Abstract:Loratadine was synthesized from 2-cyano-3-methylpyridine via Ritter reaction, alkylation and cyanidation to give 2-cyano-3-[2-(3-chlorophenyl)ethyl]piperidine, which subjected to cyclization in presence of PPA/P2O5 and then coupling with N-ethoxycarbonyl-4-piperidone. The overall yield was 39%.

Abstract:An improved process was reported to solve the problems such as low yield and high costs in the synthesis of 3-n-propyl-2, 4-dihydroxyacetophenone, which is a key intermediate of Leukotriene receptor antagonist. The target compound was synthesized starting from m-dihydroxybenzene, via acylation, allylation, Claisen rearrangement and hydrogenation reduction. The optimal reaction conditions were as follows. The acylation reaction which used acetic acid as acylation agent and ZnCl 2 as catalyst was carried out under reflux for 5 h, and 77.9% yield was obtained. The allylation reaction using allyl chloride as allylation reagent and NaBr, KI as catalyst was carried out at 40°C for 2 h and continued at 70°C for 14 h, to give 98.6% yield. In Claisen rearrangement carried out at 205°C for 4.5 h, diphenyl ether was used as reaction solvent and NaCl was used as catalyst, and the yield was improved to 67.4%. In the final step, the hydrogenated reduction with Pd/C (w(Pd) is 5%) as catalyst was carried out at 45°C for 6 h, and 88.6% yield was obtained. The process is much of industrial value because of high yield, cheap and available materials, and convenient operations. The structure of the main compounds was confirmed by 1H-NMR, GC-MS and IR.

Abstract:Desloratadine (Clarinex), the major active metabolite of loratadine (Claritin), is a nonsedating long-lasting antihistamine that is widely used for the treatment of allergic rhinitis and chronic idiopathic urticaria. For over 20 years, it has remained a mystery as to which enzymes are responsible for the formation of 3-hydroxydesloratadine, the major active human metabolite, largely due to the inability of any in vitro system tested thus far to generate this metabolite. In this study, we demonstrated that cryopreserved human hepatocytes (CHHs) form 3-hydroxydesloratadine and its corresponding O-glucuronide. CHHs catalyzed the formation of 3-hydroxydesloratadine with a Km of 1.6 μM and a Vmax of 1.3 pmol/min per million cells. Chemical inhibition of cytochrome P450 (P450) enzymes in CHHs demonstrated that gemfibrozil glucuronide (CYP2C8 inhibitor) and 1-aminobenzotriazole (general P450 inhibitor) inhibited 3-hydroxydesloratadine formation by 91% and 98%, respectively. Other inhibitors of CYP2C8 (gemfibrozil, montelukast, clopidogrel glucuronide, repaglinide, and cerivastatin) also caused extensive inhibition of 3-hydroxydesloratadine formation (73%-100%). Assessment of desloratadine, amodiaquine, and paclitaxel metabolism by a panel of individual CHHs demonstrated that CYP2C8 marker activity robustly correlated with 3-hydroxydesloratadine formation (r(2) of 0.70-0.90). Detailed mechanistic studies with sonicated or saponin-treated CHHs, human liver microsomes, and S9 fractions showed that both NADPH and UDP-glucuronic acid are required for 3-hydroxydesloratadine formation, and studies with recombinant UDP-glucuronosyltransferase (UGT) and P450 enzymes implicated the specific involvement of UGT2B10 in addition to CYP2C8. Overall, our results demonstrate for the first time that desloratadine glucuronidation by UGT2B10 followed by CYP2C8 oxidation and a deconjugation event are responsible for the formation of 3-hydroxydesloratadine.

