Abstract:Focusing particles into a tight stream is usually a necessary step prior to counting, detecting and sorting them. Meanwhile, particle spacing control in microfluidic devices could also be applied in the field of accurate cell detection, material synthesis and chemical reaction. To achieve simultaneous particle focusing and spacing control, a novel microchannel composed by Dean and sheath flow section was proposed and fabricated according to the elaborated design principle with its manipulating performance in situ visualized. Using microspheres with a few microns as a template, the trajectory of the particles was discovered to follow lateral migration and reach certain equilibrium positions at the end of the designed Dean section. After being focused, the streamline was further concentrated and centralized with a controllable interparticle distance in sheath flow section. For sheath flow section, the angle between symmetrical tributaries and the mainstream channel and abrupt constriction/expansion structure of mainstream channel as important channel geometric features were investigated to minimize the focusing streamline width and optimize spacing control. An modified analytical model for sheath flow with different tributary angles was derived and proved to well describe the microsphere spacing control process.

Abstract:Using inertia effect of fluid to realize the high flux precise control of rigid particles is a novel microfluidic method, which has important applications in materials synthesis, biochemical reactions and medical diagnosis etc.In this study, we designed and fabricated an microfiuidic chip consisted of focusing units and the sheath liquid focusing structures based on the inertial focusing principle.Employing polystyrene microspheres with a diameter of 10 μm as solid particles model to study the focusing effect under certain flow conditions.The experimental results showed that when the flow rate of ethanol reached 710 μL/min, the solid microspheres began to be focusedto form a single flow pattern after flowed through the 60th focusing units.The sheath flow under certain conditions focusing on a single array of microspherescan enhance the focusing effect and controlthe spacing of the particles.The obtainedresults provide an important reference not only for the further study of the kinematic properties of rigid particles in microfluidics, but also for the development of novel microfluidic chips.
Three-Dimensional (3d) Hydrodynamic Focusing for Continuous Sampling and Analysis of Adherent Cells

Abstract:A simple three-dimensional (3D) hydrodynamic focusing microfluidic device integrated with continuous sampling, rapid dynamic lysis, capillary electrophoretic (CE) separation and detection of intracellular content is presented. One of the major difficulties in microfluidic cell analysis for adherent cells is that the cells are prone to attaching to the channel surface. To solve this problem, a cross microfluidic chip with three sheath-flow channels located on both sides of and below the sampling channel was developed. With the three sheath flows around the sample solution-containing cells, the formed soft fluid wall prevents the cells from adhering to the channel surface. Labeled cells were 3D hydrodynamically focused by the sheath-flow streams and smoothly introduced into the cross-section one by one. The introduction of sheath-flow streams not only ensured single-cell sampling but avoided blockage of the sampling channel by adherent cells as well. The maximum rate for introduction of individual cells into the separation channel was about 151 cells min(-1). With electric field applied on the separation channel, the aligned cells were driven into the separation channel and rapidly lysed within 400 ms at the entry of the channel by sodium dodecylsulfate (SDS) added in the sheath-flow solution. The microfluidic system was evaluated by analysis of reduced glutathione (GSH) and reactive oxygen species (ROS) in single HepG2 cells. The average analysis throughput of ROS and GSH in single cells was 16-18 cells min(-1).

Abstract:State of the art acoustofluidics typically treat micro-particles in a multi-wavelength range due to the scale limitations of the established ultrasound field. Here, we report a spatial selective acoustofluidic device that allows trapping micro-particles and cells in a wavelength scale. A pair of interdigital transducers with a concentric-arc shape is used to compress the beam width, while pulsed actuation is adopted to localize the acoustic radiation force in the wave propagating direction. Unlike the traditional usage of geometrical focus, the proposed device is designed by properly superposing the convergent section of two focused surface acoustic waves. We successfully demonstrate a single-column alignment of 15-μm polystyrene particles and double-column alignment of 8-μm T cells in a wavelength scale. Through proof-of-concept experiments, the proposed acoustofluidic device shows potential applications in on-chip biological and chemical analyses, where localized handing is required.

