Abstract:The mixed ligand coordination compound of copper with norfloxacin (NFLX) and 1, 10-phen has been synthesized and characterized by means of X-ray single crystal diffraction. The structure features of the coordination compound are described. Antibacterial activities of the coordination compound have been tested against different microorganisms. The antitumor activities of the coordination compound on leukemia HL-60 cell line and liver cancer BEL-7402 cell line have been measured, respectively. The results indicated that the coordination compound has strong inhibitory effect on HL-60 and BEL-7402 cell lines.

Abstract:Molecular dynamics (MD) simulations were used to investigate the interaction of norfloxacin with the DNA oligonucleotide d[ATATCGATAT](2). The initial binding structure was built with molecular modeling based on the experimental results. A 2 ns MD calculation was performed to study the norfloxacin-DNA complex and the results indicated that norfloxacin was inserted in the minor groove of DNA, binding to the region of duplex TCGA bases. The possible H-bonds between carbonyl and carboxyl group of norfloxacin and amine group of the guanine base support a minor-groove complex for the compound in GC DNA sequence. Molecular dynamics studies complement the structural analysis and provide a clear picture of the norfloxacin-DNA complex.

Abstract:Drug-drug cocrystals can improve the physical and pharmaceutical properties of drugs and have been widely studied by many researchers. A new pharmaceutical cocrystal [C 16 H 18 FN 3 O 3 ][C 16 H 14 O 6 ] · 2(C 2 H 5 OH) (HSP-NOR) was synthesized by hesperetin (HSP) and norfloxacin (NOR) with the aim of improving the antibacterial activity and physicochemical properties. The single crystal structure shows that the asymmetric unit comprises both hesperetin and norfloxacin, as well as two ethanol molecules, in which norfloxacin exists as a zwitterion. The cocrystal formation improves the solubilities of hesperetin and norfloxacin in pure water. Preliminary antibacterial experiments showed that the antibacterial activities of the cocrystal on Streptococcus pneumoniae , Escherichia coli , Shigella dysenteriae , and Pseudomonas aeruginosa were stronger than those of norfloxacin. The in vivo study of acute toxicity showed that the minimum lethal dose of the cocrystal to mice was >3000 mg kg -1 BW, and there were no signs of toxicity after oral administration at this dosage.

Abstract:Norfloxacin (NORF) is a broad-spectrum quinolone that is widely utilized for the treatment of various bacterial infections and is considered one of the most commonly used fluoroquinolone antibiotics. However, NORF's clinical utility is limited by its poor water solubility and relatively low oral bioavailability. This study presents an optimization and synergistic enhancement approach through salt/co-crystal, aiming to maximize the biopharmaceutical properties of NORF with the use of phenolic acid. Following this strategy, two new hydrate salts of NORF with phenolic acid, namely, NORF—3,5-DBA hydrate (salt 1) and NORF—VA hydrate (salt 2), were prepared and systematically confirmed. Two hydrate salts were produced by means of the slow evaporation crystallization method, and the structures were determined through single-crystal X-ray diffraction (SCXRD). Additionally, powder X-ray diffraction (PXRD), Fourier-transform infrared (FT-IR) spectroscopy, differential scanning calorimetry (DSC), thermogravimetric analysis (TGA), and high-performance liquid chromatography (HPLC) were applied to analyze the features of the two salts. The experimental results indicated that the formation of the two salts could enhance the solubility and improve the release behavior of NORF. Interestingly, the physicochemical properties of NORF were significantly improved as a result, leading to an enhancement in its antibacterial activity. This was demonstrated by the enhanced inhibition of bacterial strains and the lower minimum inhibitory concentration values.

Abstract:Three novel rare earth complexes [N(CH3)4][Ln(NF)4]6H2O(Ln=Nd(III)(1), Sm(III)(2), Ho(III)(3)) were synthesized using hydrothermal method from norfloxacin HNF=1-ethyl-6-fluoro-1,4-dihydro-4-oxo-7-(1-piperazinyl)-3-quinoline carboxylic acid, C16H18FN3O3), imidazole and rare earth nitrates. The complexes were characterized by elemental analysis, FT-IR, TG-DTG and X-ray single crystal diffraction. Each rare earth ion was eight-coordinated with carboxyl-O atoms and keto-O atoms from norfloxacin. Four of the norfloxacin ions acted as bidentate chelate group took part in the coordination with rare earth ion. The structures of complexes were tetragonal system with space group I41/acd, which were allomerism. The interaction between complex 1 and DNA was studied by electronic absorption spectra and fluorescence spectroscopy. The binding interaction between the complex 1 and bovine serum albumin (BSA) was studied by fluorescence spectroscopy. The complex 1 bound to DNA by the mode of partial intercalation. Complex 1 had a strong ability to quench the fluorescence from BSA. The complex interaction was mainly a static quenching process with BSA together with formation of two binding sites.

Abstract:Reaction of norfloxacin (H-Norf) with AgNO3 yields an unusual mononuclear complex [Ag(H-Norf)2]NO3 (1) in which the local coordination environment around Ag+ ion is approximately linear with a N–Ag–N angle of 162.1(2)°. Larger concentration of Ag+ ions was found in water solution of 1. The unique bonding in 1 may lead to the readily release of Ag+ ion from 1, and leads to better antibacterial action in topical burn treatments. Strong blue fluorescent emission of 1 was also observed.

