Abstract:This work establishes a hyphenated methodology coupling HPLC with ICP-MS for simultaneous speciation analysis of arsenic, mercury and lead for the first time. Four arsenicals (As(III), DMA, MMA and As(V)), four mercurials (Hg(II), MeHg, EtHg and PhHg) and three lead compounds (Pb(II), TML and TEL) were simultaneously analyzed within only 8 min with acceptable resolution (2.0-8.2 for As, 1.6-6.1 for Hg and 2.7-4.0 for Pb). The detection limits were 0.036-0.20 for As-species, 0.023-0.041 for Hg-species, and 0.0076-0.14 mu g L-1 for Pb-species. The developed method was applied for the measurement of five lotus seed samples, indicating the presence of DMA (19.6-28.2 mu g kg(-1)), TML (1.4-2.9 mu g kg(-1)), MeHg (1.2-4.8 mu g kg(-1)) and EtHg (0.8-2.2 mu g kg(-1)). This method provides a promising tool for studying the toxic, metabolic and bioavailable behaviors of arsenic, mercury and lead.

Abstract:There are many kinds of micronutrient elements contained in foods that are vital to human health. The strong dependence of the toxicity and bioavailability of microelement upon their chemical forms has made the speciation and bioavailability vital to the food quality control and nutrition evaluation. A brief introduction to the high performance liquid chromatography hyphenated with inductive coupled plasma mass spectrometry ( HPLC-ICP-MS) is given. The hyphenated technique which based on several chromatographic separation mechanisms plays an important role in speciation analysis for its advantage of high sensitivity, high selectivity, wide linearity, low detection and multi-microelement determination simultaneously. The application of HPLC-ICP-MS in microelements speciation analysis and bioavailability research in foods concerning the selenium speciation analysis in selenized yeast, selenium-enriched garlic, selenium nutritional supplement and other foods, the arsenic speciation analysis in sea-products and farm products, the other microelements speciation analysis and the investigation of metabolic and bioactivity of microelement in organism is reviewed in this paper.

Abstract:Chemical speciation is a very important subject in the environmental protection, toxicology, and chemical analytics due to the fact that toxicity, availability, and reactivity of trace elements depend on the chemical forms in which these elements occur. Research on low analyte levels, particularly in complex matrix samples, requires more and more advanced and sophisticated analytical methods and techniques. The latest trends in this field concern the so-called hyphenated techniques. Arsenic, antimony, chromium, and (underestimated) thallium attract the closest attention of toxicologists and analysts. The properties of those elements depend on the oxidation state in which they occur. The aim of the following paper is to answer the question why the speciation analytics is so important. The paper also provides numerous examples of the hyphenated technique usage (e.g., the LC-ICP-MS application in the speciation analysis of chromium, antimony, arsenic, or thallium in water and bottom sediment samples). An important issue addressed is the preparation of environmental samples for speciation analysis.

Abstract:Due to the fact that metals and metalloids have a strong impact on the environment, the methods of their determination and speciation have received special attention in recent years. Arsenic, antimony, and thallium are important examples of such toxic elements. Their speciation is especially important in the environmental and biomedical fields because of their toxicity, bioavailability, and reactivity. Recently, speciation analytics has been playing a unique role in the studies of biogeochemical cycles of chemical compounds, determination of toxicity and ecotoxicity of selected elements, quality control of food products, control of medicines and pharmaceutical products, technological process control, research on the impact of technological installation on the environment, examination of occupational exposure, and clinical analysis. Conventional methods are usually labor intensive, time consuming, and susceptible to interferences. The hyphenated techniques, in which separation method is coupled with multidimensional detectors, have become useful alternatives. The main advantages of those techniques consist in extremely low detection and quantification limits, insignificant interference, influence as well as high precision and repeatability of the determinations. In view of their importance, the present work overviews and discusses different hyphenated techniques used for arsenic, antimony, and thallium species analysis, in different clinical, environmental and food matrices.

Abstract:Speciation analytics as determination of various forms of elements is becoming more and more popular and valuable in areas such as environmental protection, biochemistry, geology, medicine, pharmaceuticals, and food product quality control. Its growing importance is related to the fact that what often decides the toxicological properties of a given element or compound is not its total content but the presence of its various forms, especially ionic. Determination of low concentrations of analytes requires complex and sophisticated analytical methods and techniques. The latest trends in this area concern the so-called “hyphenated techniques,” in which the separation and various detection methods are combined. Abstract:Mercury (Hg) and its compounds are highly toxic for humans and ecosystems, and their chemical forms determine both their behavior and transportation as well as their potential toxicity for human beings. Determining the various species of an element is therefore more crucial than understanding its overall concentration in samples. For this reason, several studies focus on the development of new analytical techniques for the identification, characterization, and quantification of Hg compounds. Commercially available, hyphenated technology, such as HPLC-ICP-MS, supports the rapid growth of speciation analysis. This review aims to summarize and critically examine different approaches for the quantification of mercury species in different samples using HPLC-ICP-MS. The steps preceding the quantification of the analyte, namely sampling and pretreatment, will also be addressed. The scenarios evaluated comprehend single and multi-element speciation analysis to create a complete guide about mercury content quantification.

