Abstract:Started from 5-hydroxy-2-pentanone, (R) (E)-3, 7-dimethyl-2-octene-1, 8-dioic acid, callosobruchusic acid, was synthesized via five steps with D-(-)-camphor sultam as the chiral auxiliary. It was of good optical purity and yield.

Abstract:A parallel process for the new synthesis of 2,7-dimethyl-2,4,6-octatriene-1,8-dial was investigated.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-butenal was obtained by a process involving aldehyde protection,ester exchange and catalytic oxidation from 4-acetoxy-2-methyl-2-butenal.A new catalytic oxidation process was proposed to get 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal by oxidating 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol with copper(Ⅰ) chloride、2,2,6,6-tetramethyl-1-piperidinyloxy and oxygen.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol was chlorizated with SOCl_2 in DMF to get 2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane.2,7-Dimethyl-2,4,6-octatriene-1,8-dial was acquired in an overall yield of 48% by one-pot synthesis method from 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal,2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane and triethylphosphite.

Abstract:An insecticidal compound 6β-propoxycelangulin V was synthesized from celangulin V and 1-propanol through anhydrous iodination and Williamson reactions.The purpose of the study was to develop the microsynthetic method of 6β-propoxycelangulin V.The powdered root bark of Chinese bittersweet(Celastrus angulatus Max.)was extracted by benzene under reflux,followed by purification by macroporous-resin and silica-gel chromatography.The desired product was purified by RP-HPLC and identified by HRMS,1H NMR and 13C NMR.The total yield was about 17%,and the purity was higher than 98%.This method could be applied in radiosythesis of 6β-([1-14C]propoxy)celangulin V.

Abstract:Methyl 3-oxo-2,6,6-trimethylcyclohex-1-ene-1-carboxylate was synthesised over eight steps from the starting material of citral instead of α-ionone. KMnO4, HC(OEt)3 and O2/TEMPO/CuCl were substituted for NaIO4, CH3I and pyridinium chlorochromate, respectively. Every step was simplified and gave a 48% overall yield. The procedure is suitable for large scale production.

Abstract:The sex pheromone of the banded cucumber beetle was synthesized using Evans asymmetric alkylation, ring‐opening reaction of (R)‐2‐methyloxirane, SN2 alkylation of secondary tosylate, and coupling of chiral tosylate with Grignard reagent as central strategies. Diabrotica balteata LeConte is one of the most important polyphagous agricultural pests. The sex pheromone of this pest was synthesized using Evans asymmetric alkylation, ring‐opening reaction of (R)‐2‐methyloxirane, SN2 alkylation of secondary tosylate, and coupling of chiral tosylate with Grignard reagent as central strategies. The sex pheromone prepared herein would be useful to control D. balteata.

Abstract:The marine peptides have been a fertile source for new bioactive molecules, and many possess potent, activity, across a broad spectrum of targets. Their diversity in both biological activity and in chemical complexity has made these secondary metabolites being the focus of much work in recent years. As a fragment, (R)-2-methyloctanoic acid-7-yne was always found in those nature materials. In this article, (R)-2-methyl-7-octynoic acid was synthesized with pentane-1, 5-diol as the start material. Stereogenic carbon(C-2) was obtained through alkylation induced with Evans' chiral auxiliary, and alkyne 13 was got by use of Ohira-Bestmann reagent. Through 13-step reactions the target molecule was prepared with over yield of 17. 1%, and e. e. beyond 99%.

Abstract:Synthesis of the C7-C23 fragment and its 18R, 19S-diastereomer of iriomoteolide-1a has been accomplished from the C7-C12 allyl bromide, the C13-C16 vinyl iodide, and the C17-C23 alkyl iodide fragments. These fragments were assembled first by the B-alkyl Suzuki-Miyaura cross-coupling to give the C13-C23 intermediate. The latter, after being transformed into the C13 aldehyde, was coupled to the C7-C12 allyl bromide in the presence of indium powder in THF-H(2)O (1: 1) at 70 degrees C to the fully functionalized C7-C23 fragment with orthogonal protecting groups at C19 (PMB ether), and C9, C14, and C22 (TBS, TES, and TBS ethers, respectively). Formation of the characteristic six-membered C9/C13-hemiacetal ring has been demonstrated after global desilylation using pyridine-buffered HF.