Abstract:OBJECTIVE:To study the determination of desloratadine in human serum and its pharmacokinetics in healthy volunteers. METHODS:A single oral dose of 10 mg desloratadine was given to 18 healthy volunteers. The serum concentrations of desloratadine were determined by HPLC-MS assay. The pharmacokinetics parameters of desloratadine tablets were calculated with program 3P97. RESULT:The main pharmacokinetics parameters of desloratadine tablets were as followsút(max)(1.611 +/-0.366)h, C(max) (4.455+/-1.990)microg x L(-1), AUC(0-t) (58.50+/-21.34)microg x L(-1) x h(-1), AUC(0-infinity) (60.59+/-22.32)microg x L(-1) x h(-1), t(1/2(ke)) (20.303+/-5.833)h, Ke (0.0372+/-0.0116)h(-1) and CL(0.1838+/-0.0563)L x h(-1). CONCLUSION:Desloratadine tablet is absorbed quicker in the 18 healthy volunteers than the reports and its peak blood concentration reached at 1.5 h after oral administration with t(1/2) 20 h.

Abstract:2-Bromo-5-hydroxy-4-methoxybenzaldehyde(I)and 5-hydroxy-4-methoxy-2-nitrobenzaldehyde(II)were synthesized.The intermediates 2-bromo-4,5-dimethoxybenzaldehyde and 4,5-dimethoxy-2-nitrobenzaldehyde were synthesized firstly from veratraldehyde through bromination and nitration respectively.In the presence of molar quantity methionine,the 5th-position methoxys of the intermediates were in turn hydrolysed to hydoxy by concentrated sulfuric acid to give I and II.The chemical structures of the titled compounds were confirmed by 1H-NMR,IR and MS spectrum.The yields of compounds I and II are 85.0% and 65.0% respectively.The orientations of the bromination and nitration steps were suggested and discussed.The mechanism of methionine in the hydrolysis step may involve in the formation of the multiple hydrogen bonded association complex,and the calculated single point energy(E=-1532.5 hartree)of the multiple hydrogen bonded association complex seems to show it is possibly existed at the RHF/6-31G level.

Abstract:5-[2-(ethylthio)propyl]-1,3-cyclohexanedione, the title compound of this study, is an important intermediate for preparation of 3-hydroxy-cyclohex-2-enones herbicides, such as clethodim, sethoxydim, cloproxydim, etc.. The optimization of the synthesis route and the preparation method of the title compound were studied and the results were described concisely as follows. Firstly, 3-ethylthiobutanal was prepared with high yield by reacting crotonaldehyde with ethanethiol in triethylamine. Secondly, methyl acetoacetate was converted to 6-ethylthiohept-3-en-2-one via three steps, hydrolysis, condensation with 3-ethylthiobutanal and dehydration, then the 6-ethylthiohept-3-en-2-one was cyclocondensed with di-Me malonate by Michael addition and Clasisen condensation, and further via saponification and decarboxylation , the title compound was obtained. The products of each steps and the final title compound obtained were compared with each its own standerd samples and were confirmed by LC/MS.

Abstract:Seven target compounds were designed and synthesized according to the structure of lead compound—desloratadine.The structures of the compounds were identified by 1H NMR and HR-MS.The in vitro results based on histamine-induced contraction of guiea-pig ileum showed that zwitterionic compound 9c had stronger antihistamine activity than positive control desloratadine.The human ether-à-go-go related gene(hERG) inhibition and guinea-pig langendorff model indicated that compound 9a had low inhibitory activity on hERG and yet no effect on the period of electrocardiogram,demonstrating that compound 9a may not cause the potential cardiotoxicity.

Abstract:A rapid and simple liquid chromatographic-tandem mass spectrometric (LC/MS/MS) method was developed and validated for the simultaneous determination of desloratadine and its active metabolite 3-hydroxydesloratadine concentrations in human plasma. After liquid–liquid extraction with ethyl ether for sample preparation, the chromatographic separation was achieved on a CAPCELL PAK C18 column (50mm×2.0mm, 5μm, Shiseido). [2H4]desloratadine and [2H4]3-OH desloratadine were used as internal standards. A mobile phase consisted of 5mM ammonium formate in water, methanol and acetonitrile (50:30:20). Detection was by positive ion electrospray tandem mass spectrometry on a Sciex API3000. A quadratic regression (weighted 1/concentration) gave the best fit for calibration curves over the concentration range 0.05–10ng/mL for both desloratadine and 3-OH desloratadine. The method was shown to be accurate, rapid and sufficiently sensitive to be successfully applied to a pharmacokinetic and bioequivalent study.

