Abstract:The efficient remote sulfonylation of N-(quinolin-8-yl)benzamide derivatives at the C5 position has been well developed. The reaction generates environmentally benign byproducts by utilizing stable and safe sodium sulfinates as sulfide sources. A series of N-(5-(phenylsulfonyl) quinolin-8-yl) benzamide derivatives were successfully obtained in moderate to high yields. In particular, they are less unpleasantly odorous and more environmentally friendly than previous means.

Abstract:The first remote C-H trifluoromethylation of N-(quinolin-8-yl)-benzamide derivatives was accomplished with a user-friendly chitosan-based heterogeneous copper catalyst under mild conditions. The position-selective C-H activation protocol afforded the corresponding coupling products in good to excellent yields with excellent reusability of the catalyst by using the low-costing and stable Langlois reagent (CF3SO2Na) as the "CF3" source. Furthermore, control experiments suggested a single-electron-transfer process played a vital role in the heterogeneous C-CF3 cross-coupling.

Abstract:An efficient and convenient method is developed for the remote C-H bond chalcogenation of 8-aminoquinoline scaffolds on the C5 position that is geometrically inaccessible. The protocol makes use of inexpensive CuBr2 as mediator and shows good tolerance toward numerous disulfides/diselenides and aliphatic amides, giving the corresponding products in good to excellent yield.

Abstract:The copper‐catalyzed cross‐dehydrogenative coupling of N‐iminoquinolinium ylides with secondary amines led to ortho‐amino‐substituted quinoline derivatives with high levels of regioselectivity in good yields. This direct C–H bond amination transformation employs CuI as the catalyst without the use of a ligand, external oxidant, or base. The reaction is operationally simple and can be conducted under mild conditions. The N‐benzoyl directing group can be removed without any additional steps.

Abstract:The remote C-O coupling of quinoline amides at the C5 position has been established using cheap and readily available copper catalyst under mild conditions. The positionally selective esterification protocol afforded C-O coupling products in moderate to excellent yields. Most importantly, this method may provide a potential alternative to the existing ways to build functionalized 5-hydroxyquinoline derivatives, which are utilized as pivotal intermediates in the synthesis of drug candidates.

Abstract:An iron(II)‐catalyzed cascade reaction, involving cyanoalkylsulfonylation and cyclization, has been devised to enable the synthesis of functionalized sulfonylated quinolino‐quinazolinone alkyl nitriles. This three‐component radical transformation exhibits excellent chemo‐ and regioselectivity while functioning without the need for external oxidizing or reducing agents. The cost‐effectiveness of this catalytic system, together with its broad substrate application, which includes drug molecule derivatives, make this approach efficient and adaptable.
