Abstract:Inflammation and infection caused by implants are serious problems in the clinical application of titanium and titanium alloy. Titanium copper alloy has shown strong antibacterial properties, but the surface bioactivity is still necessary to be modification to improve biocompatibility. In this paper, a TiO2/Cu2O multifunctional coating was prepared on Ti-5Cu alloy by an anodic oxidation process in order to enhance the surface bioactivity without affecting the antibacterial ability. The surface physical and chemical properties of the anodized Ti-Cu alloys were tested by X-ray diffraction (XRD), X-ray photo-electron spectroscopy (XPS), scanning electron microscopy (SEM), Vickers hardness tester, contact angle tester and laser confocal microscope, and the corrosion properties were studied by inductively coupled plasma mass spectrometer and electrochemical workstation. The results have shown that the roughness, the hydrophilicity, the copper ion release the corrosion resistance were increased by the anodization in a voltage dependent way. The effect of the anodization was evaluated on the antibacterial properties and cell bioactivity. It was shown that pure titanium did not have antibacterial properties after the anodization. However, the anodization significantly improved the antibacterial properties of titanium copper alloy due to the formation of TiO2/Cu2O coating. No cytotoxicity was found on Ti-5Cu alloys and the anodized Ti-5Cu alloys but the treatment accelerated the cell adhesion. It was suggested that anodization treatment might be a potential surface biomodification method for the clinical application of titanium copper alloys.

Abstract:Titanium-copper (Ti-Cu) alloy is an advanced antibacterial material with excellent mechanical properties, thermodynamic stability, corrosion resistance and biocompatibility. Sandblasting and acid-etching was applied to the Ti-3Cu alloy to construct a rough surface with Ti2Cu phase on the surface in order to improve the antibacterial properties and the osseointegration. The phase constitutes and the physical properties were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM) and confocal laser scanning microscope (CLSM), and the surface chemical properties were analyzed by X-ray photoelectron spectroscopy (XPS) and electrochemical testing. The antibacterial property was assessed by the plate-count method and the cell compatibility was evaluated by the CCK-8 test in order to reveal the effect of surface characteristics on the antibacterial ability and bioactivity. The results demonstrated a rough and lamellar surface structure with many submicron Ti2Cu particles on the surface of Ti-3Cu, which could enhance the antibacterial ability and promote the cell proliferation and the initial adhesion of osteoblasts. However, the surface treatment also reduced the corrosion resistance and accelerated the Cu ion release.

Effects of Surface Roughening on Antibacterial and Osteogenic Properties of Ti-Cu Alloys with Different Cu Contents

Abstract:In order to reduce implant-related infections and improve early osseointegration, we performed HF+ anodic oxidation on the surface of the new antibacterial Ti-Cu alloy to make the titanium-based implants have a micro/submicron structure. On this basis, a series of surface-modified Ti-xCu alloys with different Cu contents (3, 5 and 7 wt%) were fabricated and a comprehensive study was conducted on MC3T3-E1 cell adhesion, proliferation, apoptosis and its antibacterial activity against Staphylococcus aureus. Results showed that the Ti-xCu alloys by HF etching + anodized possessed multifunctional characteristics of antibiofilm and antibacterial abilities, excellent biocompatibility and osteogenesis promoting abilities. Increment of Cu content significantly contributed to the antibacterial and osteogenic properties of HF etching + anodized Ti-Cu alloys. Cell proliferation rates of HF etching + anodized Ti-7Cu alloys were lower than those of Ti-3Cu and Ti-5Cu alloys, while the early cell apoptosis rates were higher than Ti-3Cu and Ti-5Cu groups. All the above results finally presented that the HF etching + anodized Ti-5Cu alloy exhibited extremely strong antibacterial properties, good biological compatibility and osteogenic ability, as well as the most excellent ductility and corrosion resistance, providing a great potential application for the future dental implantation.
Effect of Anodic Oxidation Pulse Voltage on Antibacterial Properties and Biocompatibility of Ti-Ag Alloy

