Abstract:Natural fibrillar-like macromolecules find applications in several fields, thanks to their peculiar features, and are considered perfect building blocks for natural and artificial functional materials. Indeed, fibrous proteins (such as collagen or fibroin) are commonly used in scaffold fabrication for biomedical applications, due to the high biophysical similarity with the extracellular matrix (ECM) which stimulates tissue regeneration. In the textile industry, cellulose-based fabrics are widely used in place of cotton and viscose, which both have sustainability issues related to their fabrication. With this in mind, the structural characterization of the materials at molecular scale plays a fundamental role in gaining insight into the fiber assembly process. In this work, we report on three fibers of research interest (i.e., type I collagen, silk fibroin extracted from Bombyx mori, and cellulose) to show the power of wide-angle X-ray scattering to characterize both intra- and intermolecular parameters of fibrous polymers. The latest possibilities offered in the X-ray scattering field allow one to study fibers at solid state or dispersed in solutions as well as to perform quantitative scanning X-ray microscopy of tissues entirely or partially made by fibers.

Abstract:The precisely defined geometry of phenylacetylene dendrimers and macrocycles makes it possible to investigate systematic variations in chemical architecture on the nature of microstructural organization. Here we report on transmission electron microscopy, selected-area electron diffraction and high-resolution electron microscopy studies of crystalline phases of these synthetic materials. Since the molecules are sensitive to electron beam damage, low-dose techniques were used to capture images and diffraction patterns dynamically. The data show increased crystal misorientation with increasing side-group length and molecular complexity. As the size increases, the number of conformations available to the molecule also increases, making it difficult to pack the molecules with fewer defects.

Abstract:Metallic filaments with submicrometer diametere have been fabricated. Standard diffraction techniques with conventional x-ray sources were unsuccessful in identifying the structure of these materials. However, with the use of synchrotron radiation produced on a wiggler beam line, diffraction data were obtained in measurement periods as short as 10 milliseconds. Two cylindrical single crystals of bismuth were studied, each with a diameter of 0.22 0.02 micrometer. The volume of sample illuminated for these measurements was 0.38 cubic micrometer, less than 0.5 femtoliter. The crystals are grown in glass capillaries, and, because bismuth expands on solidification, they are under a residual hoop stress. The crystallographic data indicate the presence of a linear compressive strain of about 2 percent, which is assumed to be the result of a residual stress of about 2 gigapascals.

Abstract:Microfocus X-ray diffraction has been used to analyze interfacial stress transfer in two-phase polymer systems consisting of a highly oriented polymer fiber in an isotropic resin matrix. Synchrotron radiation has been used to obtain high-quality X-ray diffraction patterns from single poly(p-phenylene benzobisoxazole) (PBO) and poly(p-phenylene terephthalamide) (PPTA) fibers in these model two-phase systems. Two different specimen geometries were studied: a fully embedded fiber composite and a microdroplet specimen. A procedure is demonstrated whereby well-defined diffraction patterns can be obtained, using a 5 μm diameter X-ray beam, from individual 12 μm diameter polymer fibers in over 1 mm thickness of resin by subtracting the scattering of the resin matrix from that of the fiber and matrix to obtain a diffraction pattern of the fiber only. Shifts of meridional diffraction peaks of the fibers were determined as a function of stress and were converted into crystal strain. The fiber stress was then determined from calibrations obtained from previous X-ray diffraction studies of fiber deformation. The point-to-point variation of fiber stress was mapped along the fibers in the specimens, and a force-balance approach was used to determine the shear stress at the fiber−matrix interface. This present study was concerned only with an optically transparent polymer matrix, but the possible extension of the technique to opaque matrices and other multiphase polymer systems is discussed.

Abstract:The mechanisms of plastic deformation are investigated using different characterization tools as SEM, TEM or synchrotron‐based X‐ray technique like Laue microdiffraction (μLaue). However, structural information can be limited to the specimen surface (SEM), to extremely thin samples (TEM) or due to depth averaging (μLaue). Until today, a non‐destructive in situ investigation of a dislocation population, and de facto, the determination of the local stress tensor in bulk samples remains challenging. To decompose the depth integrated Laue microdiffraction signals, we used the so‐called "Differential Aperture X‐Ray Microscopy" (DAXM),,allowing the 3D determination of the local structural crystal properties. Using this approach, the local crystallographic phase, orientation, and the elastic strain tensor are obtained with 1μm3 voxel size. In order to accomplish the experiment, a protocol and a new combined in situ mechanical testing rig with a DAXM microscope was created. The experiment was conducted on a severely bent focused ion beam copper single crystal microcantilever (10x10x25μm3). The local deviatoric strain tensor and the local lattice curvature in the deformed sample were analysed in 3D. The advantages and resolution limits of the technique will be discussed in detail. Exploiting synchrotron X-ray tomography for a novel insight into flax-fibre defects ultrastructure

