Abstract:Twelve new ent-abietane diterpenoids, chlorabietins A-L (), were isolated from the roots of Chloranthus oldhamii. Their structures and absolute configurations were determined by extensive spectroscopic analyses, X-ray diffraction, and experimental/calculated electronic circular dichroism (ECD) spectroscopy. Among the new isolates, chlorabietins D () and E () are the first two naturally occurring 8-spiro-fused 9,10-seco-ent-abietanes containing an unexpected cis-fused A/B ring system. Chlorabietin F () is a rare chinane-type diterpenoid featuring a hitherto unknown C-ring cleavage between C-13 and C-14, which might be derived from a common precursor of the above spiro-diterpenoid epimers and , and their biosynthetic relationships are briefly discussed. Meanwhile, chlorabietin I () is the first representative of the abietane-type diterpenoids possessing a tetrahydrofurano function bridging C-6 and C-19. Chlorabietins B (), C (), F (), and G () showed anti-neuroinflammatory effects by inhibiting the nitric oxide (NO) production in lipopolysaccharide (LPS)-activated murine BV-2 microglial cells, with IC50 values ranging from 16.4 to 33.8 μM.

Abstract:Fourteen new ent-abietane-type diterpenoids, sessilifols A-N (1-14), and three related new norditerpenoids (15-17) were isolated from Chloranthus sessilifolius. The absolute configurations were determined by single-crystal X-ray diffraction analysis, the modified Mosher's method, and/or the observed Cotton effects in their electronic circular dichroism spectra. Sessilifols A (1) and B (2) possess an uncommon five-membered C-ring rearranged by oxidative cleavage of the C-13/C-14 bond in abieta-7,13-diene followed by the formation of a new C-C bond between C-12 and C-14. Sessilifol C (3) is a rare 7,8-seco-9-spiro-fused ent-abietane, whereas sessilifol O (15) represents the first example of a naturally occurring 14-norabietane-type diterpenoid. Compounds 6 and 9 were found to have moderate antineuroinflammatory activities by inhibiting the nitric oxide production in lipopolysaccharide-stimulated murine BV-2 microglial cells, with IC50 values of 8.3 and 7.4 mu M, respectively.

Abstract:Two new ent-pimarane (1 and 2), eight new ent-abietane (3-10) diterpenoids, and eight known analogues (11-18) were isolated from the whole plants of Chloranthus henryi. The absolute configuration of 1 was determined on the basis of single-crystal X-ray diffraction data. Compound 8 represents a class of rare naturally occurring C-14 norabietanes, and compounds 9 and 10 feature rare 13,14-seco-abietane skeletons. Compounds 5, 12, 13, and 15 inhibited the yeast-to-hyphae transition of Candida albicans with IC50 values between 97.3 and 738.7 μM.

Abstract:Six new ent -abietane diterpenoids, abientaphlogatones A–F ( 1 – 6 ), along with two undescribed ent -abietane diterpenoid glucosides, abientaphlogasides A–B ( 7 – 8 ) and four known analogs were isolated from the aerial parts of Phlogacanthus curviflorus ( P. curviflorus ). The structures of these compounds were determined using high-resolution electrospray ionization mass spectrometry (HR-ESI-MS), one-dimensional and two-dimensional nuclear magnetic resonance (NMR) spectroscopy, electronic circular dichroism (ECD) spectra, and quantum chemical calculations. Notably, compounds 5 and 6 represented the first reported instances of ent -norabietane diterpenoids from the genus Phlogacanthus . In the β -hematin formation inhibition assay, compounds 2 , 4 , 7 – 10 , and 12 displayed antimalarial activity, with IC 50 values of 12.97–65.01 μmol·L –1 . Furthermore, compounds 4 , 5 , 8 , and 10 demonstrated neuroprotective activity in PC12 cell injury models induced by H 2 O 2 and MPP + .

