Abstract:Four pairs of novel enantiomeric phenanthrenes, named (+)/(−)-blephenanthropyran A [(+)/(−)- 1 ], (+)/(−)-blephenanthropyran B [(+)/(−)- 2 ], (+)/(−)-blephenanthrofuran A [(+)/(−)- 3 ], and (+)/(−)-blephenanthrofuran B [(+)/(−)- 4 ], were isolated from the tubers of Bletilla striata (Thunb.) Reichb.f. Their structures and absolute configurations were elucidated by extensive spectroscopic analysis and electronic circular dichroism calculations. Compounds (+)/(−)- 1 and (+)/(−)- 2 are 9,10,11,12-tetrahydrophenanthropyrones, while (+)/(−)- 3 and (+)/(−)- 4 are 9,10,11,12-tetrahydrophenanthrofurans. Since B. striata is known as "an excellent medicine for lung diseases" in traditional Chinese medicine, the effects of the isolates in A549 lung cancer cells were investigated. The results showed that compound (+)- 3 significantly inhibited the viability of A549 cells. Furthermore, (+)- 3 could promote the apoptosis of A549 cells by upregulating the protein expression of caspase3, cleaved caspase3, cleaved PARP, and Bax. Thus, compound (+)- 3 is an effective proliferation inhibitor and apoptosis inducer in lung cancer cells.
