Abstract:Three hiherto unknown phenylpropanoid compounds, namely (7S,8R)‐1‐(1‐ethoxy‐2‐hydroxypropyl)‐2‐methoxy‐3,4‐(methylenedioxy)benzene (1), (7S,8S)‐1‐(1‐ethoxy‐2‐hydroxypropyl)‐2‐methoxy‐3,4‐(methylenedioxy)benzene (2), and (7S,8R)‐1‐(1‐methoxy‐2‐hydroxypropyl)‐2‐methoxy‐3,4‐(methylenedioxy)benzene (3), along with 12 known compounds (4 – 15) were obtained from the extract of whole plant of Chloranthus anhuiensis. Among them, 7 and 13 were obtained from nature for the first time. The structures of these natural compounds were characterized by extensive spectroscopic analysis and calculated electronic circular dichroism (ECD) data. Furthermore, their cytotoxic and neuroprotective activities were evaluated using MDA‐MB‐231, 4T1, HepG2, and PC12 cell lines. Compounds 8 and 13 exhibited moderate cytotoxic activities against MDA‐MB‐231 cell line with the IC50 values of 39.7 and 25.8 μm, respectively. And all the isolated compounds have no neuroprotective activities.

Abstract:Two new dimeric lindenane-type sesquiterpenoids (1 and 2, named chlorasessilifols A and B, resp.), one new ent-podocarpane-type C17 norditerpenoid (3), and one new ent-torarane-type diterpenoid (4), along with seven known terpenoids, were isolated from the whole plant of Chloranthus sessilifolius. The new structures were established by means of spectroscopic methods and/or observed cotton effects in the circular dichroism spectra. Among the isolates, 3α,7β-dihydroxy-ent-abieta-8,11,13-triene (11) exhibited significant anti-neuroinflammatory activity by inhibiting the nitric oxide production in lipopolysaccharide-stimulated murine BV-2 microglial cells, with an IC50 value of 4.3 μM.

Abstract:Two new eudesmane-type sesquiterpenoids, chlojaponols A (1) and B (2), and four new lindenane-type sesquiterpenoid lactones, chlojaponilactones F-I (3⬜6), along with six known analogues were isolated from whole plants of Chloranthus japonicus. The structures of new compounds were established based on analyses of spectroscopic data including HR-MS, 1D and 2D NMR. The antifungal activities of some of the compounds were tested and compound 5 showed 82.61% inhibition to Sclerotinia sclerotiorumat the concentration of 50 μg/mL.

Abstract:We examined the composition and antimicrobial activity of two essential oils from Chloranthus japonicus Sieb. and Chloranthus multistachys Pei. GC-FID and GC-MS analyses identified 48 and 39 compounds, which represented 95.56% and 94.58%, respectively, of all components in these oils. Of these, 28 compounds were common to both, with a relatively high amount of oxygenated monoterpenes (50.95% and 39.97%). Antimicrobial properties were evaluated in vitro via disc diffusion and microbroth dilution assays. Activities were strong against most tested microorganisms, with inhibition zones ranging from 8.1 to 22.2 mm. For both species, minimum values for inhibitory and bactericidal concentrations were 0.39 to 12.50 mg/mL and 0.78 to 50.00 mg/mL, respectively. These results suggest that these essential oils are potent natural sources of antimicrobial agents for the medicinal and pharmaceutical industries.

Abstract:Mesona chinensis is a common medicinal and edible plant in the Lingnan region of China, which has extensive pharmacological activity. However, the study of its chemical constituents is not sufficient. In this study, a variety of modern chromatographic separation techniques were used to isolate two compounds from 95% ethanol extract of the grass parts of M. chinensis. Their absolute configurations were determined by ultraviolet spectroscopy(UV), infrared spectroscopy(IR), high resolution mass spectrometry(HR-ESI-MS), 1D and 2D nuclear magnetic resonance(1D NMR and 2D NMR), and single-crystal X-ray diffraction(SC-XRD). Specifically, they were two new benzoyl-sesquiterpenes and named mesonanol A and mesonanol B, respectively. The results of the pharmacological activity evaluation showed that neither of the two new compounds showed obvious antiviral and anti-inflammatory activities.

