Abstract:From Dendrobium plicatile stems, three phenanthrenes were isolated. The structures are 2,5-dihydroxy-4,9,10-trimethoxyphenanthrene, 2,5-dihydroxy-4-methoxyphenanthrene and 2,5,9-trihydroxy-4-methoxy-9,10-dihydrophenanthrene.

Abstract:From the CHCl3-soluble portion of the 70% EtOH extract of the stems and leaves of Dioscorea nipponica Makino, two new dihydrophenanthrenes were isolated, 2,7-dihydroxy-3,4,6-trimethoxy-9,10-dihydrophenanthrene (1), 4,4',7,7'-tetrahydroxy-2,2',6,6'-tetramethoxy-1,1'-bi-9,10-dihydrophenanthrenyl (2). The structures were determined by means of HRMS, 1H-NMR, 13C-NMR, and HMBC experiments.

Abstract:A new 9,10-dihydrophenanthrene,1,5-dihydroxy-3,4,7-trimethoxy-9,10-dihydrophenanthrene (1) was isolated and identified from the whole plants of Dendrobium moniliforme, as well as 24 known compounds including hircinol (2), (2R*,3S*)-3-hydroxymethyl-9-methoxy-2-(4'-hydroxy-3',5'-dimethoxyphenyl)-2,3,6,7-tetrahydro-phenanthro[4,3-b]furan-5,11-diol (3), diospyrosin (4), aloifol I (5), moscatilin (6), 3,4'-dihydroxy-3',4,5-trimethoxybibenzyl (7), gigantol (8), 3,3'-dihydroxy-4,5-dimethoxybibenzyl (9), longicornuol A (10), N-trans-cinnamoyltyramine (11), paprazine (12), N-trans-feruloyl 3'-O-methyldopamine (13), moupinamide (14), dihydroconiferyl dihydro-p-coumarate (15), dihydrosinapyl dihydro-p-coumarate (16), 3-isopropyl-5-acetoxycyclohexene-2-one-1 (17), p-hydroxybenzaldehyde (18), vanillin (19), p-hydroxyphenylpropionic acid (20), vanillic acid (21), protocatechuic acid (22), (+)-syringaresinol (23), β-sitosterol (24) and daucosterol (25). Antioxidant phenanthrenes and lignans from Dendrobium nobile

Abstract:To study the antioxidant constituents from the stems ofDendrobium nobile,and to discuss their structmre-activity relationship.Compounds were isolated from a 60% ethanolic extract by various chromatographic techniques and were identified by spectral analysis.The antioxidant activities of compounds were evaluated by DPPH free radical scavenging assay.Five phenanthrenes and four lignans were obtained from the active fractions of D.nobile.Their structures were identified as fimbriatone (1),confusarin (2),flavanthrinin (3),2,5-dihydroxy-4,9-dimethoxyphenanthrene (4),3,7-dihydroxy-2,4-dimethoxyphenanthrene (5),syringaresinol (6),pinoresinol (7),medioresinol (8) and lirioresinol-A (9),respectively.Compounds 2 and 6 exhibited more potent DPPH scavenging activities than vitamin C.All the above compounds were reported from this plant for the first time,and compounds 3,4 and 9 were reported for the first time from the genus of Dendrobium.For all phenanthrenes and lignans,an electron-donating methoxyl group in the ortho position to the phenolic hydroxyl group exhibits enhanced antioxidant activities.

Abstract:A new enantiomeric pair of spirodiketones, (+)- and (-)-denobilone A (1 and 2), three new phenanthrene derivatives (3-5), and three new biphenanthrenes (22-24), along with 11 known phenanthrene derivatives (6-16), five known bibenzyl derivatives (17-21), and four known biphenanthrenes (25-28), were isolated from Dendrobium nobile. The structures of 1-5 and 22-24 were elucidated using comprehensive spectroscopic methods. (+)-Denobilone and (-)-denobilone A (1 and 2) were isolated as a pair of enantiomers by chiral HPLC. The absolute configurations of (+)- and (-)-denobilone A (1 and 2) were determined by comparing their experimental and calculated electronic circular dichroism spectra. The absolute configuration of denobilone B (3) was determined by X-ray crystallographic analysis. The inhibitory activities of all compounds against nine phytopathogenic fungi and three cancer cell lines were evaluated.

