Abstract:A facile preparation of 3-aminofuran derivatives via multicomponent reactions of thiazole carbenes, aldehydes, and dimethyl acetylenedicarboxylate (DMAD) is reported. In this process, the thiazole carbenes, generated in situ from thiazolium salts, reacted with aldehydes and DMAD at -78 to 0 degrees C in CH2Cl2 to afford the substituted furans in moderate to good yields. Eight substituted thiazolium salts were employed as carbene precursors in the reaction. Besides aryl aldehydes, alpha beta-unsaturated aldehydes, aliphatic aldehydes, and arenedial were also investigated and found to be applicable to this reaction.

Abstract:A facile and highly efficient three-component reaction of thiazole or benzothiazole carbenes, disubstituted ketenes, and activated alkynes is disclosed. With this methodology, a polysubstituted ring system containing furo[2,3-c]thiazepine core can be constructed from simple and readily accessible starting materials in good yields. The scope and limitation of this transformation were investigated in detail by using various thiazole carbene, ketene, and alkyne components. Furthermore, the synthetic utilities of these unique polyheterocyclic compounds were demonstrated via their Diels-Alder reactions with benzynes to furnish thiazepine-fused 7-oxanorbornadiene derivatives in excellent yields.

Abstract:A Lewis-acid-promoted approach to the synthesis of highly functionalized dihydrobenzofuran derivatives was developed. A diverse range of functional groups are tolerated in this type of reaction. The reaction mechanism investigation indicates that the highly reactive phenyl cation intermediate is probably involved in this process. Abstract:A novel method has been developed for the synthesis of 1,3-thiazin-4-one compounds containing trifluoromethyl groups utilizing 2-trifluoromethyl acrylic acid and thioamides as key starting materials. This protocol is characterized by its simplicity, practicality, and tolerance toward various functional groups. Given the straightforward nature of the procedure, the ready availability of both starting materials, and the significance of drugs containing trifluoromethyl, it is anticipated that this reaction will have wide-ranging applications.

Abstract:We have developed a novel and efficient method for the preparation of 2,4-disubstituted-1,2,3-triazoles. These compounds 3a-3p were synthesized in good yield by refluxing alpha-aminoacetophenones and hydrazines in glacial acetic acid. The reaction proceeds under mild conditions and is compatible with various functional groups.

Abstract:Abstract A convenient approach for the synthesis of thiazolo[3,4-a]-benzimidazole derivatives has been developed. Three-component reaction of readily available o-phenylenediamines, aryl isothiocyanate, and 4-chloro-3-oxobutanoate in toluene–dichloromethane without any catalyst proceeds under reflux to afford the title compounds in high yields. The structure of the target molecule was confirmed by X-ray diffraction.

Abstract:The furan and thiophene skeleton structures are widely present in natural products and synthetic drugs, and display multiple bioactivities. An effective one-pot three-component reaction to access furan and thiophene derivatives from 2-bromo-5-(dichloromethyl)furan or -thiophene; cyanoacetate esters, cyanoacetamides, or malononitrile; and aliphatic secondary amines has been developed. The multicomponent reaction for the construction of new C–N and C–C bonds in one pot is simple, mild, and efficient, and the yield of the target product is as high as 96%. Abstract:A simple and efficient strategy for the chemoselective synthesis of (2 Z ,5 Z )-3-benzyl/alkyl-5-(2-oxo-5-arylfuran-3(2 H )-ylidene)-2-(phenylimino)thiazolidin-4-one derivatives is developed via a sequential three-component reaction of readily available phenacyl bromides, pyridine and methyl 2-(3-benzyl/alkyl-4-oxo-2-(phenylimino)thiazolidin-5-ylidene)acetates in acetonitrile at room temperature. The advantages of this one-pot sequential Michael addition/1,2-H shift/elimination of pyridine/intramolecular cyclization are highlighted by its low energy requirements (short reaction times at room temperature), excellent yields, the absence of a metal catalyst, and mild conditions. The product structures are characterized by spectroscopic techniques and single-crystal X-ray analysis. Abstract:A facile and efficient one-pot three-component reaction method for the synthesis of thiazine-dicarboxylates is reported. Reaction of an isocyanide and dialkyl acetylenedicarboxylate with 2-amino-4H-1,3-thiazin-4-one derivatives containing both an acidic proton and an internal nucleophile gave the products in good yields of 76–85%. The reactivity of dialkyl acetylenedicarboxylates was further tested in the synthesis of thiazole-pyrimidines where a two-component reaction of 2-aminothiazole with dialkyl acetylenedicarboxylates was successfully converted to a more efficient three-component reaction of a thiourea, α-haloketone and dialkyl acetylenedicarboxylate (DMAD/DEtAD) to give thiazole-pyrimidines in good yields of 70–91%.

Both experimental and DFT studies revealed that this Ag-catalyzed unusual transformation is initiated by a facile aza-Michael addition instead of the conventional imine condensation. Abstract:We recently reported a new class of triphenylphosphine and isocyanide based multicomponent reactions (Ph3P/DAAD and IMCRs/DAAD) mediated by R3P/DAAD and RNC/DAAD zwitter-ionic intermediates in the presence of CH, OH, and NH acids. Herein. the reactions of Huisgen 1:1 zwitterionic intermediates, generated from diallyl acetylenedicarboxylates (DAAD), trivalent phosphorus reagents, and isocyanides, with aromatic carboxylic acids as initial proton source are investigated. This novel binucleophilic (R3P-RNC/DAAD) system afforded 2-aminofuran derivatives.
