Abstract:Acute dissection of the left main coronary artery during diagnostic cardiac catheterization with selective coronary arteriography is an uncommon but recognized complication of the procedure. That similar dissection may occur during percutaneous transluminal coronary angioplasty is less well recognized. This report describes two cases of left main coronary dissection resulting in acute occlusion that occurred during percutaneous transluminal coronary angioplasty and demonstrates that survival with essentially complete functional recovery may result if immediate surgical intervention is undertaken. Recognition and treatment of this potentially catastrophic complication of angioplasty is described.

Abstract:A novel method based on gravity and electric force driving of cells was developed for flow cytometry and fluorescence activated cell sorting in a microfluidic chip system. In the experiments cells flowed spontaneously under their own gravity in a upright microchip, passed through the detection region and then entered into the sorting electric field one by one at an average velocity of 0.55 mm s(-1) and were fluorescence activated cell sorted (FACS) by a switch-off activation program. In order to study the dynamical and kinematic characteristics of single cells in gravity and electric field of microchannels a physical and numerical module based on Newton's Law of motion was established and optimized. Hydroxylpropylmethyl cellulose (HPMC) was used to minimize cell assembling, sedimentation and adsorption to microchannels. This system was applied to estimate the necrotic and apoptotic effects of ultraviolet (UV) light on HeLa cells by exposing them to UV radiation for 10, 20 or 40 min and the results showed that UV radiation induced membrane damage contributed to the apoptosis and necrosis of HeLa cells.

Abstract:Hydrogel-based microfluidic chips are more biologically relevant than conventional polydimethylsiloxane (PDMS) chips, but the inherent swelling of hydrogels leads to a decrease in mechanical performance and deformation of the as-prepared structure in their manufacture and application processing. Non-swelling hydrogel has, for the first time, been utilized to construct microfluidic chips in this study. It was fabricated by covalently cross-linking the biocompatible copolymer of di-acrylated Pluronic F127 (F127-DA). Thanks to their non-swelling property, the hydrogel-based microfluidic chips maintain their as-prepared mechanical strength and channel morphology when equilibrated in aqueous solution at 37 degrees C. Moreover, the microfluidic chips are autoclavable and show an appropriately slow degradation rate by remaining stable within 21 days of incubation. Based on these properties, a vessel-on-a-chip was established by seeding human umbilical vein endothelial cells (HUVECs) onto the microchannel surfaces inside the microfluidic chip. Under 6 days of perfusion culture with a physiologically relevant shear stress of 5 dyne per cm(2), the HUVECs in the chip show responsivity to fluid shear stress and express higher endothelial functions than the corresponding static culture. Therefore, non-swelling hydrogel-based microfluidic chips could potentially be applicable for cell/tissue-related applications, performing much better than conventional PDMS or existing hydrogel based microfluidic chips.

Abstract:The microfluidic passive control of microparticles largely relies on the hydrodynamic effects of the carrier media such as Newtonian fluids and viscoelastic fluids. Yet the viscoelastic/Newtonian interfacial effect has been scarcely investigated, especially for high-resolution particle separation. Here we report a microfluidic co-flow of Newtonian (water or PBS) and viscoelastic fluids (PEO) for the size-dependent separation of microparticles. The co-flow condition generates a stable viscoelastic/Newtonian interface, giving rise to the wall-directed elastic lift forces that compete with the center-directed lift forces, and efficiently hinders the migration of microparticles from the Newtonian to the viscoelastic fluid in a size-dependent manner. An almost complete separation of a binary mixture of 1 μm and 2 μm polystyrene particles is achieved by the co-flow of water and a very dilute PEO solution (100 ppm), whereas the sole use of water or PEO could not lead to an efficient separation. This co-flow microfluidic system is also applied for the separation of Staphylococcus aureus (1 μm) from platelets (2-3 μm) with >90% efficiencies and purities.
Microfluidic droplet-array liquid-liquid chromatography based on droplet trapping technique.

Abstract:We describe the first realization of liquid chromatographic separation in a droplet-based microfluidic system and develop a novel mode for microchip-based chromatography named as droplet-array liquid-liquid chromatography. In this system, two arrays of picoliter-scale droplets immobilized on both sidewalls of a microchannel with droplet trapping technique served as the stationary phase in chromatographic separation, while the other immiscible phase flowing in the microchannel served as the mobile phase. The chromatographic separation was achieved on the basis of multiple extraction and elution of analytes between the droplet array stationary phase and the mobile phase. The proof-of-concept study of the droplet-array LC system was performed in the separation of fluoranthene and benzo[b]fluoranthene. Under the optimum conditions, the two analytes were separated within 26 min with separation efficiencies of 112 mu m and 119 mu m plate height, respectively. The advantages of the present system include simple structure, low driving pressure, and relatively high sample capacity. It can also provide a useful platform for LC theory study and educational purposes by allowing the researchers and students to directly "see" the continuous extraction and elution process of a chromatographic separation.

