Abstract:Ligulaverin A I, an eremophilane derivative with a novel carbon skeleton, was isolated from the medicinal plant Ligularia veitchiana, along with four analogues (ligulaverin B 2, ligulaverin C 3, ligulaverin D 4 and ligulaverin E 5). Abstract:Two new eremophilane-type sesquiterpenoids eremophil-6-en-11-ol (1) and (7 alpha,9 alpha,10 alpha)-9,10-epoxy-eremophilan-11-ol (2), together with a known eremophilane-type (6 alpha,8 alpha)-6,8-dihydroxyeremophil-7(11)-en-12-oic acid 12,8-lactone (3) were isolated from the rhizomes of Ligularia veitchiana. The structures of 1 and 2 were established by spectral analysis including H-1- and C-13-NMR, HSQC, HMBC, and HR-ESI-MS data. The compounds 1 and 3 were assessed against lung-cancer (A549) and stomach-cancer (BCG823) cell lines by the MTT method. The results showed that I exhibited significant inhibiting activities on the growth of these cancer cells with IC50 values between 1 - 100 mu g/ml, whereas compound 3 had no effect on the same cell lines.

Abstract:Aim:To study the chemical constituents of the leaves of Ligularia veitchiana.Methods:Silica gel and sephadex LH-20 chromatography were used to isolate the chemical constituents whose chemical structure were identified by chemical,physical,and spectral methods .Results and Conclusion: Three compound were isolated and identified:β-sitosterol, 8β-hydroxyeremophil-7(11)-en-12,8α(14β,6α)-dioxide, and daucosterol.

Abstract:Sesquiterpenes, one type of the representative constituents for compositae plants, have been the phytochemical research focus for last decades, because of their structural and biological diversity. In recent years, systematic phytochemical investigations on five Compositae medicinal plants from Gansu and Yunnan provinces of western China, Laggera pterodonta DC. Benth. Laggera alata D. Don Sch-Bip. ex Oliv., Senecio tsoongianus Ling., Senecio saluenensis and Ligularia veitchiana (Hemsl.) Greenm were carried out and nearly 100 eudesmane and eremophilane derivatives were isolated and structurally elucidated. 56 eudesmanoids were obtained from the two species of Laggera genus, likewise 36 eremophilanoids were isolated from the Ligularia and Senecio plants of the tribe Senecioneae. Some of these sesquiterpenoids showed remarkable antibacterial, anti-inflammatory, cytotoxic and antiviral activities.

Abstract:Ligularia fischeriTurcz. is a medicinal plant for the treatment of inflammation in China and Korea. Its chemical components in anti-sepsis activity and the related molecular mechanisms remain unknown yet. In this study, two undescribed eremophilane sesquiterpenoids fischerins A (1) and B (2), together with 8 known sesquiterpenoid derivatives (3-10), were isolated from the whole plant of L. fischeri. Their structures were identified by detailed spectroscopic and ECD analyses. 3-Oxo-8-hydroxyeremophila-1,7(11)-dien-12,8-olide (6) showed the most inhibitory effect on NO production in LPS-stimulated RAW 264.7 cells with the IC50 value of 6.528 μM. Meanwhile, compound 6 also decreased the mRNA expression of pro-inflammatory factors IFN-γ, IL-1β, IL-6 and TNF-α via downregulating NF-κB signaling pathway in vitro. Furthermore, compound 6 reduced the mortality, murine sepsis score, the serum TNF-α level and organic damage in a mouse model of sepsis. These findings indicated that compound 6 possessed the potent anti-inflammatory activity and had the potential as a promising drug candidate for sepsis therapy.

Abstract:Phytochemical investigation of the roots of LIGULARIA VEITCHIANA has resulted in the characterization of two new eremophilanolides together with beta-sitosterol, stigmasterol, taraxol, and liguhodgsonal whose structure was further confirmed by the (13)C-NMR techniques. The structures of the new constituents were elucidated by spectroscopic methods and shown to be 1beta, 10beta-epoxy-6beta(2'-hydroxymethylacryloyloxy)-8beta-hydroxy-eremophila-7(11)-en-12,8alpha-olide ( 1) and 1beta, 10beta-epoxy-6beta-(2'-hydroxymethylacryloyloxy)-8beta-ethoxy-eremophila-7(11)-en-12,8alpha-olide ( 2).

Abstract:A phytochemical investigation of the roots of Ligularia atroviolacea resulted in the isolation of 24 compounds including seven new eremophilanoids named eremophila-3,7(11),8-triene-12,8;14,6alpha-diolide (1), 3beta-(angeloyloxy)eremophil-7(11)-en-12,8beta-olid-14-oic acid (2), 1alpha-chloro-10beta-hydroxy-6beta-(2-methylpropanoyloxy)-9-oxo-7,8-furoeremophilane (3), (10betaH)-8-oxoeremophila-3(4),6(7)-diene-12,14-dioic acid (4), (10alphaH)-8-oxoeremophila-3(4),6(7)-diene-12,14-dioic acid (5), 8beta-[eremophila-3',7'(11')-diene-12',8'alpha;14',6'alpha-diolide]eremophila-3,7(11)-diene-12,8alpha;14,6alpha-diolide (6), and ligulatrovine A (7), eleven known eremophilanoids, 8-18, four steroids, one glucose derivative, and one fatty acid. The structures of these compounds were elucidated by spectroscopic methods including 2D-NMR experiments. The structure of 3 was also established by an X-ray diffraction study. The in vitro cytotoxicity evaluation of selected compounds was performed on seven cultured tumor cell lines, i.e., KB, BEL-7404, A549, HL-60, HeLa, CNE, and P-388D1. The preliminary taxonomy of this species was also discussed, and the possible biogenesis of a dimer possessing a new noreremophilanoid type skeleton, 7, is presented in a preliminary form.

Abstract:Three new terpenoids, pubinernoids A-C (1-3), together with six known sesquiterpenoids, were isolated from the leaves and stems of Schisandra pubescens var. pubinervis. The structures of the new compounds were established on the basis of extensive spectroscopic analyses, including application of 2D-NMR spectroscopic techniques. A plausible formation of the sesquiterpenoid 2 is proposed (Scheme 2), starting from guaianediol (4) as the biogenetic precursor, which was also present in the extract.

Abstract:New eremophilane sesquiterpenes, 6 beta-sarracinoyloxy-1 beta,10 beta-epoxy-furanoeremophilane (1), 6 alpha-angeloyloxy-10 beta H-furanoeremophil-1-one (2), 1 alpha-hydroxy-9-deoxycacalol (3), and 1 beta-hydroxy-11(R,S)-8-oxoeremophil-6,9-dien-12-al (4a+4b), together with five known sesquiterpenes (5-9) were isolated from the roots and rhizomes of Ligularia macrophylla. The structures were elucidated by spectroscopic methods including 2D NMR techniques, and the structure of 1 was confirmed by a single-crystal X-ray diffraction experiment. The compounds were also evaluated for cytotoxic activity against human lung carcinoma (A-549) and human breast adenocarcinoma (MCF-7) and were found to show only very weak cytotoxicity.
