Abstract:6,7-Methylenedioxy (or 5-hydroxy-6-methoxy)-2-(substituted selenophenyl)quinolin-4-ones and their isosteric compounds were synthesized and evaluated for anticancer activity. Structure–activity relationships (SAR) of these compounds were established. Among all tested compounds, 6,7-methylenedioxy-2-(5-methylselenophen-2-yl)quinolin-4-one (4d) was found to be the most promising anticancer agent. In screening against NCI’s 60 human tumor cell line panel, 4d exhibited highly selective and potent inhibitory activity against MDA-MB-435 melanoma. Furthermore, the results of COMPARE analysis suggested that 4d is an antimitotic agent with a different mechanism of action from the conventional antimitotic agents, such as colchicine, vinca alkaloids and paclitaxel. Therefore, 4d was identified as a new lead compound that merits further optimization.

Abstract:In order to search for new antitumor drugs with high efficiency and low toxicity, a series of novel 4-aminoquinazoline derivatives containing 1,3,4-thiadiazole group were designed, synthesized and evaluated for antiproliferative activity against four human cancer cell lines (H1975, PC-3, MCF-7 and HGC-27) using MTT assay in vitro. Among them, compound N-(5-((3,5-dichlorobenzyl)thio)-1,3,4-thiadiazol-2-yl)-2-((4-(phenylamino)quinazolin-2-yl)thio)acetamide (15o) showed good anti-tumor proliferation activity against four tested cancer cell lines, with IC50 values of 1.96 ± 0.15 μM to PC-3 cell. The antitumor activity was significantly better than that of gefitinib. Further mechanism studies showed that compound 15o inhibited the migration ability of PC-3 tumor cells in a concentration dependent and time-dependent manner, and blocked the cell cycle in S phase. At the same time, cell cloning experiment further proved that compound 15o significantly inhibited the colony formation of PC-3 cells, The inhibition rate was as high as 92% at 2.0 μM.

Abstract:Series of novel derivatives of quinazoline containing thiosemicarbazide moiety 5 and 9 have been synthesized and tested for their antitumor activities in vitro against a panel of five human cancer cell lines. Bioassay results indicated that most of the prepared compounds exhibited cytotoxicity against various cancer cells. From the structure–activity relationships it was found that unsubstituted quinazoline ring and benzene ring or halogen substituted benzene ring in quinazoline derivatives 5 and 9 would be the most favorable for their antitumor activity.

Abstract:In order to find efficient and low toxicity anti-tumor drugs, a series of novel 4-aminoquinazoline derivatives containing benzothiazole were designed and synthesized. And their antiproliferative activities against four human cancer cell lines [human breast cancer cell line (MCF-7), human gastric carcinoma cell line (MGC-803), human prostate cancer cell line (PC-3), human gastric carcinoma cell line (HGC-27)] were evaluated by using methyl thiazolyl tetrazolium (MTT) assay. The results showed that most compounds exhibited good antiproliferative activities against the four human tumor cell lines. Among them, 2-((benzo[d]thiazol-2-ylmethyl)thio)-N-(3-chloro-4-fluorophenyl)-quinazolin-4-amine (13n) showed the best antiproliferative activity against MCF-7, MGC 803, PC-3 and HGC-27 cancer cell lines, with IC50 values of (6.01 +/- 0.54), (7.63 +/- 0.48), (6.16 +/- 0.34), (7.59 +/- 0.62) mu mol.L-1, respectively. Its activity was better than the positive control gefitinib. Molecular docking showed that compound 13n could bind well with epidermal growth factor receptor (EGFR). In a nutshell, this work provides clues to discover antitumor agent based on the quinazoline scaffold.

Abstract:A series of quinoline derivatives (4a–4o) have been synthesized and their biological activities were also evaluated as potential telomerase inhibitors. Bioassay tests demonstrated that most of the compounds exhibited substantial broad-spectrum antitumor activity against the three cancer cell lines (HepG2, SGC-7901 and MCF-7). Moreover, all the title compounds were assayed for telomerase inhibition using the TRAP-PCR-ELISA assay. Compounds 4d and 4i displayed the most potent anticancer activities, which were comparable to the positive control. Docking simulation was performed to position compounds 4d and 4i into the telomerase structure active site to determine the probable binding model. Compounds 4d and 4i with potent inhibitory activity in tumor growth inhibition may be potential anticancer agents.

Abstract:In order to find efficient and low toxicity antitumor drugs, a series of novel 2,4,6-trisubstituted quinazoline derivatives containing trifluoromethyl were synthesized and evaluated for their antitumor activities against human cancer cell lines (PC-3, MCF-7, Eca-109, MGC-803, HGC-27, A549, H1975) by using methyl thiazolyl tetrazolium (MTT) assay. Most of compounds exerted moderate to excellent antitumor activity against seven human cancer cells. Among them, N-(3-bromophenyl)-6-methoxy-2-04-(trifluoromethyl)benzyl)thio)quinazolin-4-amine (161) showed the best antitumor activity against PC-3 cancer cell line, with the IC50 values of (2.22 +/- 0.15) mu mol/L, which was better than the positive control of gefitinib. At the same time, and compound 161 could dose-dependently and time-dependently induce PC-3 cells apoptosis.