Abstract:For the application of titanium and titanium alloys in orthopedic implant materials, the antibacterial properties and cell biocompatibility determine whether the implant surgery is successful. In this study, a functional anodic oxidation (AO) coating was successfully prepared to modify the surface of Ti-Ag alloy. The surface characteristics of the anodized Ti-Ag alloy were analyzed using techniques such as X-ray diffraction (XRD), scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), and contact angle measurements. The corrosion characteristics of Ti-Ag samples were tested by an electrochemical workstation. In addition, the antibacterial properties and cell activity were studied by the plate count method and MC3T3-E1 cell staining. The results indicate that the AO process can generate a multi-functional TiO2/Ag2O coating with a large number of block and flower-like structures on the surface of a Ti-Ag alloy. When the AO voltage of the sample is 120 V, the maximum roughness is 0.73 μm and the minimum wetting degree is 23°, which improves the biocompatibility. The corrosion test results show that AO treatment can improve the corrosion resistance of a Ti-Ag alloy. The oxidation voltage is 20 V and the coating has the best corrosion resistance. The corrosion open circuit potential (Eocp) is 107.621 mV and the corrosion current density (icorr) is 2.241 × 10−8 A·cm−2. This coating can promote ion release and show more than 99% of a strong antibacterial ability against S. aureus. The results of the compatibility evaluation by cultured cells showed that the multifunctional coating formed by the anodic oxidation process did not cause cytotoxicity and promoted the adhesion of MC3T3-E1 cells.

Abstract:Ti and its alloys have received wide attention in marine engineering. However, the limited anti-biofouling capability may hinder their wide application. In the present work, micro-arc oxidation (MAO) with and without the introduction of ultrasonic vibration (UV) has been conducted on metallic Ti substrate in an aqueous solution containing Na2Cu-EDTA to produce a Cu-modified TiO2 coating. Microstructural characterization reveals that the introduction of UV increased the thickness of the coating (ranging from ~13.5 μm to ~26.2 μm) compared to the coating (ranging from ~8.1 μm to ~12.8 μm) without UV. A relatively higher Cu content (~2.13 wt.%) of the coating with UV relative to the coating (~1.39 wt.%) without UV indicates that UV enhances the incorporation of Cu into TiO2. Further, both electrochemical properties and the response to sulfate-reducing bacteria (SRB) were evaluated, revealing that UV introduction endows Cu-modified TiO2 coating with enhanced corrosion resistance and antifouling capability. The present results suggest that ultrasound-auxiliary micro-arc oxidation (UMAO) obviously enhances the surface performance of Ti alloys for promising applications in marine engineering.

Abstract:Although micro‐arc oxidation is frequently employed in producing wear‐ and corrosion‐resistant coatings on Ti and Ti alloys for biomedical applications, the formation of non‐uniform microstructure of the coatings is still unavoidable. To overcome this drawback, this work adopted ultrasonic assistance in the micro‐arc oxidation of Ti–6Al–4V and systematically investigated. The influences of ultrasonic treatment with different durations on the coating microstructures and resultant properties. The positive effects of ultrasonic, such as cavitation effect, sound flow effect, and mechanical effect, accelerate the cooling rate of electrolyte and promote the homogeneity of the solute on the sample surface. Therefore, the ultrasonic‐assisted coatings exhibit uniform microstructures with fewer cracks and therefore improved performances. For instance, compared to the counterpart without ultrasonic assistance, the ultrasonic‐assisted coating with 15‐min duration demonstrate 25.1% lower mass loss in the tribology test and half corrosion density in Hank's solution. Such results indicate that ultrasonic assistance in micro‐arc oxidation of Ti–6Al–4V can homogenize the microstructures of coatings and enhance their corrosion resistance and wear resistance in human body.This article is protected by copyright. Abstract:In this work, TiO2/CuO coating was prepared on titanium (Ti) by combination of magnetron sputtering and annealing treatment. The microstructure, biocompatibility, corrosion resistance and antibacterial property of TiO2/CuO coating were investigated in comparison with pure Ti and TiO2 coating. The results show that TiO2/CuO coating is mainly composed of TiO2 and CuO. In vitro cytocompatibility evaluation suggests that no obvious toxicity appears on the TiO2/CuO coating, and the coating stimulates the osteoblast spreading and proliferation. Compared with Ti and TiO2 coating, TiO2/CuO coating exhibits improved corrosion resistance and antibacterial ability against S.aureus. This study is the first attempt to apply the combination of magnetron sputtering and annealing treatment to introduce the Cu into TiO2 coating for surface modification of Ti-based implant materials, which may provide a research foundation for further development of bioactive multifunctional coatings to meet the better clinical demand.

