Abstract:An improved synthesis of (2S, 4S)- and (2S, 4R)-2-amino-4-methyldecanoic acids was accomplished using a glutamate derivative as starting material and Evans' asymmetric alkylation as the decisive step. The NMR data of the two diastereomers were measured and compared with those of the natural product. Abstract:Using 2-methylbutenoic acid as material, (S)-(+)-dimethylcyclopropane carboxamide was achieved via esterification, cyclopropanantion, hydrolysis, optical resolution and ammonolysis. 7-chloro-2-oxohepanoic acid was synthesized via two steps: through addition of mono-Grignard reagent of 1-bromo-5-chloro-pentane to diethyl oxalate, the ethyl 7-chloro-2-oxohepanoate was synthesized, then the ester was hydrolyzed to produce 7-chloro-2-oxohepanoic acid.(S)-(+)-dimethylcyclopropane carboxamide, 7-chloro-2-oxohepanoic acid and L-cysteine hydrochloride reacted through two steps to produce cilastatin. The yields of (S)-(+)-dimethylcyclopropane carboxamide, 7-chloro-2-oxohepanoic acid and cilastatin were 7.3%, 46.2% and 18.3%. The new synthesis route of cilastatin has the advantages of simple technics and mild react condition, which is easy for industrial production.

Abstract:A new, practical and cost-effective route for scalable synthesis of 2-[(4-dodecyloxyphenyl)sulfonyl] butanoic acid, a key intermediate of a new cyan dye-forming coupler containing a sulfone group, was developed by adopting phenol as the starting material. The synthesis was accomplished in five steps with etherification, chlorosulfonation, reduction, nucleophilic reaction by C-S coupling and hydrolyzation. An important objective of the new synthetic route was the synthesis of 2-[4-(dodecyloxyphenyl)sulfonyl]butanoate. Overall yield obtained at optimized conditions increased to 66 %. The synthetic strategy was proven to be a process enabling rapid delivery of the target product with high purity.
Synthesis and Chiral Resolution of 2,2- Dimethylcyclopropane Carboxylic Acid

Abstract:S)-(+)-2,2-dimethylcyclopropane carboxylic acid is the key intermediate of Cilastatin (an excellent inhibitor of dehydropeptidase-I). Using 2-methylbutenoic acid as raw material, 2,2-dimethylcyclopropane carboxylic acid was achieved via esterification, cyclopropanantion and hydrolysis, the yield is 44.1%. Specially, using zinc powder/copper (I) chloride-acetyl chloride as catalysts, the cyclopropanation proceedes easily with dibromomethane. Then 2,2-dimethylcyclopropane carboxylic acid is converted to acid chloride and reacted with L-carnitine oxalate (the chiral reagent for resolution). After fractional crystallization and hydrolysis, (S)-(+)-2,2-dimethylcyclopropane carboxylic acid was prepared with 16.7% yield. Thus a new synthesis route of 2,2-dimethylcyclopropane carboxylic acid was designed and its chiral resolution procedure was improved, this new route and improved procedure are clean, convenient to operate and apt for industrialization with lower cost.

Abstract:An insecticidal compound 6β-propoxycelangulin V was synthesized from celangulin V and 1-propanol through anhydrous iodination and Williamson reactions.The purpose of the study was to develop the microsynthetic method of 6β-propoxycelangulin V.The powdered root bark of Chinese bittersweet(Celastrus angulatus Max.)was extracted by benzene under reflux,followed by purification by macroporous-resin and silica-gel chromatography.The desired product was purified by RP-HPLC and identified by HRMS,1H NMR and 13C NMR.The total yield was about 17%,and the purity was higher than 98%.This method could be applied in radiosythesis of 6β-([1-14C]propoxy)celangulin V.

Abstract:Cilastatin is an important inhibitor of dehydropeptidase-I.One of its intermediate,7-chloro-2-oxohepanoic acid,was synthesized via two steps: through addition of mono-Grignard reagent of 1-bromo-5-chloro-pentane to diethyl oxalate,the ethyl 7-chloro-2-oxohepanoate was synthesized with 57% yield;then the ester was hydrolyzed to produce 7-chloro-2-oxohepanoic acid with 81% yield.The chemical structure of ethyl 7-chloro-2-oxohepanoate was approved by MS.

Abstract:A new strategy for enantioselective assembly of the trisubstituted tetrahydrofuran ring has been established for synthesis of the C1-C12 acid fragment of amphidinolide T series marine macrolides. The key steps involve the SmI2-mediated highly enantioselective reductive coupling of an aldehyde with the (1S,2R)-N-methylephedrine-derived crotonate to form the cis-3,4- disubstituted gamma-butyrolactone and the subsequent BF3-mediated 1,3-anti-selective allylation of the five-membered-ring oxocarbenium ion with allyltrimethylsilane. The desired C1-C12 acid fragment was obtained in >25% overall yield via a 9-step sequence. (C) 2009 Elsevier Ltd. All rights reserved.