Abstract:Flax fibres are valuable reinforcements for tomorrow's composites. However, defects called kink-bands, which mainly appear on fibres during the extraction and transformation phases, might affect their mechanical properties. Defects induced pores, within the kink-band are investigated in this work. They were morphologically explored using synchrotron phase-contrast X-ray microtomography, a technique that displays a sharp 3D representation of the pores. The study highlights the link between kink-bands and secondary cell wall ultrastructure. Pores are organised concentrically around the lumen, and their low thickness suggest that they are located at the interface between cellulose layers within S2 (G) layer. Moreover, the pores inclination with reference to the lumen axis follows the typical microfibrillar angle changes observed in the literature in the kink-band region. The volumes of the pores were measured, and a local increase in porosity was revealed in zones where defects are most severe along the fibre.

Abstract:Coaxial core/shell electrospun nanofibers consisting of ferroelectric P(VDF-TrFE) and relaxor ferroelectric P(VDF-TrFE-CTFE) are tailor-made with hierarchical structures to modulate their mechanical properties with respect to their constituents. Compared with two single and the other coaxial membranes prepared in the research, the core/shell-TrFE/CTFE membrane shows a more prominent mechanical anisotropy between revolving direction (RD) and cross direction (CD) associated with improved resistance to tensile stress for the crystallite phase stability and good strength-ductility balance. This is due to the better degree of core/shell-TrFE-CTFE nanofiber alignment and the crystalline/amorphous ratio. The coupling between terpolymer P(VDF-TrFE-CTFE) and copolymer P(VDF-TrFE) is responsible for phase stabilization, comparing the core/shell-TrFE/CTFE with the pristine terpolymer. Moreover, an impressive collective deformation mechanism of a two-length scale in the core/shell composite structure is found. We apply in-situ synchrotron X-ray to resolve the two-length scale simultaneously by using the small-angle X-ray scattering to characterize the nanofibers and the wide-angle X-ray diffraction to identify the phase transformations. Our findings may serve as guidelines for the fabrication of the electrospun nanofibers used as membranes-based electroactive polymers.
Space-resolved diffusing wave spectroscopy measurements of the macroscopic deformation and the microscopic dynamics in tensile strain tests

Abstract:We couple a laser-based, space-resolved dynamic light scattering apparatus to a universal testing machine for mechanical extensional tests. We perform simultaneous optical and mechanical measurements on polyether ether ketone, a semi-crystalline polymer widely used in the industry. Due to the high turbidity of the sample, light is multiply scattered by the sample and the diffusing wave spectroscopy (DWS) formalism is used to interpret the data. Space-resolved DWS yields spatial maps of the sample strain and of the microscopic dynamics. An excellent agreement is found between the strain maps thus obtained and those measured by a conventional stereo-correlation bench. The microscopic dynamics reveals both affine motion and plastic rearrangements. Thanks to the extreme sensitivity of DWS to displacements as small as 1 nm, plastic activity and its spatial localization can be detected at an early stage of the sample deformation, making the technique presented here a valuable complement to existing material characterization methods.

Abstract:Electrospinning allows the production of fibrous networks for tissue engineering, drug delivery, and wound healing in health care. It enables the production of constructs with large surface area and a fibrous morphology that closely resembles the extracellular matrix of many tissues. A fibrous structure not only promotes cell attachment and tissue formation but could also lead to very interesting mechanical properties. Poly(3-hydroxybutyrate-co-4-hydroxybutyrate) (P(3HB-co-4HB)) is a biodegradable polyester that exhibits a large (>400%) elongation before failure. In this study, synchrotron X-ray phase contrast imaging was performed during tensile deformation to failure on a non-woven fiber mat of P(3HB-co-4HB) fibers. Significant reorientation of the fibers in the straining direction was observed, followed by localized necking and eventual failure. From an original average fiber diameter of 4.3 µm, a bimodal distribution of fiber diameter (modal diameters of 1.9 and 3.7 µm) formed after tensile deformation. Extensive localized necking (thinning) of fibers between (thicker) fiber–fiber contacts was found to be the cause for non-uniform thinning of the fibers, a phenomenon that is expected but has not been observed in 3D previously. The data presented here have implications not only in tissue regeneration but for fibrous materials in general.

