Abstract:In this paper, we present a heterogeneous integration platform for bio-sensing applications, which seamlessly integrates low-power silicon-based circuits with cost-effective printed electronics. A prototype of wearable Bio-Sensing Node is fabricated to investigate the suitability of this integration approach. A 1.5 ×3.0 mm2 customized mixed-signal system-on-chip (SoC) with the size of is utilized to amplify, digitize, buffer, and transmit the sensed bio-signals. Inkjet printing technology is employed to print nano-particle silver ink on a flexible substrate to fabricate chip-on-ίex, electrodes as well as interconnections. This additive and digital fabrication technology enables fast prototype of the customized electrode pattern. Its high accuracy and fine resolution features allow the direct integration of the bare die (the pad size of 65 μm and pitch size of 90 μm) on the flexible substrate, which significantly miniaturizes the wearable system. The optimal size and layout of printed electrodes are investigated through the in vivo test for electrocardiogram recording applications. The total size of the implemented Bio-Sensing Node is 4.5 ×2.5 μm, which is comparable with a commercial electrode. This inkjet printed heterogeneous integration approach offers a promising solution for the next-generation cost-effective personalized wearable healthcare monitoring devices.

Abstract:This paper presents a hybrid wearable bio-sensing system, which combines traditional small-area low-power and high-performance System-on-Chip (SoC), flexible paper substrate and cost-effective Printed Electronics. Differential bio-signals are measured, digitized, stored and transmitted by the SoC. The total area of the chip is 1.5 × 3.0 mm2. This enables the miniaturization of the wearable system. The electrodes and interconnects are inkjet printed on paper substrate and the performance is verified in in-vivo tests. The quality of electrocardiogram signal sensed by printed electrodes is comparable with commercial electrodes, with noise level slightly increased. The paper-based inkjet printed system is flexible, light and thin, which makes the final system comfortable for end-users. The hybrid bio-sensing system offers a potential solution to the next generation wearable healthcare technology.

Abstract:This paper presents a concept of a full-printing methodology aiming at convenient and fast fabrication of microfluidic devices. For the first time, we achieved a microfluidic biochemical sensor with all functional structures fabricated by inkjet printing, including electrodes, immobilized enzymes, microfluidic components and packaging. With the cost-effective and rapid process, this method provides the possibility of quick model validation of a novel lab-on-chip system. In this study, a three-electrode electrochemical system was integrated successfully with glucose oxidase immobilization gel and sealed in an ice channel, forming a disposable microfluidic sensor for glucose detection. This fully-printed chip was characterized and showed good sensitivity and a linear section at a low-level concentration of glucose (0-10 mM). With the aid of automatic equipment, the fully-printed sensor can be massively produced with low cost.

Abstract:In recent years, 3D integrated microfluidic systems have become increasingly more popular because of their ability to incorporate multifunctional components, including porous membranes and biological scaffolds. Because of limitations in resolution, fabrication efficiency and materials, it is hard to develop complex integrated microfluidic systems with low cost and high efficiency. In this paper, we present a novel method that utilizes modular structure-based design, which could greatly reduce the time and cost for customization of complete integrated chips, compared to traditional techniques. By printing sacrificial patterns on the substrate using the 3D printing approach and subsequently covering them with PDMS prepolymer, PDMS slices with modular structures were obtained, each with specific functions. By combining different PDMS slices with specific modular structures and other functional components, such as membranes and scaffolds, the conceptual design was efficiently converted into complete integrated microfluidic chips. As proof-of-concept, customized 3D microfluidic chips were generated and successfully used for cell culture and biological analysis. Furthermore, the flexible combination with biofabrication of hydrogel beads was also presented, revealing the potential use of this technique in the fabrication of organ-on-a-chip.
Developing Microfluidic Sensing Devices Using 3D Printing

Abstract:This short perspective assesses the present landscape for desktop 3D printing to design and fabricate sensors, in particular, those associated with microfluidics and multiplexing. Lots of advanced devices have already been reported, and this article briefly surveys interesting achievements. Microfluidics can be designed and optimized faster and more easily on low cost desktop 3D printers that with competing methods. Rapid prototyping leads directly to a final (marketable) product fabricated on the same 3D printer. While resolution is not as good as lithographic approaches, very often channel and feature resolution on the order of 100 μm obtainable with SLA 3D printers is perfectly suitable for the desired sensing device. Two examples from our team's research are used to illustrate how using a 3D printer along with simple automation can reduce a complex microfluidic sensing procedure to a much simpler automated one. Future possibilities for sensor technology are discussed.

