Abstract:This work establishes a hyphenated methodology coupling HPLC with ICP-MS for simultaneous speciation analysis of arsenic, mercury and lead for the first time. Four arsenicals (As(III), DMA, MMA and As(V)), four mercurials (Hg(II), MeHg, EtHg and PhHg) and three lead compounds (Pb(II), TML and TEL) were simultaneously analyzed within only 8 min with acceptable resolution (2.0-8.2 for As, 1.6-6.1 for Hg and 2.7-4.0 for Pb). The detection limits were 0.036-0.20 for As-species, 0.023-0.041 for Hg-species, and 0.0076-0.14 mu g L-1 for Pb-species. The developed method was applied for the measurement of five lotus seed samples, indicating the presence of DMA (19.6-28.2 mu g kg(-1)), TML (1.4-2.9 mu g kg(-1)), MeHg (1.2-4.8 mu g kg(-1)) and EtHg (0.8-2.2 mu g kg(-1)). This method provides a promising tool for studying the toxic, metabolic and bioavailable behaviors of arsenic, mercury and lead.

Abstract:There are many kinds of micronutrient elements contained in foods that are vital to human health. The strong dependence of the toxicity and bioavailability of microelement upon their chemical forms has made the speciation and bioavailability vital to the food quality control and nutrition evaluation. A brief introduction to the high performance liquid chromatography hyphenated with inductive coupled plasma mass spectrometry ( HPLC-ICP-MS) is given. The hyphenated technique which based on several chromatographic separation mechanisms plays an important role in speciation analysis for its advantage of high sensitivity, high selectivity, wide linearity, low detection and multi-microelement determination simultaneously. The application of HPLC-ICP-MS in microelements speciation analysis and bioavailability research in foods concerning the selenium speciation analysis in selenized yeast, selenium-enriched garlic, selenium nutritional supplement and other foods, the arsenic speciation analysis in sea-products and farm products, the other microelements speciation analysis and the investigation of metabolic and bioactivity of microelement in organism is reviewed in this paper.

Abstract:The knowledge of elemental speciation and interaction between elemental species and ligands (including biomolecules) can provide an understanding of their transport, transformation, bioavailability, metabolic pathways and toxicity in organisms. By taking advantage of the powerful separation capabilities of capillary electrophoresis (CE) in capillaries/microchannels, and the atomic specificity, multi-elemental characteristics and extremely high sensitivity of inductively coupled plasma spectrometries (ICPs), the CE-ICP hyphenation is one of the most preferred choices in elemental speciation and element-ligand/element-biomolecule interaction as well as the indirect determination of non-metallic compounds. This article provides an overview of electrophoresis in capillaries and microchannels coupled to an ICP with respect to interfaces and applications from elemental speciation, element-ligand interaction to indirect determination.

Abstract:Multi-elemental speciation analysis in a single run reduces costs, waste and time, and also sample consumption, which is favorable to a large set of instable samples. This work demonstrates mercury and arsenic speciation in a single run using high performance liquid chromatography with inductively coupled plasma mass spectrometric detection (HPLC/ICP-MS). Thiol compounds in mobile phases interfered with the elution of arsenite because of coordination action, which was overcome by gradient elution (elution of As(III) by thiol-free mobile phases at first). Four arsenic forms (As(III), DMA, MMA and As(V)) and three mercury species (Hg(II), MeHg and EtHg) were separated and detected by HPLC/ICP-MS using 4 mM tetrabutylammonium hydroxide (TBAH) (pH 6.0) in the first 1.0 min and 4 mM TBAH + 20 mM L-cysteine (pH 6.0) in the next 9.0 min. The detection limits were obtained at 0.078 for As(III), 0.081 for DMA, 0.078 for MMA and 0.15 mu g L-1 for As(V), and 0.016 for Hg(II), 0.027 for MeHg and 0.032 mu g L-1 for EtHg, and the reproducibilities (50 mu g L-1 for each As-species and 10 mu g L-1 for each Hg-species) were all lower than 3.7%. The proposed method was successfully applied for speciation analyses of arsenic and mercury in five lotus roots, which indicated DMA (3.5-7.9 mu g kg(-1)) as the only arsenic species and MeHg (0.6-1.0 mu g kg(-1)) as the only mercury species. Analyses of two certified reference materials demonstrated good agreement between determined values and certified values. All results prove its advantages including high sensitivity and high analytical efficiency, which are beneficial for routine speciation analyses of arsenic and mercury in a single run in environmental and food fields.

