharmaceutical fields, such as treatment of diabetes.

Abstract:Glucose monitoring sensors with high softness and flexibility are critical for the developments of wearable and implantable healthcare devices that enable diagnosis, prognosis, and management of diabetes. The design and implementation of such sensors have been extensively exploited by electrochemical strategies, which, however, suffer from poor reusability and complex modification procedures, and necessitate frequent calibration or sensor replacement due to enzymatic reaction instability. Here, a soft and plasmonic hydrogel optical sensor is created for quantitative and continuous glucose monitoring under physiological conditions. The optical sensor consists of a flexible optical fiber made from composites of gold nanoparticles and glucose-responsive hydrogels. The reversible binding of glucose to the nanocomposite optical fiber results in dynamic volume expansion of the hydrogel matrix, which modulates the localized surface plasmon resonance effect, enabling glucose to be quantified from the light transmission. To achieve robust readout, a dual-wavelength differential approach is employed to endow the sensor with self calibration capability. We show that the sensor is reversible and reusable for detecting physiological glucose levels with high linearity and negligible hysteresis. The soft and flexible glucose sensor holds great promises of serving as a minimally-invasive probe for point-of-care glucose monitoring in clinics.
Surface-immobilized and Self-Shaped DNA Hydrogels and Their Application in Biosensing.

Abstract:Hydrogels are of great interest in the field of biosensing for their good biocompatibility, plasticity, and capability of providing 3D scaffolds. Nevertheless, the application of hydrogels has not been linked with broad surface biosensing systems yet. To overcome the limitations, here for the first time, surface-immobilized pure DNA hydrogels were synthesized using a surficial primer-induced strategy and adopted for biosensing applications. The DNA hydrogel 3D scaffold is successfully constructed on a transparent ITO electrode, which facilitates both colourimetric and electrochemical measurements. Results show that the hydrogel is able to wrap enzymes solidly and exhibits favourable stability under different conditions. Owing to the free diffusion of the micromolecular targets throughout the hydrogel, while isolating the enzymes from the macromolecular interferences outside the hydrogel, the direct colourimetric and electrochemical detection of hydrogen peroxide and bilirubin in serum is achieved. The detection limit of hydrogen peroxide in serum is 22 nM by colourimetric analysis and 13 nM by electrochemical measurement. The detection limit of bilirubin is 32 nM, a favourable limit that could be used in jaundice diagnosis. In addition, the enzyme@hydrogel can be easily regenerated and the catalytic activity is retained for a few cycles, thus allowing the recycling of the hydrogel-based biosensing system. The successful integration of DNA hydrogels with surface biosensing systems will greatly expand the applications of hydrogels for diagnostic and environmental monitoring purposes.

Abstract:Responsive hydrogels show obvious superiorities when being employed at detection owing to their polymeric networks and optical properties. Albeit important, the slow response of traditional hydrogels against targets limits their further applications, and enhancing the responsiveness of hydrogel is therefore imperative. Herein, a cryogel with hierarchical structures was designed through adjusting the hydrogel structure by the freezing process to improve the responsive properties, and fabricated for rapid analysis of 17β-estradiol (E2). The cryogel encapsulating the DNA hairpins rapidly adsorbed the complementary DNA isolated from the aptamer in the presence of E2 and activated an efficient hybridization chain reaction (HCR). After integration with a smartphone system, a portable colorimetric sensing platform was created for sensitive detection of E2 with a low detection limit of 0.5 nM. Our work provides a promising way for highly responsive hydrogel-based biosensor design, and broadens their applications in rapid and portable target detection.

Abstract:DNA hydrogels are hydrophilic polymer networks formed by DNA cross-linked polymer backbones or self-assembly of DNA building blocks. Following Watson-Crick base pairing principle and switchable recognition capability towards a wide range of biomolecules, programmable DNA assembly or disassociation events can be applied for the fabrication of highly sensitive biosensors. DNA hydrogels have excellent biocompatibility, degradability, stability and mechanical properties, which have attracted extensive attention. Recently, various functional DNA hydrogels have emerged with the development of nucleic acids amplification strategies and the incorporation of suitable nanomaterials. In this review, we first provide a comprehensive overview of the synthesis principles of DNA hydrogels. The functional regulations are highlighted. Recent advances on the development of DNA hydrogels-based biosensors are then discussed with selected examples. In the final section, challenges and perspectives for the biosensing applications are prospected.

