Abstract:A concise and efficient synthetic approach was developed for the synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones via a one-pot, three-component reaction of 4-aminocoumarin, arylglyoxal monohydrates, and aromatic amines promoted by KHSO4 in toluene under reflux condition. The attractive features of this protocol are operational simplicity, short reaction time, easy purification of products and good to excellent yields.

Abstract:Abstract A two-step, diversity-oriented synthesis of highly functionalized benzofuran-2-carboxamides has been developed. The Ugi four-component reaction of aromatic amines, isocyanides, 2-bromobenzaldehyde, and 2-bromoacetic acid was used to provide a collection of N-aryl 2-bromoacetamides. The latter were then reacted with selected salicyladehydes in the presence of Cs2CO3 in acetonitrile under controlled microwave heating at 90–140 °C for 15–40 minutes to furnish the benzofuran-2-carboxamides in moderate to good yields.

Abstract:A facile preparation of 3-aminofuran derivatives via multicomponent reactions of thiazole carbenes, aldehydes, and dimethyl acetylenedicarboxylate (DMAD) is reported. In this process, the thiazole carbenes, generated in situ from thiazolium salts, reacted with aldehydes and DMAD at -78 to 0 degrees C in CH2Cl2 to afford the substituted furans in moderate to good yields. Eight substituted thiazolium salts were employed as carbene precursors in the reaction. Besides aryl aldehydes, alpha beta-unsaturated aldehydes, aliphatic aldehydes, and arenedial were also investigated and found to be applicable to this reaction.

Abstract:Benzodifurazan reacts with 1,3-dibromo-5,5-dimethylhydantoin in sulfuric acid to form 5-bromo-4,5-dihydrobenzo[1,2- c :3,4- c ']bis([1,2,5]oxadiazol)-4-ol ( A ). The structure of the product was established by X-ray diffraction analysis. It was found that compound A exhibits properties of epoxides: it readily takes up nucleophiles to form alcohols, whose reaction with p -toluenesulfonyl chloride in the presence of tetrabutylammonium bromide and sodium hydroxide in benzene gives functional derivatives of benzodifurazan. On treatment with a base [Ca(OH) 2 ], compound A converts to 1a,7b-dihydrooxireno[2',3':5,6]benzo[1,2- c :3,4- c ']bis([1,2,5]oxadiazole).

Abstract:A facile and highly efficient three-component reaction of thiazole or benzothiazole carbenes, disubstituted ketenes, and activated alkynes is disclosed. With this methodology, a polysubstituted ring system containing furo[2,3-c]thiazepine core can be constructed from simple and readily accessible starting materials in good yields. The scope and limitation of this transformation were investigated in detail by using various thiazole carbene, ketene, and alkyne components. Furthermore, the synthetic utilities of these unique polyheterocyclic compounds were demonstrated via their Diels-Alder reactions with benzynes to furnish thiazepine-fused 7-oxanorbornadiene derivatives in excellent yields.

Abstract:A mild three-component synthetic approach to versatile 2-amino-1,4-dihydropyridines from terminal alkynes, sulfonyl azides, and N-sulfonyl-1-aza-1,3-butadienes was successfully developed and relied on the in situ generation of metalated ynamide intermediates Ib to achieve a formal inverse electron-demand hetero-Diels-Alder reaction. Experimental results suggest that alkali metal cations (Li(+) and Cs(+) ions) might play a critical role to achieve the cycloaddition process.

Abstract:A palladium(0)-catalyzed cascade reaction for the efficient synthesis of 2,3-disubstituted benzofuran derivatives 3 containing a 3-trisubstituted alkene functional group in moderate yields from alkynyl-substituted benzynes 1 and aryl halides 2 has been developed. This method provides an efficient and alternative approach to benzofurans which are very useful heterocyclic compounds with biological and pharmacological potentials. A plausible mechanism involving intramolecular ene reaction, intermolecular insertion, and β-H elimination is proposed.

Abstract:Although various methods for the preparation of dibenzofurans are available, an efficient, general synthesis of dibenzofurans from benzofurans remains an unsolved problem. Herein, we provide the first benzannulation of readily prepared 2‐nitrobenzofurans with alkylidene malononitriles. This methodology enables facile assembly of a wide variety of highly functionalized dibenzofurans in moderate to excellent yields under metal‐free conditions. Gram‐scale synthesis and further elaborations of the product were succesfully performed, demonstrating synthetic utility of this method.

Abstract:An efficient N-heterocyclic carbene (NHC)-catalyzed asymmetric conjugate addition reaction to afford synthetically challenging benzofuranone derivatives having vicinal all-carbon quaternary and tertiary stereocenters is presented. The reaction operates solely through noncovalent interaction between a newly designed NHC and the substrates, providing access to a series of functionalized benzofuranones in good yields and with high ee values. The protocol applies to preparative-scale synthesis. A catalytic cycle involving a noncovalent substrate–NHC interaction is implicated in the process, based on a mechanistic control study. Abstract:A Lewis-acid-promoted approach to the synthesis of highly functionalized dihydrobenzofuran derivatives was developed. A diverse range of functional groups are tolerated in this type of reaction. The reaction mechanism investigation indicates that the highly reactive phenyl cation intermediate is probably involved in this process. The chirality of substrate is retained under the reaction conditions.

Abstract:In this research, the functionalized 1,6‐naphthyridines were synthesized using multicomponent reaction of benzaldehydes, malononitrile, and 4‐aminocumarin in water at room temperature in the presence of SiO2/Fe3O4@MWCNTs as a recyclable catalyst in high yields. This study synthesized 1,6‐naphthyridine as a novel class of fused heterocyclic compounds with high efficiency using a multicomponent reaction. The reaction involved benzaldehyde or its derivatives, 2 mol of malononitrile, and 4‐aminocumarin in an aqueous solution at room temperature in the presence of SiO2/Fe3O4@MWCNTs as a recyclable catalyst. This study examines the antioxidant properties of 1,6‐naphthyridine in addition to the other research undertaken in this work. The production process of 1,6‐naphthyridine demonstrated several advantageous features, including rapid reactions, high yields of the final product, and straightforward separation of the catalyst and product from the reaction mixture.

Abstract:A novel method for the synthesis of benzofuran analogues via condensation of 2-hydroxybenzaldehyde or 2-hydroxyphenyl ketone with methyl 4-bromocrotonate was reported. Moderate to good yield was obtained with cesium carbonate as base in DMF at 70 degrees C. The new-generated double bond was (E)-configuration determined by X-ray diffraction. All benzofuran derivatives were determined by H-1 NMR, C-13 NMR and HRMS techniques.

Abstract:A series of functionalized 3-fluoro-2,3-dihydro-4H-pyran derivatives were efficiently synthesized from ethyl 2-fluoroacetoacetate 1, malononitrile 2 and aromatic aldehydes 3 via one-pot, multi-component reactions catalyzed by NaOH. The solvent ethanol played dual roles as both reactant and reaction medium, resulting in the further occurrence of intramolecular cyclization to afford cyclic products 4. The structures of product 4 were determined by spectral methods and X-ray diffraction analysis.