Abstract:It is well established that the metal content in Chinese hamster ovary (CHO) cell culture media (CCM) greatly affects process productivity and critical quality attributes (CQAs). Metals exist in diverse chemical forms in CCM, which may change during the monoclonal antibody (mAb) production cycle. Naturally, it is postulated that the chemical speciation of these metals affects their uptake and metabolism. While the targeted forms of metals in media are company-specified, their commercial sources, as well as those of the organic media constituents, may result in concentrations and speciation that differ from the intended formulae. Therefore, there is a need for a method to determine the concentration of metals and their chemical forms. Presented here is a methodology to speciate inorganic- versus -ligated metals using high-performance liquid chromatography (HPLC) employing a polypropylene capillary-channeled (C-CP) fiber column with inductively coupled plasma mass spectrometry (ICP-MS) determinations of five target metals (Mn, Fe, Co, Cu, and Zn). A 50 μL injection of CCM supernatant is used for an effective quantification method to identify metal speciation and concentration deviations from reported formula levels. Two case studies as to the utility of the methodology are presented: potential chemical contamination and species variability due to shelf-life. Further development of this method has relevance towards quality control, identification of contaminants, assessment of media stability/degradation products, and measurement of whole cell metal uptake in growth media.

Abstract:Toxicity, mobility, bioavailability and biofunctions of chromium, cadmium, mercury and lead are heavily dependent upon their specific chemical forms, leading to a high demand to metal speciation analysis rather than total quantification. Simultaneous speciation analysis of multiple metal(loid)s is attractive to a large sample capacity containing unstable analytes due to its economic and environmental advantages over the conventional single elemental strategies. In this work, an analytical method integrating online solid phase extraction into high performance liquid chromatography interfaced with inductively coupled plasma mass spectrometry (ICP-MS) to simultaneously preconcentrate and quantify Cr, Cd, Hg and Pb forms in pg L-1 levels in water was developed. Cr (III + VI), Cd(II), Hg(II), Pb(II), methylmercury (MeHg), ethylmercury (EtHg), and trimethyl (TML) and triethyl lead (TEL) were captured by the C18 adsorbent (equilibrated with 10 mL of 1.0 mM 2-hydroxyethanethiol at 10 mL min-1) and eluted by mobile phase (5.0 mM Cys at pH 2.0), then completely separated on the C18 column within 8.0 min and eventually determined by ICP-MS. Low limits of detection (0.001-0.007 ng L-1) and quantification (0.003-0.023 ng L-1), good relative standard deviations (<4%) and high enrichment factors (827-2656 folds) were obtained with good linearities. Three reference materials of total cadmium (GBW08602), total mercury (GBW08603) and total lead (GBW08601) in water were analyzed by the developed method to validate the accuracy with good agreement with certified values and satisfactory recoveries (92-100%). This method was proved feasible by the determination of Cr, Cd, Hg and Pb compounds in drinking water, river water, pond water and tap water.

Abstract:Tin and lead are a global concern considering their species-dependent toxicity, bioavailability and transformation. Simultaneous speciation analysis of tin and lead is challenging for a large food capacity containing unstable species. Herein, we developed two sensitive methods for rapid quantification of tin and lead species in Antarctic seafood by high-performance liquid chromatography and inductively coupled plasma mass spectrometry based on strong cation-exchange and Amphion columns. Inorganic tin and lead, four organotin and two organolead compounds can be analysed in 16 min on a 10-cm Amphion II column (mobile phase: 4 mM sodium dodecyl benzene sulfonate at pH 2.0) with 0.02-0.24 mu g L-1 detection limits. The method was applied to Antarctic krill and fish, demonstrating the presence of any tin and lead species down to mu g kg-1 level. Overall, the proposed methods are sensitive, efficient and environment-friendly for routine speciation analysis of tin and lead in food samples.