Abstract:The level of glucose, one of the body's energy-providing substances, is closely related to many diseases. Therefore, it remains a challenge to develop platform that can satisfy both the need for accurate detection of glucose for clinical diagnosis as well as device-free and efficient on-site monitoring of blood glucose. Herein, we synthesized Zr-MOFs@Pt nanocomposites for the first time. The peroxidase-like activity of Zr-MOFs@Pt was greatly enhanced two times compared with Zr-MOFs, due to the synergistic effect of Pt nanoparticles that was in situ growing in Zr-MOFs. Hydrogen peroxide was generated from glucose with the catalysis of glucose oxidase (GOx). H 2 O 2 was decomposed by Zr-MOFs@Pt to produce free radical, which could oxidize 3,3′,5,5′-tetramethylbenzidine (TMB) to generate blue oxTMB, and could be utilized for colorimetric sensing of glucose. By crosslinking, TMB and PVA hydrogel matrix formed the microneedle patch display layer, and PVA as well as GOx composed the tip of the needle, the sensing system was developed to hydrogel microneedles array for detection of glucose. When the tip of the needle was inserted into a solution containing glucose, it reacted with GOx in the tip sector to produce H 2 O 2 . The H 2 O 2 would react with Zr-MOFs@Pt in the display layer to produce free radicals, and further oxidize TMB causing the display layer to exhibit blue color. This work provides a new approach for designing device-free sensing platforms for glucose monitoring, which also offers great potential for development of microneedle analytical devices.

Abstract:The lack of solid-phase media limits the portability of colorimetric sensing platforms. In this study, a series of transparent polyvinyl alcohol (PVA) hydrogels encapsulated antimony tin oxide nanoparticles (ATO NPs) and 3,3′,5,5′-tetramethylbenzidine (TMB) were developed as the solid-phase sensing media for glucose and uric acid. Under the conditions of H 2 O 2 and UV light, the hydrogel presented a multicatalytic ability (photo Fenton-like and peroxidase-like activities), which accelerated the oxidation of TMB, turning the hydrogel from colorless to blue and finally enhancing the detection signal. The plasticity of the hydrogel allowed it to be designed into various shapes (membrane, microsphere etc. ) to adapt multiple detection platforms (a liquid/solid-phase UV spectrophotometer, a NanoPhotometer, and smartphone spectroscopy). The hydrogel sensing media exhibited excellent tunability and enhanced the photocatalytic ability. The proposed material was successfully applied to detect glucose and uric acids in real samples by four detection platforms to evaluate its practicability.

Three-dimensional hydrogel structures as optical sensor arrays, for the detection of specific DNA sequences

Abstract:The fabrication and characterization of surface-attached PEG-diacrylate hydrogel structures and their application as sensing platforms for the detection of specific target sequences are reported. Hydrogel structures were formed by a photopolymerization process, using substrate-bound Eosin Y molecules for the production of free radicals. We have demonstrated that this fabrication process allows for control over hydrogel growth down to the micrometer scale. Confocal imaging revealed relatively large pore structures for 25% (v/v) PEG-diacrylate hydrogels, which appear to lie in tightly packed layers. Our data suggest that these pore structures decrease in size for hydrogels with increasing levels of PEG-diacrylate. Surface coverage values calculated for hydrogels immobilized with 21-mer DNA probe sequences were significantly higher compared to those previously reported for 2- and 3-dimensional sensing platforms, on the order of 10(16)molecules cm(-2). Used as sensing platforms in DNA hybridization assays, a detection limit of 3.9 nM was achieved for hybridization reactions between 21-mer probe and target sequences. The ability of these hydrogel sensing platforms to discriminate between wild-type and mutant allele sequences was also demonstrated, down to target concentrations of 1-2 nM. A reduction in the hybridization time down to a period of 15 min was also achieved, while still maintaining confident results, demonstrating the potential for future integration of these sensing platforms within Lab-on-Chip or diagnostic devices.