Abstract:Limitation of 3D construction ability, complex preparation process, and developing customer demands have promoted efforts to find low-cost, rapid prototyping, and operation simple methods to produce novel functional devices in the near future. Amongst various techniques, 3D-printed technology is a promising candidate for the fabrication of biosensors and biomedical detection devices with a wide variety of potential applications. This review offers four important 3D printing techniques towards biosensors and biomedical detection devices and its applications. The principle and printing process of 3D-printed technologies will be generalized and the printing performance of many 3D printer will be compared. Despite the limitation of 3D-printed resolution, these 3D-printed technologies have already shown promising applications in many biosensors and biomedical detection devices such as 3D-printed microfluidic devices, 3D-printed optical devices, 3D-printed electrochemical devices and 3D-printed integrated devices. Some of the most representative examples will also be discussed here, demonstrating that the 3D-printed technology can rationally design biosensor and biomedical detection devices and achieve important applications in microfluidic, optical, electrochemical and integrated devices.

Abstract:This paper gives details on the use of 3D smart printing technology to fabricate microfluidic chips for integration into biosensors for the detection and diagnosis of diseases. Additive manufacturing, also known as 3D printing, is a process used to create complex, three-dimensional objects by adding layer upon layer of material until the desired shape is formed. Microfluidic chips are used to manipulate fluids through separation and mixing. Conventional microfluidic chip fabrication methods are expensive, require much experience to operate, and are time consuming, while 3D printing offers a solution to these challenges. The 3D printing technique prints models designed using a computer-aided design software such as Autodesk Fusion 360. In this work the authors show example microfluidic chips which were printed using a 3D printer, these include an X-channel chip, Y-channel chip and a lateral flow chip which can all be integrated with biosensing setups.

Abstract:As a kind of facile tool, microfluidic paper-based analytical devices (μPADs) have been widely used in analytical and biomedical fields. However, because we lack the ability to control the continuous perfusion of these devices, they are not generally used in fields that require continuous flow, especially biofluidics fields such as cell culturing, drug screening, and organs on chips. In this study, we designed a novel, low-cost, and compact platform that can be used to control the continuous perfusion of μPADs. As most of the parts of this platform can be created using a three-dimensional (3D) desktop printer, our platform can be easily duplicated by other researchers. We demonstrate that with our system, μPADs can be promising paper-based biofluidic platforms for cell culturing and drug screening.
High-resolution low-cost LCD 3D printing for microfluidics and organ-on-a-chip devices

Abstract:The fabrication of microfluidic devices has progressed from cleanroom manufacturing to replica molding in polymers, and more recently to direct manufacturing by subtractive (e.g., laser machining) and additive (e.g., 3D printing) techniques, notably digital light processing (DLP) photopolymerization. However, many methods require technical expertise and DLP 3D printers remain expensive at a cost ∼15-30 K USD with ∼8 M pixels that are 25-40 μm in size. Here, we introduce (i) the use of low-cost (∼150-600 USD) liquid crystal display (LCD) photopolymerization 3D printing with ∼8-58 M pixels that are 18-35 μm in size for direct microfluidic device fabrication, and (ii) a poly(ethylene glycol) diacrylate-based ink developed for LCD 3D printing (PLInk). We optimized PLInk for high resolution, fast 3D printing and biocompatibility while considering the illumination inhomogeneity and low power density of LCD 3D printers. We made lateral features as small as 75 μm, 22 μm-thick embedded membranes, and circular channels with a 110 μm radius. We 3D printed microfluidic devices previously manufactured by other methods, including an embedded 3D micromixer, a membrane microvalve, and an autonomous capillaric circuit (CC) deployed for interferon-γ detection with excellent performance (limit of detection: 12 pg mL-1, CV: 6.8%). We made PLInk-based organ-on-a-chip devices in 384-well plate format and produced 3420 individual devices within an 8 h print run. We used the devices to co-culture two spheroids separated by a vascular barrier over 5 days and observed endothelial sprouting, cellular reorganization, and migration. LCD 3D printing together with tailored inks pave the way for democratizing access to high-resolution manufacturing of ready-to-use microfluidic and organ-on-a-chip devices by anyone, anywhere.

