Abstract:In the title salt [systematic name: 4-(3-carb-oxy-1-ethyl-6-fluoro-4-oxo-1,4-di-hydro-quin-olin-7-yl)piperazin-1-ium nitrate], C16H19FN3O3 +·NO3 -, proton transfer from nitric acid to the N atom of the piperazine ring of norfloxacin has occurred to form a mol-ecular salt. In the extended structure, N-H⋯O hydrogen bonds link alternating cations and anions into [100] chains, which are reinforced by aromatic π-π stacking inter-actions between the quinoline moieties of the norfloxacinium cations.

Abstract:Norfloxacin (NORF) is a broad-spectrum quinolone that is widely utilized for the treatment of various bacterial infections and is considered one of the most commonly used fluoroquinolone antibiotics. However, NORF's clinical utility is limited by its poor water solubility and relatively low oral bioavailability. This study presents an optimization and synergistic enhancement approach through salt/co-crystal, aiming to maximize the biopharmaceutical properties of NORF with the use of phenolic acid. Following this strategy, two new hydrate salts of NORF with phenolic acid, namely, NORF—3,5-DBA hydrate (salt 1) and NORF—VA hydrate (salt 2), were prepared and systematically confirmed. Two hydrate salts were produced by means of the slow evaporation crystallization method, and the structures were determined through single-crystal X-ray diffraction (SCXRD). Additionally, powder X-ray diffraction (PXRD), Fourier-transform infrared (FT-IR) spectroscopy, differential scanning calorimetry (DSC), thermogravimetric analysis (TGA), and high-performance liquid chromatography (HPLC) were applied to analyze the features of the two salts. The experimental results indicated that the formation of the two salts could enhance the solubility and improve the release behavior of NORF. Interestingly, the physicochemical properties of NORF were significantly improved as a result, leading to an enhancement in its antibacterial activity. This was demonstrated by the enhanced inhibition of bacterial strains and the lower minimum inhibitory concentration values.

Abstract:This study demonstrates an efficient heterogeneous electro-Fenton system with a CoFe-layered double hydroxide/carbon felt (CoFe-LDH/CF) cathode for norfloxacin (NFXN) removal. Two-dimensional CoFe-LDH prepared at the optimal temperature of 90 degrees C could produce sufficient catalyst dosages and highly crystallized honeycomb hexagonal nanoplates, revealing extraordinary (at least 90%) mineralization of NFXN at all pH values after 480 min of treatment. The brilliant catalytic activity presented over similar to 97% for the mineralization of NFXN after 5 cycles experiments, indicating the good reusability of the prepared cathode. According to density functional theory (DFT) calculations, center dot OH could be generated easily since the lower energy barriers were achievable for intermediates absorbed on Co atoms rather than those absorbed on Fe atoms, demonstrating that the activity trend remained Co > Fe on CoFe-LDH. In addition, the mechanisms of H2O2 generation on practical CoFe-LDH/CF cathodes were explored. The CF in the CoFe-LDH/CF cathode is a highly active and stable 2-electron ORR catalyst for H2O2 production, which provides sufficient substrates for center dot OH generation. This study potentially provides new insight into exploring the nature of reactions in hetero-EF systems.

Abstract:The mixed ligand coordination compound of copper with norfloxacin (NFLX) and 1, 10-phen has been synthesized and characterized by means of X-ray single crystal diffraction. The structure features of the coordination compound are described. Antibacterial activities of the coordination compound have been tested against different microorganisms. The antitumor activities of the coordination compound on leukemia HL-60 cell line and liver cancer BEL-7402 cell line have been measured, respectively. The results indicated that the coordination compound has strong inhibitory effect on HL-60 and BEL-7402 cell lines.

Abstract:A new cocrystal of Norfloxacin, a poorly soluble fluoroquinolone antibiotic, has been synthetized by a solvent-mediated transformation experiment in toluene, using resorcinol as a coformer. The new cocrystal exists in both anhydrous and monohydrate forms with the same (1:1) Norfloxacin/resorcinol stoichiometry. The solubility of Norfloxacin and the hydrated cocrystal were determined by the shake-flask method. While Norfloxacin has a solubility of 0.32 ± 0.02 mg/mL, the cocrystal has a solubility of 2.64 ± 0.39 mg/mL, approximately 10-fold higher. The dissolution rate was tested at four biorelevant pH levels of the gastrointestinal tract: 2.0, 4.0, 5.5, and 7.4. In a first set of comparative tests, the dissolution rate of Norfloxacin and the cocrystal was determined separately at each pH value. Both solid forms showed the highest dissolution rate at pH 2.0, where Norfloxacin is totally protonated. Then, the dissolution rate decreases as pH increases. In a second set of experiments, the dissolution of the cocrystal was evaluated by a unique dissolution test, in which the pH dynamically changed from 2.0 to 7.4, stepping 30 min at each of the four biorelevant pH values. Results were quite different in this case, since dissolution at pH 2 affects the behavior of Norfloxacin at the rest of the pH values.

