Abstract:ective: The quinoline scaffold is widely acknowledged as a critical framework for the development of novel pharmaceuticals. The derivatives of quinoline have antifungal, antibacterial, antitumor, and anticancer effects. Over the past 30 years, bacterial and fungal infections have grown significantly. Developing new infectious illness treatments is a major issue. As antimicrobials, we produced substituted 2-chloroquinoline compounds. The antimicrobial screening shows moderate activities against used microbes. Methods: By employing microwave irradiation, 2-chloro-5,6-dimethyl-3-(((substituted-benzylidene)hydrazono)methyl)quinoline can be synthesized quickly and efficiently. One-pot reaction between ( E )-2-chloro-3-(hydrazonomethyl)-7,8-dimethylquinoline and substituted Carbaldehyde in glacial acetic acid and MeOH as a solvent yields the anticipated outcome. Results and Discussion: The MIC of produced compounds was determined by broth dilution. All fifteen ( I–XV ) synthesized compounds were evaluated for their antibacterial and antifungal activity in vitro . Compounds ( I–VII ), ( X–XIII ), and ( XIV ) have greater antibacterial activity against Staphylococcus aureus and Streptococcus pyogenes Gram-positive bacteria, Compounds ( IV ), ( VII ), ( XIII ), ( XIV ), and ( XV ) have demonstrated effective antibacterial activity against Escherichia coli and Pseudomonas aeruginosa Gram-negative bacteria. Compounds ( I ), ( IV ), ( V ), and ( XI ) exhibited moderate antifungal activity against C. albicans , A. niger , and A. clavatus . Conclusions: A powerful one-pot approach has been designed to synthesize substituted quinolones. Microwave-assisted synthesis greatly accelerates reaction rate and reduces product impurityand shows modest in vitro antimicrobial and antifungal activity.

Abstract:In our attempt towards the synthesis and development of effective antimicrobial, anticancer and antioxidant agents, a novel series of 2,3-dihydropyrido[2,3-d]pyrimidin-4-one 7a–e and pyrrolo[2,1-b][1,3]benzothiazoles 9a–e were synthesized. The synthesis of 2-(1,3-benzo thiazol-2-yl)-3-(aryl)prop-2-enenitrile (5a–e) as the key intermediate was accomplished by a microwave efficient method. Via a new variety oriented synthetic microwave pathway, these highly functionalized building blocks allowed access to numerous fused heteroaromatic such as 7-amino-6-(1,3-benzo thiazol-2-yl)-5-(aryl)-2-thioxo-2,3dihydropyrido [2,3-d]pyrimidin-4(1H)-one 7a–e and 1-amino-2-(aryl)pyrrolo[2,1-b][1,3]benzothiazole-3-carbonitrile derivatives 9a–e in order to study their antimicrobial and anticancer activity. The present investigation offers effective and rapid new procedures for the synthesis of the newly polycondensed heterocyclic ring systems. All the newly synthesized compounds were evaluated for antimicrobial, anticancer and antioxidant activity. Compounds 7a,d, and 9a,d showed higher antimicrobial activity than cefotaxime and fluconazole while the remaining compounds exhibited good to moderate activity against bacteria and fungi. An anticancer evaluation of the newly synthesized compounds against the three tumor cell lines (lung cell NCI-H460, liver cancer HepG2 and colon cancer HCT-116) exhibited that compounds 7a, d, and 9a,d have higher cytotoxicity against the three human cell lines compared to doxorubicin as a reference drug. These compounds also exhibited higher antioxidant activity and a great ability to protect DNA from damage induced by bleomycin.

Abstract:Ethyl 1-amino-3,3-dimethyl-3,4-dihydronaphthalene-2-carboxylate was used as a platform to synthesize 5,5-dimethyl-3- iso -propyl-2-thioxo-2,3,5,6-tetrahydrobenzo[ h ]quinazolin-4(1 H )-one (thioxobenzoquinazoline), which was alkylated by various halides in alkaline solution to produce 2-thio-substituted 5,5-dimethyl-3- iso -propyl-5,6-dihydrobenzo[ h ]quinazolin-4(3 H )-ones. Condensation of thioxobenzoquinazoline with hydrazine hydrate, 2-ethanolamine, and 3-propanolamine synthesized 2-hydrazinyl-5,5-dimethyl-3- iso propyl-5,6-dihydrobenzo[ h ]quinazolin-4(3 H )-one, 2-[(2-hydroxyethyl)amino]-5,5-dimethyl-3- iso -propyl-5,6-dihydrobenzo[ h ]quinazolin-4(3 H )-one, and 2-[(3-hydroxypropyl)amino]-3- iso -propyl-5,5-dimethyl-5,6-dihydrobenzo[ h ]quinazolin-4(3 H )-one, respectively. The antibacterial and antitumor properties of the synthesized compounds were studied. The research established that the studied compounds had antibacterial and antitumor activity. A comparative evaluation of the data indicated that the activity of the studied compounds was noticeably inferior to that of the antimicrobial reference drug furazolidone. The studied compounds at doses of 170 and 175 mg/kg showed weak antitumor effects, inhibiting tumor growth by 28 – 33%.

