Abstract:A new sesquiterpene, 7α-H-11-hydroxyeremophil-1(10)-en-2-one (1), together with 14 known sesquiterpenoids (2–15), were isolated from the extract of the whole plant of Ligularia fischeri. Among them, compounds 2–6 and 15 were isolated from Ligularia fischeri for the first time. The new compound was characterized by means of spectroscopic methods including 1D and 2D NMR and HR-ESI-MS, and the known ones were established by comparing their NMR data with those of the corresponding compounds in the literature.

Abstract:Phytochemical investigation of the roots of LIGULARIA VEITCHIANA has resulted in the characterization of two new eremophilanolides together with beta-sitosterol, stigmasterol, taraxol, and liguhodgsonal whose structure was further confirmed by the (13)C-NMR techniques. The structures of the new constituents were elucidated by spectroscopic methods and shown to be 1beta, 10beta-epoxy-6beta(2'-hydroxymethylacryloyloxy)-8beta-hydroxy-eremophila-7(11)-en-12,8alpha-olide ( 1) and 1beta, 10beta-epoxy-6beta-(2'-hydroxymethylacryloyloxy)-8beta-ethoxy-eremophila-7(11)-en-12,8alpha-olide ( 2).

Abstract:Two new eremophilane-type sesquiterpenoids eremophil-6-en-11-ol (1) and (7 alpha,9 alpha,10 alpha)-9,10-epoxy-eremophilan-11-ol (2), together with a known eremophilane-type (6 alpha,8 alpha)-6,8-dihydroxyeremophil-7(11)-en-12-oic acid 12,8-lactone (3) were isolated from the rhizomes of Ligularia veitchiana. The structures of 1 and 2 were established by spectral analysis including H-1- and C-13-NMR, HSQC, HMBC, and HR-ESI-MS data. The compounds 1 and 3 were assessed against lung-cancer (A549) and stomach-cancer (BCG823) cell lines by the MTT method. The results showed that I exhibited significant inhibiting activities on the growth of these cancer cells with IC50 values between 1 - 100 mu g/ml, whereas compound 3 had no effect on the same cell lines.

Abstract:An abietane diterpenoid, triptobenzene Y and six sesquiterpene polyol esters, wilforsinines C–H, together with 14 known compounds, have been isolated from the roots of Tripterygium wilfordii. The structures of the compounds were elucidated on the basis of spectroscopic analyses. The quinone reductase (QR) induction assay indicated that two compounds showed moderate QR-inducing activities at concentrations of 25μM and 50μM, respectively.

Abstract:Ligulaverin A I, an eremophilane derivative with a novel carbon skeleton, was isolated from the medicinal plant Ligularia veitchiana, along with four analogues (ligulaverin B 2, ligulaverin C 3, ligulaverin D 4 and ligulaverin E 5). Abstract:New eremophilane sesquiterpenes, 6 beta-sarracinoyloxy-1 beta,10 beta-epoxy-furanoeremophilane (1), 6 alpha-angeloyloxy-10 beta H-furanoeremophil-1-one (2), 1 alpha-hydroxy-9-deoxycacalol (3), and 1 beta-hydroxy-11(R,S)-8-oxoeremophil-6,9-dien-12-al (4a+4b), together with five known sesquiterpenes (5-9) were isolated from the roots and rhizomes of Ligularia macrophylla. The structures were elucidated by spectroscopic methods including 2D NMR techniques, and the structure of 1 was confirmed by a single-crystal X-ray diffraction experiment. The compounds were also evaluated for cytotoxic activity against human lung carcinoma (A-549) and human breast adenocarcinoma (MCF-7) and were found to show only very weak cytotoxicity.
Phytotoxic Terpenoids from Ligularia cymbulifera Roots

Abstract:Ligularia cymbulifera is one of the predominant species in the Hengduan Mountains, China, and has led to a decrease in the amount of forage grass in this area. However, little is known about the mechanism behind its predominance. In this study, two novel eremophilane sesquiterpenes, ligulacymirin A and B (1 and 2), together with seven other known terpenoids (3-9), were isolated from the roots of L. cymbulifera. The structures of 1 and 2 were determined by spectroscopic methods and single-crystal X-ray diffraction. Each compound showed phytotoxic activities against Arabidopsis thaliana, and each was detected and identified in rhizosphere soil by UHPLC-MS. Compound 3 was the most potent phytotoxin, showing remarkable inhibition against both seedling growth (EC50 = 30.33 ± 0.94 μg/mL) and seed germination (EC50 = 155.13 ± 0.52 μg/mL), with an average content in rhizosphere soil of 3.44 μg/g. These results indicate that terpenoids in L. cymbulifera roots might be released as phytotoxins in rhizosphere soil to interfere with neighboring plants.

Abstract:OBJECTIVE To investigate the chemical constituents of the roots and rhizomes of Ligularia macrophylla.METHODS The compounds were isolated through repeated column chromatography on silica gel, polyamide, RP-18 and Sephadex LH-20. And the structures were elucidated on the basis of spectroscopic data (UV, IR, MS, 1H NMR, 13C NMR, DEPT and 2D NMR). RESULTS Four flavonol glycosides and two monoterpene glycosides were obtained and determined as rhamnazin-3-O-β-D-rutinoside ( 1 ), rhamnetin-3-O-β-D-rutinoside ( 2), rutin ( 3), afzelin ( 4), betulalbuside A (5) and 3,7-dimethyloct-1 -en-3,8-diol8-O-β-D- glucopyranoside (6). CONCLUSION All the compounds were isolated from this plant for the first time.