Abstract:A micro flow injection wetting film liquid–liquid extraction system has been developed for trace analyte concentration and on-chip detection. A hydrophobic channel fabricated on a polycarbonate chip was used to support the wetting film, and hydrostatic pressure generated by the difference in liquid levels was employed to drive the fluids. Sequential injection of segments of aqueous sample solution and organic solvent was conducted by switching the sample- or solvent-containing vials to an on-chip sampling probe, and detection was performed by a co-focused, laser induced fluorescence detector. Using butyl rhodamine B as a model analyte and butanol as the solvent for both film-coating and elution, various experimental conditions such as hydrostatic pressure, coating time, channel length, sampling volume, and sample acidity were investigated. Under optimized conditions, a 24-fold enrichment factor was obtained with the consumption of about 3μL sample solution, and a detection limit (3σ) of 6.0×10−9M butyl rhodamine B was achieved at the sampling rate of 19h−1. Eleven consecutive runs of a 1.0×10−5M butyl rhodamine B solution produced a relative standard deviation of 1.5% for the detected fluorescence signals.
Fabrication of Micro/Nanofluidic Chip on Glass Substrate and Its Application for Electrokinetic Concentration of Proteins

Abstract:A simple and cost-effective double-lithography and double wet-etching method (abbriciated as double-etching technique) was developed for fabricating the full glass chips with hybrid micro- and nanochannels. The microchannels were firstly constructed on a glass substrate by standard UV photolithography and wet etching technique. After washing away the rest photoresist and spin coating of a new layer of photoresist on the microchannel-structured glass substrate, nanochannels with a depth less than 100 nm were prepared on the same substrate by second lithography and wet etching. Finally, the channel-structured glass substrate was sealed to a cover glass plate using room temperature bonding technique. Without clean room facilities and expensive facilities, fluidic chips with hybrid micro- and nanochannel network were fabricated with high yields. The analytical features of those fluidic chips were demonstrated by the successful concentration of fluorescein isothiocyanate(FITC)-labeled human serum albumin(HSA) via electrokinetic ion trapping. Enrichment factor of more than 200 was achieved for 0.5 mg/mL of FITC-HSA solutions within 30 s.

Abstract:Droplet sorting and enrichment, as a prominent field within microfluidic technology, represent a pivotal stage in the manipulation of droplets and particles. In recent times, droplet sorting methods based on lab-on-disk (LOD) have garnered significant interest among researchers for their inherent merits, including high throughput, ease of operation, seamless device integration, and independence from supplementary driving forces. This study introduces a centrifugal force-driven microfluidic chip comprising spiral microchannels. The chip incorporates microhole arrays along the sidewall of the spiral channels, enabling size-based sorting and enrichment of microdroplets under the influence of multiple forces. Firstly, a comparative analysis was performed to assess the influence of the separation port structure and rotational speed on efficiency, and a mechanical modeling approach was employed to conduct kinetic analyses of droplet behavior during instantaneous separation. Those findings demonstrated a good agreement with the experimental results at ω <100 rpm. Subsequently, sorting experiments on homogeneous droplets indicated that repetitive sorting could increase the recovery ratios, RT(α), of high-concentration droplets (20.7%) from 35.3% to over 80%. We also conducted a sorting experiment on three-component homogeneous-phase emulsions using a serially connected chip array, and the sorting throughput was 0.58 mL/min. As a result, the RT(α) for 60 and 160 μm droplets were 99.4% and 88.9%, respectively. Lastly, we conducted elution experiments and dualsample sorting on a single chip, and the fluorescence results demonstrated that this study provided an efficient and non-cross-contaminating sorting method for non-homogenous phase multi-sample microreactor units.

Abstract:A novel type of miniaturized liquid-liquid chromatography (LLC) system with droplet-array stationary phase was developed on a microfluidic chip. The droplet-trapping technique was employed to produce two arrays of stationary phase droplets in the picoliter range on both sidewalls of the microchannel. Mobile phase was. driven by gravity with a linear velocity range of 100-1.000 mu m/s. A fluorescent dye, Eosin Y, was used as model sample to demonstrate the possibility of the system in achieving chromatography separation.