Abstract:In this study, some novel benzo[c]quinoline derivatives were synthesized, their structural characteristics were described, and their in vitro anticancer efficacy was investigated. The synthesis involves an initial quaternization of the nitrogen atom from benzo[c]quinoline and then a [3+2] dipolar cycloaddition reaction of the in situ formed ylide. The effectiveness of synthesis using traditional thermal heating (TH) compared to microwave (MW) and ultrasound (US) irradiation was investigated in detail. The setup of a reaction under MW or US irradiation offers a number of additional benefits: higher yields, a reduction in the amount of solvent used compared to TH, a reduction in the reaction time from hours to minutes, and a reduction in the amount of energy consumed. The structure of all the obtained compounds was proved by several spectral techniques (FTIR, HRMS, and NMR). All benzo[c]quinoline derivatives (quaternary salts and cycloadducts) along with ten other benzo[f]quinoline derivatives (quaternary salts and cycloadducts), previously obtained, were tested in an in vitro single-dose anticancer experiment. The results demonstrated that the cycloadducts 5a–c and 6a–c exhibit stronger anticancer activity than quaternary salts 3a–c. The most active compound is compound 5a, with anticancer activity on most of the cell lines studied, while the second most active compound is 6c, showing significant lethality for the SR leukemia cell line (17%). Structure-activity relationship (SAR) correlations are also included in the study.

Abstract:In continuation of our research work on amino substituted quinoline-4-carboxylic acid derivatives, microwave irradiated and conventional heating methods were used for synthesis of target compounds. Benzaldehyde, pyruvic acid, and 3-chloro-4-fluoroaniline in absolute ethanol media reacted, condensed, and cyclized to form intermediate 7-chloro-6-fluoro-2-phenyl-quinoline-4-carboxylic acid. This intermediate reacted with various substituted amines attaining desired products 6-fluoro-2-phenyl-7-substitutedamino-quinoline-4-carboxylic acid derivatives (7a–7l). Products obtained by microwave synthesizer showed short reaction time of 110–210 s and yield of 91–96 %, demonstrating advantages of the said method. All synthesized compounds were identified and characterized by FT-IR, 1H and 13C NMR, Mass spectroscopy, and Elemental analysis. Synthesized compounds were tested for their effect on cellular viability against various carcinoma cell lines viz. MCF-7, HELA, Hep-2, NCI, HEK-293, and VERO by XTT bioassay at 24 h of drug exposure using doxorubicin and methotrexate as standard drugs. Majority of the compounds proved to be more potent than doxorubicin and compounds 7a, 7c, 7d, and 7i exhibited significant anticancer activity. Apoptotic DNA fragmentation was carried out on MCF-7 and HEK-293 cell lines and found that few compounds exhibited excellent DNA fragmentation pattern confirming apoptosis. Docking study was performed by Surflexdock to establish probable mechanism of action of synthesized compounds using X-ray crystallographic structure of the ATPase domain of hTopoIIα. Docking experiments confirmed good correlation between calculated interactions with the hTopoIIα and the observed IC50 values. The present study of quinoline-4-carboxylic acid derivatives may be considered as promising lead for future design of potent hTopoIIα inhibitors as novel anticancer agents.

Abstract:Herein, a new series of N -substituted 1,2,3-triazolylmethyl indole derivatives 4(a–u) was synthesized by rationally incorporating a pharmacophoric active heterocyclic ring containing indole and triazole moieties in one molecular frame via the conventional and microwave irradiation methods. Briefly, the new compounds 4(a–u) were synthesized via the N -alkylation of tetrahydro-1 H -carbazoles followed by click reaction and copper-catalyzed Huisgen [3 + 2] cycloaddition in the presence of copper sulphate and sodium ascorbate with various aromatic azides 3(a–m) . All the newly synthesized compounds were characterized via 1 H and 13 C NMR, mass, and IR spectroscopy and evaluated for their antimicrobial, antioxidant and anticancer activities. Among the synthesized compounds, 4d , 4j , 4n , 4p , 4s and 4r were found to exhibit good antimicrobial, antioxidant, anticancer activities. The biological activity of the synthesized compounds was further supplemented by molecular docking studies against the target receptors caspase-3 and 17-beta-hydroxy steroid dehydrogenase type 1, revealing that the reported structures best fit into the active site pocket of the target molecules.

