Abstract:An efficient method for the solution-phase parallel synthesis of 4-substituted-1,4-dihydroisoquinolin-3-ones is developed. The isoquinolinones were constructed employing an intramolecular Heck reaction, providing full regio- and stereoselectivity. Most of the synthesized compounds showed potent antiproliferative activities against tumor cell lines.

Abstract:A novel class of acetylhydrazone derivatives (5a-x) containing 2-(phenylthiomethyl)-1H-benzo-[d]-imidazole moieties are synthesizer, and their antitumor activities against A549, HCT116, HepG2, PC-9, and A375 were determined by the MTT assay. Among them are N-(2,4-dihydroxybenzylidene)-2-(2-(phenylthiomethyl)-1H-benzo[d]-imidazol-1-yl)acetohydrazide (5a) and N-(5-bromo-2-hydroxy-benzylidene)-2-(2-(phenylthiomethyl)-1H-benzo[d]-imidazol-1-yl)acetohydrazide (5d) which displayed excellent cancer inhibitory activity against the tested cancer cells (IC(50) 4-17 μM), compared with 5-FU and SU11248. The others have moderate to weak inhibitory activity against the tested cancer cell lines.

Abstract:Tetrahydroisoquinoline and its derivatives have a wide range of biological activities, such as antitumor, antimicrobial, and analgesic properties and so on. Some tetrahydroisoquinoline alkaloid derivatives have been studied as antitumor drug molecules, but 2-benzyl-7-substituted-ethoxy]-3, 4-dihydro-1(2H)-isoquinolinone series have not been reported yet. In this paper, we would like to report the design, synthesis, and biological evaluation of the novel tetrahydroisoquinoline derivatives. Among those compounds, four new tetrahydroisoquinoline derivatives have not been reported before. All the newly synthesized compounds were characterized by 1H NMR and their antitumor activities were evaluated by using HT-29, K562 and HepG2 cell line. 2-Benzyl-7-[2-(tert-butyl-4-piperazinecarboxylate) ethoxy]-3, 4-dihydro-1(2H)-isoquinolinone demonstrated good antitumor activity against K562 cancer cell with an IC50 of 1.58 μM.

Abstract:Ethyl 1-amino-3,3-dimethyl-3,4-dihydronaphthalene-2-carboxylate was used as a platform to synthesize 5,5-dimethyl-3- iso -propyl-2-thioxo-2,3,5,6-tetrahydrobenzo[ h ]quinazolin-4(1 H )-one (thioxobenzoquinazoline), which was alkylated by various halides in alkaline solution to produce 2-thio-substituted 5,5-dimethyl-3- iso -propyl-5,6-dihydrobenzo[ h ]quinazolin-4(3 H )-ones. Condensation of thioxobenzoquinazoline with hydrazine hydrate, 2-ethanolamine, and 3-propanolamine synthesized 2-hydrazinyl-5,5-dimethyl-3- iso propyl-5,6-dihydrobenzo[ h ]quinazolin-4(3 H )-one, 2-[(2-hydroxyethyl)amino]-5,5-dimethyl-3- iso -propyl-5,6-dihydrobenzo[ h ]quinazolin-4(3 H )-one, and 2-[(3-hydroxypropyl)amino]-3- iso -propyl-5,5-dimethyl-5,6-dihydrobenzo[ h ]quinazolin-4(3 H )-one, respectively. The antibacterial and antitumor properties of the synthesized compounds were studied. The research established that the studied compounds had antibacterial and antitumor activity. A comparative evaluation of the data indicated that the activity of the studied compounds was noticeably inferior to that of the antimicrobial reference drug furazolidone. The studied compounds at doses of 170 and 175 mg/kg showed weak antitumor effects, inhibiting tumor growth by 28 – 33%.

Abstract:Abstract Regioselective cyclocondensation of 2,4-diacetyl-5-hydroxy-5-methyl-3-(3-nitrophenyl/4-nitrophenyl)cyclohexanones 1a,b with cyanothioacetamide fforded the corresponiing 7-acetyl-4-cyano-1,6-dimethyl-6-hydroxy-8-(3-nitrophenyl/4-nitrophenyl)-5,6,7,8-tetrahydro-soquinoline-3(2 H )-thiones 2a,b in 93-96%. Reaction of compounds 2a,b ethyl iodide, 2-chloroacetamide or N -(naphthalen-1-yl)-2-chloroacetamide ( 5 ) in the presence of sodium acetate gave the corresponding p(5,6,7,8-tetrahydroisoquinolin-3-yl)thio derivatives 3a,b , 4a,b and 6a,b . In a similar manner, reaction of a,b with other N -aryl-2-chloroacetamides 7a-d gave 2-[(7-acetyl-4-cyano-1,6-dimethyl-6-hydroxy-8-(3-nitrophenyl/4-nitrophenyl)-5,6,7,8-tetrahydroisoquinolin-3-yl)thio]- N -arylacetamides ( 8a-g ). On heating of compounds 8a-e in ethanol containing anhydrous sodium carbonate, they converted into 7-acetyl-1-amino- N -aryl-5,8-dimethyl-8-hydroxy-6-(3-nitrophenyl/4-nitrophenyl)-6,7,8,9-tetrahydrothieno[2,3- c ]isoquinoline-2-carboxamides 9a-e . Structural formulae of all synthesized compounds were characterized on the basis of their spectroscopic data. Also, the applications of most synthesized isoquinolines as anticancer and as antioxidant agents have been carried out and the obtained results are reported herein.

