Abstract:A novel synthesis process of 5H-dibenz[b,f]azepine was studied.It is an important intermediate of 5H-dibenz[b,f]azepine-5-carboxamide,used in genetic engineering,materials science and so on.With 10,11-dihydro-5H-dibenz[b,f]azepine as raw material,5-chloroformyl-5H-dibenz[b,f]azepine was prepared by reflux with triphosgene in toluene,brominated with bromine in chlorobenzene,and dehydrobrominated at 130℃,finally,the title compound was obtained by removing chloroformyl group with NaOH in isopropanol with 80.4% overall yield.The optimal reaction conditions and yields (temperature,time,molar yield) were as follows: acyl chlorination,110℃,10 h,93.0%; bromination,dehydrobromination,100℃,2 h,130℃,5 h,88.0%; removal of chloroformyl group,40℃,3 h,98.2%. The melting point of the title compound was the same as literature reported,and its structure was confirmed by 1H NMR.The optimal technology condition to remove chloroformyl group as protection group and its application of protection for some amine compounds were also studied.

Abstract:A simple, rapid, and highly efficient method has been developed for the aza-Michael addition of acrylonitrile to 2-aryl-oxymethylbenzimidazole derivatives in the presence of anhydrous potassium carbonate under microwave irradiation. A series novel of 1-cyanoethyl-2-aryloxymethylbenzimidazole derivatives have been prepared and characterised by 1H NMR, 13C NMR, IR spectra and elemental analysis.

Abstract:The title compound, (1R,2R,3R,5S)-3-(3-chlorophenyl)-1-hydroxy-2-methyl-6-phenyl-8-oxa-6-azabicyclo[3.2.1]octan-7-one, was synthesized and characterized by H-1 and C-13 NMR and HRMS spectroscopy. Its absolute molecular configuration was investigated by X-ray crystallography. The crystal structure analysis revealed that the structure exhibits intermolecular hydrogen bonds of the type O-H horizontal ellipsis O. The methyl group and chlorophenyl group are located in the cis configuration. The conformation is stabilized by weak intermolecular C-H... O interactions.

In this route, levulinic acid was used as a starting material, and bromination followed by hydrolysis and BF3 · OEt2‐catalyzed cyclization were carried out. Abstract:In the present study, we have synthesized a series of novel glucosyl triazoles for the first time. The glucosyl triazoles 4a-e were synthesized by reaction of some azidoglycosides with various terminal alkynes via a copper-catalyzed [3+2] cycloaddition (oclick chemistryo) and were deprotected to afford the corresponding glucosyl triazoles 5a-e in good yields. The structures of the new compounds were determined by IR, NMR spectroscopy, and mass spectrometry. The antitumor (human cervical cancer cell) activity was evaluated for the target compounds.

Abstract:A new kind of amorphous molecular materials containing azobenzene chromophors has been synthesized. The precursor molecule was prepared by esterification between 1, 3, 5-benzenetricarbonyl chloride and N-ethyl- N-hydroxyethyl-aniline. The precursor molecule was then reacted with the diazonium salts of 5-aminoisophthalic acid to introduce donor-acceptor azobenzene chromophores at the peripheral groups. The azobenzene amorphous molecular material was characterized by the spectroscopic methods and thermal analysis. The azo compound synthesized can form stable amorphous glass. The amorphous molecular material was used to fabricate surface relief gratings (SRG) by exposing spin-coated thin films of the azo compound to an interference pattern of Ar+ laser beams at modest intensity ( 120 mW/cm(2)). The surface modulation was about 390 nm and the first order of diffraction efficiency was up to 23%.

Abstract:Enaminones are important synthetic intermediates for the construction of diverse heterocyclic scaffolds owing to their push-pull electronic structures. A series of novel (Z)-1-(3,5-bis(trifluoromethyl)phenyl)-3-(aryl/alkyl)prop-2-en-1-ones ( 5, 6 ) and (Z)-3-(aryl/alkylamino)-1-(4-(trifluoromethyl)phenyl)prop-2-en-1-ones ( 9 , 10 ) were synthesized with (E)-1-(3,5-bis(trifluoromethyl)phenyl)-3-(dimethylamino)prop-2-en-1-one ( 2 ) & (E)-3-(dimethylamino)-1-(4-(trifluoromethyl)phenyl) prop-2-en-1-one ( 8 ) as starting materials. The structures of the fluorinated enaminones were established with the help of spectral and analytical data. X-ray crystallographic study for a model compound ( 6a ) gave a vivid image showing the existence of the product in Z-form unlike the corresponding intermediate that exists in E-form.

