Abstract:Nanofluidic systems are attracting a great deal of interest due to their fundamental significance and potential applications in chemistry, biology and physics. However, high fabrication cost, expensive equipments and complicated fabrication process of most current fabrication techniques prevent lots of researchers from entering the nanofluidic field. Here we present a quick, simple and cost-effective method for fabricating two-dimensional (2D) nanochannel in polycarbonate (PC) substrates. Silica nanowires, taper-drawn from commercially available single-mode fiber were used as templates and embedded in the PC substrate by hot embossing. The nanochannels were created after removing the nanowires by hydrofluoric acid (HF) etching. Poly(dimethylsiloxane) (PDMS) was used to seal the nanochannel reversibly. Nanochannels with widths range from 100 to 900 nm and lengths up to several millimeters were obtained. Various nanostructures including integrated micro and nanochannels, nanochannel array, bent nanochannel and cross-shaped nanochannel were fabricated and characterized by fluorescent microscope, scanning electron microscope (SEM) and atomic force microscope (AFM).
Direct Fabrication of Freestanding and Patterned Nanoporous Junctions in a 3D Micro-Nanofluidic Device for Ion-Selective Transport

Abstract:In the field of micro-nanofluidics, a freestanding configuration of a nanoporous junction is highly demanded to increase the design flexibility of the microscale device and the interfacial area between the nanoporous junction and microchannels, thereby improving the functionality and performance. This work first reports direct fabrication and incorporation of a freestanding nanoporous junction in a microfluidic device by performing an electrolyte-assisted electrospinning process to fabricate a freestanding nanofiber membrane and subsequently impregnating the nanofiber membrane with a nanoporous precursor material followed by a solidification process. This process also enables to readily control the geometry of the nanoporous junction depending on its application. By these advantages, vertically stacked 3D micro-nanofluidic devices with complex configurations are easily achieved. To demonstrate the broad applicability of this process in various research fields, a reverse electrodialysis-based energy harvester and an ion concentration polarization-based preconcentrator are produced. The freestanding Nafion-polyvinylidene fluoride nanofiber membrane (F-NPNM) energy harvester generates a high power (59.87 nW) owing to the enlarged interfacial area. Besides, 3D multiplexed and multi-stacked F-NPNM preconcentrators accumulate multiple preconcentrated plugs that can increase the operating sample volume and the degree of freedom of handling. Hence, the proposed process is expected to contribute to numerous research fields related to micro-nanofluidics in the future.
Creating Sub-50 Nm Nanofluidic Junctions in PDMS Microfluidic Chip Via Self-Assembly Process of Colloidal Particles.

Abstract:Polydimethylsiloxane (PDMS) is the prevailing building material to make microfluidic devices due to its ease of molding and bonding as well as its transparency. Due to the softness of the PDMS material, however, it is challenging to use PDMS for building nanochannels. The channels tend to collapse easily during plasma bonding. In this paper, we present an evaporation-driven self-assembly method of silica colloidal nanoparticles to create nanofluidic junctions with sub-50 nm pores between two microchannels. The pore size as well as the surface charge of the nanofluidic junction is tunable simply by changing the colloidal silica bead size and surface functionalization outside of the assembled microfluidic device in a vial before the self-assembly process. Using the self-assembly of nanoparticles with a bead size of 300 nm, 500 nm, and 900 nm, it was possible to fabricate a porous membrane with a pore size of ~45 nm, ~75 nm and ~135 nm, respectively. Under electrical potential, this nanoporous membrane initiated ion concentration polarization (ICP) acting as a cation-selective membrane to concentrate DNA by ~1,700 times within 15 min. This non-lithographic nanofabrication process opens up a new opportunity to build a tunable nanofluidic junction for the study of nanoscale transport processes of ions and molecules inside a PDMS microfluidic chip.