Abstract:Toxicity, mobility, bioavailability and biofunctions of chromium, cadmium, mercury and lead are heavily dependent upon their specific chemical forms, leading to a high demand to metal speciation analysis rather than total quantification. Simultaneous speciation analysis of multiple metal(loid)s is attractive to a large sample capacity containing unstable analytes due to its economic and environmental advantages over the conventional single elemental strategies. In this work, an analytical method integrating online solid phase extraction into high performance liquid chromatography interfaced with inductively coupled plasma mass spectrometry (ICP-MS) to simultaneously preconcentrate and quantify Cr, Cd, Hg and Pb forms in pg L-1 levels in water was developed. Cr (III + VI), Cd(II), Hg(II), Pb(II), methylmercury (MeHg), ethylmercury (EtHg), and trimethyl (TML) and triethyl lead (TEL) were captured by the C18 adsorbent (equilibrated with 10 mL of 1.0 mM 2-hydroxyethanethiol at 10 mL min-1) and eluted by mobile phase (5.0 mM Cys at pH 2.0), then completely separated on the C18 column within 8.0 min and eventually determined by ICP-MS. Low limits of detection (0.001-0.007 ng L-1) and quantification (0.003-0.023 ng L-1), good relative standard deviations (<4%) and high enrichment factors (827-2656 folds) were obtained with good linearities. Three reference materials of total cadmium (GBW08602), total mercury (GBW08603) and total lead (GBW08601) in water were analyzed by the developed method to validate the accuracy with good agreement with certified values and satisfactory recoveries (92-100%). This method was proved feasible by the determination of Cr, Cd, Hg and Pb compounds in drinking water, river water, pond water and tap water.

Abstract:The knowledge of elemental speciation and interaction between elemental species and ligands (including biomolecules) can provide an understanding of their transport, transformation, bioavailability, metabolic pathways and toxicity in organisms. By taking advantage of the powerful separation capabilities of capillary electrophoresis (CE) in capillaries/microchannels, and the atomic specificity, multi-elemental characteristics and extremely high sensitivity of inductively coupled plasma spectrometries (ICPs), the CE-ICP hyphenation is one of the most preferred choices in elemental speciation and element-ligand/element-biomolecule interaction as well as the indirect determination of non-metallic compounds. This article provides an overview of electrophoresis in capillaries and microchannels coupled to an ICP with respect to interfaces and applications from elemental speciation, element-ligand interaction to indirect determination.

Abstract:Arsenic (As), Selenium (Se) and Mercury (Hg) are three trace elements that have been the subject of much analytical discussion and investigation over the last three decades. While Selenium (Se) is among the list of essential trace elements necessary for the regulation of metabolic processes and overall health, As and Hg are not, and have been the centre of various cases surrounding the contamination of food, water and the environment. The focus of this review is to explore the area of chemical speciation, particularly as it relates to the measurement of these elements in various clinical matrices by HPLC-ICP-MS. This review will highlight the importance of accurately identifying the various chemical species of each of these elements, especially when considering their respective toxicological impacts on human health.