Abstract:Two new dimeric lindenane-type sesquiterpenoids (1 and 2, named chlorasessilifols A and B, resp.), one new ent-podocarpane-type C17 norditerpenoid (3), and one new ent-torarane-type diterpenoid (4), along with seven known terpenoids, were isolated from the whole plant of Chloranthus sessilifolius. The new structures were established by means of spectroscopic methods and/or observed cotton effects in the circular dichroism spectra. Among the isolates, 3α,7β-dihydroxy-ent-abieta-8,11,13-triene (11) exhibited significant anti-neuroinflammatory activity by inhibiting the nitric oxide production in lipopolysaccharide-stimulated murine BV-2 microglial cells, with an IC50 value of 4.3 μM.

Abstract:Chemical investigation on the aerial parts of Euphorbia fischeriana resulted in the identification of eight abietane-type diterpenoids including three undescribed ones, named 3α-acetoxy-14-hydroxy-ent-abieta-8(9),13(15)-dien-16,12-olide (1), 3α,7β-dihydroxy-ent-abieta-11(12),13(15)-dien-16,12-olide (2), and 2β-hydroxy helioscopinolide B (3). Compound 1 exhibited moderate cytotoxic activities against the HL-60, SMMC-7721, and SGC-7901 cancer cells.

Abstract:Two diterpenoids and a few miscellaneous components (one phenethylpropanoid, one flavonoid, and one lignan) were isolated from the MeOH extract of the whole plant of the rare Chloranthaceae plant Chloranthus sessilifolius during a further phytochemical reinvestigation. Based on their spectroscopic data (i.e., IR, UV, 1D/2D NMR, ECD, and HR-ESIMS) and/or comparing with those reported physicochemical data in related literature, the structures were determined to be (3R,5S,6S,95,10S,135)-3,6,13-trihydroxy-ent-abieta-8(14)-en-7-one (1), 3 alpha,15-dihydroxy-13 alpha-methoxy-ent-abieta-8(14)-en-7-one (2), p-coumaroyl-beta-phenethyl amine (3), narigenin trimethyl ether (4), and (3R,4-R,3'R,4'R)-6,6'-dimethoxy-3,4,3',4'-tetrahydro-2H,2H'[3,3]-bichromenyl-4,4'-diol (5). Among them, compound 1 is new, while compounds 3 similar to 5 were isolated from the Chloranthus genus for the first time.

Abstract:Three labdane-type [multisins A-C (1-3)], two guaiane-type [multisins D (4) and E (5)], and one eudesmane-type [multisin F (6)] previously undescribed terpenoids, together with 14 mono- (7-20) and seven dimeric- (21-27) known terpenoids, were isolated from the 90% MeOH extract of the whole plant of Chloranthus multistachys. Their structures and absolute configurations were determined by extensive spectroscopic methods and electronic circular dichroism (ECD) calculations. Compounds 4 and 5 are rare trinor-sesquiterpenes with a de-isopropyl guaiane skeleton, whereas compound 6 is a rearranged dinor-eudesmene featuring an uncommon octahydro-1H-indene ring system. Among the isolates, the dimeric lindenane sesquiterpenoid shizukaol C (25) exhibited the most potent (IC50 = 8.04 μM) anti-neuroinflammatory activity by inhibiting the nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated murine BV-2 microglial cells.

Abstract:Three unprecedented phloroglucinol-diterpene adducts, chlorabietols A-C (1-3), were isolated from the roots of the rare Chloranthaceae plant Chloranthus oldhamii. They represent a new class of compounds, featuring an abietane-type diterpenoid coupled with different alkenyl phloroglucinol units by forming a 2,3-dihydrofuran ring. Their structures were elucidated by detailed spectroscopic analysis, molecular modeling studies, and electronic circular dichroism calculations. Compounds 1-3 showed inhibitory activity against protein tyrosine phosphatase 1B (PTP1B) with IC50 values of 12.6, 5.3, and 4.9 μM, respectively.