Abstract:Loratadine (LOR, Claritin(®)) is a long-acting antihistamine used to treat allergic rhinitis. The major active human metabolite, desloratadine (DL, Clarinex(®)), is extensively metabolized to 3-hydroxydesloratadine (3-OH-DL) (M40) and subsequently glucuronidated before elimination. This study revealed the ability of a novel, long-term hepatocyte micropatterned co-culture (MPCC) model to generate in vivo metabolites. Metabolites were detected and characterized using non-targeted MS/MS(ALL) with SWATH™ acquisition by a UHPLC-Q-TOF system. Results & methodology: Human MPCCs extensively metabolized LOR and formed 3-OH-DL-glucuronide (M13). Cross-species comparisons revealed monkey- and rat-specific metabolites with gender-specific DL-pyridine-N-oxide formation in male rats. These results demonstrate a first for an in vitro hepatocyte model to generate circulating metabolites of LOR and detect species-specific differences. Early focus on human metabolites could have spared characterization of nonhuman metabolites in preclinical species.

Abstract:Loratadine (Lor) is an antihistamine drug commonly used to relieve the symptoms of allergy. It has high permeability but low solubility under physiological conditions. To overcome the problem of low solubility, we synthesized and characterized two Loratadine multi-component crystalline phases with oxalic acid (Oxa), i.e., a 1:1 Lor-Oxa conjugate acid-base (CAB) cocrystal (Lor-Oxa CAB) and a 2:1 Lor-Oxa cocrystal monohydrate (Lor-Oxa hydrate). Both cocrystals exhibited an enhanced solubility and intrinsic dissolution rate (IDR) compared to Lor and adequate physical stability. The intrinsic dissolution rate of Lor-Oxa CAB is 95 times that of Lor, which makes it a promising candidate for tablet formulation development.

Abstract:In this work, a new form of loratadine (Form B) was prepared from a 20% methanol and 80% water mixture and characterized by polarizing microscopy (POM), scanning electron microscopy (SEM), powder X-ray diffraction (PXRD) and differential scanning calorimetry (DSC). The dissolution, solubility and interfacial energy of Form B in water were measured and compared with those of amorphous and Form A loratadine. In addition, the solid-state transformations of loratadine solid forms were investigated by hot-stage microscopy (HSM) and DSC while the non-isothermal crystallization of amorphous loratadine was studied by DSC. It was found that Form B has a unique acicular structure and PXRD pattern, which are different from Form A. Moreover, Form B has a higher equilibrium solubility compared with Form A. The results of the solid-state transformation investigation (DSC) shows that the amorphous form was changed to the new polymorph B at 106.7 degrees C and Form B converted exothermally to Form A at 93.9 degrees C during heating. In the non-isothermal crystallization of amorphous loratadine, the crystallization rate constant (Z(t)) increased with the increasing heating rate and the Avrami exponent (n) was found to be about 3. Hopefully, Form B will be considered to be a promising approach to improve the loratadine solubility/dissolution rate.