Abstract:A concise synthesis of the sex pheromones of elm spanworm as well as painted apple moth has been achieved. The key steps were the alkylation of acetylide ion, Sharpless asymmetric epoxidation and Brown's P2-Ni reduction. This approach provided the sex pheromone of the elm spanworm (1) in 31% total yield and those of the painted apple moth (2, 3) in 26% and 32% total yields. The ee values of three final products were up to 99%. The synthesized pheromones hold promising potential for use in the management and control of these pests.

Abstract:A tunable four-module coupling approach has been established for assembling the 20-membered macrolactone core related to the proposed structure of iriomoteolide-1a by using ring-closing metathesis as the key step. Two C1-C6 (2E)-diene acid fragments with (4R,5S)- and (4S,5R)-stereogenic centers, respectively, were synthesized via anti-selective aldol reaction and E-selective conjugate addition of Me2CuLi with an alkynoic ester in the presence of TMSCl. The ring-closing metathesis reaction was carried out in the presence of 10 mol% Grubbs second-generation initiator at room temperature to give exclusively the (6E)-cycloalkenes. Our four-module coupling strategy enables efficient synthesis of two diastereomers of the proposed iriomoteolide-1a with opposite chirality at C4 and C5.

Abstract:An efficient synthesis of the C3-C12 aldehyde fragment of 24-demethylbafilomycin C 1 was accomplished for assembling the 16-membered plecomacrolide skeleton according to a 1,3-diene-ene ring-closing metathesis (RCM) strategy. A boron-mediated ANTI-selective aldol condensation of Abiko’s chiral propionate was used to secure the C6 and C7 stereogenic centers while the C8 chirality was introduced from a chiral building block. The dithiane alkylation and the methyl ketone Horner-Wittig olefination using allyldiphenylphosphine oxide were employed for construction of the requisite ( E)-1,3-diene subunit.

Abstract:A new strategy for enantioselective assembly of the trisubstituted tetrahydrofuran ring has been established for synthesis of the C1-C12 acid fragment of amphidinolide T series marine macrolides. The key steps involve the SmI2-mediated highly enantioselective reductive coupling of an aldehyde with the (1S,2R)-N-methylephedrine-derived crotonate to form the cis-3,4- disubstituted gamma-butyrolactone and the subsequent BF3-mediated 1,3-anti-selective allylation of the five-membered-ring oxocarbenium ion with allyltrimethylsilane. Abstract:An alternative synthesis of (Z)-3-dodecen-1-y1 (E)-2-butenoate without use of carcinogenic ethylene oxide and HMPA is described. By coupling of the tetrahydropyranyl (THP) ether of 3-butyn-1-ol with 1-bromooctane with sodamide in liquid ammonia, 12-(2-tetrahydropyranyloxy)-9-dodecyne was obtained; subsequent hydrolysis and semihydrogenation afforded (Z)-3-dodecen-1-ol. The alcohol was then reacted with crotonyl chloride to give the desired crotonate with a total yield of 29.8%. Males of sweet-potato weevil,Cylas formicarius were strongly attracted to the synthetic sex pheromone. The attraction from the dispenser of polyethylene tube was better than the attraction from the dispenser of rubber septum in the field.

Abstract:The paulownia bagworm, Clania variegata Snell, is an economically important pest of agriculture and forests. The sex pheromone of this pest and its stereoisomers were synthesized, and two of the stereoisomers were prepared for the first time. Our strategy was efficient and mainly included the ring-opening reaction of (S)-2-methyloxirane, the coupling of chiral sulfonate, the oxidative cleavage of olefin, and Yamaguchi esterification. Moreover, the overall yields of our synthesis were 23–29%, with eight steps in the longest route.

Abstract:Abstract Lyonetia clerkella Linnaeus is one of notorious pests in orchards. A new and efficient asymmetric synthesis of the sex pheromone of this pest has been developed. The central to this approach involved DMAPO/Boc2O-mediated N-acylation, Evans’ chiral auxiliaries and the coupling of chiral tosylate with n-propyl magnesium bromide. Furthermore, we have demonstrated the scalability of the new acylation.