Abstract:Metal speciation analysis of microsamples in the clinical, biological and forensic fields is important for elucidating the metals' toxicity, mobility and metabolic behaviors of metals. Such analysis may be achieved by nanoliter high-performance liquid chromatography (nanoHPLC) hyphenated with inductively coupled plasma mass spectrometry (ICP-MS). In this work, an in-column high-pressure nebulizer (ICHPN) was developed to enhance the sensitivity and the separation efficiency of the sheathless nanoHPLC-ICP-MS coupling. The ICHPN consists of two concentric fused-silica capillaries with tapered tips connected via a PEEK tee, where C-18 silica particles with a size of 5 mu m were packed in the tapered inner capillary based on the keystone effect. Combining a heated single pass spray chamber with a makeup gas, the ICHPN was capable of independently optimizing the nebulization efficiency and the transport efficiency. The ICHPN offered high sensitivities, low detection limits and good precision at nanoflow rates. Compared with commercial nebulizers, the ICHPN fabrication was simple, rapid, reproducible and inexpensive. By implementing the ICHPN, we achieved rapid separation of four mercury species, i.e., Hg2+ and methyl-, ethyl- and phenylmercuric chloride, within 8.0 min with good resolution (2.0-13.9). Detection limits of 0.044-0.13 mu g L-1 were obtained with precisions of peak heights and areas ranging from 1.5 to 3.5% for a 50 mu g L-1 standard solution. Good agreement between the determined and certified values of mer- cury species in a certified reference material of caprine blood (SRM 955c) together with good recoveries (93-101%) validated the accuracy of the method. Abstract:Nanoliter high-performance liquid chromatography shows low consumption of solvents and samples, offering one of the best choices for arsenic speciation in precious samples in combination with inuctively coupled plasma mass spectrometry. A systematic investigation on coupling nanoliter high-performance liquid chromatography to inductively coupled plasma mass spectrometry from instrument design to injected sample volume and mobile phase was performed in this study. Nanoflow mobile phase was delivered by flow splitting using a conventional high-pressure pump with reuse of mobile phase waste. Dead volume was minimized to 60 nL for the sheathless interface based on the previously developed nanonebulizer. Capillary columns for nanoliter high-performance liquid chromatography were found to be sensitive to sample loading volume. An apparent difference was also found between the mobile phases for nanoliter and conventional high-performance liquid chromatography. Baseline separation of arsenite, arsenate, monomethylarsenic, and dimethylarsenic was achieved within 11 min on a 15 cm C-18 capillary column and within 12 min on a 25 cm strong anion exchange column. Detection limits of 0.9-1.8 mu g/L were obtained with precisions variable in the range of 1.6-4.2%. A good agreement between determined and certified values of a certified reference material of human urine (GBW 09115) validated its accuracy along with good recoveries (87-102%).

Abstract:This work described the utilization of ion-pair reversed phase liquid chromatography coupled to inductively coupled plasma mass spectrometry (RP-LC-ICP-MS) for iodine speciation analysis in urine. Considering the requirements of green analytical chemistry and the Ar ICP-MS instrument, three aqueous mobile phases were employed for the separation of seven iodine species including iodide, iodate and five iodo amino acids (monoiodotyrosine - MIT, di-iodotyrosine - DIT, tri-iodothyronine - T-3, reversed tri-iodothyronine - rT(3), and thyroxine - T-4). The aqueous mobile phases were composed of an ion-pair reagent (tetrabutylammonium hydroxide - TBAH) and an eluent (ammonium chloride, L-phenylalanine or deoxycholic acid) at low concentrations in ultrapure water. Owing to tremendous difference in retention behavior between these iodinated forms, a gradient elution mode was performed for the rapid separation of IO3- and I-, MIT and DIT, and T-3, rT(3) and T-4, respectively. Iodine species separation was achieved with a 12.5 mm C-18 guard column in 7 min. The detection limits for IO3-, I-, MIT, DIT, T-3, rT(3) and T-4 were 0.047, 0.046, 0.057, 0.072, 0.093, 0.094 and 0.081 mu g L-1, respectively. Application of the proposed method was demonstrated by the speciation analysis of iodine in four real urine samples. The developed method offered satisfactory recoveries in the 93-106% range and good repeatability, showing great potential in routine analysis of iodine speciation in environmental, food and biological samples.
Speciation and Distribution of Copper in A Mining Soil Using Multiple Synchrotron-Based Bulk and Microscopic Techniques

