Validation and Comparative In-vitro Dissolution Studies of Cefaclor in Their Powder for Oral Suspension Dosage Forms

Abstract:A novel, simple, accurate, stability-indicating RP-HPLC method and in vitro dissolution study was conducted using the following media: buffer pH 1.2, pH 4.5 and pH 6.8. For determination of Cefaclor (CEF) in powder for oral suspension (POS) dosage forms. RP-HPLC method was performed on the Thermo column Hypersil BDS C18 column (250 mm×4.6 mm, 5 μm particle size, using 5.0 mM sodium 1-pentanesulfonate in a mixture of water: triethylamine: methanol adjusted to pH 2.5 with ortho phosphoric acid (78:10:22 v/v) as the mobile phase at a flow rate of 1.5 mL/min and UV detection at 265 nm, the retention time of Cefaclor 11±0.027 min and the total run time 15 min. This method is validated according to ICH guidelines. Linear relationships were obtained in the ranges of 32.78-262.24 μg/mL for Cefaclor at different USP buffer media with correlation coefficients of 1.0, 0.9997 and 0.9998 respectively. The method was successfully applied to the in vitro dissolution studies for Cefaclor in the Generic product; Ceclor 125 and Ceclor 250 mg / 5 mL POS and hence it is considered equivalent to the Innovator product Alfatil 125 and Alfatil 250 mg / 5 mL POS at different USP buffer media and the two products are considered similar.
