Abstract:In this report, we present a highly efficient aromatic chlorination that is catalysed by 1,4-diazabicyclo[2.2.2]octane (DABCO) with N -chlorosuccinimide (NCS) as the chloro source. The mild conditions, easy-availability and stability of the catalyst and reagents, as well as good functional-group tolerance, showed the approach to be a versatile protocol for the late-stage aromatic chlorination of complex natural products and drugs. A plausible catalytic mechanism was constructed through comprehensive computational studies.

Abstract:An array of novel 1,4-diazobicyclo[2.2.2]octane (DABCO) based ionic liquids were developed and used as recyclable catalysts for the aza-Michael addition at room temperature without any organic solvent. [DABCO-PDO][OAc] was found to be the most efficient catalyst, and the amount of catalyst was only 10 mol %. Various amines reacted with a wide range of α,β-unsaturated amides, smoothly affording target products in good to excellent yields within hours. Moreover, the catalyst could be reused up to eight times, still maintaining a high catalytic activity. Finally, a plausible mechanism was proposed. FTIR and computational chemistry were used to verify the catalytic mechanism.

Abstract:Functionalized ketene dithioacetals are important intermediates in organic synthesis.α-Functionalization reactions based on them provide potential precursors for the construction of usefully carbo-and heterocyclic compounds.In this paper,a versatileα-bromination of functionalized ketene dithioacetals was developed.In the presence of H2 O,the reactions of functionalized ketene dithioacetals(1) with CuBr 2 in THF at room temperature gave variousα-bromo ketene dithioacetals(2) in excellent yields with high regioselectivities.The mechanism of thisα-bromination is proposed to undergo an addition of bromine formed in situ to compound 1 and sequontial elimination of theα-proton.The simplicity of execution,mild conditions,inexpensive,easily usable brominating agent and high yields,make this synthetic method attractive for practical applications.

Abstract:A catalytic asymmetric brominative dearomatization reaction of benzofuran derivatives was achieved by using hydroquinidine 1,4-phthalazinediyl diether [(DHQ)2PHAL] as the catalyst and N-bromoacetamide (NBAc) as the brominating reagent. A series of brominated spiro[benzofuran-2,5'-oxazoles] bearing two contiguous stereogenic centers were obtained in high yields (up to 99%) with excellent enantioselectivity (up to 97% ee).

Abstract:A mild and efficient method for the synthesis of branched N-allylic lactams and related N-heteroaromatics was reported. This strategy involves a DABCO-catalyzed SN2′-SN2′ type N-allylic alkylation with Morita−Baylis−Hillman (MBH) carbonates. A wide scope of lactams and related N-heterocycles as well as MBH carbonates are well tolerated, and various branched N-substituted products were obtained in moderate to excellent yields. The utility of this method was further demonstrated by the gram-scale synthesis and diverse transformation of the products.

Abstract:Herein, an electrophilic halogenation of di- or trisubstituted allenoates and 3-alkynoates is demonstrated with NBS and NIS reagents under DABCO catalysis. The protocol affords densely functionalized 1,4-dicarbonyl 3-haloalkenes in good yields with excellent (E)-stereoselectivity (up to 83% yield, >20:1 dr). The role of the Brønsted base in the unusual transformation is demystified through control experiments and computational studies. The deliverables are identified as good synthons for various synthetic transformations and valuable targets for biologically active pyridazine scaffolds.

Abstract:A practical, efficient Pd(OAc)(2)/Cu(II)/PPh3 cocatalytic system has been developed. With only 1 mol% Pd(OAc)(2), 20 mol% copper(II) salt, and 0.5 equiv of inexpensive PPh3 as ligand, the direct arylation of benzoazoles with aryl bromides could be performed smoothly in mild condition, affording the desired arylated benzoazoles in good yields (75-93%).

Abstract:Aromatic bromination catalysed by 0.5‒10 mol% of D-camphorsulfonic acid‒BiCl3 with N-bromosuccinimides (NBS) was carried out in MeCN under air conditions, and the procedure was extended to the reactions with N-chlorosuccinimides (NCS) and N-iodosuccinimides (NIS). The halogenation of some drugs and natural products was also attempted. One-pot bromination / Suzuki-Miyaura cross-coupling and bromination / Sonogashira coupling reactions were achieved without the removal of the solvent.

Abstract:A Lewis base catalyst Trip-SMe (Trip = triptycenyl) for electrophilic aromatic halogenation using N-halosuccinimides (NXS) is introduced. In the presence of an appropriate activator (as a noncoordinating-anion source), a series of unactivated aromatic compounds were halogenated at ambient temperature using NXS. This catalytic system was applicable to transformations that are currently unachievable except for the use of Br2 or Cl2: e.g., multihalogenation of naphthalene, regioselective bromination of BINOL, etc. Controlled experiments revealed that the triptycenyl substituent exerts a crucial role for the catalytic activity, and kinetic experiments implied the occurrence of a sulfonium salt [Trip-S(Me)Br][SbF6] as an active species. Compared to simple dialkyl sulfides, Trip-SMe exhibited a significant charge-separated ion pair character within the halonium complex whose structural information was obtained by the single-crystal X-ray analysis. A preliminary computational study disclosed that the π system of the triptycenyl functionality is a key motif to consolidate the enhancement of electrophilicity.

Abstract:On the basis of the chiral ion pair between catalytic anionic stereogenic-at-cobalt(III) complexes and halonium ion intermediates, an asymmetric halogenation/semipinacol rearrangement reaction has been established using N-bromosuccinimide as the halogen source. This protocol provides an alternative approach for accessing a diverse set of chiral β-bromocycloketones in good yields with high enantioselectivities (≤96% yield, 95.5:4.5 er).

Abstract:Deoxygenative difunctionalization of carbonyls affords a straightforward and effective route to construct geminal dual functionalized motifs. However, the research in this field is very challenging due to the strong bond dissociation energies of the C-O double bond or the subsequently formed C-O bond. Herein, we report a highly efficient deoxygenative haloboration of aldehydes to generate secondary α-haloboronates. Meanwhile, the difficult-to-obtain tertiary α-haloboronates can be also readily prepared via the same strategy with ketones. Furthermore, enantioselective chloroboration of carbonyls was successfully achieved to give chiral secondary or tertiary α-chloroboronates, the important intermediates to access enantioenrich multisubstituted stereocenters. These versatile products can be surprisingly attained through this simple and mild process with remarkable substrate scope expansion and functional group tolerance. Additionally, these reactions can proceed well on large scales, giving more practical values in the application.