Abstract:Metal speciation analysis of microsamples in the clinical, biological and forensic fields is important for elucidating the metals' toxicity, mobility and metabolic behaviors of metals. Such analysis may be achieved by nanoliter high-performance liquid chromatography (nanoHPLC) hyphenated with inductively coupled plasma mass spectrometry (ICP-MS). In this work, an in-column high-pressure nebulizer (ICHPN) was developed to enhance the sensitivity and the separation efficiency of the sheathless nanoHPLC-ICP-MS coupling. The ICHPN consists of two concentric fused-silica capillaries with tapered tips connected via a PEEK tee, where C-18 silica particles with a size of 5 mu m were packed in the tapered inner capillary based on the keystone effect. Combining a heated single pass spray chamber with a makeup gas, the ICHPN was capable of independently optimizing the nebulization efficiency and the transport efficiency. The ICHPN offered high sensitivities, low detection limits and good precision at nanoflow rates. Compared with commercial nebulizers, the ICHPN fabrication was simple, rapid, reproducible and inexpensive. By implementing the ICHPN, we achieved rapid separation of four mercury species, i.e., Hg2+ and methyl-, ethyl- and phenylmercuric chloride, within 8.0 min with good resolution (2.0-13.9). Detection limits of 0.044-0.13 mu g L-1 were obtained with precisions of peak heights and areas ranging from 1.5 to 3.5% for a 50 mu g L-1 standard solution. Good agreement between the determined and certified values of mer- cury species in a certified reference material of caprine blood (SRM 955c) together with good recoveries (93-101%) validated the accuracy of the method. Abstract:Multi-elemental speciation analysis in a single run reduces costs, waste and time, and also sample consumption, which is favorable to a large set of instable samples. This work demonstrates mercury and arsenic speciation in a single run using high performance liquid chromatography with inductively coupled plasma mass spectrometric detection (HPLC/ICP-MS). Thiol compounds in mobile phases interfered with the elution of arsenite because of coordination action, which was overcome by gradient elution (elution of As(III) by thiol-free mobile phases at first). Four arsenic forms (As(III), DMA, MMA and As(V)) and three mercury species (Hg(II), MeHg and EtHg) were separated and detected by HPLC/ICP-MS using 4 mM tetrabutylammonium hydroxide (TBAH) (pH 6.0) in the first 1.0 min and 4 mM TBAH + 20 mM L-cysteine (pH 6.0) in the next 9.0 min. The detection limits were obtained at 0.078 for As(III), 0.081 for DMA, 0.078 for MMA and 0.15 mu g L-1 for As(V), and 0.016 for Hg(II), 0.027 for MeHg and 0.032 mu g L-1 for EtHg, and the reproducibilities (50 mu g L-1 for each As-species and 10 mu g L-1 for each Hg-species) were all lower than 3.7%. The proposed method was successfully applied for speciation analyses of arsenic and mercury in five lotus roots, which indicated DMA (3.5-7.9 mu g kg(-1)) as the only arsenic species and MeHg (0.6-1.0 mu g kg(-1)) as the only mercury species. Analyses of two certified reference materials demonstrated good agreement between determined values and certified values. All results prove its advantages including high sensitivity and high analytical efficiency, which are beneficial for routine speciation analyses of arsenic and mercury in a single run in environmental and food fields.

Abstract:Tin and lead are a global concern considering their species-dependent toxicity, bioavailability and transformation. Simultaneous speciation analysis of tin and lead is challenging for a large food capacity containing unstable species. Herein, we developed two sensitive methods for rapid quantification of tin and lead species in Antarctic seafood by high-performance liquid chromatography and inductively coupled plasma mass spectrometry based on strong cation-exchange and Amphion columns. Inorganic tin and lead, four organotin and two organolead compounds can be analysed in 16 min on a 10-cm Amphion II column (mobile phase: 4 mM sodium dodecyl benzene sulfonate at pH 2.0) with 0.02-0.24 mu g L-1 detection limits. The method was applied to Antarctic krill and fish, demonstrating the presence of any tin and lead species down to mu g kg-1 level. Overall, the proposed methods are sensitive, efficient and environment-friendly for routine speciation analysis of tin and lead in food samples.

Abstract:Elemental speciation is reviewed as a new approach to trace metals analysis. The importance of metal speciation relative to total metal analysis is considered. Various types of chromatographic separation methods using inductively coupled plasma-mass spectrometry (ICP-MS) detection are discussed. A brief introduction to the instrumental techniques is given, as well a discussion of coupling the instrumentation. Various chromatographies are included with an emphasis on liquid chromatography (LC). Gas chromatography (GC) and capillary electrophoresis (CE) are also discussed. Application examples contrast and compare the advantages and disadvantages of ICP- MS with other detection methods. Summary tables are included for two of the LC methods.