Abstract:Herein, a one‐pot reaction between cyclohexanone, thiourea, and 2,5‐dimethoxybenzaldehyde allowed to prepare hexahydroquinazoline‐2(1H)‐thione 4 firstly, which dollowed by reacting with hydrazine hydrate to produce the corresponding 2‐hydrazinylhexahydroquinazoline 6. Interesting analogs of thiazolo[3,2‐a]quinazoline 7‐13 where obtained when hexahydroquinazoline‐2(1H)‐thione 4 reacted with 1,2‐dibromoethane, chloroacetyl chloride, bromoacetic acid, bromoacetic acid/4‐chlorobenzaldehyde, 2‐bromopropionic acid, ethyl bromocyanoacetate, and/or bromomalononitrile; respectively. While triazolo[4,3‐a] quinazoline 14‐16 were created when 2‐hydrazinylhexahydroquinazoline 6 reacted with triethyl orthoformate, acetic anhydride, and carbon disulfide respectively. Additionally, the new analogs were examined for their antibacterial and antifungal properties against Escherichia coli, Staphylococcus aureus, and Candida albicans. It was discovered that triazolo[4,3‐a] quinazoline analogs 14‐16 have superior bacterial and fungal activity when compared to the corresponding conventional doses of Streptomycin and Griseofulvin. Towards Candida albicans; compounds 14, 15, and 16 increase activity with 1.14%, 1.15%, and 1.21%, respectively more than griseofulvin.While, for Staphylococcus aureus; compounds 14, 15, and 16 increase activity with 1.5%, 1.5%, and 1.7%, respectively more than streptomycin. Morever, for Escherichia coli; compounds 14, 15, and 16 increase activity with 1.19%, 1.21%, and 1.22%, respectively more than streptomycin. Finally, structure activity relationships show that quinazoline derivatives exhibit higher activity when fused to pyrazole ring 14–16 as compared when fused thiophene ring 7–13.

Abstract:Background: This research centers on the development and spectroscopic characterization of new quinazolin-4(3H)-one-isoxazole derivatives (5a–e). The aim was to investigate the regioselectivity of the 1,3-dipolar cycloaddition involving arylnitriloxides and N-propargylquinazolin-4(3H)-one, and to assess the antioxidant properties of the synthesized compounds. The synthetic approach started with the alkylation of quinazolin-4(3H)-one using propargyl bromide, followed by a 1,3-dipolar cycloaddition reaction. Methods: The structural identification of the products was performed using various spectroscopic methods, such as IR, 1H, 13C, and HMBC NMR, HRMS, and single-crystal X-ray diffraction. To further examine the regioselectivity of the cycloaddition, Density Functional Theory (DFT) calculations at the B3LYP/6-31G(d) level were employed. Additionally, the antioxidant potential of the compounds was tested in vitro using DPPH (2,2-Diphenyl-1-picrylhydrazyl)radical scavenging assays. The reaction selectively produced 3,5-disubstituted isoxazoles, with the regiochemical outcome being independent of the substituents on the phenyl ring. Results: Theoretical calculations using DFT were in agreement with the experimental results, revealing activation energies of −81.15 kcal/mol for P-1 and −77.32 kcal/mol for P-2, favoring the formation of P-1. An analysis of the Intrinsic Reaction Coordinate (IRC) confirmed that the reaction proceeded via a concerted but asynchronous mechanism. The antioxidant tests demonstrated that the synthesized compounds exhibited significant radical scavenging activity, as shown in the DPPH assay. The 1,3-dipolar cycloaddition of arylnitriloxides with N-propargylquinazolin-4(3H)-one successfully resulted in novel 3,5-disubstituted isoxazoles. Conclusions: The experimental findings were well-supported by theoretical predictions, and the antioxidant assays revealed strong activity, indicating the potential for future biological applications of these compounds.

