Abstract:An efficient metal-free protocol has been developed for the sp(3) C-H bond oxidation of 2-methylquinolines in the presence of PIDA under microwave irradiation, providing 2-quinolinecarboxaldehydes with moderate to high yields and excellent selectivities. A wide range of 2-quinolinecarboxaldehydes with different functional groups have been synthesized. A tentative mechanism has been presented to explain this highly selective oxidation process. Abstract:A one-pot, microwave-assisted protocol has been developed for the synthesis of 5,6-dihydroquinazolinones that incorporate structural fragments from chalcones, acetylacetoacetate, ammonium formate and formamide. This process generates two rings, two carbon–carbon and three carbon–nitrogen bonds and does not require the use of chromatographic purification. The dihydroquinazolinones were efficiently aromatized without the need for metal-based oxidants by a microwave-assisted halogenation–elimination sequence in the presence of N-bromosuccinimide, again in the absence of chromatographic purification.

Abstract:The synthesis of bifunctional pyridine and quinolione derivatives were investigated using terephthalic and isophthalic aldehydes as a precursor. The reaction proceeds under microwave irradiation with good yield (70-92%) and short reaction time (7-9 min.). We provide a rapid and efficient method of synthesizing a range of bifunctional monocyclic and bicyclic products related to 1,4-dihydropyridines (1,4-DHPs).

Abstract:A new method for synthesis of synthesis 6-hydroxy-3, 4-dihydro-2(lH) quinolinone was investigated with higher yield and simpler operation. The title compound was prepared starting from 3-chloropropionyl chloride, via cyclizing, nitration, reduction, diazotization and hydrolysis. The total yield is 67.7%, and the yield of each step is between 86.5% -99.6%. 6-Hydroxy-3, 4-dihydro-2(1H)quinolinone was characterized by MS and 1H NMR.

Abstract:An effective synthesis of 3,4-dihydropyrimidin-2(1H)-one derivatives using ionic liquid 1-n-butyl-3-methylimmidazolium hexafluorophosphate ([bmim]PF6) as the catalyst from aldehydes, 1,3-dicarbonyl compounds and urea (thiourea) under microwave irradiation is described. This new method has the advantage of high yields (80-95% for aromatic aldehydes) and short reaction time (3-5 min).

Abstract:A series of nine new 2,3-disubstituted 4(3H)-quinazolin-4-one derivatives was furnished starting from the 2-propyl-4(3H)-quinazo-line-4-one (2). The reinvestigation of the key starting quinazolinone 2 was performed under microwave irradiation (MW) and solvent-free conditions. Combination of MW and phase-transfer catalysis using tetrabutylammonium benzoate (TBAB) as a novel neutral ionic catalyst was used for carrying out N-alkylation and condensation reactions of compound 2 as a simple, efficient and eco-friendly technique. The structure of the synthesized compounds was elucidated using different spectral and chemical analyses. In vitro antimicrobial activity of the compounds was investigated against four bacterial and two fungal strains; very modest activity was achieved. Some of the synthesized compounds were screened for their antitumor activity against different human tumor cell lines. The screened compounds exhibited a significant antitumor activity on some of the cancer cell lines, melanoma (SK-MEL-2), ovarian cancer (IGROV1), renal cancer (TK-10), prostate cancer (PC-3), breast cancer (MCF7) and colon cancer (HT29). The most active, even more active than the reference 5-fluorouracil, were found to be ethyl 4-[(4-oxo-2-propylquinazolin-3(4H)-yl)methyl]benzoate (3c), 3-{2-[6-(pyrrolidin-1-yl-sulfonyl)-1,2,3,4-tetrahydroquinoline]-2-oxoethyl}-2-propylquinazolin--4(3H)-one (3e), N'-[(E)-(2H-1,3-benzodioxo-5-yl)methylidene]-2-(4-oxo-2-propylquinazolin-3(4H)-yl)acetohydrazide (10a), N'-[(E)-(4-hydroxyphenyl)methylidene]-2-(4-oxo-2-propylquinazo-lin-3(4H) -yl)acetohydrazide (10b) and N'-[(E)-(4-nitrophenyl)methyl idene]-2-(4-oxo-2-propylquinazolin-3(4H)-yl)acetohydrazide (10c).

Abstract:A green, rapid, and efficient method was developed for synthesizing quinazolinone derivatives from substituted 2-halobenzoic acids and amidines via microwave-assisted iron-catalyzed cyclization with or without ligand in water (methods A and B) or DMF (methods C and D). With these methods, moderate to high yields of the desired products can be obtained from even inactive substrates, such as guanidines. To the best of our knowledge, this is the first report on the synthesis of N-heterocyclic compounds by iron-catalyzed C-N coupling in aqueous media.