Abstract:Quantification of ultra-trace inorganic and organic species of lead and mercury in unpolluted environmental water is crucial to estimate the mobility, toxicity and bioavailability and interactions. Simultaneous pre-concentration of Pb and Hg species in pg L-1 levels followed by multi-elemental speciation analysis makes great sense to a large set of unstable samples because of time advantages. Herein simultaneous enrichment and speciation analysis of ultra-trace lead and mercury in water was developed by online solid-phase extraction coupled with high performance liquid chromatography and inductively coupled plasma mass spectrometry (SPE-HPLC-ICP-MS) for this aim. Pb(II), trimethyl lead (TML), triethyl lead (TEL), Hg(II), methylmercury (MeHg) and ethylmercury (EtHg) were baseline separated in 11 min under gradient elution using 5 mM L-cysteine (Cys) at pH 2.5 in the 0-1 and 4-15 min and 5 mM Cys + 0.5 mM tetrabutyl ammonium hydroxide solution at pH 2.5 in the 1-4 min. Lead and mercury species in 10 mL intact water samples were adsorbed on a 1 cm C18 enrichment column preconditioned with 10 mL of 1 mM 2-mercaptoethanol at 10 mL min-1, and then directly desorbed by the mobile phases. High enrichment factors (459 for Pb(II), 1248 for TML, 1627 for TEL, 2485 for Hg(II), 1984 for MeHg and 1866 for EtHg) were obtained with good relative standard deviations (<5%), leading to low LODs (0.001-0.011 ng L-1) and LOQs (0.004-0.036 ng L-1). Good accuracy of this method was validated by two certified reference materials of total lead in water (GBW08601) and total mercury in water (GBW08603) along with spiked recoveries (89-93%). The method was applied to analyze trace lead and mercury species in river, lake, tap and rain water, and purified and mineral water. Inorganic lead of 13-68 ng L-1 and inorganic mercury of 21-49 ng L-1 were measured in the nine water samples whereas TML, TEL and MeHg were not detected with 2-5 ng L-1 EtHg presented only in one river water and tap water. Abstract:A hyphenated method for mercury speciation analysis by the coupling of high performance liquid chromatography and inductively coupled plasma mass spectrometry with the online strong anion exchange column (SAX) preconcentration was developed. The Hg analytes (Hg(+), MeHg, EtHg and Hg(2+)) were absorbed on the SAX column preconditioned with sodium 3-mercapto-1-propanesulfonate, and then rapidly eluted (less than 16s) by 5μL 3% (v/v) 2-mercaptoethanol. The enrichment factors of 1025 for Hg(+), 1084 for MeHg, 1108 for EtHg and 1046 for Hg(2+) were obtained using 6mL sample in a 1.5-min enrichment procedure. Rapid separation of the four mercurial compounds was achieved within 5min on a 50-mm C18 column using 0.5% (v/v) 2-mercaptoethanol as the mobile phase. The detection limits for Hg(+), MeHg, EtHg and Hg(2+) were 0.015, 0.010, 0.009 and 0.016ng L(-1), each, and the relative standard deviations of peak height and peak area (5ng L(-1) for each Hg species) were all below 5%. Mercury speciation in three freshwater, two drinking water and two seawater samples were then analyzed by the proposed method. MeHg and Hg(2+) concentrations down to 0.14 and 0.56ng L(-1) were detected in the drinking waters.

Abstract:The applicability of the hydride generation (HG) sample introduction technique combined with different spectrochemical detection methods for non-chromatographic speciation of toxic As species, i.e., As(III), As(V), dimethylarsinate (DMA) and monomethylarsonate (MMA), in waters and other environmental, food and biological matrices is presented as a promising tool to speciate As by obviating chromatographic separation. Different non-chromatographic procedures along with speciation protocols reported in the literature over the past 20 year are summarized. Basic rules ensuring species selective generation of the corresponding hydrides are presented in detail. Common strategies and alternative approaches are highlighted. Aspects of proper sample preparation before analysis and the selection of adequate strategies for speciation purposes are emphasized.

Abstract:The main aim of the research was to develop a complementary analytical approach consisting of bespoke speciation analysis and non-targeted speciation analysis of As, Sb, and Cr in flavored bottled drinking water samples using HPLC/ICP-DRC-MS and ESI-MS/MS. The scope of two previously developed analytical procedures, (1) multielemental speciation procedure for AsIII, AsV, CrVI, SbIII, and SbV analysis and (2) arsenic speciation procedure for AsB, AsIII, DMA, MMA, and AsV quantification, was extended to the analysis of a new sample type in terms of bespoke speciation analysis. As for the non-targeted speciation, analysis size exclusion chromatography was used with ICP-MS and a complementary technique, ESI-MS/MS, was used for the organic species of As, Sb, and Cr screening. Full validation of procedures 1 and 2 was conducted. Procedure 1 and 2 were characterized with precision values in the range from 2.5% to 5.5% and from 3.6% to 7.2%, respectively. Obtained recoveries ranged from 97% to 106% and from 99% to 106% for procedures 1 and 2, respectively. Expanded uncertainties calculated for procedures 1 and 2 ranged from 6.1% to 9.4% and from 7.4% to 9.9%, respectively. The applicability of the proposed procedures was tested on bottled drinking water samples. Results for the real samples in procedure 1 were in the range from 0.286 ± 0.027 [μg L−1] to 0.414 ± 0.039 [μg L−1] for AsIII, from 0.900 ± 0.083 [μg L−1] to 3.26 ± 0.30 [μg L−1] for AsV, and from 0.201 ± 0.012 [μg L−1] to 0.524 ± 0.032 [μg L−1] for SbV. CrVI and SbIII were not detected in any sample. As for procedure 2, results were in the range from 0.0541 ± 0.0053 [μg L−1] to 0.554 ± 0.054 [μg L−1] for AsB. Results for AsIII and AsV obtained with procedure 2 were in good accordance with results obtained with procedure 1. DMA and MMA were not detected in any sample.