Abstract:Three new phenanthrenes, thrigonosomone A (1), 6,9-O-dedimethyltrigonostemone (2), and thrigonosomone B (3) were isolated from Trigonostemon lii. Their structures were elucidated by spectroscopic methods. Thrigonosomone A (1) possessed a novel seven-membered cyclic anhydride moiety, and its relative configuration was determined by chemical computation and ROESY experiment. The cytotoxicity of compounds 1–3 was also evaluated.

Abstract:Objective To study the chemical constituents from ethanol extracts of the stems ofDendrobium aphyllum.Methods Ten compounds were isolated by silica gel,Sephadex LH-20 column chromatography and reversed-phase semi-HPLC.Results Ten compounds were identified by analyzing their spectral data and comparing with the previously reported literatures as 4,7-dihydroxy-2-methoxy-9,10-dihydroxyphenanthrene (1),2,4-dihydroxy-7-methoxy-9,10-dihydroxyphenanthrene (2),4-methoxy-2,5,7,9-tetrahydroxy-9,10-dihydrophe-nanthrene (3),5-methoxy-4,7,9-trihydroxy-9,10-dihydrophenanthrene (4),pinoresinol (5),eugenol-1-D-glucoside (6),trans-ferulicacid(7),dodecan-1-ol(8)β-sitosterol(9),andβ-daucosterol(10).Conclusion All compounds are isolated from this plant for the first time.

Abstract:One new phenanthrene, aphyllone A (1) and four new bibenzyl derivatives, aphyllone B (2) and aphyllals C–D (3–5), together with nine known compounds (6–14), were isolated from the stems of Dendrobium aphyllum (Roxb.) C. E. Fischer. The structures of these new compounds were elucidated by means of extensive spectroscopic analyses, and the absolute configuration of compound 1 was determined by single crystal X-ray diffraction and quantum calculations. Compounds 6, 8 and 14 inhibited NO production at the concentration of 25μM in LPS-stimulated RAW264.7 cells with the inhibition (%) of 32.48, 35.68, and 38.50. Compound 2 possessed significant DPPH radical scavenging activity with scavenging percentage of 87.97% at the concentration of 100μg/mL.

Abstract:One bi-bicyclic and two bi-tricyclic derivatives of coumarin-benzofuran, phenanthrene-phenanthrene and phenanthrene-phenanthraquinone, along with seven known compounds, were isolated from stems of Dendrobium thyrsiflorum Rchb.f. (Orchidaceae). On the basis of chemical, NMR (1H, 13C, HMQC, HMBC and NOESY) and mass spectrometry data, their structures were elucidated as denthyrsin [3-(5',6'-dimethoxybenzofuran-2'-yl)-6,7-dimethoxy-2H-chromen-2-one; 1], denthyrsinol (4,5'-dimethoxy-[1,1']biphenanthrenyl-2,5,4',7'-tetraol; 2), and denthyrsinone (7,4',7'-trihydroxy-2,2',8'-trimethoxy-[5,1']biphenanthrenyl-1,4-dione; 3). Compounds 1-3 and denthyrsinin (1,5,7-trimethoxyphenanthrene-2,6-diol; 4) showed significant cytotoxic activities against Hela (13.5, 9.3, 9.9 and 2.7 microM, respectively), K-562 (0.45, 1.6, 6.0 and 2.3 microM, respectively) and MCF-7 (18.1, not tested, 3.5 and 4.8 microM, respectively) cell lines.

Abstract:Three new dihydrophenanthrenes were isolated from the whole plant of Elatostema tenuicaudatum collected in Vietnam. These compounds were identified as 2,3,5-trihydroxy-9,10-dihydrophenanthrene ( 1 ), 2-methoxy-5-hydroxy-9,10-dihydrophenanthrene 3-O- β -D-glucopyranoside ( 2 ), and 2,5-dihydroxy-9,10-dihydrophenanthrene 3-O- β -D-glucopyranoside ( 3 ). Their structures were determined by HR-ESI-MS and 1D, 2D NMR spectroscopy. Furthermore, the inhibitory activities of these compounds against nitric oxide (NO) production in lipopolysaccharide-activated RAW264.7 cells were evaluated. Compound 1 exhibited significant inhibition of NO production, with an IC 50 value of 15.8 ± 1.2 μM. This study represents the first report on the chemical compositions and biological activities of E. tenuicaudatum .