Abstract:Molecular-level understanding of soil Cu speciation and distribution assists in management of Cu contamination in mining sites. In this study, one soil sample, collected from a mining site contaminated since 1950s, was characterized complementarily by multiple synchrotron-based bulk and spatially resolved techniques for the speciation and distribution of Cu as well as other related elements (Fe, Ca, Mn, K, Al, and Si). Bulk X-ray absorption near-edge structure (XANES) and extended X-ray absorption fine structure (EXAFS) spectroscopy revealed that soil Cu was predominantly associated with Fe oxides instead of soil organic matter. This agreed with the closest association of Cu to Fe by microscopic X-ray fluorescence (U-XRF) and scanning transmission X-ray microscopy (STXM) nanoanalysis, along with the non-occurrence of photoreduction of soil Cu(II) by quick Cu L3,2-edge XANES spectroscopy (Q-XANES) which often occurs when Cu organic complexes are present. Furthermore, bulk-EXAFS and STXM-coupled Fe L3,2-edge nano-XANES analysis revealed soil Cu adsorbed primarily to Fe(III) oxides by inner-sphere complexation. Additionally, Cu K-edge μ-XANES, L3,2-edge bulk-XANES, and successive Q-XANES results identified the presence of Cu2S rather than radiation-damage artifacts dominant in certain microsites of the mining soil. This study demonstrates the great benefits in use of multiple combined synchrotron-based techniques for comprehensive understanding of Cu speciation in heterogeneous soil matrix, which facilitates our prediction of Cu reactivity and environmental fate in the mining site.

Abstract:A quantitative method was established for the simultaneous speciation of arsenic (As) and selenium (Se) in rice using high performance liquid chromatography (HPLC)- dynamic reaction cell (DRC)-inductively coupled plasma mass spectrometry (ICP-MS). Hamilton PRP- X100 (150 mm × 4.6 mm × 10 μm) and gradient elution using (NH 4) 2CO 3 at pH 9.5 was allowed separation of AsB, As(III), DMA, MMA, As(V), Se(IV) and Se(VI) in less than 14 min. The ion 75As 12CHH + was determined to eliminate the interference of 75ArCl + for 75As + and the potentially interfering 40Ar 40Ar + for 80Se+ was reduced by using CH4 as reactive gas at optimal Rpq (0.35) and DRC cell gas flow rate (0.6 mL min-1). The detection limits for As and Se were in the range of 0.02 - 0.06 ng mL -1. The relative standard deviation of the peak areas for seven injections of 5 ng mL -1 As and Se mixture ranged from 0.32% to 4.67%. The method was applied to the determination of As and Se species from rice extracted by artificial gastric juice which intended to mimic the physical and biochemical process in the human digestive system. In this study, inorganic As containing As(III) and As(V) took up a high content of total As in Chinese rice samples. Se intake from rice samples is lower than the Se Recommended Dietary Allowance (RDA) in humans (70000 ng day -1). Because rice was the predominant Se source in the Chinese diet, lower Se in rice samples might lead to the Se deficiency of inhabitants in China. © 2011 IEEE.