Abstract:Drug-drug cocrystals can improve the physical and pharmaceutical properties of drugs and have been widely studied by many researchers. A new pharmaceutical cocrystal [C 16 H 18 FN 3 O 3 ][C 16 H 14 O 6 ] · 2(C 2 H 5 OH) (HSP-NOR) was synthesized by hesperetin (HSP) and norfloxacin (NOR) with the aim of improving the antibacterial activity and physicochemical properties. The single crystal structure shows that the asymmetric unit comprises both hesperetin and norfloxacin, as well as two ethanol molecules, in which norfloxacin exists as a zwitterion. The cocrystal formation improves the solubilities of hesperetin and norfloxacin in pure water. Preliminary antibacterial experiments showed that the antibacterial activities of the cocrystal on Streptococcus pneumoniae , Escherichia coli , Shigella dysenteriae , and Pseudomonas aeruginosa were stronger than those of norfloxacin. The in vivo study of acute toxicity showed that the minimum lethal dose of the cocrystal to mice was >3000 mg kg -1 BW, and there were no signs of toxicity after oral administration at this dosage.

Abstract:A novel carbon quantum dots (CQDs) sensitized 2D/2D carbon nitride nanosheets and bismuth tungstate composite (CQD-CNs/BWO) was successfully prepared via the facile hydrothermal method and used for the photocatalytic degradation of norfloxacin (NOR). During 120 min irradiation test, CQD-CNs/BWO exhibited 9 and 1.76 times higher photocatalytic activity than CNs and BWO, respectively. CQDs and constructed 2D/2D structure could not only improve the light harvesting but also promote the generation and separation of electron-holes. The existing inorganic ions in solution (e.g. bicarbonate ions, chlorine ions, and sulfate ions) could inhibit NOR degradation. Based on the electron spin resonance and free radicals inhibition tests, the holes and superoxide radicals rather than hydroxyl radicals were the main reactive species. The intermediates and possible pathways were proposed, and the antibacterial activity of the treated solution after the reaction was evaluated via bacteriostatic tests. The prepared composite material with high photocatalytic activity and stability is potentially effective for the degradation of antibiotics in wastewater.

Norfloxacin removal by ultraviolet-activated sodium percarbonate and sodium hypochlorite: process optimization and anion effect

Abstract:The efficiency of UV-activated sodium percarbonate (SPC) and sodium hypochlorite (SHC) in Norfloxacin (Norf) removal from an aqueous solution was assessed. Control experiments were conducted and the synergistic effect of the UV-SHC and UV-SPC processes were 0.61 and 2.89, respectively. According to the first-order reaction rate constants, the process rates were ranked as UV-SPC > SPC > UV and UV-SHC > SHC > UV. Central composite design was applied to determine the optimum operating conditions for maximum Norf removal. Under optimum conditions (UV-SPC: 1 mg/L initial Norf, 4 mM SPC, pH 3, 50 min; UV-SHC: 1 mg/L initial Norf, 1 mM SHC, pH 7, 8 min), the removal yields for the UV-SPC and UV-SHC were 71.8 and 72.1%, respectively. HCO3-, Cl-, NO3-, and SO42- negatively affected both processes. UV-SPC and UV-SHC processes were effective for Norf removal from aqueous solution. Similar removal efficiencies were obtained with both processes; however, this removal efficiency was achieved in a much shorter time and more economically with the UV-SHC process.

Abstract:The title compound {systematic name: 7-[(1S,6S)-8-aza-2-azoniabicyclo[4.3.0] non-8-yl]-1-cyclopropyl-6-fluoro-8-methoxy- 4-oxo-1,4-dihydroquinoline-3-carboxylic acid chloride monohydrate}, C21H25FN3O4+center dot Cl-center dot H2O, crystallizes with two moxifloxacinium cations, two chloride ions and two uncoordinated water molecules in the unit cell. The crystal structure has a pseudo-inversion center except for the chloride ions. In both moxifloxacinium cations, the quinoline rings are approximately planar, the maximum atomic deviations being 0.107 (3) and 0.118 (3) angstrom. The piperidine rings adopt a chair conformation while the pyrrolidine rings display a half-chair conformation. In the crystal, the carboxyl groups, the protonated piperidyl groups, the uncoordinated water molecule and chloride anions participate in O-H center dot center dot center dot O, O-H center dot center dot center dot Cl and N-H center dot center dot center dot Cl hydrogen bonding; weak intermolecular C-H center dot center dot center dot O and C-H center dot center dot center dot Cl hydrogen bonding is also present in the crystal structure.