Abstract:Medium energy (40--65 MeV) ..gamma.. rays following radiative capture of 50 MeV polarized protons on /sup 11/B were studied for the first time by using a NaI detector ensemble with good energy resolution and small background counts. Discrete ..gamma.. rays feeding the ground 0/sup +/, 4.44 MeV 2/sup +/, and 9.64 MeV 3/sup -/ states, and prominent ..gamma.. peaks feeding the 19 and 22 MeV excitations region in /sup 12/C were identified. The analyzing powers of the radiative capture were found to depend on the microscopic configurations of the final states.

Abstract:Titanium‑copper (TiCu) alloys showed potential application in clinical field due to their excellent antibacterial properties. A selective acid etching (SAE) was applied to TiCu alloy in order to further improve the antibacterial properties. The phase constitute and the surface microstructure were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM) and X-ray photoelectron spectroscopy (XPS), the corrosion properties and Cu ion release behavior were measured by electrochemical testing and inductively coupled plasma spectrometry (ICP), the antibacterial property was assessed by a plate-count method and the cell compatibility was evaluated by CCK-8 test. It was shown that more Ti2Cu particles were exposed on the surface of TiCu sample after SAE treatment, which increased the corrosion current density and accelerated the Cu ion release due to the galvanic corrosion between Ti-Ti2Cu couple. The plate counting results demonstrated that the antibacterial ability of TiCu alloy was significantly enhanced from 79% to >99.9% by SAE treatment due to the multifunction of increased Ti2Cu phase on the surface, Cu ion release and the formation of Cu2O. Besides, no cytotoxicity was detected to MC3T3-E1 cells on all test samples.

Abstract:Titanium (Ti) and its alloys are used extensively in implants due to their excellent biocompatibility and mechanical properties. However, Ti-based implant materials have specific complications associated with their applications, such as the loosening of implanted host interface owing to unsatisfactory cell adhesion and the susceptibility of the implants to bacterial infections. Hence, a surface that displays selective biointeractivity, i.e., enhancing beneficial host cell responses but inhibiting pathogenic microbial adhesion, would be highly desirable. This study aims to confer long-lasting antibacterial properties and good biocompatibility on Ti via the microarc oxidation technique. The biocompatibility of the Ti surface was evaluated by cytotoxicity test, and the bacteriostasis rate was evaluated by antibacterial efficacy. The results showed that the implant surface might be nontoxic to cell and its long-lasting antibacterial properties could be significantly improved. These results indicate that such microarc oxidation coatings are expected to have good potential in transcutaneous implant applications.