Abstract:Chemical speciation is a very important subject in the environmental protection, toxicology, and chemical analytics due to the fact that toxicity, availability, and reactivity of trace elements depend on the chemical forms in which these elements occur. Research on low analyte levels, particularly in complex matrix samples, requires more and more advanced and sophisticated analytical methods and techniques. The latest trends in this field concern the so-called hyphenated techniques. Arsenic, antimony, chromium, and (underestimated) thallium attract the closest attention of toxicologists and analysts. The properties of those elements depend on the oxidation state in which they occur. The aim of the following paper is to answer the question why the speciation analytics is so important. The paper also provides numerous examples of the hyphenated technique usage (e.g., the LC-ICP-MS application in the speciation analysis of chromium, antimony, arsenic, or thallium in water and bottom sediment samples). An important issue addressed is the preparation of environmental samples for speciation analysis.

Abstract:It has been challenging to analyze inorganic arsenic (iAs) with anion exchange HPLC-Electrospray Ionization-Mass spectrometry (HPLC-ESI-MS), because arsenite (As(III)) is difficult to retain on column and the salts in mobile phase causes ionization suppression of iAs. To address these issues, a method has been developed involving the determination of arsenate (As(V)) with mixed mode HPLC-ESI-MS and the conversion of As(III) to As(V) for total iAs. As(V) was separated from other chemicals on Newcrom B, a bi-modal HPLC column involving anion exchange and reverse phase interaction. The elution employed a two-dimensional gradient, including a formic acid gradient to elute As(V) and a concurrent alcohol gradient to elute organic anions used in sample preparations. As(V) was detected by Selected Ion Recording (SIR) in negative mode at m/z = 141 with a QDa (single quad) detector. As(III) was quantitatively converted to As(V) by mCPBA oxidation and measured for total iAs. By replacing salt with formic acid in elution, the ionization efficiency for As(V) was greatly enhanced in ESI interface. The limit of detection (LOD) for As(V) and As(III) were 0.0263 μM (1.97 ppb) and 0.0398 μM (2.99 ppb), respectively. The linear range was 0.05-1 μM. The method has been used to characterize iAs speciation change in the solution and precipitation in a simulated iron-rich groundwater caused by air exposure.

Abstract:Background Mercury (Hg) is a persistent pollutant occurring in the environment able to transition between different species. It can therefore be found in air, soil and water reservoirs becoming a present concern for the general population but also sensitive populations like pregnant women. Therefore, investigating organ-specific transfer mechanisms of Hg is mandatory for Hg toxicity testing. For this, an in vitro system using microporous inserts to monitor the transfer across an in vitro placental barrier has been used. However, due to the cytotoxicity of Hg only low concentrations (1.26 ×10 -4 – 1.36 ×10 -2 μg/μL Hg) can be applied making Hg determination in cell culture medium using inductively coupled plasma-optical emission spectrometry challenging. Especially when these trace amounts should be determined alongside other trace elements which are naturally occurring in cells and cell culture medium like the essential metals manganese (Mn), iron (Fe), copper (Cu), and zinc (Zn). Additionally, Hg analysis on an ICP system holds also a number of challenges like a persistent memory effect and instability of Hg standard solutions. Methods The development of a rapid and sensitive ICP-OES method to determine Hg in different matrices like cell culture medium and cells has been performed on an Avio 220 Max ICP-OES (Perkin-Elmer) equipped with a cyclonic spray chamber and MicroMist® nebulizer. Cell lysates and cell culture medium were diluted in a mixture of 0.2% L-cysteine, 2% HNO 3 and 0.1% HCl and directly introduced into the ICP-OES system. Further method development included the suitability of the analysis of multiple elements like Mn, Fe, Cu, and Zn as well as the determination of the limit of detection and limit of quantification. Results The combination of 0.2% L-cysteine, 2% HNO 3 and 0.1% HCl is able to bind and stabilize Hg ions in standard solutions and in biological matrices over a wide dynamic concentration range (1 – 500 μg/L) also alongside other metals like Mn, Fe, Cu and Zn without losses of sensitivity. A short run time of 3 min enables high throughput analysis. Additionally, the high salt and carbon concentrations in the culture medium do not affect Hg sensitivity using the ICP-OES. Conclusion This method is a useful tool for the quantification of Hg in a variety of complex matrices including cells and cell culture media (high salt and carbon-rich (~1% each)) with high sensitivity and minimal sample preparation allowing high throughput. Furthermore, not only Hg can be determined in biological matrices, but even multiple elemental analysis can be carried out to address the effect of Hg on other metals homeostasis.