Abstract:A new phenanthrene derivative 3,7-dihydroxy-2,4,6-trimethoxyphenanthrene was isolated from the all plant of Bulbophyllum odoratissimum, and its structure was elucidated by extensive spectral studies and chemical transformation. The compound displayed cytotoxicity against the growth of human leukemia cell lines K562 and HL-60, human lung adenocarcinoma A549, human hepatoma BEL-7402 and human stomach cancer cell lines SGC-7901 with IC50 values of 14.23, 10.02, 3.42, 15.36 and 1.13 mg/ml respectively.

Abstract:Three biphenanthrene compounds (1–3) and a triphenanthrene compound (4), together with six known biphenanthrene compounds (5–10), were isolated from the tubers of Monomeria barbata Lindl. Their structures were elucidated on the basis of extensive spectroscopic analysis (1D-, 2D-NMR, and HR-ESI-MS). These four new compounds were tested in vitro for the cytotoxic activity against liver carcinoma (HepG-2), promyelocytic leukaemia (HL60), ovarian carcinoma (Skov-3) and epidermoid carcinoma (A431) cancer cell lines and the antioxidant activity in DPPH radical scavenging. Compounds 1–4 exhibited significant cytotoxic activity against HepG-2 and HL60 cell lines, and potent antioxidant activity in DPPH radical scavenging.

Abstract:Two phenanthrenes were isolated from the aerial part of Dendrobium nobile Lindl. and their structures were identified to be 4,7-dihydroxy-2-methoxy-9,10-dihydrophenanthrene (1) and denbinobin (2), among which the former has been first isolated from this plant. These two compounds were found to be cytotoxic against A549 (human lung carcinoma), SK-OV-3 (human ovary adenocarcinoma), and HL-60 (human promyelocytic leukemia) cell lines. Compound 1 also showed antitumor activity on the life span of ICR mice intraperitoneally implanted with 1 x 10(6) cells of sarcoma 180.

Abstract:Chemical investigation of Flickingeria fimbriata (Bl.) Hawkes (Orchidaceae) resulted in the isolation and identification of one new dihydrophenanthrene, 1,2,5,6,7-pentamethoxy-9,10-dihydrophenanthrene (1), together with seven known dihydrophenanthrenes, erianthridin (2), coelonin (3), 4-methoxy-9,10-dihydrophenanthrene-1,2,7-triol (4), lusianthridin (5), ephemeranthol A (6), flavanthridin (7) and hircinol (8), four known phenanthrenes, epheranthol B (9), nudol (10), denthyrsinin (11) and confusarin (12), and one known bibenzyl, batatasin III (13). The structure of the new compound was elucidated by spectroscopic analysis (HRMS, 1D and 2D NMR). All the compounds were isolated from F. fimbriata for the first time except for compounds 5 and 12, and compounds 1, 3, 4, 8, 10, 11 and 13 were obtained from this genus for the first time. Compounds 1-4 showed moderate cytotoxic activity against HepG2 cells.

Abstract:Two novel compounds, (9R)-4-metboxy-9H-fluorene-2,5,9-triol (1a) and 2,7-dihydroxy-8-methoxyphenanthro[4,5-bcd]pyran-5(5H)-one (2) were isolated from the stems of the orchid Dendrobium chrysotoxum Lindl. Their structures were established spectroscopically, by chemical derivatization, and by single-crystal X-ray diffraction analysis. The absolute configuration of la was determined by Mosher's method. Neither compound showed antitumoral activity.

Abstract:Dendroochreatene ( 1 ), a new phenanthrene derivative with a spirolactone ring, was isolated from the whole Dendrobium ochreatum plant together with 11 known compounds ( 2 – 12 ). The structure of the new compound was elucidated spectroscopically and phenolic compounds were firstly reported from D. ochreatum. Moscatilin ( 4 ), major compound isolated from D. ochreatum, was found to be cytotoxic toward H460 lung-cancer cells, with an IC 50 value of 147.3 ± 0.9 μM, while loddigesiinol C ( 7 ), C- α -methoxy derivative was inactive.
