Abstract:The concept and characteristics of reverse micelles and the principle of reverse micelles extraction protein were briefly introduced in the this paper. The factors of influencing reversed micelles extraction protein, including pH, ionic strength, the type of cation, the molecular weight and concentration of protein, the type and concentration of surfactant were introduced. Abstract:Mixed-mode chromatography is a new kind of chromatography technology for bioseparation.The special ligand combines two or more interaction modes,commonly hydrophobic and electrostatic interactions.Mixed-mode chromatography can directly capture target protein from moderate-ionic-strength feedstock without the need of dilution or other additives,which certainly improves the process efficiency and reduces the cost of primary recovery in the downstream process.This paper reviews the current advances in mixed-mode chromatography,as well as technical history and some applications.The future development and application areas are also discussed.

Abstract:Reversed micellar system (RMS) is an innovative technique used for the isolation, extraction and purification of proteins and enzymes. Studies have demonstrated that RMS is an efficient purification technology for extracting proteins and enzymes from natural plant materials or fermentation broth. Lately, reverse micelles have wider biological applications and the ease of scaling up and the possibility for the continuous process have made RMS a vital purification technique in various fields. In this study, an extensive review of RMS with the current application in biotechnology is examined. This review provides insights into the fundamental principles, key variables and parameters of RMS. In addition, a comparative study of RMS with other liquid-liquid extraction techniques is also included. The present review aims to provide a general overview of RMS by summarising the research works, since the introduction of the technology to current development.

Abstract:Reverse micelles bring mild and effective microenvironments in organic solvent that contain bio-molecules, which have attracted immense attention for application in the isolation of proteins, protein refolding, and enzymatic reaction. In this review, the application of reverse micelles for protein separation and refolding has been briefly summarized and various reverse micellar systems composed of different surfactants, including ionic, nonionic, mixed, and affinity-based reverse micelles, have been highlighted. It illustrates especially the potential application of the novel affinity-based reverse micelles consisting of biocompatible surfactant coupled with affinity ligands. Moreover, the importance to develop universal affinity-based reverse micelles for protein separation and refolding in the downstream processing of biotechnology has been pointed out.

Abstract:Biomolecules produced by living organisms can perform vast array of functions and play an important role in the cell. Important biomolecules such as lysozyme, bovine serum albumin (BSA), and bromelain are often studied by researchers due to their beneficial properties. The application of reverse micelles is an effective tool for protein separation from their sources due to the special system structure. Mechanisms of transferring biomolecules and factors that influence the extraction of biomolecules are reviewed in this paper. The enhancement of biomolecule extraction could be achieved depending on the properties of reverse micelles. This paper provides an overall review on lysozyme, BSA, and bromelain extraction by reverse micelle for various applications.

Abstract:This paper aims to discuss the use of reverse micelles in downstream processing of biotechnological products. The interest in this technology is piqued firstly by various advantages of a liquid-liquid extraction process, among which are cost effectiveness, and ease of scaling up and implementing a continuous process for whole broth processing. The use of reverse micelles is thought to be among the most promising due to the high efficiency and selectivity being achieved in some systems. However, there are various issues that have impeded the widespread use of reverse micelles such as the identification and development of suitable surfactants and ligands; as well as difficulties in the back extraction process. These issues, as well as latest developments and applications of reverse micelles in downstream processing of biotechnological products will be discussed in this paper.

Backward extraction of reverse micellar encapsulated proteins using a counterionic surfactant

Abstract:The back-extraction of proteins encapsulated in AOT reverse micelles was performed by adding a counterionic surfactant, either TOMAC or DTAB. This novel backward transfer method gave higher backward extraction yields compared to the conventional method with high salt and high pH of the aqueous stripping solution. The protein activity was maintained in the resulting aqueous phase, which in this case had a near neutral pH and low salt concentration. A sharp decrease of the water content was observed in the organic phase corresponding to protein back-extraction using TOMAC. The backward transfer mechanism was postulated to be caused by electrostatic interaction between oppositely charged surfactant molecules, which lead to the collapse of the reverse micelles. The back-extraction process with TOMAC was found to be very fast; more than 100 times faster than back-extraction with the conventional method, and as much as 3 times faster than forward extraction. The formation of 1:1 complexes of AOT and TOMAC in the solvent phase was observed, and these hydrophobic complexes could be efficiently removed from the solvent using adsorption onto Montmorillonite in order for the organic solvent to be reused. A second cationic surfactant, DTAB, confirmed the general applicability of counterionic surfactants for the backward transfer of proteins.