Abstract:Speciation analysis has become an invaluable tool in human health risk assessment, environmental monitoring or food quality control. Another step is to develop reliable multielemental speciation methodologies, to reduce costs, waste and time needed for the analysis. Separation and detection of species of several elements in a single analytical run can be accomplished by high performance liquid chromatography hyphenated to inductively coupled plasma mass spectrometry (HPLC/ICP-MS). Our review assembles articles concerning multielemental speciation determination of: As, Se, Cr, Sb, I, Br, Pb, Hg, V, Mo, Te, Tl, Cd and W in environmental, biological, food and clinical samples analyzed with HPLC/ICP-MS. It addresses the procedures in terms of following issues: sample collection and pretreatment, selection of optimal conditions for elements species separation by HPLC and determination using ICP-MS as well as metrological approach. The presented work is the first review article concerning multielemental speciation analysis by advanced hyphenated technique HPLC/ICP-MS.
Simultaneous Arsenic and Selenium Speciation Analysis Using HPLC-ICP-MS

Abstract:Four arsenic species and three selenium species simultaneous analysis method was build with High performance liquid chromatography and Inductively coupled plasma-mass spectrometry(HPLC-ICP-MS).The column of Hamilton PRP-X100 was used for chromatogram seperation,and Collision cell technology(CCT) was used for ICP-MS analysis.The detection limits for arsenite(As(Ⅲ)),dimethylarsinic acid(DMA),monomethylarsonic acid(MMA),arsenate(As(Ⅴ)),selenocystine(SeCys),selenite(Se(Ⅳ)) and selenomethionine(SeMet) were 0.2 μg/L,0.2 μg/L,0.2 μg/L,0.3 μg/L,0.5 μg/L,0.4 μg/L and 2.5 μg/L,respectively.The line range was 10~200 μg/L for all species except the SeMet,all linear correlation coefficients were not less than 0.9995.The species of arsenic and selenium in fresh mushroom and fish was detected with this method,good spiked recoveries from 81.6% to 103.7% were obtained.

Abstract:In order to obtain a well understanding of the toxicity and ecological effects of trace elements in the environment, it is necessary to determine not only the total amount, but also their existing species. Speciation analysis has become increasingly important in making risk assessments of toxic elements since the toxicity and bioavailability strongly depend on their chemical forms. Effective separation of different species in combination with highly sensitive detectors to quantify these particular species is indispensable to meet this requirement. In this paper, we present the recent progresses on the speciation analysis of trace arsenic, mercury, selenium and antimony in environmental and biological samples with an emphasis on the separation and detection techniques, especially the recent applications of high performance liquid chromatography (HPLC) hyphenated to atomic spectrometry or mass spectrometry.

Abstract:The technique of coupled in situ electrodeposition-electrothermal atomic absorption spectrometry (ED-ETAAS) is applied to the analytes Bi, Pb, Ni and Cu. Bi, Pb, Ni and Cu are deposited quantitatively from their EDTA complexes at E(cell)=1.75, 2.0, 3.0 and 2.5 V, respectively (E(cell)=E(anode)-E(cathode)+iR). By varying the cell potential, selective reduction of free metal ions could be achieved in the presence of the EDTA complexes. For Bi(3+) and Pb(2+) this utilised the voltage windows E(cell)=0.6-1.0 and 1.8-2.0 V, respectively. For Ni, deposition at E(cell)=1.7-2.0 V achieved substantial, but not complete, differentiation between Ni(2+) (ca. 90-100% deposition) and Ni(EDTA)(2-) (ca. 12-20% deposition). An adequate voltage window was not obtained for Cu. The ability of ED-ETAAS to differentiate between electrochemically labile and inert species was demonstrated by application of both ED-ETAAS and anodic stripping voltammetry to the time-dependent speciation of Pb in freshly mixed Pb(2+)-NaCl media. Application to natural water samples is complicated by adsorption of natural organic matter to the graphite cathode.