Abstract:In this article, we demonstrate single-layered, "microfluidic drifting" based three-dimensional (3D) hydrodynamic focusing devices with particle/cell focal positioning approaching submicron precision along both lateral and vertical directions. By systematically optimizing channel geometries and sample/sheath flow rates, a series of "microfluidic drifting" based 3D hydrodynamic focusing devices with different curvature angles are designed and fabricated. Their performances are then evaluated using confocal microscopy, fast camera imaging, and side-view imaging techniques. Using a device with a curvature angle of 180, we have achieved a standard deviation of +/- 0.45 mu m in particle focal position and a coefficient of variation (CV) of 2.37% in flow cytometric measurements. To the best of our knowledge, this is the best CV that has been achieved using a microfluidic flow cytometry device. Moreover, the device showed the capability to distinguish 8 peaks when subjected to a stringent 8-peak rainbow calibration test, signifying the ability to perform sensitive, accurate tests similar to commercial flow cytometers. We have further tested and validated our device by detection of HEK-293 cells. With its advantages in simple fabrication (i.e., single-layered device), precise 3D hydrodynamic focusing (i.e., submicrometer precision along both lateral and vertical directions), and high detection resolution (i.e., low CV), our method could serve as an important basis for high-performance, mass-producible microfluidic flow cytometry.

Abstract:High-throughput, high-precision single-stream focusing of microparticles has a potentially wide range of applications in biochemical analysis and clinical diagnosis. In this work, we develop a sheathless three-dimensional (3D) particle-focusing method in a single-layer microchannel. This novel microchannel consists of periodic high-aspect-ratio curved channels and straight channels. The proposed method takes advantage of both the curved channels, which induce Dean flow to promote particle migration, and straight channels, which suppress the remaining stirring effects of Dean flow to stabilize the achieved particle focusing. The 3D particle focusing is demonstrated experimentally, and the mechanism is analyzed theoretically. The effects of flow rate, particle size, and cycle number on the focusing performance were also investigated. The experimental results demonstrate that polystyrene particles with diameters of 5-20 μm can be focused into a 3D single file within seven channel cycles, with the focusing accuracy up to 98.5% and focusing rate up to 98.97%. The focusing throughput could reach up to ∼105 counts/min. Furthermore, its applicability to biological cells is also demonstrated by 3D focusing of HeLa and melanoma cells and bovine blood cells in the proposed microchannel. The proposed sheathless passive focusing scheme, featuring a simple channel structure, small footprint (9 mm × 1.2 mm), compact layout, and uncomplicated fabrication procedure, holds great promise as an efficient 3D focusing unit for the development of next-generation on-chip flow cytometry.

Abstract:Efficient and reliable manipulation of biological particles is crucial in medical diagnosis and chemical synthesis. Inertial microfluidic devices utilizing passive hydrodynamic forces in the secondary flow have drawn considerable attention for their high throughputs, low costs, and harmless particle manipulation. However, as the dominant mechanism, the inertial lift force is difficult to quantitatively analyze because of the uncertainties of its magnitude and direction. The equilibrium position of particles varies along the migration process, thus inducing the instabilities of particle separation. Herein, we present a designable inertial microfluidic chip combining a spiral channel with periodic expansion structures for the sheathless separation of particles with different sizes. The stable vortex-induced lift force arising from the periodic expansion and the Dean drag force significantly enhanced the focusing process and determined the final equilibrium position. The experimental results showed that over 99% of target particles could be isolated with the high target sample purity of 86.12%. In the biological experiment, 93.5% of the MCF-7, 89.5% of the Hela, and 88.6% of the A549 cells were steadily recovered with excellent viabilities to verify the potential of the device in dealing with biological particles over a broad range of throughputs. The device presented in this study can further serve as a lab-on-chip platform for liquid biopsy and diagnostic analysis.