Abstract:The connection of microfluidic devices to the outer world by tubes and wires is an underestimated issue. We present methods based on 3D printing to realize microfluidic chip holders with reliable fluidic and electric connections. The chip holders are constructed by microstereolithography, an additive manufacturing technique with sub-millimeter resolution. The fluidic sealing between the chip and holder is achieved by placing O-rings, partly integrated into the 3D-printed structure. The electric connection of bonding pads located on microfluidic chips is realized by spring-probes fitted within the printed holder. Because there is no gluing or wire bonding necessary, it is easy to change the chip in the measurement setup. The spring probes and O-rings are aligned automatically because of their fixed position within the holder. In the case of bioanalysis applications such as cells, a limitation of 3D-printed objects is the leakage of cytotoxic residues from the printing material, cured resin. This was solved by coating the 3D-printed structures with parylene-C. The combination of silicon/glass microfluidic chips fabricated with highly-reliable clean-room technology and 3D-printed chip holders for the chip-to-world connection is a promising solution for applications where biocompatibility, optical transparency and accurate sample handling must be assured. 3D printing technology for such applications will eventually arise, enabling the fabrication of complete microfluidic devices.

Abstract:Over the last two decades, microfluidic devices have rapidly emerged as revolutionary platforms for research and medicine. Various kinds of materials and fabrication strategies have been used to manufacture microfluidic devices; however, most of these strategies face challenges including complexity, time consumption, and cost in recreating delicate and intricate structural organizations. Herein, a silicone composite (Si-Co) was developed and employed as a functional support bath in which a sacrificial ink was 3D printed using embedded printing, enabling freeform fabrication of complex-shaped microfluidic devices. Si-Co was a soft material that had high optical transparency and tunable mechanical properties and yield stress with self-recovering ability. Sacrificial ink filaments formed inside the Si-Co support bath were found to be stable and circular with a wide range of resolution reaching up to 50 μm in channel size. The strategy was exemplified by fabricating microfluidic devices for generation of stable microgels of various sizes. To confirm biocompatibility of Si-Co, induced-pluripotent stem cell-derived endothelial cells were lined up inside microfluidic channels to obtain vascular mimics. Additionally, proof-of-concept spheroid fabrication devices were generated. Overall, the presented strategy allows for facile, rapid, cost-effective, and high-resolution printing and presents potential for the development of microfluidic devices for various applications, such as but not limited to organ-on-a-chip devices, 3D bioprinting and drug testing.

Microfluidic Actuation via 3D-Printed Molds toward Multiplex Biosensing of Cell Apoptosis

Abstract:Multiplexed analysis of biochemical analytes such as proteins, enzymes, and immune products using a microfluidic device has the potential to cut assay time, reduce sample volume, realize high-throughput, and decrease experimental error without compromising sensitivity. Despite these huge benefits, the need for expensive specialized equipment and the complex photolithography fabrication process for the multiplexed devices have, to date, prevented widespread adoption of microfluidic systems. Here, we present a simple method to fabricate a new microfluidic-based multiplexed biosensing device by taking advantage of 3D-printing. The device is an integration of normally closed (NC) microfluidic valving units which offer superior operational flexibility by using PDMS membrane (E ∼ 1-2 MPa) and require minimized energy input (1-5 kPa). To systematically engineer the device, we first report on the geometrical and operational analysis of a single 3D-printed valving unit. Based on the characterization, we introduce a full prototype multiplexed chip comprising several microfluidic valves. The prototype offers-for the first time in a 3D-printed microfluidic device-the capability of on-demand performce of both a sequential and a parallel biochemical assay. As a proof of concept, our device has been used to simultaneously measure the apoptotic activity of 5 different members of the caspase protease enzyme family. In summary, the 3D-printed valving system showcased in this study overcomes traditional bottlenecks of microfabrication, enabling a new class of sophisticated liquid manipulation required in performing multiplexed sensing for biochemical assays.
All-printed Multiplexed Electrocatalytic Biosensors with Rationally Designed Nanoparticle Inks

