Abstract:Quinoproteins are an important type of redox enzymes for biological oxidation processes. Inspired by the quinone cofactors, particularly from copper amine oxidases, a number of small molecular quinone catalysts have been developed for C-H functionalizations of amines. Bio-inspired quinone catalysts have significantly expanded the substrate scope to include branched primary amines, secondary amines and tertiary amines, far beyond the scope of quinoproteins. This review summarizes the evolution of quinone catalysts, their mechanism and catalytic applications.

Abstract:Catalytic asymmetric electrochemical C-H functionalization of simple ketones has been developed. The transformation is realized by the combination of electrochemical oxidation and chiral primary amine catalysis. This metal- and oxidant-free method furnishes diverse C1-alkylated tetrahydroisoquinolines in high yields and with excellent enantioselectivities under very mild conditions.

Abstract:The direct oxidative N-acylation reaction of primary amides with aryl/alpha,beta-unsaturated aldehydes was achieved in the presence of azolium salt C3 and an inorganic base using 3,3',5,5'-tetra-tert-butyldipherioquinone as the oxidant, thus providing an efficient approach for the synthesis of three types of imide compounds including N-sulfonylcarboxamides, N-sulfinylcarboxamides, and dicarboxyimides in good yield.

Abstract:An unprecedented chiral secondary amine-catalyzed formal [4 + 2] annulation of aldehydes and oxidation-generated β-unsubstituted o-QMs is reported. This asymmetric protocol allows direct functionalization of the benzylic C–H bonds and furnishes [4 + 2] cycloadducts, chromanols, with excellent enantioselectivity and in up to 92% yield. The usability of this approach was further demonstrated by the enantioselective synthesis of anticancer Rhinacanthins derivative NKPLS8.

Abstract:Metal-free carbocatalysis has been widely utilized for aerobic oxidative reactions. Here, we report that oxygen-rich carbon quantum dots (O-CQDs) demonstrate a catalytic performance superior to graphene oxide, if used as a metal-free nanocatalyst for the direct transformation of amines and alcohols, under mild and solvent-free conditions. O-CQDs show a yield of 75% for the oxidative coupling of amine to imine (with 5wt% catalyst loading) and a conversion of 3.8% for benzyl alcohol (with 2wt% catalyst loading). The catalytic activities of thermally treated O-CQDs are further improved for benzylamine, for example, indicated by a yield of up to 98% with 4wt% catalyst loading. In addition, O-CQDs show a photoenhanced catalytic ability of amine (98% yield with 5wt% catalyst loading for 6h reaction). Characterizations and simulations show that numerous carboxyl oxygen functional groups and unpaired electrons at the edge sites of O-CQDs are likely involved in the aerobic oxidation of amines.

Abstract:Biocatalytic preparation of chiral amines is a large and burgeoning field in organic chemistry. Many enzymes and routes have been published, including transaminases, imine reductases, reductive aminases, amine dehydrogenases and others. However, all these routes rely on some sacrificial substrate, in the form of either amine donor or cofactor regeneration substrate. Herein, we report the direct oxidative amination of p‐substituted phenols catalyzed by an evolved flavoprotein oxidase, with the consumption of only substrate and O2, and release of H2O2. The substrate scope of the reaction is studied, and is tolerant of a diverse panel including ammonia, primary and secondary amines, and amino acids. The reaction is later employed at preparative scale to generate aminated products in 50‐80% yield. This report establishes flavoprotein oxidase as a new and economical member of the chemist's toolkit for biocatalytic generation of chiral amines, acting as oxidative aminase.

Abstract:The development of heterogeneous catalysts for the synthesis of pharmaceutically relevant compounds is always important for chemistry research. Here, we report a selective aerobic oxidation of aromatic and aliphatic amines to corresponding amides over a nanorod manganese oxide (NR-MnOx) catalyst. The kinetic studies reveal that the NR-MnOx catalyzed amine-to-amide reaction proceeds the oxidative dehydrogenation of the amines into nitriles, followed by hydrolysis of nitrile into amides. The NR-MnOx exhibits fast kinetics and high selectivities in these steps, as well as hinders the by-product formation. More importantly, the NR-MnOx catalyst is stable and reusable in the continuous recycle tests with water as a sole by-product, exhibiting superior sustainability and significant advancement to outperform the traditional amide production route in acidic or basic media with toxic by-products.

Abstract:A convenient and efficient copper-catalyzed aerobic oxidation of primary amines to aryl nitriles was described. Various benzylic and allylic amines were selectively oxidized to the corresponding nitriles in high yields using CuBr2/NMI as the catalyst and O2 as the oxidant. The oxidation reaction profiles monitored by 1H NMR disclosed the scenario of the reaction path as well as the role of the additives. The addition of NMI increased the rate of reaction and suppressed the hydrolysis and the deamination.