Abstract:Conventional and microwave irradiation (MWI) methods were applied to synthesize a series of thirteen carbazole-thiazole-Schiff base hybrids ( 2a-2m ). MWI assisted synthesis was a benign approach to green synthesis on account of its miniscule reaction time and use of solvent. The structures of the synthesized derivatives were confirmed by means of spectral analytical data. All the compounds were evaluated for dual biological potency i.e., antimicrobial and antioxidant activity. The tested analogues had shown some modest antimicrobial activity and compounds 2f and 2i exhibited higher antioxidant activity compared to standard ascorbic acid. Oral bioavailability and adverse effects of the new entities were considered from druglikeness descriptors, having no violation of Lipinski's rule of five and Veber's rule for the synthesized compounds except for 2i, 2l , and 2m . The compounds which exhibited relatively high activity were explored further for molecular docking studies to understand the mode of binding interactions with the receptor protein. Molecular dynamics (MD) simulation was further studied to predict the stability of selective compounds inside the receptors which was well agreed with in vitro results and antioxidant value.

Abstract:Condensation reactions of 2-thioxo-2,3-dihydro-1 H -spiro[benzo[ h ]quinazoline-5,1′-cycloheptan]-4(6 H )-one with 2-aminoethanol and 3-aminopropanol were used to synthesize 2-[(2-hydroxyethyl)amino]-3 H spiro[benzo[ h ]quinazoline-5,1′-cycloheptan]-4(6 H )-one and 2-[(3-hydroxypropyl)amino]-3 H -spiro[benzo-[ h ]quinazoline-5,1′-cycloheptan]-4(6 H )-one respectively. Interaction of this thioxoquinazoline with 2-chloromethylbenzoquinazolines with different structures yielded dibenzoquinazoline systems connected at position 2 by an -SCH 2 - linker. Reactions using methylene iodide produced 2,2'-[methylenebis(sulfanediyl)]-bis(3 H -spiro[benzo[ h ]quinazoline-5,1′-cycloheptan]-4(6 H )-one, while use of 1,2-dibromoethane, 2,3-dibromoacetic acid ethyl ester, and 1,3-dibromopropane led to the production of 9,10-dihydrospiro[benzo-[ h ]thiazolo[2, 3- b ]quinazoline-6,1′-cycloheptan]-7(5 H )-one, ethyl-7-oxo-5,7,9,10-tetrahydrospiro[benzo[ h ]-thiazolo[2,3- b ]quinazoline-6,1′-cycloheptan]-9-carboxylate, and 10,11-dihydro-5 H -spiro[benzo[ h ][1,3]-thiazino[2,3- b ]quinazoline-6,1′-cycloheptan]-7(9 H )-one respectively. The study compounds were found to have activity which was significantly inferior to that of reference antimicrobial drug furazolidone. Study compounds at doses of 150–185 mg/kg exhibited antitumor activity, inhibiting tumor growth in mice by 20–40%.

Abstract:Quinazolinone is a preferred structural motif with notable pharmacological activity that is present in a wide range of naturally occurring compounds. A microwave assisted tandem cyclooxidative method has been developed to afford quinazolinones via recyclable ionic liquid supported copper catalyst. This sustainable method exhibits operational simplicity, rapid, clean, energy efficient route and a variety of 2-substituted quinazolinones are obtained in excellent yields. In addition, this innovative approach enables us to develop a library of nitriles in an environment friendly synthetic protocol. Moreover, the catalyst can be recycled and reused up to three consecutive cycles without any significant loss of catalytic activity. Further organic transformation of the synthesized quinazolinones was carried out to afford reported as well as novel bioactive heterocyclic compounds.

Abstract:In the presented work, a series of 22 hybrids of 8-quinolinesulfonamide and 1,4-disubstituted triazole with antiproliferative activity were designed and synthesised. The title compounds were designed using molecular modelling techniques. For this purpose, machine-learning, molecular docking, and molecular dynamics methods were used. Calculations of the pharmacokinetic parameters (connected with absorption, distribution, metabolism, excretion, and toxicity) of the hybrids were also performed. The new compounds were synthesised via a copper-catalysed azide-alkyne cycloaddition reaction (CuAAC). 8-N-Methyl-N-{[1-(7-chloroquinolin-4-yl)-1H-1,2,3-triazol-4-yl]methyl}quinolinesulfonamide was identified in in silico studies as a potential strong inhibitor of Rho-associated protein kinase and as a compound that has an appropriate pharmacokinetic profile. The results obtained from in vitro experiments confirm the cytotoxicity of derivative 9b in four selected cancer cell lines and the lack of cytotoxicity of this derivative towards normal cells. The results obtained from silico and in vitro experiments indicate that the introduction of another quinolinyl fragment into the inhibitor molecule may have a significant impact on increasing the level of cytotoxicity toward cancer cells and indicate a further direction for future research in order to find new substances suitable for clinical applications in cancer treatment.