Abstract:Regioselective cyclocondensation of 2,4-diacetyl-5-hydroxy-5-methyl-3-(3-nitrophenyl/4-nitrophenyl)cyclohexanones 1a,b with cyanothioacetamide afforded the corresponding 7-acetyl-4-cyano-1,6-dimethyl-6-hydroxy-8-(3- and -4-nitrophenyl)-5,6,7,8-tetrahydrosoquinoline-3(2H)-thiones 2a,b. Reaction of compounds 2a,b with ethyl iodide, 2-chloroacetamide (4a), or its N-aryl derivatives 4b-e in the presence of sodium acetate trihydrate gave 3-ethylthio-5,6,7,8-tetrahydroisoquinoline 3 and (5,6,7,8-tetrahydroisoquinolin-3-ylthio)acetamides 5a-i, respectively. Cyclization of compounds 5b-d,f,g into their isomeric 1-amino-6,7,8,9-tetrahydrothieno[2,3-c]isoquinoline-2-carboxamides 6b-d,f,g was achieved by heating in ethanol containing a catalytic amount of sodium carbonate. Structures of all synthesized compounds were characterized on the basis of their elemental analyses and spectroscopic data. The crystal structure of 5,6,7,8-tetrahydroisoquinoline 5d was determined by X-ray diffraction analysis. In addition, the biological evaluation of some synthesized compounds as anticancer agents was performed, and only six compounds showed moderate to strong activity against PACA2 (pancreatic cancer cell line) and A549 (lung carcinoma cell line). Moreover, the antioxidant properties of most synthesized compounds were examined. The results revealed high antioxidant activity for the most tested compounds.

Abstract:In light of the importance of N-fused heterocycles in pharmaceuticals, there is continuing interest in research on N-fused heterocycles and their preparation. A new and efficient reductive dearomatization-initiated intramolecular cyclization reaction with a broad scope has been developed, affording 3,4,6,7-tetrahydro-2H-pyrimido[1,6-c]quinazolin-2-imine derivatives. Notably, this type of compound showed good inhibitory activity against specific kinases and human cancer cell lines. These results might mean a new molecular scaffold for the development of new antitumor agents.

Abstract:Transition‐metal‐catalyzed inert C−H bond functionalization using sulfoxonium ylides as metal carbene surrogates to replace the explosive and labile diazo compounds is synthetically attractive. Presented herein is a divergent synthesis of (CF3)‐indazolo[3,2‐a]isoquinoline derivatives based on C−H bond activation‐initiated cascade reactions of 3‐aryl‐1H‐indazoles with sulfoxonium ylides. The formation of products involves Rh(III)‐catalyzed C(sp2)−H bond metalation, carbene formation, migratory insertion, proto‐demetalation and intramolecular condensation. In general, this newly developed protocol features simple substrates, structurally diverse products, step‐economy and scalability. In addition, evaluation of the photophysical properties and anti‐proliferative activity against three human cancer cell lines of the products thus obtained showed that some of them possess potent anticancer activity with potential self‐tracking capability through fluorescence detection.

Abstract:Cancer, as a public health issue, is the leading cause of death worldwide. Tetrahydroisoquinoline derivatives have effective biological activities and can be used as potential therapeutic agents for antitumor drugs. In this work, we designed and synthesized a series of novel tetrahydroisoquinoline compounds and evaluated their antitumor activity in vitro on several representative human cancer cell lines. The results showed that the vast majority of compounds showed good inhibitory activities against the cancer cell lines of HCT116, MDA-MB-231, HepG2, and A375.

Abstract:Based on the structural feature of nonclassical antifolates, a series of 4 (3H)-quinazolinone derivatives bearing aromatic amine side chains have been designed by incorporating trimethoxyphenyl, an antitumor pharmacophore, with 4 (3H)-quinazolinone. N-alkylation of 3,4,5-trimethoxyphenylamine with appropriate amount of alkyl halides afforded 4 kinds of N-substituted 3,4,5-trimethoxyphenylamines in 30.3%~60.6% yields. The target compounds 8a 8m were obtained through the reactions of 6-bromo-2-methyl-4 (3H)-quinazolinone with 3,4,5-trimethoxyphenylamine, N-substituted 3,4,5-trimethoxyphenylamines or other arylamines at room temperature in 30.8%~71.9% yields, and their structures were confirmed by means of ESIMS, 1H NMR, elemental analysis or HRMS. The in vitro cytotoxicities against A-549 (human non-small cell lung cancer), HCT-8 (human colon cancer) and Bel-7402 (human liver cancer) cell lines of the synthesized compounds 8a~8m were tested with colorimetric MTT assay, and the results indicated that the percent growth inhibition against A-549, HCT-8 and Bel-7402 cell lines at 5×10^(-6)g/mL concentration of the compounds 8a~8m were all lower than 25%.

Abstract:AIM:To design and synthesize new arylsubstituted imidazolin-2-one analogues as antitumor compounds.METHODS:Arylsubstituted imidazolin-2-ones were prepared by condensation the appropriate omega-amino-acetophenone hydrochloride with arylisocyanate in toluene. The target compounds prepared in this study were tested for cytotoxicity against PC-3, A549, HO-8910, Hela, HL60, K562 and HL60R cancer cell lines, and mechanism of one of the products 4y was further evaluated with its mechanium.RESULTS:Thirty-six new compounds were synthesized and confirmed by 1H NMR, MS and elemental analysis. One of the synthesized products, compound 4y, displayed an encouraging selective activity against HL60 cells, and it was partlydue to the cell cycle arrest and cell apoptosis.CONCLUSION:Compound 4y is worthy to be intensively studied.
