Abstract:Organohalides are vital organic building blocks with applications spanning various fields. However, direct halogenation of certain neutral or unreactive substrates by using solely the regular halogenating reagents has proven challenging. Although various halogenation approaches via activating halogenating reagents or substrates have emerged, a catalytic system enabling broad substrate applicability and diverse halogenation types remains relatively underexplored. Inspired by the halogenation of arenes via thianthrenation of arenes, here we report that thianthrene, in combined use with trifluoromethanesulfonic acid (TfOH), could work as an effective catalytic system to activate regular halogenating reagents (NXS). This new protocol could accomplish multiple types of halogenation of organic compounds including aromatics, olefins, alkynes and ketones. The mechanism study indicated that a highly reactive electrophilic halogen thianthrenium species, formed in situ from the reaction of NXS with thianthrene in the presence of TfOH, was crucial for the efficient halogenation process.

Abstract:Aromatic bromination catalysed by 0.5‒10 mol% of D-camphorsulfonic acid‒BiCl3 with N-bromosuccinimides (NBS) was carried out in MeCN under air conditions, and the procedure was extended to the reactions with N-chlorosuccinimides (NCS) and N-iodosuccinimides (NIS). The halogenation of some drugs and natural products was also attempted. One-pot bromination / Suzuki-Miyaura cross-coupling and bromination / Sonogashira coupling reactions were achieved without the removal of the solvent.

Abstract:A mild and general procedure for the difluoromethylation of organochalcogen compounds has been accomplished via sulfoxide and selenoxide-directed [3,3]-sigmatropic rearrangement/Haller-Bauer reaction with difluoroenoxysilanes. The reactions showed good functional group compatibility, wide substrate applicability, and facile scalability. The synthetic utility is highlighted by the iterative use of this protocol to construct multi-CF2H-containing chalcogenides and the late-stage modification of pharmaceuticals.

Abstract:Utilizing simple precursors for the exquisite construction of molecules with diversity and complexity is urgent. Herein, a novel A3-X type reaction for functionalized 3-haloquinolines has been developed by PhI (OAc)2-mediated four-component tandem addition/cyclization/oxidation/aromatization process with simple aromatic amine, aldehyde, alkyne and halide salt. Broad functional group tolerance, very mild condition, easy for further transformation illustrate the concision, high efficiency and practicability of this A3-X type reaction.

Abstract:We have successfully developed a highly enantioselective hydrogenation of various 3-aryl and 3-methyl maleinimides to access enantiomerically pure 3-substituted succinimides catalyzed by Rh/bisphosphine-thiourea (Zhao-Phos). This efficient catalytic system furnished the desired 3 substituted succinimide products with high yields and enantioselectivities (up to 99% yield, full conversions, almost all 3-aryl succinimide products up to 99% ee, and 3-methyl succinimide with 83% ee). Our catalytic system has a strong substrate tolerance and generality. Whether the N-substituted group of maleinimides is H or other protecting groups, the maleinimides were hydrogenated well (up to >99% ee, 99% yield). Moreover, the hydrogenation succinimide products can be readily utilized for the construction of biologically active molecules, such as chiral amides and pyrrolidines.

Abstract:Comprehensive Summary Palladium‐catalyzed regioselective amination of unactivated alkene remains a challenge and is of great interest. Herein, a three‐component coupling of unactivated alkene, sulfonamide and N‐halo compound accessing to vicinal haloamine has been conceived. This aminohalogenation represents a modular and regioselective strategy. The electrophilic halogenating agent enables regioselective anti‐Markovnikov aminopalladation and facilitates subsequent halogenation event. And This protocol is characterized by gram‐scale syntheses and late‐stage functionalizations. Of note, the recovered byproduct phthalimide allows for reusing by conversion to starting material. Abstract:A highly regioselective iodosulfonylation of allenes in the presence of CuI and 1,10-phenanthroline has been developed for the synthesis of various useful ( E)-α-iodomethyl vinylsulfones in moderate to excellent yields. This practical reaction is fast, operationally simple, and in particular, proceeds under very mild conditions to afford the target products with high regio- and stereoselectivity. The selectivity was illustrated by a conceptual DFT analysis.

Abstract:Aminohalogenation reaction of β-nitrostyrenes with N-halosuccinimides (N-chloro and N-bromosuccinimides) has been successfully conducted by using nickel acetate as a catalyst in the presence of potassium carbonate and succinimide as co-additives. The reaction was easily performed at room temperature under nitrogen gas protection to give dihalorinaed haloamino products in good to excellent yields (60%–98%). The structure has been confirmed by X-ray crystal structure analysis.

Triptycenyl Sulfide: A Practical and Active Catalyst for Electrophilic Aromatic Halogenation Using N-Halosuccinimides

Abstract:A Lewis base catalyst Trip-SMe (Trip = triptycenyl) for electrophilic aromatic halogenation using N-halosuccinimides (NXS) is introduced. In the presence of an appropriate activator (as a noncoordinating-anion source), a series of unactivated aromatic compounds were halogenated at ambient temperature using NXS. This catalytic system was applicable to transformations that are currently unachievable except for the use of Br2 or Cl2: e.g., multihalogenation of naphthalene, regioselective bromination of BINOL, etc. Controlled experiments revealed that the triptycenyl substituent exerts a crucial role for the catalytic activity, and kinetic experiments implied the occurrence of a sulfonium salt [Trip-S(Me)Br][SbF6] as an active species. Compared to simple dialkyl sulfides, Trip-SMe exhibited a significant charge-separated ion pair character within the halonium complex whose structural information was obtained by the single-crystal X-ray analysis. A preliminary computational study disclosed that the π system of the triptycenyl functionality is a key motif to consolidate the enhancement of electrophilicity.

Abstract:We herein describe a nickel-catalyzed cyanation reaction of aryl/alkenyl halides with alkyl isocyanides. Both aryl/alkenyl iodines and bromides were found to be competent electrophiles that reacted with alkyl isocyanides, affording nitrile compounds in moderate to good yields. A range of functional groups including halogens as well as hydroxyl, formyl, and acetamino groups were fairly compatible with the nickel catalysis. This protocol featured broad functional group tolerance, simple reaction conditions, and gram-scale synthesis.

Abstract:The most prevalent strategies for preparing halogenated alkyl aromatics face one remarkable challenge: low regioselectivity. Therefore, the highly regioselective halogenation of alkyl arenes is still an unsolved problem. Herein, a highly regioselective halogenation of alkyl aromatics through nitromethane-assisted Bronsted acid catalysis is disclosed. Under the mild reaction conditions, a wide diversity of halogenated alkyl aromatics is obtained in high yields with excellent regioselectivity. The hydrogen bonding of protonic acid with the solvent nitromethane promotes this transformation.

Abstract:Herein, we describe a nickel-catalyzed divergent formylation and carboxylation reaction of aryl halides with isocyanides. A rich array of aromatic aldehydes and carboxylic acids can be, respectively, accessed in moderate to good yields. Some sensitive functional groups such as hydroxyl, iodine, cyano, and indolyl are fairly tolerant of nickel catalysis. In the carboxylation reactions, the combination of isocyanide and H2O is first employed as a promising carbonyl surrogate instead of gaseous CO and CO2.