Abstract:A series of new compounds based on 8-methyl-2-phenyl quinazolinone Schiff's base were synthesized from 3-amino quinazolinone intermediates. These compounds were evaluated for their potential as anticancer and antimicrobial agents through docking analysis. Anticancer evaluation was done against sixty different cancer cell lines. Compounds 7b and 7 h showed strong efficacy against Melanoma (MDA-MB-435 Cell lines) and Non-Small Cell Lung Cancer (NCI-H522), respectively. Compound 7 g was found to be predominantly effective against both breast cancer (HS-578T) and central nervous system cancer (SNB-19). The antimicrobial activity results showed that the newly synthesized compounds 7d and 7j, which contain halogen, exhibited potential inhibiting action against selected bacterial and fungal microorganisms. The SAR study revealed that compounds with EDGs like hydroxyl and meta -substituted chloro group were found to be more potent in anticancer studies; while compounds with EWGs substituted in the para position (7d and 7j) demonstrated higher antimicrobial activity. Moreover, Antimicrobial docking analysis indicated that compounds 7d and 7j have a high affinity towards molecular targets found in both bacteria and fungi, with negative binding energies ranging from -8 to -12.5 kcal/mol. We endorse further evaluation of these compounds in combination with standard antibiotics to potentially increase their synergistic effect.

Abstract:In the present study, we report a facile synthesis of a new series of 3-subtituted-2-thioxo-2,3-dihydroquinazolin-4(1 H )-one derivatives via the cascade reaction of various isothiocyanate derivatives and anthranilic acid. The reaction was assisted by microwave irradiation and finished after 1 h affording the target products with the yield within the range 40–72%. The synthetic compounds were characterized by spectroscopic methods including IR, NMR ( 1 H, 2D, and 13 C), and mass spectrometry. All spectral data indicated good accordance with expected structures. In addition, the complex of quinazoline derivatives with Candida albicans dihydrofolate reductase ( C. albicans DHFR) were studied in silico . The results showed that all quinazoline compounds ( 2a–f ) have a greater ability to inhibit C. albicans DHFR than 5-(phenylsulfanyl)-2,4-quinazolinediamine. Therefore, they are promising candidates for further studies in finding the lead structure for antimicrobial activity.

Abstract:A highly efficient microwave assisted synthesis of new quinolone and coumarin molecular hybrids has been developed in the present work by direct approach. The synthesis is based on dehydrohalogenation and dehydration followed by the condensation of amine derivatives with oxygen atom of the chromone intermediate ring and CCl bonds. A series of 7-(2‐chloro ethoxy)-4-methyl-1 H -quinolones 5(a-b), 6(a-c) and 7-(2-substituted ethoxy)-4-methyl-chromones 7(a-f) derivatives have been explored as dual antagonists using molecular modelling simulations targeting bacterial and fungal infections. The results clearly demonstrated the stronger binding affinity of compounds 6b (-8.5 kcal/mol, -9.4 kcal/mol) and 7d (-7.9 kcal/mol, -9.3 kcal/mol) towards peptide formylase and tubulin alpha beta heterodimer proteins which were comparatively higher as compared to standards ciprofloxacin (-7.1 kcal/mol) and griseofulvin (-7.6 kcal/mol) respectively. The in-vitro antimicrobial studies carried out using broth dilution method on various bacterial and fungal strains were found to be consistent with the docking results. Out of the synthesized derivatives compounds 5b, 6b and 7d revealed significant antibacterial activity against E.coli (MIC: 15.62, 15.62 and 15.62 μg/ml), P.aeruginosa (MIC: 31.25, 31.25 and 15.62 μg/ml), S.pyogenes (MIC: 31.25, 15.62 and 31.25 μg/ml) and antifungal activity against C.albicans (MIC: 31.25, 31.25 and 31.25 μg/ml) and A.niger (MIC: 31.25, 31.25 and 62.5 μg/ml) respectively. The results obtained from in-vitro antimicrobial studies clearly depicts the potential antimicrobial activity of outperformed compounds 5b, 6b and 7d in the management of bacterial and fungal infections.