Abstract:This work presents the effect of anodic oxidation (AO) on the microstructure and properties of Ti‐13Nb‐13Zr‐3Cu (TNZ‐3Cu). The AO treatment provides TNZ‐3Cu alloy with a dense inner and porous outer layer, which is composed of TiO2, Nb2O5, ZrO2, and CuO. The AO treatment improves the corrosion resistance and antibacterial properties of TNZ‐3Cu alloy and promotes the early adhesion of cells. Ti‐13Nb‐13Zr (TNZ), a representative of β‐type titanium alloys, has garnered significant interest in biomedical implant materials recently because of its high strength, and closer elastic modulus to human bone. However, TNZ alloy does not have antibacterial properties and has the potential to cause implant‐related infection, even implantation failure. Herein, TNZ alloy is alloyed by Cu element and the surface is modified by anodic oxidation (AO) to enhance the biocompatibility and antimicrobial qualities. The corrosion resistance, antimicrobial properties, and cytotoxicity of the TNZ‐3Cu alloy before and after anodization are studied by X‐ray diffraction, X‐ray photoelectron spectroscopy, scanning electron microscopy, electrochemical test, mechanical property test, and cell test. The experimental results show that the primary constituents of the TNZ‐3Cu alloy are α′′ martensite and β phase after solid solution treatment. The elastic modulus of TNZ‐3Cu alloy is (65 ± 1.33) GPa, 9 GPa lower than that of TNZ alloy. The AO treatment significantly improves the hydrophilicity and antibacterial properties as well as the corrosion resistance by the formation of a dense oxides composites coating. The cell test results show that the coating improves the initial adhesion of cells by the high hydrophilic surface and promotes the proliferation of cells by the release of copper ions.

Abstract:In order to solve the challenging problem of microbial infections caused by microorganisms on medical implants, it is imperative to develop novel antimicrobial biomaterials. This work demonstrated that 317L-Cu stainless steel (SS), created by adding copper through a solution and aging heat treatment process, exhibited good antibacterial properties against staphylococcus aureus, achieving 2 log reduction of planktonic cells after 5 days of incubation. In this study, the antibacterial test was performed using the plate count method, the fluorescence cell staining method and the quantitative polymerase chain reaction (qPCR) method. It is well known that a high concentration of copper ion can lead to cytotoxicity. This work explored the cytotoxicity of 317L-Cu SS through real-time cell analysis (RTCA). Experimental results demonstrated that the 317L-Cu SS possessed a satisfactory antibacterial ability against S. aureus, and the antibacterial rate based on the reduction of sessile cell count reached 98.3% after 24-hour treatment. The bacterial adhesion and the biofilm thickness were considerably reduced by the 317L-Cu SS. The results of RTCA suggested that 317L-Cu SS did not introduce cytotoxicity to mouse cells, indicating its suitability as a medical implant material.

Abstract:Titanium (Ti) and its alloys have good biocompatibility, mechanical properties and corrosion resistance, making them attractive for biomedical applications. However, their biological inertness and lack of antimicrobial properties may compromise the success of implants. In this review, the potential of micro-arc oxidation (MAO) technology to create bioactive coatings on Ti implants is discussed. The review covers the following aspects: 1) different factors, such as electrolyte, voltage and current, affect the properties of MAO coatings; 2) MAO coatings affect biocompatibility, including cytocompatibility, hemocompatibility, angiogenic activity, corrosion resistance, osteogenic activity and osseointegration; 3) antibacterial properties can be achieved by adding copper (Cu), silver (Ag), zinc (Zn) and other elements to achieve antimicrobial properties; and 4) MAO can be combined with other physical and chemical techniques to enhance the performance of MAO coatings. It is concluded that MAO coatings offer new opportunities for improving the use of Ti and its alloys in biomedical applications, and some suggestions for future research are provided.
Characteristics of novel Ti-40Nb-xCu alloy and surface treatment with superior antibacterial property and biocompatibility using micro-arc oxidation for dental implants