Abstract:Microwave-assisted organic synthesis (MAOS) has emerged as a transformative technique in organic chemistry, significantly enhancing the speed, efficiency, and selectivity of chemical reactions. In our research, we have employed microwave irradiation to expedite the synthesis of quinazolinones, using water as an eco-friendly solvent and thereby adhering to the principles of green chemistry. Notably, the purification of the product was achieved without the need for column chromatography, thus streamlining the process. A key innovation in our approach is using aldehyde bisulfite adducts (Bertagnini's salts) as solid surrogates of aldehydes. Bertagnini's salts offer several advantages over free aldehydes, including enhanced stability, easier purification, and improved reactivity. Green metrics and Eco-Scale score calculations confirmed the sustainability of this approach, indicating a reduction in waste generation and enhanced sustainability outcomes. This methodology facilitates the synthesis of a diverse array of compounds, offering substantial contributions to the field, with potential for widespread applications in pharmaceutical research and beyond.

Abstract:An efficient ring-closing reaction was developed for the synthesis of spirocyclopropane quinolinediones from 2,2-disubstituted 2-benzoylacetamides in the presence of [bis-(trifluoroacetoxy)iodo]benzene (PIFA). This reaction proceeds at room temperature in the absence of a metal catalyst to provide spirocyclopropane quinolinediones in good to excellent yields. Furthermore, these compounds were readily converted to the corresponding pyrrolo[3,2-c] quinolinones to provide tricyclic ring structures similar to those found in the natural products martinellic acid and martinelline via an intermolecular amine ringopening cyclization reaction.

Abstract:Benzotriazole-activated biotin was utilized as a versatile building block for the efficient and convenient synthesis of novel biotin conjugates with quinolone antibiotics via amino acid linkers under microwave irradiation. The products are obtained in yields of 70–91% with retention of chirality.

Abstract:Atom- and step-economy in IBX assisted diversity-oriented synthesis is achieved with a versatile AQ auxiliary α-amino acid analogs offering rapid access to polycyclic spiro-quinolines featuring a quaternary stereocenter in 20%–91% yields under mild conditions via 7,8-dearomatization of quinolines. Free of a preinstalled activation group is highlight of this intramolecular oxidation spiroannulation tandem reaction. This type of N-heterospirocycles, traditionally difficult to access, may open the door to a potentially interest scaffold for synthetic and medicinal chemistry.

Abstract:ective: The quinoline scaffold is widely acknowledged as a critical framework for the development of novel pharmaceuticals. The derivatives of quinoline have antifungal, antibacterial, antitumor, and anticancer effects. Over the past 30 years, bacterial and fungal infections have grown significantly. Developing new infectious illness treatments is a major issue. As antimicrobials, we produced substituted 2-chloroquinoline compounds. The antimicrobial screening shows moderate activities against used microbes. Methods: By employing microwave irradiation, 2-chloro-5,6-dimethyl-3-(((substituted-benzylidene)hydrazono)methyl)quinoline can be synthesized quickly and efficiently. One-pot reaction between ( E )-2-chloro-3-(hydrazonomethyl)-7,8-dimethylquinoline and substituted Carbaldehyde in glacial acetic acid and MeOH as a solvent yields the anticipated outcome. Results and Discussion: The MIC of produced compounds was determined by broth dilution. All fifteen ( I–XV ) synthesized compounds were evaluated for their antibacterial and antifungal activity in vitro . Compounds ( I–VII ), ( X–XIII ), and ( XIV ) have greater antibacterial activity against Staphylococcus aureus and Streptococcus pyogenes Gram-positive bacteria, Compounds ( IV ), ( VII ), ( XIII ), ( XIV ), and ( XV ) have demonstrated effective antibacterial activity against Escherichia coli and Pseudomonas aeruginosa Gram-negative bacteria. Compounds ( I ), ( IV ), ( V ), and ( XI ) exhibited moderate antifungal activity against C. albicans , A. niger , and A. clavatus . Conclusions: A powerful one-pot approach has been designed to synthesize substituted quinolones. Microwave-assisted synthesis greatly accelerates reaction rate and reduces product impurityand shows modest in vitro antimicrobial and antifungal activity.

Abstract:γ-Carbolines were obtained in a simple one-pot Fischer arting from N-acetyl-3-bromo-4-piperidone hydrobro-reaction starting from N-acetyl-3-bromo-4-piperidone hydrobromide and substituted arylhydrazine hydrochlorides in acidic media under microwave irradiation or by a thermal process. The optimization procedures are reported in detail, and the results from micro-wave processes are compared with conventional ones.

Abstract:Quinazolinone is a preferred structural motif with notable pharmacological activity that is present in a wide range of naturally occurring compounds. A microwave assisted tandem cyclooxidative method has been developed to afford quinazolinones via recyclable ionic liquid supported copper catalyst. This sustainable method exhibits operational simplicity, rapid, clean, energy efficient route and a variety of 2-substituted quinazolinones are obtained in excellent yields. In addition, this innovative approach enables us to develop a library of nitriles in an environment friendly synthetic protocol. Moreover, the catalyst can be recycled and reused up to three consecutive cycles without any significant loss of catalytic activity. Further organic transformation of the synthesized quinazolinones was carried out to afford reported as well as novel bioactive heterocyclic compounds.