Abstract:Euphoroids A-C (1-3), three new ent-abietane-type diterpenoids, together with ten known analogues (4-13) were obtained from the roots of Euphorbia ebracteolata. The structures of these compounds were determined by extensive spectroscopic data analysis, including UV, HRESIMS, 1D-, and 2D-NMR data. The inhibitory effects of compounds 1-13 on human cancer cells were determined using the MTT assay. The results revealed that new compounds 2 and 3 showed moderate cytotoxic activities against human cancer cell lines. Especially, compound 3 displayed selective cytotoxic effect agains cancer cell lines.

Abstract:Two novel ent-abietane diterpenoids, 3alpha,20-epoxy-6beta-hydroxy-1,7-dioxo-ent-abiet-15(17)-en-16-oic acid (1) and ent-abieta-7,15(17)-diene-3beta,16,18-triol (2) were isolated from Isodon eriocalyx var. laxiflora. Their structures were determined by extensive spectroscopic analysis and confirmed by X-ray crystallography. Compound 1 is an unprecedented example that establishes that a naturally occurring ent-abietane diterpenoid can have an oxygenation pattern almost identical to those of 3alpha,20-epoxy-ent-kaurane diterpenoids.

Abstract:Three hiherto unknown phenylpropanoid compounds, namely (7S,8R)‐1‐(1‐ethoxy‐2‐hydroxypropyl)‐2‐methoxy‐3,4‐(methylenedioxy)benzene (1), (7S,8S)‐1‐(1‐ethoxy‐2‐hydroxypropyl)‐2‐methoxy‐3,4‐(methylenedioxy)benzene (2), and (7S,8R)‐1‐(1‐methoxy‐2‐hydroxypropyl)‐2‐methoxy‐3,4‐(methylenedioxy)benzene (3), along with 12 known compounds (4 – 15) were obtained from the extract of whole plant of Chloranthus anhuiensis. Among them, 7 and 13 were obtained from nature for the first time. The structures of these natural compounds were characterized by extensive spectroscopic analysis and calculated electronic circular dichroism (ECD) data. Furthermore, their cytotoxic and neuroprotective activities were evaluated using MDA‐MB‐231, 4T1, HepG2, and PC12 cell lines. Compounds 8 and 13 exhibited moderate cytotoxic activities against MDA‐MB‐231 cell line with the IC50 values of 39.7 and 25.8 μm, respectively. And all the isolated compounds have no neuroprotective activities.

Abstract:Ten new ent-kaurane diterpenoids, including two pairs of epimers 1/2 and 4/5 and a 6,7-seco-ent-kauranoid 10, were obtained from the aerial parts of Isodon serra. The structures of the new compounds were confirmed by extensive spectroscopic methods and electronic circular dichroism (ECD) data analysis. An anti-inflammatory assay was applied to evaluate their nitric oxide (NO) inhibitory activities by using LPS-stimulated BV-2 cells. Compounds 1 and 9 exhibited notable NO production inhibition with IC50 values of 15.6 and 7.3 μM, respectively. Abstract:Hupelactones A (1) and B (2), two new eudesmanolide-type enantiomers of the corresponding compounds, along with four mono- (3-6) and nine dimeric- (7-15) known sesquiterpenoids were isolated from the whole plant of Chloranthus henryi var. hupehensis (syn. C. henryi). The new structures including the absolute configurations were determined by comparison with previously reported enantiomers, extensive spectroscopic methods in combination with electronic circular dichroism (ECD) calculations. All the isolates were evaluated for their inhibitory activities against the lipopolysaccharide (LPS)-induced nitric oxide (NO) production in murine BV-2 microglial cells. Among them, the dimeric lindenane sesquiterpenoids shizukaols F (8) and G (11) exhibited the most potent activities, with IC50 values of 2.65 and 4.60 mu M, respectively.