Abstract:A series of new substituted derivatives of 2-cyano-3-methvlthio-3'-aminoacrylates(acrylamides) have been designed and synthesized. Abstract:(1-Chloro-2-trifluoromethylcyclohexyl)azo compounds 3a-d, prepared by oxidation of the hydrazones 2a-d of 2-trifluoromethylcyclohexanone (1) with a slightly excess amount of tertbutyl hypochlorite, react with antimony pentachloride and a nitrile to afford the respective 2,3-disubstituted-9-trifluoromethyl-6,7,8,9-tetrahydro-5H-[1,2,4]triazolo[1,5-a]azepinium hexachloroantimonates 6a-j. The reaction mechanism comprises of the initial cycloaddition of the in situ generated 1-aza-2-azoniaallene salts 4 to the triple bond of nitrile, giving the spiro-substituted 5a-j, followed by rearrangement and concomitant ring enlargement. From 3a-c where R-1 = aryl, the salts 6a-h are isolated in high yields. However, if ethyl (1-chloro-2-trifluoromethyl) azocarboxy late (3d) is employed, the neutral N(3)-unsubstituted analogues 7a and 7b are obtained and after usual basic work-up, are converted to picrates 8a and 8b respectively. For one compound 6f, the X-ray diffraction analysis has been carried out in order to confirm the structural assignment. A practical modified procedure for preparation of the crucial starting ketone 1 is also described.

Abstract:We have developed a novel palladium-catalyzed arylative dearomatization and subsequent aromatization/dearomatization/aza-Michael addition process of Ugi adducts, enabling the rapid construction of diverse zephycarinatine and zephygranditine scaffolds containing two adjacent quaternary carbon stereocenters with excellent chemoselectivity and stereoselectivity in a rapid, step-economical, and highly efficient manner. This approach shows broad substrate scope and excellent functional-group tolerance with diverse electron-rich and electron-deficient aromatic substrates. Abstract:A new fluorescent functional monomer DMAAN has been designed and synthesized for the first time. The monomer was expected to bind with cytosine via complementary hydrogen bonding under certain experimental conditions. The synthetic procedure of DMAAN is as follows. A suspension of 2,6-diaminopyridine in phosphoric acid was added to acetylacetone. The mixture was warmed for thirty minutes on a steam-bath. Then 2,4-dimethyl-7-amino-1,8-naphthyridine(DMAN, mp 216-218 degrees C) was obtained. DMAAN was prepared by the reaction of DMAN with acryloyl chloride in chloroform and triethylether mixed solvent. Acryloyl chloride in anhydrous chloroform was added dropwise over 30 minutes with constant stirring at 4 degrees C. The melting point of the final product was 239-241 degrees C. Mass spectrometry and proton NMR were used to confirm the chemical structure of DMAN and DMAAN. The products were also characterized by UV and fluorescence spectrometry.

Abstract:The N-(3-((1-(5-bromo-2-(pyrrolidin-1-yl)phenyl)-5-oxo-[1,2,4]triazolo[4,3-a]quinazolin- 4(5H)-yl)methyl)phenyl)-2-thiomorpholinoacetamide is a novel compound having the [1,2,4]triazolo[4,3-a]quinazolinone. It was synthesized by nine steps and confirmed by 13C NMR, 1H NMR, FT-IR and MS spectroscopies. Meanwhile, the single crystal of the title compound was subjected to crystallographic analysis and conformation determination. Moreover, density functional theory (DFT) was used to calculate the optimized structures of the molecule, which was compared with the X-ray measurement. The result of the molecular structure optimized by DFT is similar to the crystal structure determined by X-ray single crystal diffraction. In order to further investigate some physical properties of the title compound, by using the B3LYP/6-311+G(2d, p) method, molecular electrostatic potential and frontier molecular orbitals was calculated. Furthermore, the binding mode of the title compound and SHP2 protein (PDB: 6BMR) was analyzed by molecular docking. Finally, hirshfeld surfaces analyses can explain the atom pair contacts of the crystal and the quantitative analysis of intermolecular interactions.

This protocol is characterized by its simplicity, practicality, and tolerance toward various functional groups. Given the straightforward nature of the procedure, the ready availability of both starting materials, and the significance of drugs containing trifluoromethyl, it is anticipated that this reaction will have wide-ranging applications.
