Abstract:I am a bioanalytical chemist using cryo-electron microscopy to investigate biological and nanomaterials. I am interested in the mechanisms by which peptides specifically bind nanoparticles. Additionally, I am interested in the mechanisms by which peptides template the coalescence of atoms into nanoparticles with defined properties. I have always been interested in how drugs work at a molecular level, especially antibiotics. In my opinion, one of the most interesting mechanism-of-action stories in the literature centers around nisin, whose pore forming ability was difficult to recreate in vitro until it was discovered to require lipid II. This paper began as a passion project, for many, to discover new antifungal compounds from nature and diversify our antifungal drug arsenal. I loved working on this project because it was an exciting interdisciplinary story passed on from one researcher to anothereach adding their own diverse perspectives. The aberrant formation of insoluble plaques and fibrils by proteins comprises a pathogenic process in numerous neurodegenerative diseases. Recent proposals link protein aggregation to liquid–liquid phase separation (LLPS). In our current study, we developed a bioluminescent equivalent of a fluorescent intercalating dye, “LUMID”, that switches from blue- to green-light emission upon binding nonspecifically to DNA. LUMID is easily combined with isothermal amplification to enable sensitive, sequence-specific detection of DNA with a smartphone-based readout. My Ph.D. work focused on developing water-soluble azobenzene photoswitches and applying them to photocontrol biological systems. My goal is to work on the interface of chemistry and biology and to use both fields to manipulate and control biological targets to help us unravel their mechanisms or design new therapeutics. Since 1921, insulin has been a life-saving drug for diabetes treatment, but its intricate structure has hindered the development of new types. In my Ph.D. research, we pioneered a phage display method for rapidly selecting novel insulin analogs, using omniligase-1. I have long been fascinated by the tripartite relationship among a host, gut microbes, and small molecules... In this paper, we mined multiple novel gut-derived nisin-like lantibiotics from public gut bacterial genomes and expressed and tested them against human gut pathogens and commensal bacteria. Growing evidence now suggests that RNA plays an active role in genetic regulation via various RNA modifications. My research interest lies in the functional impact of modifications like inosine, m5C, m6A, m1A, and phosphorothioate, both in coding and noncoding RNAs.
