Abstract:Cyanoguanidine derivatives of loratadine (3a–i) were synthesized and screened for antitumor and anti-inflammatory activity. The most promising compound 3c (R=n-C8H17) possessed at least twofold higher in vitro cytotoxicity than 5-fluorouracil against mammary (MCF-7 and MDA-MB 231) as well as colon (HT-29) carcinoma cells. The mode of action, however, is so far unclear. The participation of the COX-1/2 enzymes on the cytotoxicity, however, is very unlikely. Nevertheless all compounds showed stronger in vivo anti-inflammatory activity than ibuprofen in the xylene-induced ear swelling assay in mice.

Abstract:Loratadine was synthesized from 2-cyano-3-methylpyridine via Ritter reaction, alkylation and cyanidation to give 2-cyano-3-[2-(3-chlorophenyl)ethyl]piperidine, which subjected to cyclization in presence of PPA/P2O5 and then coupling with N-ethoxycarbonyl-4-piperidone. The overall yield was 39%.

Abstract:Seven target compounds were designed and synthesized according to the structure of lead compound—desloratadine.The structures of the compounds were identified by 1H NMR and HR-MS.The in vitro results based on histamine-induced contraction of guiea-pig ileum showed that zwitterionic compound 9c had stronger antihistamine activity than positive control desloratadine.The human ether-à-go-go related gene(hERG) inhibition and guinea-pig langendorff model indicated that compound 9a had low inhibitory activity on hERG and yet no effect on the period of electrocardiogram,demonstrating that compound 9a may not cause the potential cardiotoxicity.

Abstract:Three quinoline carbonothioate compounds were synthesized using o-fluoroaniline and ethyl 2-methylacetoacetate as starting materials via multi-steps. The structures of three compounds were confirmed using H-1 NMR and HRMS. The structure of O-ethyl S-(8-fluoro-2,3-dimethylquinolin-4-yl) carbonothioate (3a) was further determined through X-ray single-crystal diffraction. Single-crystal X-ray diffraction analysis indicated that compound 3a crystallizes in the monoclinic space group C2/c, with unit cell parameters: a = 14.3284(6) & Aring;, b = 19.2857(8) & Aring;, c = 10.3990(5) & Aring;, beta = 108.1220(10)degrees, V = 2731.0(2) & Aring;(3). In addition, the physicochemical properties of compound 3a were calculated and discussed using molecular electrostatic potential, frontier molecular orbital and Hirshfeld surface analysis. The antifungal activity of the three compounds was evaluated at 50 mu g/mL, with compound 3b exhibiting good activity (75.8%) against R. solani.

Abstract:AIMTo search for novel antiasthmatic agents.METHODSCoupling seratrodast (SD), an antiasthmatic drug, with several different types of NO donors including oxatriazoles, N-hydroxyguanidines and furoxans; evaluating the antiasthmatic effects of coupled compounds by determining their inhibitory activity of guinea pig asthma induced by acetylcholine and histamine; and assessing NO releasing ability.RESULTSNine novel target compounds (I1-9) were synthesized, and their structures were established by IR, NMR, MS and elemental analysis. Preliminary pharmacological test showed that most of the compounds showed high antiasthmatic activities (the latent period of induced asthma was prolonged from 10 s (SD) to 26-62 s), among which 3 compounds (I4, I6, I7) were more potent than SD (P < 0.05, P < 0.01) and released more NO than others. The maximum concentrations (Cmax) of NO-release in vitro were 0.1878, 0.1393 and 0.2473 mg x L(-1), respectively.CONCLUSIONNO donating-SD derivatives are worthy to be futher investigated.

Abstract:Loratadine (Lor) is an antihistamine drug commonly used to relieve the symptoms of allergy. It has high permeability but low solubility under physiological conditions. To overcome the problem of low solubility, we synthesized and characterized two Loratadine multi-component crystalline phases with oxalic acid (Oxa), i.e., a 1:1 Lor-Oxa conjugate acid-base (CAB) cocrystal (Lor-Oxa CAB) and a 2:1 Lor-Oxa cocrystal monohydrate (Lor-Oxa hydrate). Both cocrystals exhibited an enhanced solubility and intrinsic dissolution rate (IDR) compared to Lor and adequate physical stability. The intrinsic dissolution rate of Lor-Oxa CAB is 95 times that of Lor, which makes it a promising candidate for tablet formulation development.