Abstract:Sodium bis (2-ethylhexyl) sulfosuccinate (AOT)-sodium dodecyl sulfate (SDS)/isooctane-octanol reverse micelle extraction was tested an efficient and effective approach to separate peanut protein from full-fat peanut powder. Here, important kinetic factors including pH, ion strength, and temperature were studied during reverse micelle backward extraction. The extraction conditions were obtained by response surface experiments as follows: pH 7.5, ion concentration 1.1 mol/L at temperature 35°C. Under these optimum extraction conditions, the extraction rate of protein reached 79.03%. A model on the kinetic partitioning of peanut protein was also developed. The backward extraction in this reverse micelle system was controlled by interfacial resistance instead of diffusion resistance in reverse micelle and aqueous phase with the total mass transfer rate of 0.8×10−5 m3·s−1. A two-film theory may be the mechanism for flat interface. Results of mass transfer process are helpful for creating an reverse micelle extraction process, and used for purification of peanut proteins, promoting the development of food industry.

Abstract:The reversed micellar solution formed by Sorbitan trioleate (Span 85) coupled with Cibacron Blue F3G-A (CB) as an affinity ligand show good potential for use in reversed micellar protein extraction operations. In this work, the extraction behavior of lysozyme and ovalbumin with the reversed micellar phase has been investigated at extensive pH values and CB concentrations. The partitioning isotherms of lysozyme and ovalbumin were expressed by the Langmuir equation. By addition of 3vol.% hexanol to the reversed micellar phase, the extraction capacity of lysozyme reached 6.38mg/mL, over seven times higher than that of ovalbumin (0.83mg/mL). Thus, lysozyme was separated from crude chicken ovalbumin solutions with a purification factor over 20. Moreover, the reversed micellar solution was recycled three times for the separation, and the purification factor was kept nearly 20 in the recycling. Incomplete back extraction of lysozyme to the stripping solution compromised the total recovery yields in the following recycles, so more effective back extraction method needs to be developed to make the extraction system more attractive. In general, the present results have shown the potential of the affinity-based reversed micellar system for application in protein separations.

Abstract:Protein was extracted from Xanthoceras sorbifolia bunge kernel by using reversed micelles.According to extraction conditions of Xanthoceras sorbifolia bunge kernel protein,the back-extraction was also investigated.The following parameters such as pH of back extraction aqueous phase,ion concentration,temperature,back extraction time were examined.According to the experiments,the optimum condition of backward extraction are: KCl concentration 1.2 mol/L,pH 4.0,temperature 60 ℃,and time 60min.Analysis of variance result indicated that the three factors of salt concentration,temperature and pH value significantly contribute to the protein extraction rate.Under these conditions,the back-protein extraction rate and the oil extraction rate reached 77.58% and 63.7%,respectively.

Abstract:The article discussed the kinetics of reextraction process of soybean protein through systemically studying the effect of main factors(pH values,ionic strength in the aqueous phase,initial protein concentration of organic phase,shaking speed and temperature) on total mass transfer coefficient in AOT(sodium bis-(2-ethylhexy) sulfosuccinate)-isooctane reverse micelle system.The results indicate that the total mass transfer coefficient increases with the increasing of temperature and pH value of aqueous phase;the total mass transfer coefficient increases and then decreases with the increasing of ionic strength;it is almost not influenced by protein concentration in organic phase and the shaking speed.It shows that the diffusion resistance of soybean protein in the aqueous phase and organic phase can be ignored and reextraction process is controlled by the interface resistance.Therefore,it can be concluded that the reextraction process belongs to the interface control type,and the velocity is controlled by coalescence process of 'full reversed micelles'.The mechanism of soybean protein reextraction process is understanded,which can effectively control and enhance extraction process,increase the extraction rate and provide reliable data and theoretical reference for further research.

Abstract:The extraction and back extraction of Bovine Serum Albumin (BSA) using a new reversed micelle system of CTAB/isooctane/1-pentanol have been investigated. Under the optimal operating conditions in this study, the extraction ratio of BSA reaches as high as 98%. The back extraction ratio can reach 80% as pH in stripping phase approaches to the isoelectric point (pI) of BSA. UV-spectra indicated that there was no significant change in the structure of BSA enriched in stripping phase. Furthermore, the reversed micelle system was repeatedly used to extract BSA from a mixed solution of protein.
