Abstract:The first nickel-catalyzed alkenylation of unactivated C(sp(3))-H bonds with vinyl iodides is described. The catalytic system comprises an inexpensive and air-stable Ni(acac)(2) as the catalyst and BINOL as the ligand, which is highly efficient for the alkenylation of beta-methyl C(sp(3))-H bonds of a broad range of aliphatic carboxamides. The resulting olefins can serve as versatile handles for further preparation. Additionally, we also demonstrated the synthesis of functionalized carboxamides bearing alpha-quaternary carbon centers from simple pivalamide via nickel-catalyzed sequential C(sp(3))-H bond functionalization.

Abstract:A nickel-catalyzed stereoselective alkenylation of an unactivated β-C(sp3)-H bond in aliphatic amide with terminal alkynes using 8-aminoquinoline auxiliary is reported for the first time. This reaction displays excellent functional group tolerance with respect to both aliphatic amides and terminal alkynes and features a cheap nickel catalytic system. The 8-aminoquinolyl directing group could be smoothly removed, and the resultant β-styrylcarboxylic acid derivatives could serve as versatile building blocks for further transformation.

Abstract:We herein describe a nickel-catalyzed cyanation reaction of aryl/alkenyl halides with alkyl isocyanides. Both aryl/alkenyl iodines and bromides were found to be competent electrophiles that reacted with alkyl isocyanides, affording nitrile compounds in moderate to good yields. A range of functional groups including halogens as well as hydroxyl, formyl, and acetamino groups were fairly compatible with the nickel catalysis. This protocol featured broad functional group tolerance, simple reaction conditions, and gram-scale synthesis.

Abstract:In this work, we demonstrated Ni-catalyzed oxidative coupling of unactivated C(sp(3))-H bonds with terminal alkynes for construction of C(sp(3))-C(sp) bonds to synthesize alkyl-substituted internal alkynes. Different amides exhibited good compatibility. Preliminary mechanistic studies were conducted to account for this alkynylation.

Abstract:C(sp3) centers adjacent to (hetero)aryl groups are widely present in physiologically active molecules. Metal‐hydride‐catalyzed hydroalkylation of alkenes represents an efficient means of forging C(sp3)‐C(sp3) bonds, boasting advantages as a wide source of substrates, mild reaction conditions, and facile selectivity manipulation. Nevertheless, the hydroalkylation of vinylarenes encounters constraints in terms of substrate scope, necessitating the employment of activated alkyl halides or alkenes containing chelating groups, remains a challenge. In this context, we report a general nickel‐hydride‐catalyzed hydroalkylation protocol for vinylarenes. Remarkably, this system enables α‐selective hydroalkylation of both aryl and heteroaryl alkenes under an extra ligand‐free condition, demonstrating excellent coupling efficiency and selectivity. Furthermore, through the incorporation of chiral bisoxazoline ligands, we have achieved regio‐ and enantioselective hydroalkylation of vinylpyrroles, thereby facilitating the synthesis of α‐branched alkylated pyrrole derivatives.

Abstract:The first Pd(II)-catalyzed enantioselective alkynylation of unbiased methylene β-C(sp3)-H bonds is reported. The readily accessible and tunable BINOL derivatives are used as chiral ligands in C-H activation for the first time. 3,3'-Fluorinated-BINOL proved crucial in determining both the reactivity and enantioselectivity. A wide range of carboxylic acid derivatives are well tolerated with high enantioselectivities (up to 96% ee). Mechanistic studies suggest that multiple ligands may participate in the stereodetermining C-H palladation step, and a chiral amplification effect is observed.

Abstract:Decarboxylative cross-coupling of aliphatic acid anhydrides with vinyl triflates or halides was accomplished via nickel catalysis. This methodology works well with a broad array of substrates and features abundant functional group tolerance. Notably, our approach addresses the issue of safe and environmental installation of methyl or ethyl group into molecular scaffolds. The method possesses high chemoselectivity toward alkyl groups when aliphatic/aromatic mixed anhydrides are involved. Furthermore, diverse ketones could be modified with our strategy.

Abstract:Herein, an electrochemically driven NiH-catalyzed reductive coupling of alkyl halides and alkyl alkenes for the construction of Csp3-Csp3 bonds is firstly reported. Notably, alkyl halides serve dual function as coupling substrates and as hydrogen sources to generate NiH species under electrochemical conditions. The tunable nature of this reaction is realized by introducing an intramolecular coordinating group to the substrate, where the product can be easily adjusted to give the desired branched product. The method proceeds under mild conditions, exhibits a broad substrate scope, and affords moderate to excellent yields with over 70 examples, including late-stage modification of natural products and drug derivatives. Mechanistic insights offer evidence for an electrochemically driven coupling process. The sp3-carbon-halogen bonds can be activated through single electron transfer (SET) by the nickel catalyst in its low valence state, generated by cathodic reduction, and the generation of NiH species from alkyl halides is pivotal to this transformation.

Abstract:A methodology for radical cross-coupling with vinylsiloxanes and pyridinium salts under nickel catalysis is described. Easily implemented from inexpensive and abundant primary amines and terminal alkynes, this modern Hiyama coupling provides efficient access to (E), (Z), and 1,1’-alkenes with configuration retention. Operating under mild conditions, this robust strategy applies to a broad range of functional groups with diverse double bond stereochemistries. This versatile reaction is scalable and straightforward, accommodating both secondary and primary alkyl groups. A radical mechanism involving hypervalent silicon intermediates is proposed.

Abstract:Methods for incorporating alkynes into molecular skeletons have been the subject of continued research. Compared to the cross -couplings used to form C-C(sp3) and C-C(sp2) bonds, couplings that form C-C(sp) bonds, particularly those catalyzed by nickel, remain less explored. In this study, we present a single nickel -cata-lyzed cross-coupling of terminal alkynes with aryl iodides or bro-mides for constructing C(sp2)-C(sp) bonds. Our method provides high functional group tolerance and broad substrate scope, result-ing in good yields. This nickel-catalyzed coupling reaction is a straightforward way to prepare optoelectronic materials and key building blocks for chemical biology. Notably, our method dis-plays decent selectivity when multi-halide-substituted arenes are used as substrates. Finally, a plausible mechanism is postulated based on experimental studies and density functional theory calculations.

Abstract:While traditional Sonogashira reaction requires a palladium catalyst and a copper co-catalyst, some recent variants were reported being promoted by single transition metals. Here we report a single nickel-catalyzed tandem Heck-Sonogashira annulation/alkynylation for enantioselectively constructing C(sp3)-C(sp) bond. In addition, using the same catalytic system, Sonogashira C(sp2)-C(sp) cross-coupling has also been achieved. The alkynylations described in this report are important for the three reasons: 1. C(sp3)/(sp2)-C(sp) bonds exist in many bioactive natural products and drug molecules as well as their key synthetic intermediates; 2. There was no precedent for single nickel-catalyzed Sonogashira reaction owing to the difficulties caused by strong coordination of nickel to the triple bond to inactivate the catalyst; 3. Isolation and characterization of single-crystal structure of a resting state intermediate, di-phosphine chelated σ-alkyl-NiII-I complex, which provided crucial evidence to support the mechanistic postulation and guided DFT calculations.