Abstract:Based on the structural feature of nonclassical antifolates, a series of 4 (3H)-quinazolinone derivatives bearing aromatic amine side chains have been designed by incorporating trimethoxyphenyl, an antitumor pharmacophore, with 4 (3H)-quinazolinone. N-alkylation of 3,4,5-trimethoxyphenylamine with appropriate amount of alkyl halides afforded 4 kinds of N-substituted 3,4,5-trimethoxyphenylamines in 30.3%~60.6% yields. The target compounds 8a 8m were obtained through the reactions of 6-bromo-2-methyl-4 (3H)-quinazolinone with 3,4,5-trimethoxyphenylamine, N-substituted 3,4,5-trimethoxyphenylamines or other arylamines at room temperature in 30.8%~71.9% yields, and their structures were confirmed by means of ESIMS, 1H NMR, elemental analysis or HRMS. The in vitro cytotoxicities against A-549 (human non-small cell lung cancer), HCT-8 (human colon cancer) and Bel-7402 (human liver cancer) cell lines of the synthesized compounds 8a~8m were tested with colorimetric MTT assay, and the results indicated that the percent growth inhibition against A-549, HCT-8 and Bel-7402 cell lines at 5×10^(-6)g/mL concentration of the compounds 8a~8m were all lower than 25%.

Abstract:In this communication, we explored the synthesis of novel alkoxy-functionalised dihydropyrimido[4,5- b ]quinolinones using a microwave-assisted multicomponent reaction. All the synthesized molecules were screened for anti-proliferative and anti-invasive activity against glioblastoma cells. 5c shows the most potent anti-proliferative activity with a half maximal effective concentration of less than 3 μM against primary patient-derived glioblastoma cells. 5c effectively inhibited invasion and tumor growth of 3D primary glioma cultures in a basement membrane matrix. This suggests that the novel compounds could inhibit both the proliferation and invasive spread of glioma and they were selected for further study.

Abstract:Microwave irradiation promotes the rapid O,N-acylation-cyclodehydration cascade reaction of oximes and acid chloride. Twenty novel 2,4,5-trisubstituted oxazole derivatives containing heterocycle moiety were synthesized and evaluated for their antiproliferative activity. The twenty compounds are all first reported and their structures were established by elemental analysis, (1)H NMR and (13)C NMR spectra. The bioassay tests showed that compounds 2-(2-(2-fluorophenyl)-4-(2,3,4-trimethoxyphenyl)oxazol-5-ylthio)benzo[d]thiazole (6af), 2-(2-(pyridin-3-yl)-4-(2,3,4-trimethoxyphenyl)oxazol-5-ylthio)pyrimidine (6bg) and 2-(2-(2-fluorophenyl)-4-(2,3,4-trimethoxyphenyl)oxazol-5-ylthio)-5-methyl-1,3,4-thiadiazole (6cf) displayed good antiproliferative activity in vitro, which were comparable to the positive control (5-fluorouracil).

Abstract:Methyl (1S,3S and 1R,3S)-1-(2,2-dimethoxyethyl)1,2,3,4-tetrahydrocarboline-3-carboxylate (3) was hydrolyzed in the presence of sodium hydroxide to give (1S,3S and 1R, 3S)-1-(2,2-dimethoxyethyl)- 1,2,3,4-tetrahydrocarboline-3-carboxylic acid (4) which was reduced with LiAlH4 to provide (1S,3S)- and (1R,3S)-1-(2,2-dimethoxyethyl)-3-hydroxymethyl-1,2,3,4-tetrahydrocarboline (10) and then amidated in ammonia containing methanol to obtain (1S,3S)and (1R,3S)-1-(2,2-dimethoxyethyl)- 1,2,3,4-tetrahydrocarboline-3-carboxamide (14). Abstract:A single crystal of new 2-substitued quinazoline derivative has been successfully synthesized through a simple and novel process based on a two-step reaction involving cyclisation, oxidation and hydrasination. The structure of the newly synthesized product was fully characterized by spectroscopic analysis NMR ( 1 H & 13 C), IR and HRMS. This intriguing compound crystalizes crystallises in a monoclinic system with P21/c space group. Notably, the stability of crystal packing was assured via hydrogen bonds, π- stacking and van der Waals interactions. An intermolecular interaction examination of the crystal structure was performed using Hirshfeld surface analysis and two-dimensional fingerprint plots. In addition, the electrostatic surface potential was calculated using density functional theory. In addition, the 3-amino-2-(2-thienyl)-4(3H)-quinazolinone, was applied to selected bacterial strains, and its antibacterial activity was assessed. In addition, a molecular docking study has been realised.
