Synthesis and Content Determination of Cefepime E-isomer

Abstract:Objective:To study the synthetic method of cefepime E-isomer and using as work standard,its quanti- tative method was developed by HPLC in cefepime.Method:The separation was performed on a Alltima C_(18) column (4.6 mm×150 mm,5 μm),the mobile phase was composed of 0.057% ammonium phosphate monobasic and ace- tonitrile with gradient elution.The gradient condition was 0-25 min,acetonitrile 0%-25 %.The flow rate was 1.0 mL·min~(-1).The absorbance was measured at 254 nm.Result :A good linearity was obtained over the range of 0.2 -4.06 μg·mL~(-1);the regression equation were Y=-1.3×10~4×9.8×10~4X(r=0.9981),the detection limit was 20.5 ng;the average recovery was 99.4%,RSD 1.5%.Conclusion:The method is proved to be suitable for detecting the content of cefepime E-isomer,which offered the advantages of specifity,speed and sensitivity.
