Abstract:Diazo compounds had been widely employed as a class of environment friendly and highly efficient coupling reagents in transition-metal-catalyzed C-H bonds insertion reactions.Recently, C-H alkylation of various arenes and heteroarenes has been achieved enormous successes using diazo compounds as the alkylation reagents under a new catalytic model.The recent progress in diazo compounds mediated transition-metal-catalyzed C-H alkylation and tandem C-H alkylation/cyclization is reviewed, including reaction mechanism and synthetic applications.Furthermore, the prospects of this reaction are also discussed.

Abstract:Utilizing simple precursors for the exquisite construction of molecules with diversity and complexity is urgent. Herein, a novel A3-X type reaction for functionalized 3-haloquinolines has been developed by PhI (OAc)2-mediated four-component tandem addition/cyclization/oxidation/aromatization process with simple aromatic amine, aldehyde, alkyne and halide salt. Broad functional group tolerance, very mild condition, easy for further transformation illustrate the concision, high efficiency and practicability of this A3-X type reaction.

Abstract:On the basis of the chiral ion pair between catalytic anionic stereogenic-at-cobalt(III) complexes and halonium ion intermediates, an asymmetric halogenation/semipinacol rearrangement reaction has been established using N-bromosuccinimide as the halogen source. This protocol provides an alternative approach for accessing a diverse set of chiral β-bromocycloketones in good yields with high enantioselectivities (≤96% yield, 95.5:4.5 er).

Abstract:Herein, an electrophilic halogenation of di- or trisubstituted allenoates and 3-alkynoates is demonstrated with NBS and NIS reagents under DABCO catalysis. The protocol affords densely functionalized 1,4-dicarbonyl 3-haloalkenes in good yields with excellent (E)-stereoselectivity (up to 83% yield, >20:1 dr). The role of the Brønsted base in the unusual transformation is demystified through control experiments and computational studies. The deliverables are identified as good synthons for various synthetic transformations and valuable targets for biologically active pyridazine scaffolds.

Abstract:The textbook alkene halogenation reaction establishes straightforward access to vicinal dihaloalkanes. However, a robust catalytic method for dihalogenizing electron-deficient olefins in an enantioselective manner is still under development, and its mechanism remains controversial. Herein, we disclose efficient regio-, anti-diastereo-, and enantioselective dibromination, bromochlorination, and dichlorination reactions of enones catalyzed by a chiral N,N'-dioxide/Yb(OTf)3 complex. With the combination of electrophilic halogen and halide salts as halogenating agents, an array of homo- and heterodihalogenated derivatives is achieved in moderate to good enantioselectivities. Moreover, DFT calculations reveal that a novel triplet halo-radical pylon intermediate is probable in accounting for the exclusive regio- and anti-diastereoselectivity.

Abstract:A series of new dihydrobenzooxophosphole-based Lewis base organocatalysts were designed and synthesized. They are shown to be effective in trichlorosilane-mediated stereoselective conjugate reductions of C=C bonds. DFT calculations reveal that the strong hydrogen bonds between the amide linker and the chloride on silicon in the transition state contribute to the high reactivity of the catalyst.

Abstract:An electrophilic thiocyano semipinacol rearrangement of allylic alcohols has been achieved for the first time by using N-thiocyano-dibenzenesulfonimide (NTSI). This approach provides a direct, simple, and efficient strategy for the formation of thiocyano carbonyl compounds with moderate to excellent yields. Meanwhile, an all-carbon quaternary center was rapidly constructed. In addition, an asymmetric version of this tandem reaction was preliminarily investigated.

Abstract:An efficient [4+2] annulation reaction of o‐acylamino‐aryl Morita‐Baylis‐Hillman carbonates with isatylidene malononitriles is reported. Under mild conditions, we have successfully obtained a series of underreported spiro[indolin‐3,2′‐quinoline] framework compounds. A novel [4+2] annulation reaction between o‐acylamino‐aryl MBH carbonates and isatylidene malononitriles has been successfully developed. This methodology exhibits excellent substrate tolerance, affording a series of spiro[indolin‐3,2′‐quinoline] framework compounds with yields up to 98%. The scalability of the reaction is also noteworthy, as evidenced by the 95% yield of the final product upon a 50‐fold scale‐up.