Abstract:A general and efficient method for the synthesis of azabicyclo[4,3,0]nonanone and azabicyclo[5,3,0]clecanone amino acid derivatives was developed with the palladium-catalyzed coupling of intermediates 9 and 10 with BocNH(2) and Boc(2)NH and the following reduction of the C-C double bond by hydrogenation as the key steps. The exploration of the synthesis of C-6-substituted dipeptide mimetics from 9 and 10 using Suzuki coupling as the key reaction has also been performed.

The oxygenated chiral quaternary carbon was assembled by asymmetric dihydroxylation in high enantioselectivity and the tetrahydrofuran ring was constructed by an acid-catalyzed 5-endo ring-opening cyclization of the epoxide possessing a vinyl moiety.

Abstract:A highly stereocontrolled synthesis of the C18–C28 ketone fragment of the 16-membered plecomacrolide micromonospolide B has been accomplished. The C21–C23 syn–anti stereotriad is secured by the anti-selective aldol condensation of the ephedrine-derived chiral propionate with (E,E)-hexa-2,4-dienal and Sharpless asymmetric allylic epoxidation–regioselective reductive epoxide ring opening, respectively. The overall yield of this 14-step sequence is 18.4% and the target C18–C28 ketone was obtained in enantiomerically pure form.

Abstract:Aim To synthesize protected aminoalkyl sulfmyl dilactones which were useful as the synthetic intermediates or the Cterminal pharmacophores of potential peptidomimetic proteasome inhibitors. Methods Organic reactions such as reduction, oxidation, olefination, and dihydroxylation were used. Results A convenient synthetic procedure to afford a series of aminoalkyl sulfinyl dilactones was presented, which would be useful in the synthesis of five- or six-member sulfinyl dilactones. Conclusion Four aminoalkyl sulfinyl dilactones connecting different α-amino acids were synthesized.

Abstract:The use of pre-prepared diaminodiacids has been established as an effective approach for the chemical synthesis of peptide disulfide bond mimics. A technical problem often encountered in the implementation of the diaminodiacids strategy is the use of heavy metal reagents to remove the side-chain protecting groups. In the present work, we reported the development of diaminodiacid that carry 4-(N-[1-(4,4dimethy1-2,6-dioxocyclo-hexylidene)-3-methylbutyl]amino)benzyl (Dmab) and 1-(4,4-dimethyl-2,6dioxocyclohex-1-ylidene)-3-methylbutyl (ivDde) protecting groups. This pair of protecting groups can be readily removed by mild hydrazinolysis during the solid-phase synthesis on resin. We demonstrated the use of Dmab/ivDde protected diaminodiacids in the successful synthesis of a disulfide surrogate of oxytocin. (C) 2017 Published by Elsevier Ltd.

Abstract:The marine peptides have been a fertile source for new bioactive molecules, and many possess potent, activity, across a broad spectrum of targets. Their diversity in both biological activity and in chemical complexity has made these secondary metabolites being the focus of much work in recent years. As a fragment, (R)-2-methyloctanoic acid-7-yne was always found in those nature materials. In this article, (R)-2-methyl-7-octynoic acid was synthesized with pentane-1, 5-diol as the start material. Stereogenic carbon(C-2) was obtained through alkylation induced with Evans' chiral auxiliary, and alkyne 13 was got by use of Ohira-Bestmann reagent. Through 13-step reactions the target molecule was prepared with over yield of 17. 1%, and e. e. beyond 99%.

Abstract:AbstractWe report herein the first total syntheses of four natural antibiotics, vermisporin, PF1052/AB4015‐A, AB4015‐L, AB4015‐B, and one hydrogenated natural product derivative, AB4015‐A2, that all feature a tetramic acid bearing cis‐decalin ring. The construction of the functionalized cis‐decalin ring was achieved by a diastereoselective intramolecular Diels‐Alder (IMDA) reaction, which proceeded via a rare endo‐boat transition state. Through an intramolecular neighboring‐group‐oriented strategy, the sterically hindered epoxy group in vermisporin, PF1052/AB4015‐A and AB4015‐L was installed efficiently. A one‐pot aminolysis/Dieckmann condensation cascade using l‐amino acid derivatives afforded the desired tetramic acid structure. The total synthesis led to the unambiguous verification of the absolute configuration of these natural products.
