Abstract:A sustainable and simple catalytic system for the atom-economical alkynylation of benzamides with low nickel loadings is described. No organic or metallic oxidants and expensive ligands are required. A broad range of benzamides and bromoalkynes bearing various synthetically useful functional groups are compatible with this reaction. The versatility of this operationally simple protocol has been further demonstrated by the controllable mono-and di-alkynylation. Importantly, substrate/catalyst ratios of up to 200, and a turnover number of 196 were achieved, highlighting the potential of this protocol for synthetic applications.

Abstract:DFT calculations have been conducted to elucidate the mechanistic details of a novel Ni-catalyzed hydrocarboxylation reaction of alkynes, in which formic acid is atom-economically used through a catalytic CO recycling manner. On the basis of our theoretical investigations, the bisphosphine (dppbz, 1,2-bis(diphenylphosphino)benzene) ligated nickel monocarbonyl complex (dppbz)NiCO was located as the active catalytic species for this process, and such a carbonyl ligand is found to be critical for the final reductive elimination. Our studies also revealed the addition of H to alkynes proceeds via a proton transfer process directly from formic acid (i.e., outer-sphere pathway) rather than through a proposed hydrometalation process (i.e., direct hydride shift from Ni-H). The thermal decomposition of formic anhydrides was found to be vital to a successful reaction, and its barrier must be slightly higher than the energetic span of the Ni catalytic cycle. Fast release of CO can poison the Ni catalyst, so that the reaction would be shut down. Other intriguing experimental observations, such as ligand effect, regioselectivity, and extraordinary compatibility of C-X (X = halogen) bonds, are also discussed in this article.

Abstract:While traditional Sonogashira reaction requires a palladium catalyst and a copper co-catalyst, some recent variants were reported being promoted by single transition metals. Here we report a single nickel-catalyzed tandem Heck-Sonogashira annulation/alkynylation for enantioselectively constructing C(sp3)-C(sp) bond. In addition, using the same catalytic system, Sonogashira C(sp2)-C(sp) cross-coupling has also been achieved. The alkynylations described in this report are important for the three reasons: 1. C(sp3)/(sp2)-C(sp) bonds exist in many bioactive natural products and drug molecules as well as their key synthetic intermediates; 2. There was no precedent for single nickel-catalyzed Sonogashira reaction owing to the difficulties caused by strong coordination of nickel to the triple bond to inactivate the catalyst; 3. Isolation and characterization of single-crystal structure of a resting state intermediate, di-phosphine chelated σ-alkyl-NiII-I complex, which provided crucial evidence to support the mechanistic postulation and guided DFT calculations.

Abstract:The cheap nickel catalysts are more reactive than the corresponding noble metal catalysts in the catalytic additions of a variety of P(O)-H bonds and an S-H bond to alkynes, affording regio- and stereoselectively both the Markovnikov and the anti-Markovnikov adducts, respectively, in high yields. A related five-coordinate hydrido nickel complex in the catalysis is successfully isolated, which can react readily with an alkyne to give the addition products.

Abstract:Methods for incorporating alkynes into molecular skeletons have been the subject of continued research. Compared to the cross -couplings used to form C-C(sp3) and C-C(sp2) bonds, couplings that form C-C(sp) bonds, particularly those catalyzed by nickel, remain less explored. In this study, we present a single nickel -cata-lyzed cross-coupling of terminal alkynes with aryl iodides or bro-mides for constructing C(sp2)-C(sp) bonds. Our method provides high functional group tolerance and broad substrate scope, result-ing in good yields. This nickel-catalyzed coupling reaction is a straightforward way to prepare optoelectronic materials and key building blocks for chemical biology. Notably, our method dis-plays decent selectivity when multi-halide-substituted arenes are used as substrates. Finally, a plausible mechanism is postulated based on experimental studies and density functional theory calculations.

Abstract:We report nickel-catalyzed cyanation and hydrocyanation methods to prepare aryl nitriles and vinyl nitriles from aryl halides and alkynes, respectively. Using inexpensive and nontoxic 4-cyanopyridine N-oxide as the cyano shuttle, the methods provide an efficient approach to prepare aryl cyanides and vinyl nitriles under mild and operationally simple reaction conditions with a broad range of functional group tolerances. In hydrocyanation of alkynes, the method demonstrated good regioselectivity, producing predominantly E- or Z-alkenyl nitriles in a controlled manner and exclusively Markovnikov vinyl nitriles when internal diaryl alkynes and terminal alkynes were applied as the substrates, respectively. The preliminary mechanistic investigation indicated that the C-CN bond cleavage process is promoted by oxidative addition to the nickel(I) complex in the cyanation of aryl halides, and further studies via a series of deuterium exchange experiments indicated that water serves as the hydrogen source for the hydrocyanation of alkynes.