Abstract:Two new mononuclear complexes, [Ni(cepha)(2)]center dot 6H(2)O (1) and [Zn(cepha)(2)]center dot 6H(2)O (2) (cepha=cephalosporoate) have been synthesized and characterized by elemental analysis, infrared spectroscopy, thermogravimetric analysis and X-ray powder diffraction analysis. The structures of complexes have also been determined by X-ray single crystal diffraction analysis, and the results show that the complexes 1 and 2 are both in orthorhombic system with space group P22(1)2(1). The cephalexin hydrolyzes during the coordination process to produce cephalosporin intermediate, cephalosporoate (cepha), which participates in coordination. Metal ion acts as six-coordinated center forming a distorted octahedral geometry, and the complexes are further extended to a three-dimensional network via intermolecular hydrogen bonds. The antitumor activity in vitro of cephalexin, metal acetate and complexes were carried out by MTT assay. The results reveal that complexes 1 and 2 have a certain inhibiting activity against MCF-7 and HepG-2 cell lines. CCDC: 1497051, 1; 1497052, 2.

Abstract:The synthesis of the complex Zn(ofo)2(H2O)·2H2O is reported.Its crystal structure is presented and discussed.The title compound crystallizes in the triclinic space group P-1 with a=9.306(1),b=11.331(1),c=17.787(2),α=92.667(1)°,β=94.898(2)°,γ=92.071(2)°,V=1865.1(3)A3,Z=2,and Dc=1.496 mg·m-3.The Zn(Ⅱ) ion is in a slightly distorted square pyramid environment of oxygen atoms.The IR bands of carboxylic and ketonic group at 1 713 and 1 622 cm-1,respectively,shift to 1 620 and 1 584 cm-1 in the coordination compound.Thermo-gravimetric(TG) and its differential(DTG) for ofo and the coordination compound are carried out and explained in detail.Both ligand and the complex are assayed against gram-positive and gram-negative bacteria by in vitro doubling dilutions method,the complex shows the same minimal inhibitory concentration(MIC) as the corresponding ligand against S.Aureus,M.Lutens,E.Coli and P.Aeruginosa.

Abstract:Molecular dynamics were used to investigate the interaction between norfloxacin and DNA duplex. The results showed that norfloxacin was situated in the minor groove of DNA, binding to the TCGA region of d [ATATCGATAT](2). Specific hydrogen bonds were formed between norfloxacin and guanine base of DNA during the 2 ns MD, which may be the reason for the preferentiality of quinolone antibacterial towards the guanine base of DNA duplex.

Abstract:In this work, a new magnesium(II) complex was synthesized by solvothermal method using marbofloxacin (MB) as the medicinal ligand. The crystal structure of this complex ([Mg(MB)2(H2O)2], MB-Mg) determined by X-ray single crystal diffraction analysis demonstrates a mononuclear and six-coordinated octahedral configuration, in which two MB ligands chelate with Mg(II) through the 3-carboxyhydroxyl O atom and the 4-carbonyl O atom of MB, respectively, and two water molecules occupy the axial positions of the octahedron. The study on the solution chemistry by means of mass spectroscopy and molar conductivity further indicate that the coordinated species of MB-Mg could be still maintained in aqueous solution. The minimal inhibitory concentration (MIC) of MB and MB-Mg against seven typical species of pathogenic bacteria were tested in vitro. The results showed that MB-Mg exhibited better inhibitory effects on most the tested strains than MB. Furthermore, the acute toxicity was tested by acute toxicity test in mice. The results showed that the LD50 of MB-Mg was 9184.25 mg/Kg, which could be classified to Grade-II toxicity (practical non-toxic) according to the standard of toxicity classification standard. The acute toxicity in vivo, represented by LD50, is significantly lower than which of MB ligands (LD50 = 1249 mg/Kg, Grade-III toxicity). This result strongly suggests that the coordination of MB with the biosafe Mg(II) can effectively reduce its toxicity in vivo, contributing to the research of new metal-based veterinary drug products, with important practical value and scientific significance.

Abstract:A new cocrystal of Norfloxacin, a poorly soluble fluoroquinolone antibiotic, has been synthetized by a solvent-mediated transformation experiment in toluene, using resorcinol as a coformer. The new cocrystal exists in both anhydrous and monohydrate forms with the same (1:1) Norfloxacin/resorcinol stoichiometry. The solubility of Norfloxacin and the hydrated cocrystal were determined by the shake-flask method. While Norfloxacin has a solubility of 0.32 ± 0.02 mg/mL, the cocrystal has a solubility of 2.64 ± 0.39 mg/mL, approximately 10-fold higher. The dissolution rate was tested at four biorelevant pH levels of the gastrointestinal tract: 2.0, 4.0, 5.5, and 7.4. In a first set of comparative tests, the dissolution rate of Norfloxacin and the cocrystal was determined separately at each pH value. Both solid forms showed the highest dissolution rate at pH 2.0, where Norfloxacin is totally protonated. Then, the dissolution rate decreases as pH increases. In a second set of experiments, the dissolution of the cocrystal was evaluated by a unique dissolution test, in which the pH dynamically changed from 2.0 to 7.4, stepping 30 min at each of the four biorelevant pH values. Results were quite different in this case, since dissolution at pH 2 affects the behavior of Norfloxacin at the rest of the pH values.
