Abstract:DMSO‐TsCl has been developed for direct methylthiolation of various electron‐rich heteroarenes (more than 40 examples) with high regioselectivity in moderate to excellent yields (up to 96%). Especially, pyrroles, furans and thiophenes can be efficiently mono‐methylthiolated. This practical method features scalable, metal‐free, mild conditions which are compatible with air and moisture. Several applications of methylthiolated product were demonstrated for the first time. Based on controlled experimental results, a plausible mechanism was proposed with two key intermediates involved in mechanism being detected by HRMS.

Abstract:An efficient copper-catalyzed tandem oxidation/alpha-formylation reaction of 2,3-dihydro-1H-pyrroles in an oxygen atmosphere has been developed. The new reaction is conducted in dimethyl sulfoxide (DMSO), which serves as both the carbonyl carbon source and solvent, in the presence of trifluoroacetic acid to directly generate structurally diverse alpha-formylpyrrole derivatives in one pot in moderate to good yields. Sensitive aldehyde groups remain intact in the presence of an oxidant, trifluoroacetic acid and a transition metal salt. A possible mechanism is presented.

Abstract:A copper-catalyzed O-methylation of carboxylic acids using dimethyl sulfoxide (DMSO) as the methyl source is disclosed. This transformation exhibits a broad substrate scope and excellent functional group tolerance. Mechanistic studies indicate that a methyl radical is generated from dimethyl sulfoxide in the reaction process.

Abstract:The expansion of cross-coupling components in Cu-catalyzed C-X bond forming reactions have received much attention recently. A novel Cu-catalyzed trifluoromethylthiolation of aryl bromides and iodides with the assistance of versatile directing groups such as pyridyl, methyl ester, amide, imine and oxime was reported. CuBr was used as the catalyst, and 1,10-phenanthroline as the ligand. By changing the solvent from acetonitrile to DMF, the coupling process could even take place at room temperature.

Abstract:The methanesulfenylation of various mercaptans and β-dicarbonyls has been investigated using in-situ generated methyl methanethiosulfonate (MMTS) from DMSO, facilitated by a catalytic amount of (COCl)2. Employing the MMTS solution alongside Et3N led to the synthesis of a range of unsymmetrical disulfides with good yields. Furthermore, the introduction of [2H6]-DMSO enabled the successful preparation of various [2H3]-disulfides. In the case of most β-dicarbonyls, successful methanesulfenylation was accomplished using Et3N and DBU, resulting in favorable yields.

Abstract:Organic thioethers play an important role in the discovery of drugs and natural products. However, the green synthesis of organic sulfide compounds remains a challenging task. The convenient and efficient synthesis of 5-alkoxy-3-halo-4-methylthio-2(5H)-furanones from DMSO is performed via the mediation of 1,3-dibromo-5,5-dimethylhydantoin (DBDMH), affording a facile route for the sulfur-functionalization of 3,4-dihalo-2(5H)-furanones under transition metal-free conditions. This new approach has demonstrated the functionalization of non-aromatic Csp2-X-type halides with unique structures containing C-X, C-O, C=O and C=C bonds. Compared with traditional synthesis methods using transition metal catalysts with ligands, this reaction has many advantages, such as the lower temperature, the shorter reaction time, the wide substrate range and good functional group tolerance. Notably, DMSO plays multiple roles, and is simultaneously used as an odorless methylthiolating reagent and safe solvent.

Abstract:An efficient copper-mediated ortho-C(sp2)-H thiolation of aromatic amides directed by a novel directing group [4-chloro-2-(1H-pyrazol-1-yl)phenyl]amine has been developed without the need of other additives or oxidants, allowing for an increased usefulness. With the high compatibility of sterically demanding substrates, this reaction is scalable and can tolerate a wide scope of functional groups to provide alkyl and aryl thioethers in good to excellent yields (up to 93%). Furthermore, the protocol has been successfully implemented for the selenylation as well.

Abstract:A family of electrophilic deuterated methylthiolating reagents, S-(methyl-d3) arylsulfonothioates, was developed in two or three steps from cheap d4-MeOH in high yields. S-(Methyl-d3) arylsulfonothioates represent a kind of powerful deuterated methylthiolating reagent and allow modular trideuteromethylthiolation with a variety of nucleophiles or electrophiles including aryl(hetero) iodides, boronic acids esters, terminal alkynes, diazonium salts, β-ketoester, and oxindole under mild reaction conditions. A structure-reactivity research (SAR) study was conducted and provided a new avenue for the development of deuterated methylthiolating reagents and efficient methodology for trideuteromethylthiolation.

Abstract:A new method for the formation of 1,1-difluoroethyl copper species ("CuCF2 CH3 ") with 1,1-difluoroethylsilane (TMSCF2 CH3 ) has been developed. The "CuCF2 CH3 " species can be applied to the efficient 1,1-difluoroethylation of diaryliodonium salts under mild conditions, affording (1,1-difluoroethyl)arenes in good to excellent yields. This convenient procedure tolerates a wide range of functional groups and thus serves as a practical synthetic tool for the introduction of CF2 CH3 group(s) into complex molecules.