Abstract:The development of efficient sample introduction and pretreatment systems for microfluidic chip-based analytical systems is important for their application to real-life samples. In this work, world-to-chip interfacing was achieved by a novel flow-through sampling reservoir featuring a guided overflow design. The flow-through reservoir was fabricated on a 30 x 60 x 3 mm planar glass chip of crossed-channel design used for capillary electrophoresis separations. The 20-microL sample reservoir was produced from a section of plastic pipet tip and fixed at one end of the sampling channel. Sample change was performed by pumping 80-microL samples sandwiched between air segments at approximately 0.48 mL/min flow rate through the flow-through reservoir, introduced from an access hole on the bottom side of the chip. A filter paper collar wrapped tightly around the reservoir guided the overflowing sample solution into a plastic trough surrounding the reservoir and then to waste. The performance of the system was demonstrated in the separation and determination of FITC-labeled arginine, glycine, phenylalanine, and glutamic acid with LIF detection, by continuously introducing a train of different samples through the system without electrical interruption. Employing a separation channel of 4 cm (2-cm effective separation length) and 1.4-kV separation voltage, maximum throughputs of 80/h were achieved with <4.1% carryover and precisions ranging from 1.5% for arginine to 2.6% RSD (n = 11) for glycine. The sampling system was tested in the continuous monitoring of the derivatizing process of amino acids by FITC over a period of 4 h, involving 166 analytical cycles. An outstanding overall precision of 4.8% RSD (n = 166) was achieved for the fluorescein internal standard.
Comprehensive Two‐dimensional Separations Based on Capillary High‐performance Liquid Chromatography and Microchip Electrophoresis

Abstract:A novel comprehensive two-dimensional (2-D) separation system coupling capillary high-performance liquid chromatography (cHPLC) with microchip electrophoresis (chip CE) is demonstrated. Reversed-phase cHPLC was used as the first dimension, and chip CE acted as the second dimension to perform fast sample transfers and separations. A valve-free gating interface was devised simply by inserting the. outlet-end of LC column into the cross-channel on a specially designed chip. A home-made confocal laser-induced fluorescence detector was used to perform on-chip high-sensitive detection. The cHPLC effluents were continuously delivered to the chip and pinched injections of the effluents every 20 seconds were employed for chip CE separation. Gradient elution of cHPLC was carried out to obtain the high-efficiency separation. Free-zone electrophoresis was performed with triethylamine buffer to achieve high-speed separation and prevent sample adsorption. Such a simple-made comprehensive system was proved to be effective. The relative standard deviations for migration time and peak height of rhodamine B in 150 sample transfers were 3.2% and 9.8%, respectively. Peptides of the fluorescein isothiocyanate (FITC)-labeled tryptic digests of bovine serum albumin were fairly resolved and detected with this comprehensive 2-D system.
Sub-micrometer-precision, three-dimensional (3D) hydrodynamic focusing via "microfluidic drifting"

Abstract:In this article, we demonstrate single-layered, "microfluidic drifting" based three-dimensional (3D) hydrodynamic focusing devices with particle/cell focal positioning approaching submicron precision along both lateral and vertical directions. By systematically optimizing channel geometries and sample/sheath flow rates, a series of "microfluidic drifting" based 3D hydrodynamic focusing devices with different curvature angles are designed and fabricated. Their performances are then evaluated using confocal microscopy, fast camera imaging, and side-view imaging techniques. Using a device with a curvature angle of 180, we have achieved a standard deviation of +/- 0.45 mu m in particle focal position and a coefficient of variation (CV) of 2.37% in flow cytometric measurements. To the best of our knowledge, this is the best CV that has been achieved using a microfluidic flow cytometry device. Moreover, the device showed the capability to distinguish 8 peaks when subjected to a stringent 8-peak rainbow calibration test, signifying the ability to perform sensitive, accurate tests similar to commercial flow cytometers. We have further tested and validated our device by detection of HEK-293 cells. With its advantages in simple fabrication (i.e., single-layered device), precise 3D hydrodynamic focusing (i.e., submicrometer precision along both lateral and vertical directions), and high detection resolution (i.e., low CV), our method could serve as an important basis for high-performance, mass-producible microfluidic flow cytometry.
Digital light processing 3D printing of microfluidic devices targeting high-pressure liquid-phase separations

Abstract:This paper focuses on 3D printing using digital light processing (DLP) to create microchannel devices with inner diameters of 100, 200, and 500 µm and cater flow-through applications within the realm of analytical chemistry, in particular high-pressure liquid chromatographic separations. Effects of layer thickness and exposure time on channel dimensions and surface roughness were systematically investigated. Utilizing a commercially accessible 3D printer and acrylate resin formulation, we fabricated 100-500 µm i.d. squared and circular channel designs minimizing average surface roughness (< 20%) by applying a 20-µm layer thickness and exposure times ranging from 1.1 to 0.7 s. Pressure resistance was measured by encasing microdevices in an aluminum chip holder that integrated flat-bottom polyetheretherketon (PEEK) nanoports allowing to establish the micro-to-macro interface to the HPLC instrument. After thermal post-curing and finetuning the clamping force of the chip holder, a maximum pressure resistance of 650 bar (1.5% RSD) was reached (n = 3). A polymer monolithic support structure was successfully synthesized in situ with the confines of a 500 µm i.d. 3D printed microchannel. A proof-of-concept of a reversed-phase chromatographic gradient separation of intact proteins is demonstrated using an aqueous-organic mobile-phase with isopropanol as organic modifier.