Abstract:A new synthetic method of 8-chloro-10,11-dihydro-4-aza-5H-dibenzo[a,d]cyclohepten-5-one,an important intermediate of loratadine,was reported.Using 2-cyano-3-methylpyridine as the starting material,through radical reaction,Wittig-Horner reaction,hydrolysis,reduction and cyclization it was synthesized.The total yield was 20%.The chemical structure was confirmed by means of()~1H-NMR,MS and elementary analysis.

Abstract:In this work, a new form of loratadine (Form B) was prepared from a 20% methanol and 80% water mixture and characterized by polarizing microscopy (POM), scanning electron microscopy (SEM), powder X-ray diffraction (PXRD) and differential scanning calorimetry (DSC). The dissolution, solubility and interfacial energy of Form B in water were measured and compared with those of amorphous and Form A loratadine. In addition, the solid-state transformations of loratadine solid forms were investigated by hot-stage microscopy (HSM) and DSC while the non-isothermal crystallization of amorphous loratadine was studied by DSC. It was found that Form B has a unique acicular structure and PXRD pattern, which are different from Form A. Moreover, Form B has a higher equilibrium solubility compared with Form A. The results of the solid-state transformation investigation (DSC) shows that the amorphous form was changed to the new polymorph B at 106.7 degrees C and Form B converted exothermally to Form A at 93.9 degrees C during heating. In the non-isothermal crystallization of amorphous loratadine, the crystallization rate constant (Z(t)) increased with the increasing heating rate and the Avrami exponent (n) was found to be about 3. Hopefully, Form B will be considered to be a promising approach to improve the loratadine solubility/dissolution rate.

Abstract:A series of novel chalcone guanidine derivatives (4a-4q) have been designed and synthesized, and their biological activity were also evaluated as potential antiproliferative and antitubulin polymerization inhibitors. Compound 4q showed the most potent biological activity (IC50 = 0.09 +/- 0.01 mu M for MCF-7 and IC50 = 8.4 +/- 0.6 mu M for tubulin), which is comparable to the positive controls. Docking simulation was performed to position compound 4q into the colchicine binding site to determine the probable binding model, which suggested probable inhibition mechanism. Abstract:Three series of ursolic acid derivatives containing an aminoguanidine moiety were designed, synthesized, and evaluated for anti-bacterial and anti-inflammatory activity. Some compounds displayed potent anti-bacterial activity against Gram-positive bacterial strains (including multidrug-resistant clinical isolates) and Gram-negative bacterial strains, with minimum inhibitory concentration (MIC) values in the range of 2–64 µg/mL. Compounds 3a, 5a, and 7l showed significant inhibitory activity against the Gram-positive bacterial strain Staphylococcus aureus RN 4220, the Gram-negative bacterial strain Escherichia coli 1924, and four multidrug-resistant Gram-positive bacterial strains, with MIC values of 2 and 4 µg/mL. In anti-inflammatory tests, most of the compounds exhibited potent activity, in particular compound 3a displayed the most potent activity with 81.61% inhibition after intraperitoneal administration, which was more potent than ursolic acid and the reference drugs (ibuprofen and indomethacin). The cytotoxic activity of compound 3a was assessed in HeLa, Hep3B, and A549 cells.

Abstract:Seven novel 3'-mono-substituted chalcone derivatives were synthesized from 2,4-dihydroxyacetophenone in 4 steps in 18-31% overall yield. Their structures were confirmed by H-1 NMR. The anticancer activity of these new target compounds (4 similar to 10) against human leukemia K562 cells were evaluated by MTT assay in vitro. This study leads to the identification of one highly active molecule, compounds 10 (IC50 = 5.60 mu M) against K562.