Abstract:Many fabrication technologies have been used to build nano/mesoporous materials/filters with a good size control, but the integration of these systems into a microsystem format has been a challenge. Microfabricated nanofilters suffer from small open volume and low-throughput. In this paper, we developed a novel fabrication strategy for generating massively-parallel, regular vertical nanochannel membranes with a uniform, well-controlled gap size of approximately 50 nm and a depth up to approximately 40 microm, by using only standard semiconductor fabrication techniques. The vertical nanofilter membranes were fabricated into an anisotropic nanofilter array, which demonstrates the ability to integrate nanofilters and micron-sized channels/pores seamlessly. We demonstrated efficient continuous-flow separation of large DNAs and small molecules in a two-dimensional vertical nanochannel array device. These ultra-high-aspect-ratio nanochannels have the advantage of large open volume, enabling high-throughput applications.
Micro-patternable Nanoporous Polymer Integrated with Microstructures for Molecular Filtration.

Abstract:This paper proposes a facile method to fabricate nanoporous microstructures by a photo-patternable SU-8 photoresist, to serve as a molecular filter in microfluidic systems. The fabrication process involves solvent-controlled nanoporous structure formation combined with standard photolithography steps for microstructure fabrication. The self-formed nanoporous morphology embedded inside the microstructure exhibits a sufficient mechanical strength and eliminates complex processes or protocols for integration/assembly of nano- and microstructures. Field emission gun scanning electronic microscopy (FEGSEM) images showed the fabricated nanoporous morphologies with embedded nanogaps of about 6-10 nm. Atomic force microscopy (AFM) images also depicted a clear difference on the degree of porosity between the solvent-controlled SU-8 and the standard resist. Fluorescent dyes, namely Rhodamine-B and Rhodamine-6G, were employed to estimate the diffusivity of the fabricated SU-8 based nanofilter and demonstrated that the Rhodamine based fluorescent molecules can penetrate these nanosized filtration structures. The fabricated nanofilter was capable of providing a molecular weight cut-off range up to 70 kDa, estimated roughly for a molecule with a diameter of 6-10 nm. This simple process provides a novel way to integrate the nanofiltration capability into microstructures while maintaining a sufficient mechanical strength for molecular level filtration in lab-on-chip (LOC) systems.

Abstract:Long microchannels with thin walls, small width, and nanoholes or irregular shaped microgaps, which are similar to capillaries or cancerous vessels, are urgently needed to simulate the physiological activities in human body. However, the fabrication of such channels remains challenging. Here, microchannels with designable holes are manufactured by combining laser printing with line-contact capillary-force assembly. Two microwalls are first printed by femtosecond laser direct-writing, and subsequently driven to collapse into a channel by the capillary force that arises in the evaporation of developer. The channel can remain stable in solvent due to the enhanced Van der Waals' force caused by the line-contact of microwalls. Microchannels with controllable nanoholes and almost arbitrary patterns can be fabricated without any bonding or multistep processes. As-prepared microchannels, with wall thicknesses less than 1 µm, widths less than 3 µm, lengths more than 1 mm, are comparable with human capillaries. In addition, the prepared channels also exhibit the ability to steer the flow of liquid without any external pump.

Abstract:Integrated micro-nanochannel networks in fluidic devices are desirable in a number of applications ranging from self-healing/cooling materials to bioengineering. The conventional micro-manufacturing techniques are capable of either producing microchannel or nanochannel networks for a fluidic application but lack proficiency in the production of an integrated micro-nanochannel network with a smooth transition from micro-to-nano scale dimension. In addition, these techniques possess limitations such as heavy initial investment, sophistication in operation and scale-up capabilities. Therefore, the current paper demonstrates the combination of micro/nanotechnologies to design and develop a biomimetic 3-D integrated micro-nanochannel network in PDMS device for solute exchange. We have used 3-D printer, a scalable technology, to design and manufacture micro-mold having fractal-shaped features. Further, electrospinning was used to deposit nanofibrous network on the fractal mold. Subsequent micro-molding with PDMS was used to obtain fractal-shaped microchannels integrated with embedded nanofibers. Henceforth, solvent etching of nanofibers followed by bonding of thin PDMS membrane generated by spin coating to open end of channels leads to the formation of functional microdevices. These PDMS devices mimic the natural vasculature of a living system, where fractal-shaped microchannels will assist in efficient fluid flow and the site of nanovascular network participates in heat/mass transport operations. Further, dye flow propounds the functionality of such devices. Our study hence proposes a simple and scalable hybrid microtechnolgy to fabricate fluidic devices having multiscale architecture. This will also facilitate the rapid fabrication of microfluidic devices for biomedical, diagnostics, sensors and micro-TAS applications.