Abstract:In this study, 14 previously undescribed terpenoids were isolated from the Chinese liverwort Heteroscyphus coalitus (Hook.) Schiffner, including a rare harziane type diterpenoid, heteroscyphsic acid A; eight ent-clerodane diterpenoids, heteroscyphsic acids B-I; four labdane diterpenoids, heteroscyphins A-D; and one guaiane sesquiterpene, heteroscyphin E; as well as a known ent-junceic acid. Their structures were determined by a combination of MS, NMR spectroscopy, electronic circular dichroism (ECD) and single crystal X-ray diffraction analyses. The anti-virulence activity of the isolated compounds against Candida albicans DSY654 demonstrated that most of them could block hyphal growth at concentrations ranging from 4-32 itg/ml. Further investigation of the most active compound, heteroscyphin D, revealed that it could suppress the ability of C. albicans DSY654 to adhere to A549 cells and form biofilms, and modulate the transcription of related genes in this fungus.

Abstract:Four new sesquiterpenoids, 4,8-dioxo-6 beta-hydroxyl-7 beta,11-epoxycarabrane (1), 4,8-dioxo-6 beta-hydroxyl-7 alpha,11-epoxycarabrane (2), wenyujinins Q and R (3-4), and nine known sesquiterpenoids (5-13) were isolated from the Curcuma wenyujin (C wenyujin) dreg. Their structures and relative configurations were elucidated using 1D, 2D NMR, and HR-ESI-MS data. All the compounds were isolated for the first time from the C wenyujin dreg and evaluated for their antibacterial and antifungal activities. Compounds 3, 58 exhibited strong broad-spectrum antifungal activities against tested nine pathogenic fungi. Abstract:The phytoconstituents of the whole plants of Chloranthus holostegius were investigated. As a result, thirteen undescribed sesquiterpenes (chloranholosins A–M, 1 – 13 ), including ten acorane-type sesquiterpenes ( 1 − 10 ), one germacrene-type sesquiterpene ( 11 ), and two lindenane-type sesquiterpenes ( 12 − 13 ), together with fifteen known sesquiterpenes were isolated. Their structures and absolute configurations were elucidated by a comprehensive method including the spectroscopic data, electronic circular dichroism (ECD) calculations, and single-crystal X-ray diffraction. Chloranholosin L ( 12 ) was elucidated as a rare lindenane-type sesquiterpene featuring 14 α -Me and 5-OH moieties. And chloranholosin M ( 13 ) was the first lindenane-type sesquiterpene possessing β -cyclopropane, 14 α -Me, and 5 β -H configuration from the family Chloranthaceae. Furthermore, twelve new isolates and some known sesquiterpenes were evaluated for their inhibitory activity against LPS-induced nitric oxide (NO) production in RAW 264.7 macrophage cells. Among them, compounds 12 , 16 , and 23 showed comparable inhibitory activity to that of the positive control, with IC 50 values of 47.9, 41.5, and 48.3 μM, respectively.

Abstract:Investigation of the leaves of Chloranthus tianmushanensis resulted in the isolation and characterization of two new sesquiterpene dimers with a rare 18-membered triester ring, tianmushanol (1) and 8-O-methyltianmushanol (2), and four known sesquiterpenes. Abstract:Chemical investigation of the culture extract of an endophyte Xylaria curta YSJ-5 from Alpinia zerumbet (Pers.) Burtt. et Smith resulted in the isolation of eight previously undescribed compounds including five eremophilane sesquiterpenes xylarcurenes A-E, one norsesquiterpene xylarcurene F, and two α-pyrone derivatives xylarpyrones A-B together with eight known related derivatives. Their chemical structures were extensively established based on the 1D- and 2D-NMR spectroscopic analysis, modified Mosher's method, electronic circular dichroism calculations, single-crystal X-ray diffraction experiments, and the comparison with previous literature data. All these compounds were tested for in vitro cytotoxic, anti-inflammatory, α-glucosidase inhibitory, and antibacterial activities. As a result, 6-pentyl-4-methoxy-pyran-2-one was disclosed to display significant antibacterial activity against Staphylococcus aureus and methicillin-resistant S. aureus with minimal inhibitory concentration value of 6.3 μg/mL.