Abstract:Flow focusing is an important hydrodynamic technique for cytometric analysis, enabling the rapid study of cellular samples to identify a variety of biological processes. To date, the majority of flow-focusing devices are fabricated using conventional photolithography or flame processing of glass capillaries. This article presents a suite of low-cost, millifluidic, flow-focusing devices that were fabricated using a desktop sterolithgraphy (SLA) 3D printer. The suite of SLA printing strategies consists of a monolithic SLA method and a hybrid molding process. In the monolithic SLA approach, 1.3 mm square millifluidic channels were printed as a single piece. The printed device does not require any post processing, such as bonding or surface polishing for optical access. The hybrid molding approach consists of printing a mold using the SLA 3D printer. The mold is treated to an extended UV exposure and oven baked before using PDMS as the molding material for the channel. To demonstrate the viability of these channels, we performed a series of experiments using several flow-rate ratios to show the range of focusing widths that can be achieved in these devices. The experiments are validated using a numerical model developed in ANSYS.

Abstract:Abstract Microfluidic focusing of particles (both synthetic and biological), which enables precise control over the positions of particles in a tightly focused stream, is a prerequisite step for the downstream processing, such as detection, trapping and separation. In this study, we propose a novel hydrodynamic focusing method by taking advantage of open v-shaped microstructures on a glass substrate engraved by femtosecond pulse (fs) laser. The fs laser engraved microstructures were capable of focusing polystyrene particles and live cells in rectangular microchannels at relatively low Reynolds numbers (Re). Numerical simulations were performed to explain the mechanisms of particle focusing and experiments were carried out to investigate the effects of groove depth, groove number and flow rate on the performance of the groove-embedded microchannel for particle focusing. We found out that 10-μm polystyrene particles are directed toward the channel center under the effects of the groove-induced secondary flows in low-Re flows, e.g. Re < 1. Moreover, we achieved continuous focusing of live cells with different sizes ranging from 10 to 15 μm, i.e. human T-cell lymphoma Jurkat cells, rat adrenal pheochromocytoma PC12 cells and dog kidney MDCK cells. The glass grooves fabricated by fs laser are expected to be integrated with on-chip detection components, such as contact imaging and fluorescence lifetime-resolved imaging, for various biological and biomedical applications, where particle focusing at a relatively low flow rate is desirable.

This approach avoids the selec on bias of looking at firms in or near default and therefore permits theories of ex ante capital structure determina on to be tested. We find that asset vola lity and growth op ons have significant posi ve impacts, while tangibility and size have nega ve impacts. This nega ve impact is in addi on to that of other firm characteris cs such as asset intangibility and asset vola lity. The results provide strong support for the tradeoff theory of capital structure.

Abstract:In recent decades, driven by the needs of industry and medicine, researchers have been investigating how to remove carefully from the main flow microscopic particles or clusters of them. Among all the approaches proposed, crossflow filtration is one of the most attractive as it provides a non‐destructive, label‐free and in‐flow sorting method. In general, the separation performance shows capture and separation efficiencies ranging from 70% up to 100%. However, the maximum flow rate achievable (μL/min) is still orders of magnitude away from those suitable for clinical or industrial applications mainly due to the low stiffness of the materials typically used. In this work, we propose an innovative hydrodynamic‐crossflow hybrid filter geometry, buried in a fused silica substrate by means of the femtosecond laser irradiation followed by chemical etching technique. The material high stiffness combined with the accuracy of our manufacturing technique allows the 3D fabrication of non‐deformable channels with higher aspect ratio posts, while keeping the overall device dimensions compact. The filter performance has been validated through experiments with both Newtonian (water‐based solution of microbeads) and non‐Newtonian fluids (blood), achieving separation efficiencies of up to 94% and large particles recovery rates of 100%, even at very high flow rates (mL/h).