Abstract:A series of novel quinazolin-4(3H)-one derivatives 3a similar to 3e and 4a possessing 3,4-benzo[c] coumarin and amino side chain were designed and synthesized by the condensation reaction of 2-aminobenzamide and 3,4-benzo[ c] coumarin aldehyde prepared by Heck coupling reaction, followed by S(N)2 substitution reaction with haloalkane. The compounds were evaluated for their anti proliferation activities against four tumor cells and antimicrobial activities. The results showed that 3e showed moderate anti Hela activity with the IC50 value of 22.63 and 23.35 mu mol/L for 4a against MCF-7. Most tested compounds exhibited significant anti Escherichia coli activities, and the inhibition rate was above 89% at the concentration of 50 mu g/mL. The inhibition rates of 2-phenyl-4-(2-(piperidin-1-yl) ethoxy) quinazoline (1b) against Fusarium oxysporum and Rhizoctonia solani and 3d against Verticillium dahliae are 100%.

The isoquinolinones were constructed employing an intramolecular Heck reaction, providing full regio- and stereoselectivity. Abstract:A series of new quinoline derivatives was designed, synthesized and evaluated as potential antitumor agents. The results indicated that most compounds exhibited potent antiproliferative activity, and 7-(4-fluorobenzyloxy)N-(2-(dimethylamino)- ethyl)quinolin-4-amine 10g was found to be the most potent antiproliferative agent against human tumor cell lines with an IC50 value of less than 1.0 μM. Preliminary structure-activity relationships analysis suggested that (1) the large and bulky alkoxy substituent in position-7 might be a beneficial pharmacophoric group for antiproliferative activity; (2) the amino side chain substituents in position-4 facilitated the antiproliferative activity of this class of compounds; and (3) the length of the alkylamino side chain moiety affected the antiproliferative potency, with two CH2 units being the most favorable. Further investigation of the mechanism of action of this class of compounds demonstrated that the representative compound 10g triggered p53/Bax-dependent colorectal cancer cell apoptosis by activating p53 transcriptional activity. Moreover, the results showed that compound 10g effectively inhibited tumor growth in a colorectal cancer xenograft model in nude mice. Thus, these quinoline derivatives might serve as candidates for the development of new antitumor drugs.

Abstract:OBJECTIVE To synthesize a novel quinazoline derivatives and study their antitumor activity in vitro.METHODS The target compounds were synthesized from 5-nitroanthranilonitrile by cyclization,halogenation,amino substitution,nitro reduction,N-acylation etc..The anti-tumor activity in vitro of these compounds have been studied by MTT assay with prostate carcinoma PC-3 cells,ovarian carcinoma SKOV-3 cells,human colorectal cancer HCT116 cells,human epithelial carcinoma A431 cells and human erythroleukemia K562 cells.RESULTS and CONCLUSION Three new compounds were synthesized,and the chemical structures were characterized by 1HNMR and MS.Preliminary anti-tumor activity in vitro showed that compound Ⅵa has a good anti-erythroleukemia and anti-colorectal carcinoma activity.

Abstract:Based on the structural feature of nonclassical antifolates, a series of 4 (3H)-quinazolinone derivatives bearing aromatic amine side chains have been designed by incorporating trimethoxyphenyl, an antitumor pharmacophore, with 4 (3H)-quinazolinone. N-alkylation of 3,4,5-trimethoxyphenylamine with appropriate amount of alkyl halides afforded 4 kinds of N-substituted 3,4,5-trimethoxyphenylamines in 30.3%~60.6% yields. The target compounds 8a 8m were obtained through the reactions of 6-bromo-2-methyl-4 (3H)-quinazolinone with 3,4,5-trimethoxyphenylamine, N-substituted 3,4,5-trimethoxyphenylamines or other arylamines at room temperature in 30.8%~71.9% yields, and their structures were confirmed by means of ESIMS, 1H NMR, elemental analysis or HRMS. The in vitro cytotoxicities against A-549 (human non-small cell lung cancer), HCT-8 (human colon cancer) and Bel-7402 (human liver cancer) cell lines of the synthesized compounds 8a~8m were tested with colorimetric MTT assay, and the results indicated that the percent growth inhibition against A-549, HCT-8 and Bel-7402 cell lines at 5×10^(-6)g/mL concentration of the compounds 8a~8m were all lower than 25%.

Abstract:A series of 2,4,6-trisubstituted quinazoline derivatives containing thiazole group were designed and synthesized, and their antiproliferative activities against human tumor cell lines (PC-3, H1975, MGC-803 and A549) were determined. 3-((2-(((2-chlorothiazol-5-yl)methyl)thio)-6-methoxyquinazolin-4-yl)amino)benzonitrile (14i) was identified as the most potent compound in antiproliferation against MGC-803 cells, with the value was (4.54 +/- 0.32) mu mol/L, which was better than the positive control 5-fluorouracil (5-FU) [(8.14 +/- 0.68) mu mol/L]. Further mechanistic studies showed that compound 14i could inhibit the clonal proliferation and migration of MGC-803 cells. In addition, compound 14i could induce apoptosis of MGC-803 cells and arrest the cell cycle at late period of DNA synthesis/mitotic phase (G2/M) phase. These results indicated that compound 14i could as a potential antitumor drug.

Abstract:A new series of substituted benzylamino‐ and heterocyclylmethylamino carbodithioate derivatives of 4‐(3H)‐quinazolinone were synthesized via four steps starting from 2‐amino‐5‐methylbenzoic acid and initially screened against A‐549 (human non‐small cell lung cancer), HCT‐8 (human colon cancer), and Bel‐7402 (human liver cancer) cell lines at the single concentration of 5 μg/mL using the colorimetric MTT assay. The IC50 values were determined for the compounds reaching ⪈70% inhibition in primary screening by serial dilution. Among the newly synthesized compounds, 9n exhibited potent in vitro cytotoxicity against A‐549, HCT‐8, and Bel‐7402 cell lines with the IC50 values of 1.65, 0.93, and 1.43 μM, respectively.