Abstract:2-Azabicyclo[3.1.0]hexane-3-carboxylic acids were obtained via amino protection, 4-dimethylaminopyridine (DMAP) catalytic cyclization, reduction-dehydration into alkene, asymmetric Simmons-Smith reaction and hydrolysis reaction from glutamic acid. The intermediates and target products were characterized by 1H NMR and 13C NMR. (1R,3S,5R)-2-(tert-Butoxycarbonyl)-2-azabicyclo[3.1.0]hexane-3-carboxylic acid and (1S,3S,5S)-2-(tert-butoxycarbonyl)-2-azabicyclo-[3.1.0]hexane-3-carboxylic acid were analyzed by X-ray diffraction as well. The process conditions about DMAP catalytic cyclization reaction, Simmons-Smith reaction and hydrolyzation were discussed. The results show that the yield of (R)-1,2-di-tert-butyl 5-oxopyrrolidine- 1,2-dicarboxyate was 82% at the ratio of n(DMAP)∶n(di-tert-butyl dicarbonate)∶n(pyridine)=0.40∶4.0∶1.0. The ratio of cis/trans-form varied with the reaction time in Simmons-Smith reaction, which gave a best ratio of 6∶1 at 19.5 h. The total yield is 30%, and the de value is 72%.

Abstract:As a result of its unique fragrance and wider role in traditional medicine, agarwood produced in Aquilaria spp. and certain other trees has been harvested to near extinction as a natural phenomenon. Artificially induced agarwood production in Aquilaria plantations has sated some of the demand although the product quality is variable. Synthetic chemistry may have a role to play in providing sustainable routes to many of the fragrant components identified in agarwood and its smoke when burnt as incense. In this work, we report efforts towards a total synthesis of the guaiane sesquiterpene α-bulnesene, which is found, along with its more fragrant oxidised derivatives, in agarwood. Following the ring-expansion of (R)-carvone using reported procedures, α-butenylation gave a substrate for samarium diiodide mediated reductive cyclisation, the two butenyl epimers of the substrate each leading to a single bicyclic alcohol (24 and 25). Overall homoconjugate hydride reduction of one of these alcohols was achieved by Lewis acid-mediated ionisation and then hydride transfer from triethylsilane to complete an overall seven-step synthesis of 5-epi-α-bulnesene. This new synthesis paves the way for short routes to both α-bulnesene enantiomers and a study of their aerial and enzymatic oxidation products.

Abstract:A synthesis of chiral dimethylcyclopropane derivatives, useful for the synthesis of casbane, lathyrane, and ingenane-type diterpenoids, from easily available (+)-3-carene is described. The silyl enol ether (8) was derived from (-)-cis-4-caranone (7b), which was obtained from (+)-3-carene (6). Ozonolysis of 8 gave 9a and 10. The right half segment (11b) for a synthesis of crotonitenone (3) was formed from 9a in three steps. The epoxide (12) was isomerized to a mixture of the allylic alcohols (13a) and (14a), which was transformed to the ketone (20) in five steps. Methylation of 20 followed by phosphorylation and reduction gave (+)-cis-4-caranone (23) in about 40% overall yield from 12. The methylester (+)-(32b) and its enantiomer (-)-(34b) were synthesized from (+)-6. Ozonolysis of a mixture of the silyl enol ether (30) and (31) followed by methylation with diazomethane gave 32a, which was hydrolyzed to give the desired (+)-32b. The enantiomer (-)-34b was derived from 32a in five steps.

Abstract:The Wittig reaction was widely used in the synthesis of natural products particularly in the area of insect pheromore synthesis. But unfortunately when a moderated triphenylphosphonium ylid is used, the resultant compound is a impractical E, Z, mixture. For instance, the title compound was once synthesized by the Wittig between butenylidene triphenylphosphorane and 8-oxo-octanoate, followed by hydrolysis and reduction. The product was a mixture containing 75% 8E,10E, isomer and 25% 8Z,10E isomer. Recently we have reported that a simple change in phosphorus substituents from Ph3P=CHR to Ph2(RCH2)P=CHR(R′=alkenyl or H) dramatically increases the proportion of Eolefin formed from moderated ylides(R=alkenyl) and aliphatic aldehydes. According to this fact, here we report a stereoselective synthesis of E,E,-8,10-dodecadien-1-ol from butenylidene methyl diphenylphosphorane and 8-oxo-octanoate in the absence of lithium salt(Schemel). After hydrolysis and reduction, the title compound containing 94.7% 8E, 10E isomer was obtained. This approach affords a convenient synthesis for insect pheromones bearing a E,E,conjugated diene moiety. This experimental fact once again illustrates the stereochemistry of allylic diphenylphosphonium ylid is quite different from the corresponding triphenylphosphonium one.
