Abstract:A novel class of acetylhydrazone derivatives (5a-x) containing 2-(phenylthiomethyl)-1H-benzo-[d]-imidazole moieties are synthesizer, and their antitumor activities against A549, HCT116, HepG2, PC-9, and A375 were determined by the MTT assay. Among them are N-(2,4-dihydroxybenzylidene)-2-(2-(phenylthiomethyl)-1H-benzo[d]-imidazol-1-yl)acetohydrazide (5a) and N-(5-bromo-2-hydroxy-benzylidene)-2-(2-(phenylthiomethyl)-1H-benzo[d]-imidazol-1-yl)acetohydrazide (5d) which displayed excellent cancer inhibitory activity against the tested cancer cells (IC(50) 4-17 μM), compared with 5-FU and SU11248. The others have moderate to weak inhibitory activity against the tested cancer cell lines.

Abstract:A series of novel N-1-(5-substituted-1,3,4-thiadiazole-2-yl)-N-3-substituted phenyl-urea derivatives have been synthesized and evaluated for their anti-tumor activity. The effect of the title compounds on tumor metastasis was analyzed by Lewis-lung-carcinoma model. The bioassay showed that some title compounds dramatically reduced the numbers of lung metastasis and depressed the primary tumor growth. The structures of compounds were identified from the elemental analysis, IR and H-1 NMR.

Abstract:A series of 2-(benzylthio)-5-aryloxadiazole derivatives have been designed and synthesized, and their biological activities are also evaluated for EGFR inhibitory activity. Fourteen compounds among the twenty compounds are reported for the first time. Their chemical structures are characterized by 1H NMR, MS, and elemental analysis. Anti-proliferative and EGFR inhibition assay results have demonstrated that compound 3e shows the most potent biological activity (IC50 = 1.09 μM for MCF-7 and IC50 = 1.51 μM for EGFR). Docking simulation has been performed to position compound 3e into the EGFR active site to determine the probable binding model, with an estimated binding free energy value of −10.7 kcal/mol. Compound 3e with potent inhibitory activity in tumor growth inhibition may be a promising anti-tumor leading compound for the further research.

Abstract:A series of 4 beta N-indole-substituted podophyllotoxin derivatives were synthesized. Nine target compounds were evaluated against human cancer cell lines (HeLa, K562, and K562/A02) using MTT assay including three imine derivatives 8, 9, and 10in vitro. The result showed that the three compounds had higher antitumor activity than their reduced forms. Among them, compounds 8, 9, 11, and 16 were superior to the positive control VP-16.

Abstract:Ten novel benzoyl ureas containing a 1,3,4-thiadiazole group were synthesized by the reaction of 2-amino-5-substituted phenyl-1,3,4-thiadiazole and 2,6-difluorobenzoyl isocyanate, and structurally confirmed by H-1 NMR, IR, ESI-MS techniques and elemental analysis. Preliminary bioassay test indicated that some compounds showed insecticidal activity. Compounds 3d (3,5-(CH3)(2)) and 3g (4-C4H9) exhibit good insecticidal activity against Aphis craccivora with higer than 90% mortality.

Abstract:Fifteen new 3-[(4-hydroxy-6-methly-2(1H)-pyridinones)-3-yl]-4-substituted- (1H)-1,2,4-triazole-5-thione compounds were synthesized from methyl 4-hydroxy-6- methyl-2-oxo-1,2-dihydropyridine-3-carboxylate in good overall yields by four step reactions. Molecular structure of these compounds was confirmed by IR, NMR, MS, elemental analysis and X-ray diffraction. Their antitumor activities were evaluated in vitro by the SRB and MTT methods. The results show that there are two compounds which are highly effective against KB cells and one compound which is highly effective against Bel-7402 cells.

Abstract:A series of diacylhydrazine derivatives containing 1,4-benzodioxan 1-17 has been designed, synthesized and evaluated for antitumor activity. Most of the synthesized compounds demonstrated potent antitumor activity and low toxicity. Compound 10 demonstrated the most potent biological activity against MCF-7 cancer cell line, which was comparable with the positive control 5-fluorouracil. Docking simulation by positioning compound 10 into the MetAP2 structure active site was performed to explore the possible binding model.

Abstract:Ten novel benzotriazole compounds were synthesized. Their chemical structures were confirmed by 1 H NMR, IR, and elemental analyses, coupled with three selected single-crystal structures (compounds A2 , B3 , and B5 ). Their antimycotic and antitumor activities were also investigated. The title compounds showed some antitumor activities, especially in the case of A3 and A4 , which showed the most potent activity of propagation inhibition in liver and galactophore cancer cells.

Abstract:Two substituted-3-phenylthiourea derivatives have been synthesized and characterized by elemental analysis,1H NMR,MS and single-crystal X-ray diffraction.Structural stabilization of them followed by intramolecular as well as intermolecular H-bonds makes these molecules as perfect examples in molecular recognition with self-complementary donor and acceptor units within a single molecule.The antitumor activities of compounds 1 and 2 against breast adeno carcinoma cell line MCF-7 have been evaluated by MTT method.Docking simulations have also been performed to position compounds into the telomerase active sites so as to determine their probable binding models.

