Abstract:An efficient and convenient method is developed for the remote C-H bond chalcogenation of 8-aminoquinoline scaffolds on the C5 position that is geometrically inaccessible. The protocol makes use of inexpensive CuBr2 as mediator and shows good tolerance toward numerous disulfides/diselenides and aliphatic amides, giving the corresponding products in good to excellent yield.

Abstract:An efficient method for the synthesis of substituted quinolines is described. By FeCl3-promoted C–C bond formation, 2,3-disubstituted and 2,3,4-trisubstituted quinolines were successfully synthesized from the reaction between imines and electron-deficient alkynes in good yields. The reaction can also be performed in a sequential three-component manner by using aromatic amines and aromatic aldehydes instead of the corresponding imines.

Abstract:Depending on the metal complexes employed, regiodivergent C–H direct chlorination on 8-acylaminoquinolines could be realized. While the stoichiometric CuCl2/base persistently gave the C5-monochlorination, the Pd(II)-catalyzed chlorination by using N-chlorosuccinimide (NCS) as a chlorine source led to variable results mainly relying on the substitution pattern of the 8-acylaminoquinolines and the loading of NCS. This work demonstrated not only an unprecedented access to divergently halogenated quinolines, but also a less studied C(sp2)–H functionalization mode, namely the single-electron transfer (SET) mediated C(sp2)–H functionalization.

Abstract:A selective copper(I)-catalyzed benzylic C(sp3)–H geminal difunctionalization reaction of a benzylic-type sp3 carbon was developed. This novel strategy allowed simultaneous introduction of amide and hydroxyl group in a highly selective and efficient way via successive oxidative intramolecular amidation and hydroxylation. This method was also applied to the synthesis of 3-hydroxy-2,3-dihydro-1H-pyrrolo[3,4-b]quinoxalinones from readily available 3-methyl-N-substituted quinoxaline-2-carboxamides in moderate to good yields. The five-membered cyclic hemiaminal moiety of the pyrrolo[3,4-b]quinoxalinones can serve as an intermediates for subsequent transformations into other useful functional groups.

Abstract:A Cu-catalyzed tandem synthesis of azole-fused pyrimido[1,2-c]quinazolines and imidazo[1,2-c]quinazolines has been developed. The reaction is based on a C-N cross-coupling/C-H functionalization reaction of 2-(2-bromophenyl)-1,4,5,6-tetrahydropyrimidines with azoles.