Abstract:The exploration of strong chemical bonds as synthetic handles offers new disconnection strategies for the synthesis of functionalized molecules via transition metal catalysis. However, the slow oxidative addition rate of these covalent bonds to a transition metal center hampers their synthetic utility. Here, we report a C(sp3)-N bond activation strategy that bypasses thermodynamically challenging 2e- or 1e- oxidative addition via a distinct pathway in nickel catalysis. This strategy leverages a previously unknown activation pathway of photoinduced inner-sphere charge transfer of low-valent nickel(isonitriles), triggering a C(sp3)-N bond cleavage distal to the metal-ligand interaction to deliver nickel(cyanide) and versatile alkyl radicals. Utilizing this catalytic strategy, the selective intermolecular 1,2-carbocyanation reaction of alkynes with alkyl isonitriles as both alkylating and cyanating agents can be achieved, delivering a wide array of trisubstituted alkenyl nitriles with excellent atom-economy, regio-, and stereoselectivity under mild conditions. Furthermore, Markovnikov-selective hydrocyanation of aliphatic alkynes can be accomplished through the synergistic action of a photocatalyst utilizing isonitriles as the cyanation agents. Mechanistic investigations support the photogeneration of low-valent Ni(isonitrile) complexes that undergo photochemical homolysis of the C(sp3)-N bond to engage catalytic cyanation with alkynes.

Abstract:Functionalization of C-H bonds has emerged as a powerful strategy for converting inert, nonfunctional C-H bonds into their reactive counterparts. A wide range of C-H bond functionalization reactions has become possible by the catalysis of metals, typically from the second row of transition metals. First-row transition metals can also catalyze C-H functionalization, and they have the merits of greater earth-abundance, lower cost and better environmental friendliness in comparison to their second-row counterparts. C-H bond alkylation is a particularly important C-H functionalization reaction due to its chemical significance and its applications in natural product synthesis. This review covers Ni-catalyzed C-H bond alkylation reactions using alkyl halides and olefins as alkyl sources.

Abstract:Nickel(0) nanoparticles coordinated to NHC ligands bearing N-coordinated cinnamyl moieties were readily prepared by reduction of a [NiCpBr(NHC-cinnamyl)] complex with methyl magnesium bromide. The combination of a strong σ-donor NHC ligand with a π-coordinating appended cinnamyl moiety likely prevents nickel(0) nanoparticle aggregation to larger inactive species, and allows the effective and (Z)-selective semi-hydrogenation of alkynes and ynamides.

Abstract:Herein, an electrochemically driven NiH-catalyzed reductive coupling of alkyl halides and alkyl alkenes for the construction of Csp3-Csp3 bonds is firstly reported. Notably, alkyl halides serve dual function as coupling substrates and as hydrogen sources to generate NiH species under electrochemical conditions. The tunable nature of this reaction is realized by introducing an intramolecular coordinating group to the substrate, where the product can be easily adjusted to give the desired branched product. The method proceeds under mild conditions, exhibits a broad substrate scope, and affords moderate to excellent yields with over 70 examples, including late-stage modification of natural products and drug derivatives. Mechanistic insights offer evidence for an electrochemically driven coupling process. The sp3-carbon-halogen bonds can be activated through single electron transfer (SET) by the nickel catalyst in its low valence state, generated by cathodic reduction, and the generation of NiH species from alkyl halides is pivotal to this transformation.

Abstract:The myriad nickel-catalyzed cross-coupling reactions rely on the formation of an organonickel intermediate, but limitations in forming monoalkylnickel species have limited options for C(sp 3 ) cross-coupling. The formation of monoalkylnickel(II) species from abundant carboxylic acid esters would be valuable, but carboxylic acid derivatives are primarily decarboxylated to form alkyl radicals that lack the correct reactivity. In this work, we disclose a facile oxidative addition and decarbonylation sequence that forms monoalkylnickel(II) intermediates through a nonradical process. The key ligand, bis(4-methylpyrazole)pyridine, accelerates decarbonylation, stabilizes the alkylnickel(II) intermediate, and destabilizes off-cycle nickel(0) carbonyl species. The utility of this new reactivity in C(sp 3 )-C(sp 3 ) bond formation is demonstrated in a reaction that is challenging by purely radical methods—the selective cross-coupling of primary carboxylic acid esters with primary alkyl iodides.
