Abstract:Fluoroalkylated quinoxlines with various groups were efficiently synthesized via a one-pot tandem Michael addition/azidation/cycloamination process. Under the mild and metal-free conditions, a bis-imine intermediate (4a) was detected and isolated for the first time. KI played a crucial role in this reaction. The mechanism was described.

Abstract:A TfOH-promoted synthesis of fluorinated poly-fused heterocycles via the cascade cyclization of azadienes and difluoroenoxysilanes has been developed, leading to the facile construction of fluorinated benzofuro[3,2-b]pyridines, 5H-indeno-[1,2-b]pyridines, and 5,6-dihydrobenzo[h]quinolines. This one-pot formal [4 + 2] approach involves 1,4-difluoroalkylation, desulfo-nylation, cyclization, and dehydrated and dehydrofluorinated aromatization and represents the first application of difluoroenoxysilane in cascade transformations. Furthermore, this methodology is highlighted by the synthesis of three fluoro analogues of bioactive molecules with potent topoisomerase inhibitory activities.

Abstract:A hexafluoroisopropanol（HFIP）-catalyzed highly diastereoselective formal [4 + 2] cyclization between ortho-hydroxyphenyl para-quinone methides and difluoroenoxysilanes is developed. This tandem protocol provides a simple and straightforward approach to assemble diverse multiply functionalized difluorinated chromans with high to excellent diastereoselectivity by employing difluoroenoxysilane as a new C2 synthon.

Abstract:Convenient and efficient synthesis of polyfluorinated 4-hydroxyquinolin-2(1H)-ones from the corresponding oalkynylaniline derivatives and CO2 (1 atm), mediated by 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU) and AgNO3 in acetonitrile was performed. This synthetic methodology may be used to prepare fluorinated heterocycles containing peripheral alkynyl and amino groups but is not suitable for silylethynyl derivatives that give indoles as the main products. The reaction takes place under mild conditions (60 degrees C) and involves readily available starting materials that include cheap and renewable carbon dioxide.

Abstract:The switchable synthesis of 3-non, 3-mono, 3,3′-disubstituted 3,4-dihydroquinolin-2(1H)-ones was developed through a redox-neutral hydride-transfer/N-dealkylation/N-acylation strategy from o-aminobenzaldehyde with 4-hydroxycoumarin, and Meldrum's acid, respectively. The unprecedented strategy for the synthesis of 3,3′-highly functionalized 3,4-dihydroquinolin-2(1H)-one has been realized with the in situ utilization of the released HCHO via the o-QM involved Michael addition.