Abstract:Peri-implantitis and insufficient osseointegration are the principal challenges faced by dental implants at present. In order to fabricate dual-function dental implant materials possessing both antibacterial and osteogenic capabilities, this study incorporates the antimicrobial element Cu into the Ti40Nb alloy, developing a novel Ti40Nb-xCu alloy with antibacterial properties. Among them, Ti40Nb3Cu has the best overall performance. Compared to Ti40Nb, the tensile strength increased by 27.97%, reaching 613 MPa. Although the elongation rate has decreased from 23% to 13.5%, the antibacterial rates against S. aureus and P. gingivalis both exceed 85%. Furthermore, the surface of Ti40Nb-xCu alloy was then treated with micro-arc oxidation to enhance its bioactivity, thereby accelerating osseointegration. The results indicated that the MAO treatment retains the antibacterial properties of the Ti40Nb3Cu alloy while significantly promoting bone formation through its introduced porous coating, thus heralding it as a propitious candidate material for dental implant applications.

Abstract:Ti-15Mo-9Ag (TM-9Ag) alloy was designed and prepared in order to produce a titanium alloy with low elastic modulus and excellent antibacterial properties. Anodic oxidation was used to surface modify and improve the surface bioactivity of TM-9Ag and the effect on the microstructure, corrosion resistance, antimicrobial properties and biocompatibility was investigated in an oxidation voltage range of 10–80 V. The results showed that TiO 2 , MoO 2 and Ag 2 O were formed in the oxidized layer on TM-9Ag surface after the anodic oxidation. The antibacterial properties were evaluated by the plate counting method, and it was found that the antibacterial properties of the alloy were significantly improved, and the corrosion resistance was also enhanced. Cellular experiments showed that the anodic oxidized TM-9Ag alloys exhibited good cytocompatibility.

Abstract:Implant surgeries are increasingly challenging due to their rising number. Achieving the desired biomaterial surface properties to ensure a strong bond with human tissue is a significant issue. This study investigates the influence of ultrasound (US) during the micro-arc oxidation (MAO) process on Ti13Zr13Nb bio-alloy, an area not previously explored, to enhance titanium alloy coatings' properties for biomedical applications. Porous calcium-phosphate-based coatings were successfully deposited on Ti13Zr13Nb using MAO and ultrasound micro-arc oxidation (UMAO). Various properties such as morphology, chemical composition, topography, wettability, surface free energy, thickness, adhesion to the substrate, as well as mechanical and corrosion characteristics were thoroughly analyzed. Cytocompatibility was assessed using human osteoblasts. Using US during the MAO process increased coating roughness (up to ~ 17%), core height (up to 22%), isotropy (up to 17%), thickness (up to ~ 46%), and hardness (up to ~ 18%), depending on MAO parameters and US mode. Optimal coating performance was achieved at 136 mA, 600 s, and a sinusoidal US setting, resulting in the highest isotropy (~ 79%) and rutile quantity (2.6%), the lowest elastic modulus (~ 57 GPa), and the contact angle of ~ 70°, all of which could have contributed to enhancing osteoblast viability in vitro. This study, for the first time, underscores the importance of using the US during the MAO in tailoring the Ti13Zr13Nb for specific biomedical applications.

Electrochemical Deposition of Copper on Bioactive Porous Titanium Dioxide Layer: Antibacterial and Pro-Osteogenic Activities

