Abstract:Reverse micelles bring mild and effective microenvironments in organic solvent that contain bio-molecules, which have attracted immense attention for application in the isolation of proteins, protein refolding, and enzymatic reaction. In this review, the application of reverse micelles for protein separation and refolding has been briefly summarized and various reverse micellar systems composed of different surfactants, including ionic, nonionic, mixed, and affinity-based reverse micelles, have been highlighted. It illustrates especially the potential application of the novel affinity-based reverse micelles consisting of biocompatible surfactant coupled with affinity ligands. Moreover, the importance to develop universal affinity-based reverse micelles for protein separation and refolding in the downstream processing of biotechnology has been pointed out.

Abstract:Mixed-mode chromatography is a new kind of chromatography technology for bioseparation.The special ligand combines two or more interaction modes,commonly hydrophobic and electrostatic interactions.Mixed-mode chromatography can directly capture target protein from moderate-ionic-strength feedstock without the need of dilution or other additives,which certainly improves the process efficiency and reduces the cost of primary recovery in the downstream process.This paper reviews the current advances in mixed-mode chromatography,as well as technical history and some applications.The future development and application areas are also discussed.

Abstract:The reversed micellar solution formed by Sorbitan trioleate (Span 85) coupled with Cibacron Blue F3G-A (CB) as an affinity ligand show good potential for use in reversed micellar protein extraction operations. In this work, the extraction behavior of lysozyme and ovalbumin with the reversed micellar phase has been investigated at extensive pH values and CB concentrations. The partitioning isotherms of lysozyme and ovalbumin were expressed by the Langmuir equation. By addition of 3vol.% hexanol to the reversed micellar phase, the extraction capacity of lysozyme reached 6.38mg/mL, over seven times higher than that of ovalbumin (0.83mg/mL). Thus, lysozyme was separated from crude chicken ovalbumin solutions with a purification factor over 20. Moreover, the reversed micellar solution was recycled three times for the separation, and the purification factor was kept nearly 20 in the recycling. Incomplete back extraction of lysozyme to the stripping solution compromised the total recovery yields in the following recycles, so more effective back extraction method needs to be developed to make the extraction system more attractive. In general, the present results have shown the potential of the affinity-based reversed micellar system for application in protein separations.

Abstract:Biomolecules produced by living organisms can perform vast array of functions and play an important role in the cell. Important biomolecules such as lysozyme, bovine serum albumin (BSA), and bromelain are often studied by researchers due to their beneficial properties. The application of reverse micelles is an effective tool for protein separation from their sources due to the special system structure. Mechanisms of transferring biomolecules and factors that influence the extraction of biomolecules are reviewed in this paper. The enhancement of biomolecule extraction could be achieved depending on the properties of reverse micelles. This paper provides an overall review on lysozyme, BSA, and bromelain extraction by reverse micelle for various applications.

Abstract:Reversed micellar two-phase extraction is a developing technique for protein separation. Introduction of an affinity ligand is considered to be an effective approach to increase the selectivity and capacity of reversed micelles. In this article, Cibacron Blue F3G-A (CB) as an affinity ligand was immobilized to reversed micelles composed of soybean lecithin by a two-phase reaction. The affinity partitioning of lysozyme and bovine serum albumin (BSA) to the CB-lecithin micelles was studied. Formation of mixed micelles by additionally introducing a nonionic surfactant, Tween 85, to the CB-lecithin micelles was effective to increase the solubilization of lysozyme due to the increase of W-0 (water/surfactant molar ratio)/micellar size. The partitioning isotherms of lysozyme to the CB-lecithin micelles with and without Tween 85 were expressed by the Langmuir equation. The dissociation constants in the Langmuir equation decreased on addition of Tween 85, indicating the increase of the effectiveness of lysozyme binding to the immobilized CB. On addition of 20 g/L Tween 85 to 50 g/L lecithin/hexane micellar phase containing 0.1 mmol/L CB, the extraction capacity for lysozyme could be increased by 42%. Moreover, the CB-lecithin micelles with or without Tween 85 showed significant size exclusion for BSA due to its high molecular weight. Thus, lysozyme and BSA were separated from artificial solutions containing the two proteins. In addition, the affinity-based reversed micellar phase containing Tween 85 was recycled three times for lysozyme purification from crude egg-white solutions. Lysozyme purity increased by 16-18-fold, reaching 60-70% in the recycled use.