Abstract:A series of novel 4H-benzo[h]chromenes 4, 6-11, 13, 14; 7H-benzo[h]chromeno[2,3-d]pyrimidines 15-18, 20, and 14H-benzo[h]chromeno[3,2-e][1,2,4]triazolo[1,5-c]pyrimidine derivatives 19a-e, 24 was prepared. The structures of the synthesized compounds were characterized on the basis of their spectral data. Some of the target compounds were examined for their antiproliferative activity against three cell lines; breast carcinoma (MCF-7), human colon carcinoma (HCT-116) and hepatocellular carcinoma (HepG-2). The cytotoxic behavior has been tested using MTT assay and the inhibitory activity was referenced to three standard anticancer drugs: vinblastine, colchicine and doxorubicin. The bioassays demonstrated that some of the new compounds exerted remarkable inhibitory effects as compared to the standard drugs on the growth of the three tested human tumor cell lines. The structure-activity relationships (SAR) study highlights that the antitumor activity of the target compounds was significantly affected by the lipophilicity of the substituent at 2- or 3- and fused rings at the 2,3-positions.

Abstract:Seven novel 4-(tert-butyl)-5-(1,2,4-triazole-1-yl)thiazole derivatives containing benzoxazinone (B1-B7) were obtained by cyclization of 4-(tert-butyl)-5-(1H-1,2,4-triazol-1-yl)-N-(2-hydroxy-benzyl)thiazol-2-amine. The chemical structures of the target compounds were confirmed by 1H NMR and 13C NMR. The single crystal X-ray diffraction revealed that B1 crystallizes in a triclinic system with the space group P 1 ¯ . Hirshfeld surfaces and two dimensional (2D) fingerprint plots were employed to quantitatively characterize the influence of intermolecular interactions within the crystal lattice of B1. And the molecular electrostatic potential (MEP) surface was calculated employing Becke-3-Lee Yang Parr (B3LYP)/6-311++G (d,p) theoretical approach. Meanwhile, the Frontier-molecular orbitals of B1 and B2 were calculated using density functional theory (DFT) B3LYP technique with the def2-SVP basis set in the ground state. Preliminary biological tests revealed that the title compounds had certain antitumor activities against three cell lines MCF-7, Hela and A549, and compound B2 presented the most potent antitumor efficacy against A549.

Abstract:In this study, several novel coumarin derivatives, 7-hydroxy-2-oxo-2H-chromene-3-carboxyl-Trp-Trp-AA-OBzl compounds, were designed and synthesized as potential anticancer agents. Their in vitro cytotoxic activities were evaluated using methylthiazoltetrazolium (MTT) assay. The anti-tumor activity of the newly coumarin derivatives was determined in a S180 bearing mouse model and some of the compounds demonstrated tumor growth inhibition similar to the positive control, doxorubicin. Compared to doxorubicin, most of the compounds exhibited enhanced immunologic function suggesting a relatively minor toxic effect. The intercalation of the coumarin derivatives synthesized with calf thymus (CT) DNA was also studied.

Abstract:Thiourea derivatives (6a–e) were developed and screened for antitumor and anti-inflammatory activity. Most of the compounds exhibited growth inhibitory effects comparable to 5-fluorouracil in vitro against mammary (MCF-7 and MDA-MB 231) as well as colon (HT-29) carcinoma cells. They also showed stronger anti-inflammatory activity than ibuprofen in vivo in the xylene-induced ear swelling assay in mice.

Abstract:In our continuing search for novel small-molecule anticancer agents, we designed and synthesized a series of novel (E)-N'-(3-allyl-2-hydroxy)benzylidene-2-(4-oxoquinazolin-3(4H)-yl)acetohydrazides (5), focusing on the modification of substitution in the quinazolin-4(3H)-one moiety. The biological evaluation showed that all 13 designed and synthesized compounds displayed significant cytotoxicity against three human cancer cell lines (SW620, colon cancer; PC-3, prostate cancer; NCI-H23, lung cancer). The most potent compound 5l displayed cytotoxicity up to 213-fold more potent than 5-fluorouracil and 87-fold more potent than PAC-1, the first procaspase-activating compound. Structure-activity relationship analysis revealed that substitution of either electron-withdrawing or electron-releasing groups at positions 6 or 7 on the quinazolin-4(3H)-4-one moiety increased the cytotoxicity of the compounds, but substitution at position 6 seemed to be more favorable. In the caspase activation assay, compound 5l was found to activate the caspase activity by 291% in comparison to PAC-1, which was used as a control. Further docking simulation also revealed that this compound may be a potent allosteric inhibitor of procaspase-3 through chelation of the inhibitory zinc ion. Physicochemical and ADMET calculations for 5l provided useful information of its suitable absorption profile and some toxicological effects that need further optimization to be developed as a promising anticancer agent.

Abstract:AIM:To design and synthesize new arylsubstituted imidazolin-2-one analogues as antitumor compounds.METHODS:Arylsubstituted imidazolin-2-ones were prepared by condensation the appropriate omega-amino-acetophenone hydrochloride with arylisocyanate in toluene. The target compounds prepared in this study were tested for cytotoxicity against PC-3, A549, HO-8910, Hela, HL60, K562 and HL60R cancer cell lines, and mechanism of one of the products 4y was further evaluated with its mechanium.RESULTS:Thirty-six new compounds were synthesized and confirmed by 1H NMR, MS and elemental analysis. One of the synthesized products, compound 4y, displayed an encouraging selective activity against HL60 cells, and it was partlydue to the cell cycle arrest and cell apoptosis.CONCLUSION:Compound 4y is worthy to be intensively studied.