Abstract:Ti surfaces must exhibit antibacterial activity without cytotoxicity to promote bone reconstruction and prevent infection simultaneously. In this study, we employed a two-step electrochemical treatment process, namely, microarc oxidation (MAO) and cathodic electrochemical deposition (CED), to modify Ti surfaces. During the MAO step, a porous TiO2 (pTiO2) layer with a surface roughness of approximately 2.0 μm was generated on the Ti surface, and in the CED step, Cu was deposited onto the pTiO2 layer on the Ti surface, forming Cu@pTiO2. Cu@pTiO2 exhibited a similar structure, adhesion strength, and crystal phase to pTiO2. Moreover, X-ray photoelectron spectroscopy (XPS) confirmed the presence of Cu in Cu@pTiO2 at an approximate concentration of 1.0 atom %. Cu@pTiO2 demonstrated a sustained release of Cu ions for a minimum of 28 days in a simulated in vivo environment. In vitro experiments revealed that Cu@pTiO2 effectively eradicated approximately 99% of Staphylococcus aureus and Escherichia coli and inhibited biofilm formation, in contrast to the Ti and pTiO2 surfaces. Moreover, Cu@pTiO2 supported the proliferation of osteoblast-like cells at a rate comparable to that observed on the Ti and pTiO2 surfaces. Similar to pTiO2, Cu@pTiO2 promoted the calcification of osteoblast-like cells compared with Ti. In summary, we successfully conferred antibacterial and pro-osteogenic activities to Ti surfaces without inducing cytotoxic effects or structural and mechanical alterations in pTiO2 through the application of MAO and CED processes. Moreover, we found that the pTiO2 layer promoted bacterial growth and biofilm formation more effectively than the Ti surface, highlighting the potential drawbacks of rough and porous surfaces. Our findings provide fundamental insights into the surface design of Ti-based medical devices for bone reconstruction and infection prevention.
Effect of Ta(Cu)–O composite film on mechanical properties, osteoblasts proliferation and antibacterial behavior of medical titanium alloy

Abstract:Although titanium alloy (Ti–6Al–4V) has good mechanical and biological properties as a bone implant material, it still has defects of biologically inertness and poor antibacterial ability. In this study, a multi-target magnetron sputtering method was used to prepare Ta(Cu)–O composite films with three different ratios (Ta(Cu 10.82 )-O; Ta(Cu 5.68 )-O; Ta(Cu 3.43 )-O) on the surface of Ti–6Al–4V to solve the problems above. In vitro osteoblast proliferation experiments showed that the Ta(Cu)–O composite films significantly accelerated the proliferation, adhesion and spreading of osteoblasts, especially for Ta(Cu 3.43 )-O. And due to the excellent antibacterial activity of Cu, the Ta(Cu)–O composite films also showed good antibacterial effect. In conclusion, the results showed that the Ta(Cu)–O films can enhance the antibacterial properties of Ti–6Al–4V while accelerating the growth of osteoblasts, displaying great clinical potential in bone-implants applications.

Abstract:Bacterial infection and toxic metal ions releasing are the challenges in the clinical application of Ti6Al4V alloy implant materials. Copper is a kind of long-acting, broad-spectrum and safe antibacterial element, and Ta2O5 has good corrosion resistance, wear-resistance and biocompatibility, they are considered and chosen as a potential coating candidate for implant surface modification. In this paper, magnetron sputtering technology was used to prepare copper doped Ta2O5 multilayer composite coating Cu-Ta2O5/Ta2O5/Ta2O5-TiO2/TiO2/Ti (Cu-MTa2O5 for short) on Ti6Al4V alloy surface, for studying the effect of copper incorporation on the microstructure, wettability, anticorrosion and antibacterial activities of the composite coating. The results showed that Cu-MTa2O5 coating obviously improves the hydrophobicity, corrosion resistance and antibacterial property of Ti6Al4V alloy. In the coating, both copper and Ta2O5 exhibit an amorphous structure and copper mainly presents as an oxidation state (Cu2O and CuO). With the increase of the doping amount of copper, the grain size, roughness, and hydrophobicity of the modified surface of Ti6Al4V alloy are increased. Electrochemical experiment results demonstrated that the corrosion resistance of Cu-MTa2O5 coated Ti6Al4V alloy slightly decreased with the increase of copper concentration, but this coating still acts strong anticorrosion protection for Ti6Al4V alloy. Moreover, the Cu-MTa2O5 coating can kill more than 97% of Staphylococcus aureus in 24 h, and the antibacterial rate increases with the increase of copper content. Therefore, Cu-MTa2O5 composite coating is a good candidate for improving anticorrosion and antibacterial properties of Ti6Al4V alloy implant medical devices.