Abstract:Efficient and reliable manipulation of biological particles is crucial in medical diagnosis and chemical synthesis. Inertial microfluidic devices utilizing passive hydrodynamic forces in the secondary flow have drawn considerable attention for their high throughputs, low costs, and harmless particle manipulation. However, as the dominant mechanism, the inertial lift force is difficult to quantitatively analyze because of the uncertainties of its magnitude and direction. The equilibrium position of particles varies along the migration process, thus inducing the instabilities of particle separation. Herein, we present a designable inertial microfluidic chip combining a spiral channel with periodic expansion structures for the sheathless separation of particles with different sizes. The stable vortex-induced lift force arising from the periodic expansion and the Dean drag force significantly enhanced the focusing process and determined the final equilibrium position. The experimental results showed that over 99% of target particles could be isolated with the high target sample purity of 86.12%. In the biological experiment, 93.5% of the MCF-7, 89.5% of the Hela, and 88.6% of the A549 cells were steadily recovered with excellent viabilities to verify the potential of the device in dealing with biological particles over a broad range of throughputs. The device presented in this study can further serve as a lab-on-chip platform for liquid biopsy and diagnostic analysis.

Abstract:A kind of PH gradient microfluidic chips through soft-lithography microfabrication for isoelectric focusing (IEF) and high performance liquid chromatography (HPLC) is introduced here. These pH gradient chips have the advantages such as easy fabrication, controllable pH range and precision, isoelectric focusing and separation at the same time, low voltage for isoelectric focusing, and time stable pH gradient. This method has potential application to sample preparation, separation and analysis of microfluidic chips.

Abstract:Micromachined capillary electrophoresis systems with integrated sample injector have been fabricated on glass chips using standard photolithography and chemical wet etching techniques. Experiments have been done to find proper ways of obtaining regular and smooth channels, and the performance of the chip was evaluated. Voltages of 4 kV could be applied over an 20 mm long capillary channel with sufficient dissipation of the Joule heat generated (1.12 W/cm). Electroosmotic flow was used to "load" the sample into the microchip and to "pump" the mobile phase during the experiments. Using laser-induced fluorescence detection and CCD imaging, the mixture of FITC-OH and FITC-labeled amino acid were injected and separated. Two methods of sample introduction ("floating" and "pinched") were evaluated. The results showed that applications of potentials to the intersecting channels could cut the sample plug clearly during the process of separation which made the tailing unserious. Frequent flushing of channels with alkaline solution and water was demonstrated to be important to keep the reproducibility of sample loading and separation.

Abstract:A simple method for injecting well-defined non-biased sample plugs into the separation channel of a microfluidic chip-based capillary electrophoresis system was developed by a combination of flows generated by negative pressure, electrokinetic and hydrostatic forces. This was achieved by using only a single syringe pump and a single voltage supply at constant voltage. In the loading step, a partial vacuum in the headspace of a sealed sample waste reservoir was produced using a syringe pump equipped with a 3-way valve. Almost instantaneously, sample was drawn from the sample reservoir across the injection intersection to the sample waste reservoir by negative pressure. Simultaneously, buffer flow from the remaining two buffer reservoirs pinched the sample flow to form a well-defined sample plug at the channel intersection. In the subsequent separation stage, the vacuum in headspace of the sample waste reservoir was released to terminate all flows generated by negative pressure, and the sample plug at the channel intersection was electrokinetically injected into the separation channel under the potential applied along the separation channel. The liquid levels of the four reservoirs were optimized to prevent sample leakage during the separation stage. The approach considerably simplified the operations and equipment for pinched injection in chip-based CE, and improved the throughput. Migration time precisions of 3.3 and 1.5% RSD for rhodamine123 (Rh123) and fluorescein sodium (Flu) in the separation of a mixture of Flu and Rh123 were obtained for 56 consecutive determinations with peak height precisions of 6.2% and 4.4% RSD for Rh123 and Flu, respectively.

Abstract:In biologic and chemical analysis system, both of high-speed analysis and real time data processing is important. A kind of microfludic sample, separation and dynamic process instrument has been built and tested. A typical sample stacking and separation image obtained with this instrument Preliminary on-chip reaction experiments are imaged using this detection system. The system based on focusing an expanded laser beam onto chip using a cylindrical lens then detecting the separated signal with a photomultiplier tube (PMT) is illustrated. Excellent signal/noise ratio was obtained while compared with traditional electrophoresis methods.
