Abstract:A facile preparation of 3-aminofuran derivatives via multicomponent reactions of thiazole carbenes, aldehydes, and dimethyl acetylenedicarboxylate (DMAD) is reported. In this process, the thiazole carbenes, generated in situ from thiazolium salts, reacted with aldehydes and DMAD at -78 to 0 degrees C in CH2Cl2 to afford the substituted furans in moderate to good yields. Eight substituted thiazolium salts were employed as carbene precursors in the reaction. Besides aryl aldehydes, alpha beta-unsaturated aldehydes, aliphatic aldehydes, and arenedial were also investigated and found to be applicable to this reaction.

Abstract:A concise and efficient synthetic approach was developed for the synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones via a one-pot, three-component reaction of 4-aminocoumarin, arylglyoxal monohydrates, and aromatic amines promoted by KHSO4 in toluene under reflux condition. The attractive features of this protocol are operational simplicity, short reaction time, easy purification of products and good to excellent yields.

Abstract:A mild, efficient, and environmentally benign one-pot synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones by a three-component domino reaction of 4-aminocoumarins, arylglyoxal monohydrates, and 1,3-dicarbonyl compounds in refluxing ethanol without the use of catalyst is reported. Abstract:We have developed a novel and efficient method for the preparation of 2,4-disubstituted-1,2,3-triazoles. These compounds 3a-3p were synthesized in good yield by refluxing alpha-aminoacetophenones and hydrazines in glacial acetic acid. The reaction proceeds under mild conditions and is compatible with various functional groups.

Abstract:A novel method has been developed for the synthesis of 1,3-thiazin-4-one compounds containing trifluoromethyl groups utilizing 2-trifluoromethyl acrylic acid and thioamides as key starting materials. This protocol is characterized by its simplicity, practicality, and tolerance toward various functional groups. Given the straightforward nature of the procedure, the ready availability of both starting materials, and the significance of drugs containing trifluoromethyl, it is anticipated that this reaction will have wide-ranging applications.

Abstract:A facile and efficient one-pot three-component reaction method for the synthesis of thiazine-dicarboxylates is reported. Reaction of an isocyanide and dialkyl acetylenedicarboxylate with 2-amino-4H-1,3-thiazin-4-one derivatives containing both an acidic proton and an internal nucleophile gave the products in good yields of 76–85%. The reactivity of dialkyl acetylenedicarboxylates was further tested in the synthesis of thiazole-pyrimidines where a two-component reaction of 2-aminothiazole with dialkyl acetylenedicarboxylates was successfully converted to a more efficient three-component reaction of a thiourea, α-haloketone and dialkyl acetylenedicarboxylate (DMAD/DEtAD) to give thiazole-pyrimidines in good yields of 70–91%.

Abstract:Abstract A convenient approach for the synthesis of thiazolo[3,4-a]-benzimidazole derivatives has been developed. Three-component reaction of readily available o-phenylenediamines, aryl isothiocyanate, and 4-chloro-3-oxobutanoate in toluene–dichloromethane without any catalyst proceeds under reflux to afford the title compounds in high yields. The structure of the target molecule was confirmed by X-ray diffraction.

Abstract:A novel and straightforward strategy for the synthesis of functionalized thiazoles from thioamides, ynals, and alcohols via a copper(I)-catalyzed three-component reaction has been described. Through the formation of new C-S, C-N, and C-O bonds in one pot, it is easy to produce various valuable thiazoles fixed with aryl or heteroaryl groups. In addition, the reaction also exhibits other unique advantages, such as high step economics, good functional group tolerance, and good regioselectivity.

Abstract:We reported copper-catalyzed the 1,4- or 1,6-conjugate additions of arylidenemalonitriles or para-quinone methides with 2-bromodifluoromethylbenzo-1,3-thiazole for the preparation of benzothiazolyldifluoroalkyl containing (di)arylmethine compounds in moderate to excellent yields, providing a facile route for the synthesis of gem-difluoroalkylatated (di)arylmethine compounds. The synthetic utility of the current method was also demonstrated by a gram-scale reaction.

Abstract:We recently reported a new class of triphenylphosphine and isocyanide based multicomponent reactions (Ph3P/DAAD and IMCRs/DAAD) mediated by R3P/DAAD and RNC/DAAD zwitter-ionic intermediates in the presence of CH, OH, and NH acids. Herein. the reactions of Huisgen 1:1 zwitterionic intermediates, generated from diallyl acetylenedicarboxylates (DAAD), trivalent phosphorus reagents, and isocyanides, with aromatic carboxylic acids as initial proton source are investigated. This novel binucleophilic (R3P-RNC/DAAD) system afforded 2-aminofuran derivatives.

Abstract:The synthesis of 4-(trifluoromethyl)-2,3-dihydrothiazoles from alpha-enolic dithioesters and imidoyl sulfoxonium ylides has been achieved under thermal conditions. This transformation is catalyst-free, additive-free, and operationally simple, and it proceeds via a formal insertion of alkenyl S-H generated in situ from dithioesters into ylides followed by an intramolecular annulation. This approach further expands the synthetic application of two versatile sulfur-containing building blocks of fluorinated imidoyl sulfoxonium ylides and alpha-enolic dithioesters.

Abstract:A series of novel urea derivatives containing the 1,2,3-thiadiazole moiety were synthesised using microwave-assisted condensation reactions of 4-methyl-1,2,3-thiadiazole-5-carbohydrazide with various substituted isocyanatobenzenes in acetonitrile. Their chemical structures were characterised by 1H NMR, MS and elemental analysis. This method provides several advantages such as high yields, facile work-up and is environmentally friendly.