Abstract:Poly(dimethylsiloxane) has attracted much attention in soft lithography and has also been preferred as a platform for a photochemical reaction, thanks to its outstanding characteristics including ease of use, nontoxicity, and high optical transmittance. However, the low stiffness of PDMS, an obvious advantage for soft lithography, is often treated as an obstacle in conducting precise handling or maintaining its structural integrity. For these reasons, a Glass-PDMS-Glass structure has emerged as a straightforward alternative. Nevertheless, several challenges are remaining in fabricating Glass-PDMS-Glass structure through the conventional PDMS patterning techniques such as photolithography and etching processes for master mold. The complicated techniques are not suitable for frequent design modifications in research-oriented fields, and fabrication of perforated PDMS is hard to achieve using mold replication. Herein, we utilize the successive laser pyrolysis technique to pattern thin-film PDMS for microfluidic applications. The direct use of thin film at the glass surface prevents the difficulties of thin-film handling. Through the precise control of photothermal pyrolysis phenomena, we provide a facile fabrication process for perforated PDMS microchannels. In the final demonstration, the laminar flow has been successfully created owing to the smooth surface profile. We envision further applications using rapid prototyping of the perforated PDMS microchannel.

Abstract:Biological ion channels can realize high-speed and high-selective ion transport through the protein filter with the sub-1-nanometer channel. Inspired by biological ion channels, various kinds of artificial subnanopores, subnanochannels, and subnanoslits with improved ion selectivity and permeability are recently developed for efficient separation, energy conversion, and biosensing. This review article discusses the advanced fabrication and functionalization methods for constructing subnanofluidic pores, channels, tubes, and slits, which have shown great potential for various applications. Novel fabrication methods for producing subnanofluidics, including top-down techniques such as electron beam etching, ion irradiation, and electrochemical etching, as well as bottom-up approaches starting from advanced microporous frameworks, microporous polymers, lipid bilayer embedded subnanochannels, and stacked 2D materials are well summarized. Meanwhile, the functionalization methods of subnanochannels are discussed based on the introduction of functional groups, which are classified into direct synthesis, covalent bond modifications, and functional molecule fillings. These methods have enabled the construction of subnanochannels with precise control of structure, size, and functionality. The current progress, challenges, and future directions in the field of subnanofluidic are also discussed.

Dry-Release Self-assembling of Microtube Arrays for Catalytic Micropump Application

Abstract:Efficient tiny pumps, mixers and stirrers are essential elements of the microfluidic “factories” on a chip. Bending of strain-engineered nanomembranes into self-assembled round-shape microchannels and wrinkling networks have making the microfluidic integration more promising. For the strain-engineered wrinkles and tubes, common process involves the wet chemical etching of sacrificial layers, which frequently causes the process-induced adhesion and diffusion etching. Here we present a new method to make the predefined stained nanomembranes self-assembled into microtubular pump arrays by a simple thermal induced dry-release process. With a catalytic Pt layer inside, the micropumps can be activated by the catalytic decomposition of peroxide into oxygen microbubbles and water.

Abstract:Micropore arrays have attracted a substantial amount of attention due to their strong capability to separate specific cell types, such as rare tumor cells, from a heterogeneous sample and to perform cell assays on a single cell level. Micropore array filtration has been widely used in rare cell type separation because of its potential for a high sample throughput, which is a key parameter for practical clinical applications. However, most of the present micropore arrays suffer from a low throughput, resulting from a low porosity. Therefore, a robust microfabrication process for high-porosity micropore arrays is urgently demanded. This study investigated four microfabrication processes for micropore array preparation in parallel. The results revealed that the Parylene-C molding technique with a silicon micropillar array as the template is the optimized strategy for the robust preparation of a large-area and high-porosity micropore array, along with a high size controllability. The Parylene-C molding technique is compatible with the traditional micromechanical system (MEMS) process and ready for scale-up manufacture. The prepared Parylene-C micropore array is promising for various applications, such as rare tumor cell separation and cell assays in liquid biopsy for cancer precision medicine.

