Abstract:New phenanthrenes, loddigesiinols A (1) and B (7), and stilbenes, loddigesiinols C (8) and D (9), were isolated from 80% ethanol extract of Dendrobium loddigesii ROLFE (Orcharedceae) along with known compounds, including five phenanthrenes, three stilbenes, two lignans, and three sterols. Their structures were determined by spectroscopic studies. These compounds were evaluated for their inhibitory activities against nitric oxide (NO) production and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging. Phenanthrenes (1-3, 7) and dihydrophenanthrenes (4-6) showed significant inhibitory activity against NO production. In particular, 2 showed stronger inhibitory activity against NO production than 5 and 6. This suggested that this phenanthrene is a stronger inhibitor of NO production than dihydrophenanthrene. The new phenanthrenes, loddigesiinols A (1) and B (7), significantly inhibited NO production with IC50 values of 2.6 and 10.9 mu M, respectively. One new dihydrostilbene, loddigesiinol D (9), showed weak inhibitory activity against NO production with an IC50 value of 69.7 mu M, but the other new dihydrostilbene, loddigesiinol C (8), did not show this activity at 100 mu M.

Abstract:One new phenanthrene, aphyllone A (1) and four new bibenzyl derivatives, aphyllone B (2) and aphyllals C–D (3–5), together with nine known compounds (6–14), were isolated from the stems of Dendrobium aphyllum (Roxb.) C. E. Fischer. The structures of these new compounds were elucidated by means of extensive spectroscopic analyses, and the absolute configuration of compound 1 was determined by single crystal X-ray diffraction and quantum calculations. Compounds 6, 8 and 14 inhibited NO production at the concentration of 25μM in LPS-stimulated RAW264.7 cells with the inhibition (%) of 32.48, 35.68, and 38.50. Compound 2 possessed significant DPPH radical scavenging activity with scavenging percentage of 87.97% at the concentration of 100μg/mL.
Antioxidant phenanthrenes and lignans from Dendrobium nobile

Abstract:To study the antioxidant constituents from the stems ofDendrobium nobile,and to discuss their structmre-activity relationship.Compounds were isolated from a 60% ethanolic extract by various chromatographic techniques and were identified by spectral analysis.The antioxidant activities of compounds were evaluated by DPPH free radical scavenging assay.Five phenanthrenes and four lignans were obtained from the active fractions of D.nobile.Their structures were identified as fimbriatone (1),confusarin (2),flavanthrinin (3),2,5-dihydroxy-4,9-dimethoxyphenanthrene (4),3,7-dihydroxy-2,4-dimethoxyphenanthrene (5),syringaresinol (6),pinoresinol (7),medioresinol (8) and lirioresinol-A (9),respectively.Compounds 2 and 6 exhibited more potent DPPH scavenging activities than vitamin C.All the above compounds were reported from this plant for the first time,and compounds 3,4 and 9 were reported for the first time from the genus of Dendrobium.For all phenanthrenes and lignans,an electron-donating methoxyl group in the ortho position to the phenolic hydroxyl group exhibits enhanced antioxidant activities.

Their structures were established on the basis of extensive spectroscopic analyses. In addition, some compounds were tested for their cytotoxic and neurite outgrowth-promoting activities, as well as their antagonistic activity toward FXR ligand.

Abstract:Dendroochreatene ( 1 ), a new phenanthrene derivative with a spirolactone ring, was isolated from the whole Dendrobium ochreatum plant together with 11 known compounds ( 2 – 12 ). The structure of the new compound was elucidated spectroscopically and phenolic compounds were firstly reported from D. ochreatum. Moscatilin ( 4 ), major compound isolated from D. ochreatum, was found to be cytotoxic toward H460 lung-cancer cells, with an IC 50 value of 147.3 ± 0.9 μM, while loddigesiinol C ( 7 ), C- α -methoxy derivative was inactive.

Abstract:Five new dibenzocyclooctadiene lignans, schisphenlignans A-E (1-5), together with eight known ones, were isolated from the stems of Schisandra sphenanthera. The structures of 1-5 were elucidated based on the analysis of their NMR, MS and circular dichroism (CD) spectra. Some isolates were tested for their acute activities on insulin sensitivity in 3T3-L1 differentiated adipocytes, but none of them showed significant bioactivity with 10 mu M administration of the tested compounds.