Abstract:Focusing particles and cells into a tight stream is often required in order for continuous flow detection, counting and sorting. So far a variety of particle focusing methods have been developed in microfluidic devices, among which magnetic focusing is still relatively new. We develop in this work an approach to embedding symmetrically two repulsive permanent magnets about a straight rectangular microchannel in a PDMS-based microfluidic device. The closest distance between the magnets is limited only by the sizes of the embedded and holder magnets involved in the fabrication process. The developed device is used to implement and investigate the three-dimensional magnetic focusing of polystyrene particles in ferrofluid microflow with both the top- and side-view visualizations. The effects of flow speed and particle size on the particle focusing effectiveness are studied. The developed device is also applied to magnetically focus yeast cells in ferrofluid, which proves to be biocompatible as verified by a cell viability test. In addition, an analytical model is developed and found to be able to predict the experimentally observed particle and cell focusing behaviors with reasonable agreement.

Abstract:Microfluidic chips with the function of hydrodynamic focusing is widely used in the field of cell electroporation, droplet formation, drug screening and particle enrichment. Due to the small channel size of microfluidic chip, the existing processing methods have the defects of high fabrication cost or low dimensional consistency, the drawback is particularly prominent at small fabrication quantity. In this study, hydrodynamic focusing microfluidic chip was successfully fabricated by Liquid Crystal Display (LCD) mask photo-curing method with high efficiency and precision, the mechanism of curing parameters on the accuracy of microstructure was investigated, the hot pressing process was proposed to realize the bonding of cured photosensitive resin and glass substrate. By analyzing the focusing width of typical cross structure channel, a hydrodynamic focusing chip was designed. The effects of exposure time and single layer thickness on whole curing time was studied, the factors affecting the surface roughness were investigated, and the dimensional accuracy control method was proposed. The effect of bonding pressure on cured resin temperature was calculated and tested. The focusing width was verified by theoretical calculation and experiment. The mechanism for the inconsistency between the experimental focusing width and the theoretical focusing width was analyzed. The research proposed a solution of microfluidic design, fabrication, bonding and testing, which could provide a theoretical and methodological support for the efficient fabrication of microfluidic chips.

Abstract:The present work describes a novel microfluidic free-flow electrophoresis device developed by applying three-dimensional (3D) printing technology to rapid prototype a low-cost chip for micro- and nanoparticle collection and analysis. Accurate reproducibility of the device design and the integration of the inlet and outlet ports with the proper tube interconnection was achieved by the additive manufacturing process. Test prints were performed to compare the glossy and the matte type of surface finish. Analyzing the surface topography of the 3D printed device, we demonstrated how the best reproducibility was obtained with the glossy device showing a 5% accuracy. The performance of the device was demonstrated by a free-flow zone electrophoresis application on micro- and nanoparticles with different dimensions, charge surfaces and fluorescent dyes by applying different separation voltages up to 55 V. Dynamic light scattering (DLS) measurements and ultraviolet−visible spectroscopy (UV−Vis) analysis were performed on particles collected at the outlets. The percentage of particles observed at each outlet was determined in order to demonstrate the capability of the micro free-flow electrophoresis (μFFE) device to work properly in dependence of the applied electric field. In conclusion, we rapid prototyped a microfluidic device by 3D printing, which ensured micro- and nanoparticle deviation and concentration in a reduced operation volume and hence suitable for biomedical as well as pharmaceutical applications.

Abstract:This paper proposed a novel single-layer planar design of a microfluidic chip which utilizes inertial forces to realize continuous separating and three-dimensional (3D) focusing of particles. The theories of inertial microfluidics were analyzed and numerical simulation with COMSOL Multi-physics was verified by reported work. The results showed that both 3D focusing and separating of particles with a diameter of 1.0 μm and 9.9μm were obtained by changing the flow rates of inlets. The novel microfluidic design was robust, simple to perform and could easily be manufactured for laboratory and biomedical applications.
Fabrication of Micro/Nanofluidic Chip on Glass Substrate and Its Application for Electrokinetic Concentration of Proteins