Abstract:Inkjet printing, capable of rapid and template-free fabrication with high resolution and low material waste, is a promising method to construct electrochemical biosensor devices. However, the construction of fully inkjet-printed electrochemical biosensor remains a challenge owing to the lack of appropriate inks, especially the sensing inks of bioactive materials. Herein, we demonstrate a fully inkjet-printed, integrated and multiplexed electrochemical biosensor by combining rationally designed nanoparticle Inks. The stable gold (Au) nanoparticles ink with lower sintering temperature is prepared by using L-cysteine as stabilizer, and it is used to print the interconnects, the counter electrodes, and the working electrodes. The SU-8 ink is used to serve as dielectric layer for the biosensor, whereas the silver electrode is printed on the Au electrode by using commercially silver nanoparticles ink before it is chlorinated to prepare Ag/AgCl reference electrode. Moreover, we synthesize an inkjet-printable and electroactive ink, by the "one-pot method", which is composed of conductive poly 6-aminoindole (PIn-6-NH2) and gold-palladium (Au-Pd) alloy nanoparticle (Au-Pd@PIn-6-NH2) to enhance the sensing performance of gold electrode towards hydrogen peroxide (H2O2). Especially, the amino groups in PIn-6-NH2 canbe further used to immobilizing glucose oxidase (GOx) and lactic acid oxidase (LOx) by glutaraldehyde to prepare printable sensing ink for the detection of glucose and lactate. The fully inkjet-printed electrochemical biosensor enabled by advanced inks can simultaneously detect glucose and lactate with good sensitivity and selectivity, as well as facile and scalable fabrication, showing great promise for metabolic monitoring.

Abstract:Electrochemical sensors provide an affordable and reliable approach towards the detection and monitoring of important biological species ranging from simple ions to complex biomolecules. The ability to miniaturize electrochemical sensors, coupled with their affordability and simple equipment requirements for signal readout, permits the use of these sensors at the point-of-care where analysis using non-invasively obtainable biofluids is receiving growing interest by the research community. This paper describes the design, fabrication, and integration of a 3D printed Mg2+ potentiometric sensor into a 3D printed microfluidic device for the quantification of Mg2+ in low-sample volume biological fluids. The sensor employs a functionalized 3D printable photocurable methacrylate-based ion-selective membrane affixed to a carbon-mesh/epoxy solid-contact transducer for the selective determination of Mg2+ in sweat, saliva and urine. The 3D printed Mg2+ ion-selective electrode (3Dp-Mg2+-ISE) provided a Nernstian response of 27.5 mV per decade with a linear range of 10 mM to 39 μM, covering the normal physiological and clinically relevant levels of Mg2+ in biofluids. 3Dp-Mg2+-ISEs selectively measure Mg2+ over other biologically present cations - sodium, potassium, calcium, ammonium - as well as provide high stability in the analytical signal with a drift of just 13 μV h-1 over 10 hours. Comparison with poly(vinylchloride)-based Mg2+-ISEs showed distinct advantages to the use of 3Dp-Mg2+-ISEs, with respect to stability, resilience towards biofouling and importantly providing a streamlined and rapid approach towards mass production of selective and reliable sensors. The miniaturization capabilities of 3D printing coupled with the benefits of microfluidic analysis (i.e., low sample volumes, minimal reagent consumption, automation of multiple assays, etc.), provides exciting opportunities for the realization of the next-generation of point-of-care diagnostic devices.
3-D printed microfluidics for rapid prototyping and testing of electrochemical, aptamer-based sensor devices under flow conditions

Abstract:We demonstrate the ability to rapidly prototype and fabricate an epoxy-embedded electrode platform and microfluidic device suitable for using electrochemical biosensors under flow conditions. We utilize three-dimensional (3-D) printing to rapidly prototype molds to fabricate epoxy-embedded electrodes in addition to molds for rapid prototyping of PDMS microfluidic components. We characterize the bare gold epoxy-embedded electrodes using ferricyanide as a redox indicator and then characterize the performance of an adenosine triphosphate (ATP) specific electrochemical, aptamer-based (E-AB) sensor. We then incorporate the ATP specific E-AB sensors into the microfluidic device to study and take advantage of the dynamic response this class of sensor offers. We were able to flow varying concentrations of target analyte and monitor the dynamic response of the sensors to the changing concentration. This work demonstrates the ability to rapidly prototype E-AB sensors under flow conditions using 3-D printing which can lead to rapid and affordable point-of-care or fieldable applications where dynamic measurements of concentration, specificity and sensitivity and multiplex detection are necessary.