Abstract:Molecular orientations of the crystalline and the amorphous phases are studied in relation to morphology using all instrument constructed for simultaneous kinetic measurement of microscopic infrared (MicIR) dichroism from a predetermined sampling area and macroscopic stress of isotactic polypropylene (W) thin film uniaxially stretched at a constant strain rate at room temperature. The morphology imposes important influences oil profiles of MicIR dichroism as a function of stretching, time and also on a relationship between the orientation functions, f(c) and f(am), of the crystalline and the amorphous phases. Linear relationships between f(c) and f(am) are found for the local areas without giant spherulite inclusions when f(am) <0.3 similar to 0.4, suggesting the harmonious developments of amorphous chain deformation and lamellar ant orientation during the necking propagation. The proportionality constant between and f(c) and f(am) is not universal but is strongly dependent on the morphology of the surrounding area where the local area under examination is located. The linear relationship between f(c) and f(am) is completely invalidated for describing the relationship between the amorphous chain deformation and the lamellar orientation inside the spherulites, which reveals that the deformation mechanism in the spherulite is different from that in the microcrystalline region.
In situ synchrotron X-ray multimodal experiment to study polycrystal plasticity

Abstract:The microstructure of polycrystals is known to govern the performance of structural materials. This drives the need for mechanical characterization methods capable of probing large representative volumes at the grain and sub-grain scales. In this paper, the use of in situ diffraction contrast tomography (DCT) along with far-field 3D X-ray diffraction (ff-3DXRD) at the Psiché beamline of Soleil is presented and applied to study crystal plasticity in commercially pure titanium. A tensile stress rig was modified to comply with the DCT acquisition geometry and used for in situ testing. DCT and ff-3DXRD measurements were carried out during a tensile test of a tomographic Ti specimen up to 1.1% strain. The evolution of the microstructure was analyzed in a central region of interest comprising about 2000 grains. Using the 6DTV algorithm, DCT reconstructions were successfully obtained and allowed the characterization of the evolution of lattice rotation in the entire microstructure. The results are backed up by comparisons with EBSD and DCT maps acquired at ESRF-ID11 that allowed the validation of the orientation field measurements in the bulk. Difficulties at the grain boundaries are highlighted and discussed in line with increasing plastic strain during the tensile test. Finally, a new outlook is provided on the potential of ff-3DXRD to enrich the present dataset with access to average lattice elastic strain data per grain, on the possibility of performing crystal plasticity simulations from DCT reconstructions, and ultimately on comparisons between experiments and simulations at the scale of the grain.
Studies of High Performance Fibers by Atomic Force Microscopy and Molecular Simulation

Abstract:Atomic force microscopy (AFM) images obtained from cleaved surfaces of both as-spun and annealed fibers of poly(p-phenylene terephthalamide) (PPTA) have been interpreted with the aid of results from molecular simulations and wide angle X-ray scattering (WAXS). Angstrom-scale periodicities observed in AFM images of the annealed fiber are in accord with those reported in the literature based on WAXS data. For the as-spun fiber, AFM confirms the presence of at least two polymorphs. Among these, a new lattice structure is observed, which has eluded deconvolution from experimental X-ray data. This new lattice exhibits chain conformations and hydrogen bonding structure which is distinct from that seen in either of the two previously reported crystal modifications of PPTA. Interpretation of the AFM images in this case is greatly facilitated by the independent data from molecular simulations. The new crystal facet observed is fully consistent with a third crystal modification previously suggested by molecular simulation.
Biopolymer Filament Entanglement Softens then Hardens with Shear.

Abstract:It is unsatisfactory that regarding the problem of entangled macromolecules driven out of equilibrium, experimentally based understanding is usually inferred from the ensemble average of polydisperse samples. Here, confronting with single-molecule imaging this common but poorly understood situation, over a wide range of shear rate we use single-molecule fluorescence imaging to track alignment and stretching of entangled aqueous filamentous actin filaments in a homebuilt rheo-microscope. With increasing shear rate, tube "softening" is followed by "hardening." Physically, this means that dynamical localization first weakens from molecular alignment, then strengthens from filament stretching, even for semiflexible biopolymers shorter than their persistence length.