Abstract:A highly enantioselective cascade carbonylation/annulation of benzyl bromides, CO, and vinyl benzoxazinanones under mild conditions has been established by Pd/chiral Lewis base relay catalysis, providing an efficient method to assemble chiral quinolinones from readily available starting materials in good yields with excellent diastereo- and enantioselectivities. The palladium catalyst plays two roles in this reaction, enabling both the carbonylation process and the generation of the zwitterionic π-allyl palladium intermediate.
An Efficient New Methodology for the Synthesis of 1-Functionalized 2-Halo-2-alkenes via Hydrohalogenation Reaction of Electron-Deficient Allenes

Abstract:The hydrohalogenation reactions of electron-deficient allenes, such as 1,2-allenic ketones, carboxylic acids, esters, amides, nitriles, sulfoxides (with the use of AlX3/H2O), sulfones, and phosphine oxides with MX (X = I, Br, Cl, M = Na, Li) in different solvents, such as HOAc, CF3COOH-HOAc (1 : 1), CF3COOH and THF were studied. The reaction is nucleophilic conjugate addition in nature and provides efficient entries to 1-functionalized 2-halo-2-alkenes. The β,γ- over α,β-selectivity and stereoselectivity depend on the solvent(s) and the temperature applied. In most cases, the Z- and E-isomers can be easily separated with chromatography on silica gel. The subsequent cross coupling reaction with organometallic reagents and terminal alkynes showed the synthetic potential of these products.

Abstract:A novel copper(II) trifluoromethanesulfonate-catalyzed intermolecular cascade annulation strategy for the construction of a great variety of pentacyclic compounds possessing valuable carbazole fragments was developed employing propargylic alcohols and (Z)-2-styryl-1H-indoles as the initial substrates. This protocol, which entails a sequential Meyer-Schuster rearrangement/isomerization/-cyclization cascade, enables facile and atom-economical access to various pentacyclic compounds with broad functional-group tolerance in good yields under mild conditions. The conversion could be efficiently scaled up to gram quantities, accentuating a potential application of this work.

Abstract:Mono and dichloro‐substituted organic molecules are shown to possess significant roles in various fields, especially in medicinal chemistry. One approach to synthesizing these compounds involves the selective dechlorination of easily prepared trichloromethyl compounds. Nevertheless, developing a practical approach that allows selective mono‐ and dihydrodechlorination is a challenging task. Herein, we introduced a method for selective mono‐ and dihydrodechlorination of trichloroacetamides and acetates which was promoted by two different Lewis base‐boryl radicals. Accordingly, 4‐dimethylaminopyridine (DMAP)‐borane enabled mono‐substitution of chlorine atom by hydrogen while dihydro‐substitution of chlorine atoms was promoted by N‐heterocyclic carbene (NHC)‐boryl radical. Using deuterated Lewis base‐borane as deuterium atom source, mono‐ and dideuterodechlorination were also successfully took place. This protocol features broad substrate scope and operates under mild reaction conditions.

Abstract:Asymmetric synthesis of 3-hydroxyl-2-alkanones is achieved via one-pot, tandem organocatalytic aminoxylation of aldehydes and chemoselective CH2N2-induced homologation. Accelerating effect of water is observed in α-aminoxylation. MgCl2, a Lewis acid additive, improves the chemoselectivity of the diazomethane homologation to 6:1 in favor of ketone.

Abstract:We report a photochemical method for the semipinacol rearrangement of unactivated allylic alcohols. Aliphatic as well as aromatic groups participate as migrating groups, yielding a variety of α,α-disubstituted ketones. The reaction proceeds under mild conditions and is compatible with ethers, esters, halides, nitriles, carbamates, and substituted arenes. The operationally simple and fully catalytic conditions prescribe 1 mol% benzothiazinoquinoxaline as organophotocatalyst, 0.5 mol% Co-salen, and 10 mol% lutidinium triflate and, importantly, display reactivity complementary to procedures employing Brønsted acid. We showcase the utility of the protocol in late-stage drug diversifications.