Abstract:A simple and cost-effective double-lithography and double wet-etching method (abbriciated as double-etching technique) was developed for fabricating the full glass chips with hybrid micro- and nanochannels. The microchannels were firstly constructed on a glass substrate by standard UV photolithography and wet etching technique. After washing away the rest photoresist and spin coating of a new layer of photoresist on the microchannel-structured glass substrate, nanochannels with a depth less than 100 nm were prepared on the same substrate by second lithography and wet etching. Finally, the channel-structured glass substrate was sealed to a cover glass plate using room temperature bonding technique. Without clean room facilities and expensive facilities, fluidic chips with hybrid micro- and nanochannel network were fabricated with high yields. The analytical features of those fluidic chips were demonstrated by the successful concentration of fluorescein isothiocyanate(FITC)-labeled human serum albumin(HSA) via electrokinetic ion trapping. Enrichment factor of more than 200 was achieved for 0.5 mg/mL of FITC-HSA solutions within 30 s.
Optofluidic Flow Cytometer with In-Plane Spherical Mirror for Signal Enhancement

Abstract:Statistical analysis of the properties of single microparticles, such as cells, bacteria or plastic slivers, has attracted increasing interest in recent years. In this regard, field flow cytometry is considered the gold standard technique, but commercially available instruments are bulky, expensive, and not suitable for use in point-of-care (PoC) testing. Microfluidic flow cytometers, on the other hand, are small, cheap and can be used for on-site analyses. However, in order to detect small particles, they require complex geometries and the aid of external optical components. To overcome these limitations, here, we present an opto-fluidic flow cytometer with an integrated 3D in-plane spherical mirror for enhanced optical signal collection. As a result, the signal-to-noise ratio is increased by a factor of six, enabling the detection of particle sizes down to 1.5 µm. The proposed optofluidic detection scheme enables the simultaneous collection of particle fluorescence and scattering using a single optical fiber, which is crucial to easily distinguishing particle populations with different optical properties. The devices have been fully characterized using fluorescent polystyrene beads of different sizes. As a proof of concept for potential real-world applications, signals from fluorescent HEK cells and Escherichia coli bacteria were analyzed.

Abstract:High efficiency fabrication and integration of three-dimension (3D) functional devices in Lab-on-a-chip systems are crucial for microfluidic applications. Here, a spatial light modulator (SLM)-based multifoci parallel femtosecond laser scanning technology was proposed to integrate microstructures inside a given ‘Y’ shape microchannel. The key novelty of our approach lies on rapidly integrating 3D microdevices inside a microchip for the first time, which significantly reduces the fabrication time. The high quality integration of various 2D-3D microstructures was ensured by quantitatively optimizing the experimental conditions including prebaking time, laser power and developing time. To verify the designable and versatile capability of this method for integrating functional 3D microdevices in microchannel, a series of microfilters with adjustable pore sizes from 12.2 μm to 6.7 μm were fabricated to demonstrate selective filtering of the polystyrene (PS) particles and cancer cells with different sizes. The filter can be cleaned by reversing the flow and reused for many times. This technology will advance the fabrication technique of 3D integrated microfluidic and optofluidic chips.
Efficient Microfluidic Particle Separation Using Inertial Pre-Focusing and Pre-Separation Structures Without Sheath Flow

Abstract:This study introduces an innovative microfluidic particle separation technique that integrates inertial focusing with deterministic lateral displacement (DLD) on a single chip, significantly enhancing the efficiency of particle separation. This new method completes flow rate matching through a designed pre-focusing and pre-separation inertial structure, avoiding the use of sheath flow. The process involves a sequence of channels: a rectangular channel, a contraction-expansion array (CEA), a lateral separation channel, and another rectangular channel. This three-stage inertial method shortens the focusing channel length and reduces the pressure on subsequent separation stages, streamlining the separation process. It has been demonstrated to separate 20 μm particles from a mixture containing both 10 μm and 20 μm particles with remarkable precision. The technique achieves a 100% separation efficiency, ensuring all target particles are correctly isolated, and a 96.1% separation purity, indicating that the isolated particles are almost entirely free from contaminants. By eliminating the need for sheath flow, this method simplifies the apparatus and reduces operational complexity, offering significant advantages over traditional particle separation techniques. The high efficiency and purity levels achieved by this method highlight its potential for a wide range of applications in fields requiring precise particle separation, such as medical diagnostics, environmental monitoring, and industrial processing.