Abstract:We present a detailed electron diffraction study of individual single-walled carbon nanotubes. A novel sample preparation procedure provides well-separated, long and straight individual single-shell nanotubes. Diffraction experiments are carried out at 60kV, below the threshold for knock-on damage in carbon nanotubes. We describe experimental parameters that allow single-tube electron diffraction experiments with widely available thermal emission transmission electron microscopes. Further, we review the simulation of diffraction patterns for these objects.

Abstract:Serial diffraction data collected at the Linac Coherent Light Source from crystalline amyloid fibrils delivered in a liquid jet show that the fibrils are well oriented in the jet. At low fibril concentrations, diffraction patterns are recorded from single fibrils; these patterns are weak and contain only a few reflections. Methods are developed for determining the orientation of patterns in reciprocal space and merging them in three dimensions. This allows the individual structure amplitudes to be calculated, thus overcoming the limitations of orientation and cylindrical averaging in conventional fibre diffraction analysis. The advantages of this technique should allow structural studies of fibrous systems in biology that are inaccessible using existing techniques.

Abstract:This paper experimentally investigates the tensile properties of Kevlar29 filaments and fiber bundles, revealing the size and strain rate effects on the mechanical properties of fiber bundles. The fracture modes and mechanisms of the fibers were analyzed, and viscoelastic constitutive models at the fiber bundle scale and constitutive models for filaments-bundles based on the Weibull distribution were established. The results show that Kevlar29 fiber bundles exhibit significant strain rate effects: as the strain rate increases, tensile strength and modulus increase, while fracture strain and toughness decrease. Under quasi-static loading, the fracture modes of the fibers are mainly fibrillation or shear flow, but as the strain rate increases, the failure modes shift towards brittle fracture. Both constitutive models can accurately predict the tensile properties of fiber bundles at different strain rates. The accuracy and applicability of the filament-to-bundle constitutive model were verified through numerical simulations, demonstrating that the model better describes the size effect of fibers and quantifies the damage to the fiber bundles.

Abstract:Small angle X-ray scattering and diffraction methods can provide information about chromatin structure at the levels of nucleosomes, chromatin fibre and whole nuclei. Making use of the high flux and point focusing achieved on synchrotron radiation cameras it has been possible to carry out systematic static and kinetic investigations of chromatin structure with minimum damage or perturbation on fresh samples. The results provide a strong evidence for a preformed 3-D zigzag structure at low ionic strength which folds instantaneously into the “30 nm filament” upon increasing the ionic strength. The extent of folding at a given ionic strength depends on the nature of the cations and can be influenced by other substances which bind to DNA. Models of chromatin fibre structure are discussed in the context of various experimental results.

Abstract:Using a quantum electrodynamic framework, we calculate the off-resonant scattering of a broad-band X-ray pulse from a sample initially prepared in an arbitrary superposition of electronic states. The signal consists of single-particle (incoherent) and two-particle (coherent) contributions that carry different particle form factors that involve different material transitions. Single-molecule experiments involving incoherent scattering are more influenced by inelastic processes compared to bulk measurements. The conditions under which the technique directly measures charge densities (and can be considered as diffraction) as opposed to correlation functions of the charge-density are specified. The results are illustrated with time- and wavevector-resolved signals from a single amino acid molecule (cysteine) following an impulsive excitation by a stimulated X-ray Raman process resonant with the sulfur K-edge. Our theory and simulations can guide future experimental studies on the structures of nano-particles and proteins.

Photoluminescence Spectral Imaging of Ultralong Single-Walled Carbon Nanotubes: Micromanipulation-Induced Strain, Rupture, and Determination of Handedness

Abstract:We have applied photoluminescence (PL) microscopy with scanning laser excitation wavelength for imaging and characterizing individual, millimeter-long, single-walled carbon nanotubes (SWNTs) grown by chemical-vapor deposition on structured Si/SiO2 substrates. Trenches etched into the substrates allowed a direct comparison of the PL properties of air-suspended (across the trenches) and on-SiO2 segments of the same semiconducting nanotubes. For the on-SiO2 segments, we found an similar to 10-20-fold decrease in PL intensity and redshifts of the emission and excitation transitions by 7-27 meV and 5-24 meV, respectively, compared to air-suspended regions of the same SWNTs. Furthermore, PL imaging was applied to SWNTs fractured by dragging an atomic force microscope tip across on-SiO2 segments. Strong, localized changes in the emission properties were observed. These included the appearance of PL blinking at the fracture site and evidence for residual axial and to a lesser extent torsional strain extending tens of microns away from the fracture site. We also discuss how PL measurements of torsional strain can be used to determine the handedness of a luminescent nanotube.