Abstract:Abstract Fabrication of 3D microfluidic devices is normally quite expensive and tedious. A strategy was established to rapidly and effectively produce multilayer 3D microfluidic chips which are made of two layers of poly(methyl methacrylate) (PMMA) sheets and three layers of double-sided pressure sensitive adhesive (PSA) tapes. The channel structures were cut in each layer by cutting plotter before assembly. The structured channels were covered by a PMMA sheet on top and a PMMA carrier which contained threads to connect with tubing. A large variety of PMMA slides and PSA tapes can easily be designed and cut with the help of a cutting plotter. The microfluidic chip was manually assembled by a simple lamination process.The complete fabrication process from device design concept to working device can be completed in minutes without the need of expensive equipment such as laser, thermal lamination, and cleanroom. This rapid frabrication method was applied for design of a 3D hydrodynamic focusing device for synthesis of gold nanoparticles (AuNPs) as proof-of-concept. The fouling of AuNPs was prevented by means of a sheath flow. Different parameters such as flow rate and concentration of reagents were controlled to achieve AuNPs of various sizes. The sheet-based fabrication method offers a possibility to create complex microfluidic devices in a rapid, cheap and easy way.

Abstract:BACKGROUND Endothelial progenitor cell (EPC) capture stent is designed to promote rapid endothelization and healing and is potentially useful in patients undergoing primary percutaneous coronary intervention (PCI) for acute myocardial infarction (AMI). We studied the intermediate-term efficacy and safety of EPC stent and compared that with sirolimus-eluting bioabsorbable polymer stent (CURA) and bare metal stent (BMS) in AMI patients. METHODOLOGY Patients presenting with AMI who underwent primary PCI with the respective stents between January 2004 and June 2006 were enrolled in the single-center clinical registry. The study end-points were major adverse cardiac events (MACE) and stent thrombosis. RESULTS A total of 366 patients (EPC = 95, CURA = 53, BMS 218) were enrolled. Baseline demographics including age, gender, diabetes, renal impairment, predischarge left ventricular ejection fraction, and creatinine kinase level were comparable among the groups. Procedural success rate was 99.5%. Post-procedural thrombolysis in myocardial infarction (TIMI) 3 flow was achieved in EPC 91.6%, CURA 96.2%, and BMS 88.5% (P = 0.209). At 2 years, the MACE rate was EPC 13.7%, CURA 15.1%, and BMS 19.7% (P = 0.383). Target vessel revascularizations (TVR) were EPC 4.2%, CURA 9.4%, and BMS 6.0% (P = 0.439). Nonfatal myocardial infarctions were EPC 1.1%, CURA 3.8%, and BMS 4.1% (P = 0.364). One patient in the EPC group had acute stent thrombosis. There was no late stent thrombosis in the EPC group. CONCLUSION EPC stent appeared to be safe and had comparable clinical efficacy with a BMS when used in the AMI setting. At 2-year follow-up, the EPC group showed favorable, single-digit TVR rate and stent thrombosis remained a low-event occurrence.
Formation of Nanoparticle-Containing Multilayers in Nanochannels Via Layer-by-Layer Assembly

Abstract:Arrays of silicon nanochannels were used to investigate the layer-by-layer (LbL) assembly of nanoparticle-containing multilayers in confined geometries. LbL assembly of poly(vinylsulfonic acid)/titania (PVS/TiO2) and poly(diallyldimethylammonium chloride)/silica (PDAC/SiO2) resulted in conformal, uniform thickness multi layers throughout the high-aspect-ratio nanochannels. These multilayers were also calcinated to form nanoporous coatings within the nanochannels. Confined and unconfined multilayers were compared using scanning electron microscopy (SEM), showing significantly lower bilayer thicknesses when layers were assembled in confined geometries. Deposition was found to essentially stop at a nanochannel gap size of 55 nm for the PVS/TiO2 system and at 210 nm for the PDAC/SiO2 system, even though the channels were not plugged. These results provide further evidence of surface charge-induced depletion of unadsorbed species during LbL assembly, which occurs because of the closely spaced walls with like surface charge inherent in confined geometry LbL. This approach offers a simple but powerful new pathway to functionalize nanochannels with nanoporous coatings made from LbL-assembled nanoparticle-containing multilayers. It has applications in the production of functionalized nanofluidic devices for advanced separations utilizing size, charge, and potentially chemical or biological selectivity.

