Abstract:Phytochemical reinvestigation on Ligularia nelumbifolia afforded four novel sinapyl alcohol analogues named nelumols B-E (1-4) and three known sinapyl alcohol derivatives (5-7). Their structures were elucidated by NMR techniques. Total syntheses of cytotoxic geranyloxy sinapyl alcohol (6) and geranyloxy sinapyl aldehyde (7) were carried out via two different paths. The 4-O-benzyl-substituted analogues (20 and 27) as well as the 4-O-(2-methylbutenyl) derivatives (34 and 35) were also synthesized. The cytotoxicities of 6 and 7 were measured using A-549, HL-60, and KB cancer cell lines.

Abstract:The C–3 2-methyl-but-2-enoic acid ester of 2-(1-1-acetoxyisopropenyl)-5-acetyl-3,6-dihydroxy-2,3-dihydro-benzofuran (1) and 6-acetyl-7-hydroxy-2-isopropylidene-benzo[1,4]dioxin-3-one (2), together with nine known compounds (3–11) were isolated from the roots of Ligularia stenocephala. Abstract:Previous phytochemical study of Ligularia duciformis, a special weed in China, which has long been used traditionally in folk medicine for reducing inflammation and for curing apoplexy 1, dealt with the isolation of coniferyl and sinapyl alcohol derivatives. Further investigation of the chemical constituents of this plant afforded two new si napyl alcohol derivatives named as 4-0-[6,7-dihydro-5,6£-dehydro7(9)-dehydrogeranyl]-sinapyl alcohol 1 and 4-0-[5oxogeranyl]-sinapyl alcohol 2. The isolation and structure elucidation of the compounds are discussed here.

Abstract:Phytochemical investigation of the roots of Ligularia dentata has resulted in the isolation of three new bisabolane sesquiterpenes: 1alpha,3beta,10,11 -tetrahydroxy-2alpha,5alpha,8-triangeloyl-oxybisabola-(7)14-en-4-one, 1alpha,3beta,10-trihydroxy-11 -methoxy-2alpha,5alpha,8-triangeloyloxybisabola-7(14)-en-4-one, and lalpha, 3beta,10-trihydroxy-2alpha,5alpha,8-triangeloyloxybisabola-7(14),11(12)-dien-4-one. Their structures were elucidated with spectro-scopic methods and 2D-NMR techniques as well as chemical transformations. In addition, three known phenolic norsesquiterpenes, liguhodgsonal, ligujapone, and 2-hydroxy-platyphyllide have also been obtained.

Abstract:AIM:To study the chemical constituents of Ligularia dictyoneura.METHODS:Various chromatographic techniques were used to separate and purify the compounds. Their spectral data (MS, IR, NMR) were measured for structure elucidation.RESULTS:Four sesquiterpenes were isolated from Ligularia dictyoneura and their structures were identified as ligudicin A (1), ligudicin C (2), ligudicin D (3) and isopetasin (4).CONCLUSION:Compound 1 is a new compound, compound 2 is a new natural product, and compounds 3 and 4 are obtained from Ligularia dictyoneura for the first time.

Their structures were elucidatied as 2-hydroxplatyphyllide (1) , liguhodgsonal ( 2), ligujapone ( 3) , 6 '-linoleic-β-daucosterol (4) ,lupeol(5) ,6,7-dihydroxy-coumarin(6), β-sitosterol ( 7), daucosterol (8), (2S,3S,4K, 12E)-N-[2'-( R)-hydroxydocosanoate]-1,3,4-trihydroxy-2-amino-eicosanoate-12-ene ( 9) , monostearin (10), α-hydroxy-lignoceric acid (11) ,1,3-dipalmitin(12) ,hepta-cosanol(13). The structures of 1-4 were elucidated as 3 beta,4-diacetoxy-8 alpha-(2-methylbutyryloxy)-9 alpha-(4- methylsenecioyloxy)-11 alpha,12-epoxyoplop-10 (14)-ene (1), 3 beta,4-diacetoxy-9 alpha-(4-acetoxy-4-methylsenecioyloxy)-2 beta,8 alpha-di (2-methylbutyryloxy)-11 alpha,12-epoxyoplop-10 (14)-ene (2), 2 alpha-hydroxy-1 beta H,7 alpha H,10 alpha H-guai-4,11 (12)-dien-3-one (3) and 1 alpha,2 beta,3 alpha,6 alpha-tetrahydroxy-p-menthane (4) by spectroscopic methods. 1 and 2 were evaluated for their in vitro cytotoxic activity against cultured SMMC-7721 (human hepatoma), L02 (human hepatocyte), and HL-60 (human promyelocytic leukaemia) cell lines.