Abstract:Background: Bioactive compounds from various natural sources have been attracting more and more attention, owing to their broad diversity of functionalities and availabilities. However, many of the bioactive compounds often exist at an extremely low concentration in a mixture so that massive harvesting is needed to obtain sufficient amounts for their practical usage. Thus, effective fractionation or separation technologies are essential for the screening and production of the bioactive compound products. The applicatons of conventional processes such as extraction, distillation and lyophilisation, etc. may be tedious, have high energy consumption or cause denature or degradation of the bioactive compounds. Membrane separation processes operate at ambient temperature, without the need for heating and therefore with less energy consumption. The "cold" separation technology also prevents the possible degradation of the bioactive compounds. The separation process is mainly physical and both fractions (permeate and retentate) of the membrane processes may be recovered. Thus, using membrane separation technology is a promising approach to concentrate and separate bioactive compounds. Methods: A comprehensive survey of membrane operations used for the separation of bioactive compounds is conducted. The available and established membrane separation processes are introduced and reviewed. Results: The most frequently used membrane processes are the pressure driven ones, including microfiltration (MF), ultrafiltration (UF) and nanofiltration (NF). They are applied either individually as a single sieve or in combination as an integrated membrane array to meet the different requirements in the separation of bioactive compounds. Other new membrane processes with multiple functions have also been developed and employed for the separation or fractionation of bioactive compounds. The hybrid electrodialysis (ED)-UF membrane process, for example has been used to provide a solution for the separation of biomolecules with similar molecular weights but different surface electrical properties. In contrast, the affinity membrane technology is shown to have the advantages of increasing the separation efficiency at low operational pressures through selectively adsorbing bioactive compounds during the filtration process. Conclusion: Individual membranes or membrane arrays are effectively used to separate bioactive compounds or achieve multiple fractionation of them with different molecule weights or sizes. Pressure driven membrane processes are highly efficient and widely used. Membrane fouling, especially irreversible organic and biological fouling, is the inevitable problem. Multifunctional membranes and affinity membranes provide the possibility of effectively separating bioactive compounds that are similar in sizes but different in other physical and chemical properties. Surface modification methods are of great potential to increase membrane separation efficiency as well as reduce the problem of membrane fouling. Developing membranes and optimizing the operational parameters specifically for the applications of separation of various bioactive compounds should be taken as an important part of ongoing or future membrane research in this field.

Abstract:Membrane proteins are generally of natural low abundance and insoluble to aqueous solutions. Thus, investigation of membrane proteins is relatively hampered since efficient separation of membrane proteins are rather challenging. Microseparation approaches certainly play an essential role in fulfilling such demanding investigations due to their significant advantages of high throughput, reduced separation time, and low sample consumption. In this review, recent developments of microseparation are documented with aspects of three key separation methods, including CE, micro-LC and microchip. Preparation of membrane proteins prior to separation is also discussed, including solubilization and preconcentration. Certainly, more applications of microseparation approaches in membrane protein separations can be expected in the near future.

Abstract:Reversed micellar system (RMS) is an innovative technique used for the isolation, extraction and purification of proteins and enzymes. Studies have demonstrated that RMS is an efficient purification technology for extracting proteins and enzymes from natural plant materials or fermentation broth. Lately, reverse micelles have wider biological applications and the ease of scaling up and the possibility for the continuous process have made RMS a vital purification technique in various fields. In this study, an extensive review of RMS with the current application in biotechnology is examined. This review provides insights into the fundamental principles, key variables and parameters of RMS. In addition, a comparative study of RMS with other liquid-liquid extraction techniques is also included. The present review aims to provide a general overview of RMS by summarising the research works, since the introduction of the technology to current development.

Abstract:This paper aims to discuss the use of reverse micelles in downstream processing of biotechnological products. The interest in this technology is piqued firstly by various advantages of a liquid-liquid extraction process, among which are cost effectiveness, and ease of scaling up and implementing a continuous process for whole broth processing. The use of reverse micelles is thought to be among the most promising due to the high efficiency and selectivity being achieved in some systems. However, there are various issues that have impeded the widespread use of reverse micelles such as the identification and development of suitable surfactants and ligands; as well as difficulties in the back extraction process.