Abstract:A perfluoropolyether (PFPE)-based microfluidic device with cross-junction microchannels was fabricated with the purpose of producing uniform droplets. The microchannels were developed using CO2 laser engraving. PFPE was chosen as the main material because of its excellent solvent resistance. Polyethylene glycol diacrylate (PEGDA) was mixed with PFPE to improve the hydrophilic properties of the inner surface of the microchannels. The microchannels of the polydimethylsiloxane microfluidic device had a blackened and rough surface after laser engraving. By contrast, the inner surface of the microchannels of the PFPE-PEGDA microfluidic device exhibited a smooth surface. The lower power and faster speed of the laser engraving resulted in the development of microchannels with smaller dimensions, less than 30 μm in depth. The PFPE and PFPE-PEGDA microfluidic devices were used to produce uniform water and oil droplets, respectively. We believe that such a PFPE-based microfluidic device with CO2-laser-engraved microchannels can be used as a microfluidic platform for applications in various fields, such as biological and chemical analysis, extraction, and synthesis.

Fabrication of Silicon Nanopore Arrays Using A Combination of Dry and Wet Etching

Abstract:This paper presents a novel method for the fabrication of silicon nanopore arrays. The proposed method is based on inductive coupled plasma (ICP) etching and a two-step anisotropic wet etching, and can be used to fabricate individual nanopores, and massive nanopore arrays, with lower cost and less time compared with conventional fabrication processes. To optimize this nanopore fabrication process, the size of the ICP etch window was determined in a series of ICP experiments, and a theoretical analysis of the two-step wet etching was conducted. A nanopore array with an average feature size of 130 nm and a rectangular nanopore with a feature size of 38 nm were successfully obtained using this method. These results indicate the potential of this method for the large-scale production arrays of nanopores with desired sizes and shapes.

Abstract:Fabrication process of a novel multi-scaled filtration membrane for sample pretreatment in microfluidic system is presented in this paper. The membrane has a combinatorial structure with feature size ranging from hundred nanometers to micrometers. A nano-filtration membrane is mosaically embedded into the pores of a microfabricated membrane with crystal structure to get a high filtering flux at a reasonable mechanical strength. After assembling Polystyrene microspheres into the micro-pores and electrodepositing Cu in the interstices of the sphere crystal, a nano-porous structure with feature size of 450nm can be obtained after releasing the sacrificial micro-spheres off in oxygen plasma. All the process of the present micro/nano filtration unit is compatible with the micro-electro-mechanical systems (MEMS) technology and thereby is ready for the multifunction integration monolithically to construct a micro total analysis system.
Ultramicrotomy‐Assisted Fabrication of Nanochannels for Efficient Ion Transport and Energy Generation

Abstract:Ultramicrotomy enables precise nanochannels wit 2D materials for nanofluidic devices. Integrated with microfluidic chips, these membranes allow ion transport studies under confinement. Sliced membranes of vermiculite laminate channels show promise for pressure‐driven energy due to their pressure tolerance and ionic current generation. Pristine channels achieve exceptional efficiency in osmotic power generation, exceeding state‐of‐the‐art systems. Nano‐ and angstrom‐scale channels fabricated from 2D‐layered materials provide a unique platform for studying fluidic behavior at atomic‐scale confinement, with applications in desalination, osmotic power generation, and fuel cells. While various fabrication methods exist, achieving precision, scalability, and minimal fabrication time is challenging. Ultramicrotomy‐based nanofabrication is shown here as an efficient approach to create nanofluidic membranes containing nanochannels with atomically flat walls. This approach is compatible with both bottom–up nanolaminates and top–down nanochannels, produces multiple devices from the same 2D assembly and allows swift variation of channel length. Integration of these membranes into macroscopic silicon fluidic‐chips is achieved maintaining the structural and functional properties. The robustness of the sliced membranes is demonstrated through restacked vermiculate laminates that sustain applied pressures and generate ionic streaming currents. Sliced pristine vermiculite channels exhibit charge‐selective ion transport, leading to osmotic power density (Posm/A) ranging from 9.2 to 300.4 Wm−2 under various KCl concentration gradients. Maximum conversion efficiency of 23.3% is obtained at a 100‐fold gradient, yielding Posm/A of 234.6 Wm−2. Short channel lengths sliced by ultramicrotomy contribute to high Posm/A, promising applications of miniature energy devices for pressure‐driven electricity generation and osmotic power generation.