Abstract:BackgroundLoratadine is a commonly used selective non-sedating antihistaminic drug. Desloratadine is the active metabolite of loratadine and, in addition, a potential impurity in loratadine bulk powder stated by the United States Pharmacopeia as a related substance of loratadine. Published methods for the determination of both analytes suffer from limited throughput due to the time-consuming steps and tedious extraction procedures needed for the analysis of biological samples. Therefore, there is a strong demand to develop a simple rapid and sensitive analytical method that can detect and quantitate both analytes in pharmaceutical preparations and biological fluids without prior sample extraction steps.ResultsA highly-sensitive and time-saving micellar liquid chromatographic method is developed for the simultaneous determination of loratadine and desloratadine. The proposed method is the first analytical method for the determination of this mixture using a monolithic column with a mobile phase composed of 0.15 M sodium dodecyl sulfate, 10% n-Butanol and 0.3% triethylamine in 0.02 M phosphoric acid, adjusted to pH 3.5 and pumped at a flow rate of 1.2 mL/min. The eluted analytes are monitored with fluorescence detection at 440 nm after excitation at 280 nm. The developed method is linear over the concentration range of 20.0–200.0 ng/mL for both analytes. The method detection limits are 15.0 and 13.0 ng/mL and the limits of quantification are 20.0 and 18.0 ng/mL for loratadine and desloratadine, respectively. Validation of the developed method reveals an accuracy of higher than 97% and intra- and inter-day precisions with relative standard deviations not exceeding 2%.ConclusionsThe method can be successfully applied to the determination of both analytes in various matrices including pharmaceutical preparations, human urine, plasma and breast milk samples with a run-time of less than 5 min and without prior extraction procedures. The method is ideally suited for use in quality control laboratories. It can be synthesized with the material of lovastatin, hydroxy protecting agent of trimethylchlorosilane, and t-butyl lithium instead of pyrrole lithium. The total yield is about 78%.The structure is identified by UV,IR,MS,NMR spectrums.This method is very easy and the cost is very low, but the yield is high. Most of organic solvents can be recovered in order to be used next time. So this method will have perfect industrialization foreground.

Abstract:Desloratadine, a second-generation histamine H1 receptor antagonist, has established itself as a first-line drug for the treatment of allergic diseases. Despite its effectiveness, desloratadine exhibits an antagonistic effect on muscarinic M3 receptor, which can cause side effects such as dry mouth and urinary retention, ultimately limiting its clinical application. Herein, we describe the discovery of compound Ⅲ-4, a novel H1 receptor antagonist with significant H1 receptor antagonistic activity (IC<sub>50</sub> = 24.12 nM) and enhanced selectivity towards peripheral H1 receptor. In particular, Ⅲ-4 exhibits reduced M3 receptor inhibitory potency (IC<sub>50</sub> > 10,000 nM) and acceptable hERG inhibitory activity (17.6 ± 2.1 μM) compare with desloratadine. Additionally, Ⅲ-4 exhibits favorable pharmacokinetic properties, as well as in vivo efficacy and safety profiles. All of these reveal that Ⅲ-4 has potential to emerge as a novel H1 receptor antagonist for the treatment of allergic diseases. More importantly, the compound Ⅲ-4 (HY-078020) has recently been granted clinical approval.

Abstract:In this paper, a new angiotensin Ⅱ receptor antagonist, (5-methyl-2-oxo-1,3-dioxol-4-yl) methyl-4-(1-hydroxy-1-methylethyl)-2-propyl-1-[(2′-1H-tetrazol-5-yl-biphenyl-4-yl) methyl] imdazole-5-carboxylate(Olmesartan medoxomil), was synthesized using tartaric acid as starting material via nitration, cyclization, esterifcation, Grignard reaction, N-alkylation, hydrolyzation, O-alkylation and N-deprotection in 32.7% overall yield. The structure of title compound has been characterized by MS(mass spectrum), (()~1H)-NMR (nuclear magnetic resonance) and IR (ifrared spectrum). The condensation of 4-(1-hydroxy-1-methylethyl)-2-propylimidazole-5-carboxylic acid ethyl ester with 5-[4′-(bromomethyl) biphenyl-2-yl]-1-(triphenylmethyl) tetrazole by means of potassium hydroxide in DMF, instead of potassium tert-butoxide in dimethylacetamide or sodium hydroxide in DMF, gives the corresponding N-alkylated product, which is hydrolyzed with sodium hydroxide, instead of lithium hydroxide, in dioxane-water to the desired acid. The improved process is more practical.