Abstract:Six new stilbenoids, a (bibenzyldihydrophenanthrene) ether designated phoyunnanin D (1), a bis(dihydrophenanthrene) ether designated phoyunnanin E (2), and four stilbenes designated phoyunbene A-D (3-6), were isolated from the air-dried whole plant of Pholidota yunnanensis ROLFE. The new compounds were identified as 7-[2-(3-hydroxyphenethyl)-4-hydroxy-6-methoxyphenoxy]-4-hydroxy-2-methoxy-9,10-dihydrophenanthrene (1), 1-[(9,10-dihydro-4-hydroxy-2-methoxy-7-phenanthrenyl)oxy]-4,7-dihydroxy-2-methoxy-9,10-dihydrophenanthrene (2), trans-3,3'-dihydroxy-2',4',5-trimethoxystilbene (3), trans-3,4'-dihydroxy-2',3',5-trimethoxystilbene (4), trans-3,3'-dihydroxy-2',5-dimethoxystilbene (5), and trans-3-hydroxy-2',3',5-trimethoxystilbene (6) based on spectroscopic evidence. Furthermore, the inhibitory effects of compounds 1-6 on nitric oxide production in a murine macrophage-like cell line (RAW 264.7) activated by lipopolysaccharide and interferon-gamma were examined.

All structures were elucidated by spectroscopic methods (NMR, MS, UV and IR). Anti-platelet aggregation activities of compounds 1 - 5 were also tested.

Abstract:A new enantiomeric pair of spirodiketones, (+)- and (-)-denobilone A (1 and 2), three new phenanthrene derivatives (3-5), and three new biphenanthrenes (22-24), along with 11 known phenanthrene derivatives (6-16), five known bibenzyl derivatives (17-21), and four known biphenanthrenes (25-28), were isolated from Dendrobium nobile. The structures of 1-5 and 22-24 were elucidated using comprehensive spectroscopic methods. (+)-Denobilone and (-)-denobilone A (1 and 2) were isolated as a pair of enantiomers by chiral HPLC. The absolute configurations of (+)- and (-)-denobilone A (1 and 2) were determined by comparing their experimental and calculated electronic circular dichroism spectra. The absolute configuration of denobilone B (3) was determined by X-ray crystallographic analysis. The inhibitory activities of all compounds against nine phytopathogenic fungi and three cancer cell lines were evaluated.

Abstract:Two novel compounds, (9R)-4-metboxy-9H-fluorene-2,5,9-triol (1a) and 2,7-dihydroxy-8-methoxyphenanthro[4,5-bcd]pyran-5(5H)-one (2) were isolated from the stems of the orchid Dendrobium chrysotoxum Lindl. Abstract:Bioassay-guided fractionation of the 60% ethanol extract of the stems of Dendrobium nobile using the DPPH assay led to the isolation of two new bibenzyl derivatives, nobilin D (1) and nobilin E (2), and a new fluorenone, nobilone (3), together with seven known compounds (4-10). Their structures were determined on the basis of spectroscopic analyses. Compounds 1, 2, 4, 7, 8, and 10 exhibited significant antioxidant activity higher than or equivalent to vitamin C in the DPPH assay, and compounds 1, 3, 4, and 7-10 displayed higher antioxidant activity than vitamin C in the ORAC assay. Compounds 1, 2, and 10 also exhibited stronger inhibitory effects on NO production than resveratrol.

Abstract:Two new phenylpropanoid derivatives (1-2), together with eight known compounds (3-10) were isolated from the stems of Dendrobium sonia. The structures of these compounds were elucidated on the basis of spectroscopic analyses, including HRESIMS, 1 D and 2 D NMR experiments. All of the isolated compounds were tested for their Nitric Oxide (NO) Inhibitory Activities. The results of bioactive screening showed that compounds 2, 8, 9 and 10 exerted inhibitory effects on NO production with IC50 values in the range of 26.3 to 31.6 μM. Compound 8 and 9 exhibited stronger anti-inflammatory activities with IC50 values 26.3 and 27.7 μM, comparable to that of the positive control.

Abstract:A new phenanthraquinone, 7,9-dihydroxy-2-methoxy-1,4-phenanthraquinone ( 1 ), and nine known compounds were isolated and identified from the whole plants of Dendrobium lindleyi . The structure of the new compound was determined on the basis of NMR (1D and 2D) and mass spectroscopic analysis. The new compound exhibited potent antimicrobial activity against Bacillus subtilis and Escherichia coli , and its activity was comparable with that of the well-known antibacterial drug berberine.
